Effects of Storage Methods and Periods on Root Hardness and Content of Saponin in Platycodon grandiflorum Radix (저장방법과 기간이 도라지 뿌리의 경도와 사포닌함량 미치는 영향)

  • The research was conducted to provide basic information for store temperature which were low and room temperature and packing methods which were vacuum, packing and nitrogen of 3 year-old Platycodon grandiflorum. We investigated hardness and content of saponins, 1) platycodin D3, 2) polygalacin D and 3) deapioplatycodin D, in Platycodon grandiflorum and hardness of Platycodon grandiflorum, which were reduced by increasing storage period and decreased with increasing storing temperature, respectively. The packed storing method was better than others storing methods in low temperature. The high root hardness was significantly related with storing temperature and methods. The content of saponins in Platycodon grandiflorum, i.e., platycodin D3 and polygalacin D were reduced during storing period, however, the content of deapioplatycodin D was increased during storing period.

  • Platycodin D was isolated from n-butanol extract of Platycodi radix(Platycodon grandiflorum and identified by the spectroscopic analysis using $^1H$ and $^{13}C$ NMR techniques. A new method of analysis of platycodin D by high performance liquid chromatography(HPLC) was established using a reversed phase system with YMC-Pack ODS-AM( 250 X 4.6 mm) column and 30% acetonitrile as a mobile phase. Evaporative light scattering detector was used as detector. The kinds of extraction solvents and methods were examined to determine the efficient extraction condition and HPLC analysis was carried out to establish the optimum drying condition for the root of Platycodon grandiflorum. The contents of Platycodin D was highest as 0.083% when platycodon roots were dried at $60^{\circ}C$ using dry oven.

Effect of Saponin Fraction from Platycodon grandiflorum on Clinical Chemical Changes in TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)-induced Rat Toxicity

  • This study was carried out to investigate the protective effect of crude saponin from Platycodon grandiflorum on Clinical chemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD). Crude saponin was prepared from Korean Platycodon grandiflorum with Diaion HP-20 adsorption chromatography after extraction of 80% ethanol at 75℃. The crude saponin was confirmed by thin layer chrmatography. When compared with ginseng saponins, the crude saponin had both a few number of saponins and a broad distribution. Forty male rats (200±20 g) were divided into 4 groups. Normal control (NC) group received vehicle and saline; TCDD-treated (TT) group received TCDD (40 ㎍/kg, single dose) intraperitoneally; Platycodon grandiflorum saponin (PG5 and PG10) groups received crude saponin 5 mg/kg and 10 mg/kg (p.o), respectively, for 2 weeks before 1 week of TCDD-exposure. Increase of body weight was retarded greatly by TCDD-exposure. Body weight of animals in TT group was significantly decrease after 2 days of TCDD-exposure. However, body weights of animals in PG groups increased through the experimental perimental period, although the increasing rate was slower than that of NC group. Increases in contents of blood glucose, total cholesterol and triglyceride (TG) and activities of amylase, lipase, AST, ALT and LDH by toxic action of TCDD were significantly attenuated by crude saponin from Platycodon grandiflorum (P<0.05). In conclusion, these results suggest that crude saponin prepared from Korean Platycodon grandiflorum might be a member of useful protective agents against TCDD, which is one of the environmental hormones.

Plant Regeneration via Adventitious Shoot Formation in Platycodon grandiflorum (Jacq. A. DC.) (도라지 (Platycodon grandiflorum (Jacq.) A. DC.) 부정아 형성을 통한 식물체 재분화)

  • To investigate optimal conditions for plant regeneration in Platycodon grandiflorum (Jacq. A. DC.).Both leaf and hypocotyl explants were cultured on Murashige& Skoog's (MS) medium supplemented with combinations of 0.1, 0.5, 1.0, or 2.0 mg/L cytokinins (BA and kinetin) and 1.0 mg/L 2,4-D for 6 weeks, respectively. According to the type of explant, the total shoot organogenesis (56.38%) in leaf explants was higher than in hypocotyls (28.20%). In comparison with kinetin and BA for the plant regeneration, the frequency (70.38%) of leaf explants was higher in combination with kinetin and 2,4-D than of BA with 2,4-D (42.38%), whereas the frequency (35.56%) of hypocotyls explants was higher in BA combination than kinetin combination (20.83%). Thehighest frequency (94.20%) was observed from the cultures of leaf explants on the MS medium supplemented with 1.0 mg/L kinetin and 1.0 mg/L 2,4-D. Upon transfer onto 1/2 MS basal medium containing 3% sucrose, shoots developed into plantlets with roots, and were well grown in soil in the greenhouse. These results lead us to speculate that the optimization of culture conditions was responsible for the mass propagation from in vitro cultures of Platycodon grandiflorum (Jacq. A. DC.).

  • To investigate the cytotoxic effect of Platycodon grandiflorum DC, petroleum ether extract of Platycodon grandiflorum DC was partially purified by a silica gel column chromatography. Among several fractions, fraction D which was obtained under the elution with a 7:3 mixture of petroleum ether and ethyl ether, showed patent cytotoxicity against mouse leukemia cell line (L1210), human rectum cancer cell line (HRT-18) and human colon cancer cell lint (HCT-48).

Physio-chemical impacts on efficient germination in Platycodon grandiflorum for. duplex pollen culture

  • The highly valued ornamental plant, Platycodon grandiflorum for. duplex was generated by petaloidy of a calyx of Platycodon grandiflorum. The present investigation was executed to explore the several factors having effects on the germination of pollens with a view to acquire the underlying data for the artificial crossing to cultivate the species of Platycodon grandiflorum for. duplex. Both low and high temperature impaired the germination of Platycodon grandiflorum for. duplex pollens. The good germination rate was observed at the temperature of $25^{\circ}C$. The types and concentrations of carbon sources induced the differences in germination rate. The germination rate increased with the increasing concentration of sucrose and glucose except for fructose. Sucrose and glucose showed the highest results at the concentration of 20%. While fructose demonstrated the similar tendency to sucrose and glucose, it reduced the germination rate at the concentration of 20%. The highest germination rate was observed at the concentration of 15%. The appropriate carbon course for germination of pollens of Platycodon grandiflorum for. duplex was glucose of which germination rate was twice as high as that of sucrose and fructose. The germination rate was reduced substantially when the pH was close to alkali, and the potential germination rate was obtained at pH 6. Boric acid enhanced the germination rate at a lower concentration than the higher concentration.

Estimated Daily Intake of Aluminum from Platycodon grandiflorum A. De Candolle (도라지(Platycodon grandiflorum A. De Candolle) 섭취에 따른 알루미늄의 노출량 평가)

  • This study was performed to estimate daily intakes of aluminum from Platycodon grandiflorum A. De Candolle and evaluate their potential health risks for Koreans. Estimated daily intake of aluminum was calculated, whereas the actual level of aluminum in Platycodon grandiflorum A. De Candolle was analyzed using inductively coupled plasma mass spectrometry. Food consumption amount was drawn from Korea National Health and Nutrition Examination Survey (KNHANES VI-1). In analysed samples, aluminum values ranged from 0.54~564.38 mg/kg in peeled Platycodon grandiflorum A. De Candolle samples (n=53) and from 0.72~28.05 mg/kg in unpeeled ones (n=40). Statistically significant difference (P<0.001) was detected according to the type of skin. To estimate the dietary intake of Platycodon grandiflorum A. De Candolle, a total of 7,242 respondents (scenario I) were compared to 227 Platycodon grandiflorum A. De Candolle consumption subjects (scenario II). Estimated daily intake of aluminum was calculated based on point estimates. Level of safety for aluminum was evaluated by comparison with Provisional Tolerable Weekly Intake (PTWI), 1 mg/kg bw, set by the Joint FAO/WHO Expert Committee on Food Additives. For scenario I, mean estimated daily intake of aluminum was 0.001 mg/kg bw/d. For scenario II, mean estimated daily intake of aluminum was 0.033 mg/kg bw/d, and 95th percentile estimated daily intake was 0.610 mg/kg bw/d. For scenario II, aluminum from Platycodon grandiflorum A. De Candolle had a mean weekly intake that was the 23.1% of PTWI.

Saponin isolated from Platycodon grandiflorum induces cell cycle arrest in hepatic stellate cells

  • Activation of hepatic stellate cell has been identified as a critical step in hepatic fibrogenesis and is regulated by several factors including cytokines and oxidative stress. In this study, we assayed effects of saponin (CKS), inulin (CKI) and oligo-sugars (CKO) isolated from Platycodon grandiflorum A. DC, changkil (CK) on experimental cell cycle arrest and apoptosis in hepatic stellate cell line (HSC-T6). CKS induced cell arrest at G$_1$. CKS also reduced intercellular reactive oxygen species and collagen synthesis in hydrogen peroxide-induced oxidative stress and acetaldehyde-stimulated collagen synthesis, respectively, in HSC-T6 cells.
