Effect of Platycodon grandiflorum DC Extract on the Growth of Cancer Cell Lines (도라지(Platycodon grandiflorum DC) 추출 성분의 암세포 증식 억제효과)

  • To investigate the cytotoxic effect of Platycodon grandiflorum DC, petroleum ether extract of Platycodon grandiflorum DC was partially purified by a silica gel column chromatography. Among several fractions, fraction D which was obtained under the elution with a 7:3 mixture of petroleum ether and ethyl ether, showed patent cytotoxicity against mouse leukemia cell line (L1210), human rectum cancer cell line (HRT-18) and human colon cancer cell lint (HCT-48).

  • Induction and culture of hairy roots by Agrobacterium rhizogenes A4 were carried out in Platycodon grandiflorum DC. After 2-4 weeks of inoculation with Agrobacterium rhizogenes hairy roots were formed at root segments in the balloon flower. Optimized growth of hairy roots was obtained in hormone-free MS medium, 6% sucrose and pH 5.8. The pattern of ginsenoside in the transformed roots was not different with that in the ordinary roots.

  • This study was observed to the effect of the feeding Platycodon grandiflorum A. DC (3 years) extract on the bronchus diseases bacteria ( C. diphtheriae, S. aureus, Mycobacterium sp., F. nucleatum, S. pygogenes, K. pneumoniae and N. gonorrhoeae) and fungi(A. fumicatus). Platycodon grandiflorum A. DC was extracted ethanon, water, ethyl ether and petroleum ether. The extraction rate of Platycodon grandiflorum A. DC to the extract solution was identified 71.8%, 100%, 15.4% and 14.1%. Each extract solution was injected culture media into several concentrations and then investigated the bacteria cell growth during 32 hours. As a result antimicrobial activity was excellent an extract by ethyl ether and petroleum ether. Among several concentrations, bacteria cell growth inhibition was observed from 0.06% to 0.14%. The rate of antimicrobial activity was over 70%. The cell growth inhibition rate of each bacteria was appeared in order of ethyl ether > petroleum ether > water > ethanol.

  • This study was pe바ormed to observe the effects of Platycodon grandiflorum A.   • This study investigates the quality characteristics of soybean dasik made with various levels(0, 3, 6, 9 and 12%) of Platycodon grandiflorum(Jacq.) A. DC. flour additions. In soybean dasik added with Platycodon grandiflortum(Jacq.) A. DC flour, moisture contents range from $24.47{\pm}0.35$ to $26.31{\pm}0.81%$ As Platycodon grandiflorum(Jacq.) A. DC flour increased, lightness values were decreased and redness and yellowness values were increased significantly. The result of the mechanical evaluation showed that hardness increased after significantly decreasing while adhesiveness, gumminess and chewiness decreased with the amount of Platycodon grandiflorum(Jacq.) A. DC flour increased. Cohesiveness and springiness did not change significantly according to Che level of Platycodon grandiflorum(Jacq.) A. DC flour. In conclusion, soybean dasik with the addition of 6% Platycodon grandiflorum(Jacq.) A. DC flour has the most propper taste, flavor and overall acceptability on sensory evaluation.

  • Broadening the genetic base of Platycodon grandiflorum DC. cultivar to sustain improvement requires assessment of genetic diversity available in P. grandiflorum DC.. The objective of this study was to analyze the genetic variation, genetic relationship among 48 samples collected from East-Asian Area by means of RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) markers. From the 18 primers tested, produced total 211 bands with an average of 11.7 bands per primer and obtained 103 polymorphic band with an average of 5.7 bands per primer,s revealed relatively high percentage of polymorphic bands (48.8%). The genetic similarities calculated from RAPD data varied from 0.688 to 0.994 and were clustered to six major groups on a criterion of 0.78 similarity coefficient. The present study has revealed the significant genetic similarity among the samples tested. The analysis of genetic relationships in P. grandiflorum using RAPD-PCR banding data can be useful for the breed improvement.

Plant Regeneration via Adventitious Shoot Formation in Platycodon grandiflorum (Jacq. A. DC.) (도라지 (Platycodon grandiflorum (Jacq.) A. DC.) 부정아 형성을 통한 식물체 재분화)

  • To investigate optimal conditions for plant regeneration in Platycodon grandiflorum (Jacq. A. DC.).Both leaf and hypocotyl explants were cultured on Murashige& Skoog's (MS) medium supplemented with combinations of 0.1, 0.5, 1.0, or 2.0 mg/L cytokinins (BA and kinetin) and 1.0 mg/L 2,4-D for 6 weeks, respectively. According to the type of explant, the total shoot organogenesis (56.38%) in leaf explants was higher than in hypocotyls (28.20%). In comparison with kinetin and BA for the plant regeneration, the frequency (70.38%) of leaf explants was higher in combination with kinetin and 2,4-D than of BA with 2,4-D (42.38%), whereas the frequency (35.56%) of hypocotyls explants was higher in BA combination than kinetin combination (20.83%). Thehighest frequency (94.20%) was observed from the cultures of leaf explants on the MS medium supplemented with 1.0 mg/L kinetin and 1.0 mg/L 2,4-D. Upon transfer onto 1/2 MS basal medium containing 3% sucrose, shoots developed into plantlets with roots, and were well grown in soil in the greenhouse. These results lead us to speculate that the optimization of culture conditions was responsible for the mass propagation from in vitro cultures of Platycodon grandiflorum (Jacq. A. DC.).

  • To investigate the antimutagenic effect of Platycodon grandiflorum DC, methanol extract of Platycodon grandiflorum DC was investigated. In Ames test, the methanol extract showed inhibitory effects of 80-90% on the mutagenicity induced by indirect mutagen of IQ(2-amino-3-methylimidazo[4,5-f]quinoline) and direct mutatgen of MNNG(N-methyl-N'-nitro-N-nitrosoguanidine) in Salmonella typhimurium TA 98 and TA 100. And then the methanol extract was further fractionated. Among the solvent extracted fractions from the methanol extract, the ethyl acetate fraction and butanol fraction exhibited the greatest antimutagenic effect suppressing the mutagenicity IQ and MNNG with inhibition rate of 99% and 98%.

  • The immunostimulatory and host resistance effects of the Platycodon grandiflorum A. DC, changkil (CK) and inulin (CKI) isolated from CK were investigated in rats. SD rat were exposed to CK or CKI by gavages for 7days and isolated peritoneal macrophages and splenocyte were used for these studies. CK and CKI significantly enhanced peritoneal macrophages activities such as ROS production and phargocytosis. (omitted)

  • The protective effects on hepatic fibrosis of an aqueous extract from the roots of Platycodon grandiflorum A. DC (Campanulaceae), Changkil (CK), in hepatic stellate cell line, CFSC-2G. The increased deposition of extracellular matrix by hepatic stellate cells following liver injury in a process known as activation is considered a key mechanism for increased collagen content of liver during the development of liver fibrosis.
