수인성 아질산 급성 노출에 따른 잉어, Cyprinus carpio의 반수치사농도, 혈액학적 성상 및 혈장성분의 영향 (Effects of lethal concentration, hematological parameters and plasma components of common carp, Cyprinus carpio by waterborne acute nitrite exposure)

  • Common carp (Cyprinus carpio) (Weight 23.05 ± 6.98 g, Length 12.65 ± 1.38 cm) were exposed to waterborne nitrite at 0, 50, 100, 200, 400 and 800 mg NO2 -/L for 96 hours. The lethal concentration 50 (LC50) of C. carpio exposed to waterborne nitrite was 398.6 mg NO2 -/L. Hemoglobin, hematocrit and RBC count were significantly decreased by waterborne nitrite exposure. The MCV (mean mean corpuscular volume) (µl), MCH (mean corpuscular hemoglobin) (pg) and MCHC (mean corpuscular hemoglobin concentration) (%) were significantly increased. The inorganic component, plasma calcium, was significantly increased, and the organic components such as plasma cholesterol and total protein were significantly increased showing a similar tendency with calcium. In enzymatic components, the AST and ALT were also significantly increased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of C. carpio.

Lotus leaf alleviates hyperglycemia and dyslipidemia in animal model of diabetes mellitus

  • The purpose of this study was to investigate the effects of lotus leaf on hyperglycemia and dyslipidemia in animal model of diabetes. Inhibitory activity of ethanol extract of lotus leaf against yeast ${\alpha}$-glucosidase was measured in vitro. The effect of lotus leaf on the postprandial increase in blood glucose levels was assessed in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without lotus leaf extract (500 mg/kg) was administered to the rats after an overnight fast, and postprandial plasma glucose levels were monitored. Four-week-old db/db mice were fed a basal diet or a diet containing 1% lotus leaf extract for 7 weeks after 1 week of acclimation to study the chronic effect of lotus leaf. After sacrifice, plasma glucose, insulin, triglycerides (TG), total cholesterol (CHOL), high-density lipoprotein (HDL)-CHOL, and blood glycated hemoglobin levels were measured. Lotus leaf extract inhibited ${\alpha}$-glucosidase activity by 37.9%, which was 1.3 times stronger than inhibition by acarbose at a concentration of 0.5 mg/mL in vitro. Oral administration of lotus leaf extract significantly decreased the area under the glucose response curve by 35.1% compared with that in the control group (P < 0.01). Chronic feeding of lotus leaf extract significantly lowered plasma glucose and blood glycated hemoglobin compared with those in the control group. Lotus leaf extract significantly reduced plasma TG and total CHOL and elevated HDL-CHOL levels compared with those in the control group. Therefore, we conclude that lotus leaf is effective for controlling hyperglycemia and dyslipidemia in an animal model of diabetes mellitus.

  • Mirror carp, Cyprinus carpio nudus (weight 26.2±3.1 g, length 11.8±0.5 cm) were exposed to waterborne total ammonia nitrogen (TAN) at 0, 2, 4, 8, 16, 32, 64 and 128 mg TAN/L. The lethal concentration 50 (LC50) of mirror carp, C. carpio nudus exposed to waterborne ammonia was 60.38 mg TAN/L. The red blood cell (RBC) count was significantly reduced by ammonia exposure, whereas there was no significant changes in the hemoglobin concentration and hematocrit value. In the plasma components, glucose, total protein, aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by ammonia exposure. The results of this study suggest that the ammonia exposure to C. carpio nudus affects the survival rates, hematological parameters and plasma components as toxicity.

Supplementation of Pork Patties with Bovine Plasma Protein Hydrolysates Augments Antioxidant Properties and Improves Quality

  • This study investigated the effects of bovine plasma protein (PP) hydrolysates on the antioxidant and quality properties of pork patties during storage. Pork patties were divided into 4 groups: without butylated hydroxytoluene (BHT) and PP hydrolysates (control), 0.02% BHT (T1), 1% PP hydrolysates (T2), and 2% PP hydrolysates (T3). Pork patty supplemented with PP hydrolysates had higher pH values and lower weight loss during cooking than the control patties. Results showed that lightness and hardness both decreased upon the addition of PP hydrolysates. All samples containing BHT and PP hydrolysates had reduced TBARS and peroxide values during storage. In particular, 2% PP hydrolysates were more effective in delaying lipid oxidation than were the other treatments. It was concluded that treatment with 2% PP hydrolysates can enhance the acceptance of pork patty.

Antihyperlipidemic Effect of Dietary Lentinus edodes on Plasma, Feces and Hepatic Tissues in Hypercholesterolemic Rats

  • We investigated diet supplementation with shiitake mushroom fruiting bodies on biochemical and histological changes in hypercholesterolemic rats. Six-wk old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. A diet containing 5% Lentinus edodes fruiting bodies given to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and the LDL/high-density lipoprotein ratio by 34.33, 53.21, 75.00, 34.66, 25.73, and 71.43%, respectively. Feeding mushroom also significantly reduced body weight in hypercholesterolemic rats. However, it had no detrimental effects on plasma albumin, total bilirubin, direct bilirubin, creatinine, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that L. edodes significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red-O staining showed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that shiitake mushrooms could be recommended as a natural cholesterol lowering substance in the diet.

Effects of Dietary Supplementation with Branched-chain Amino Acids (BCAAs) during Nursing on Plasma BCAA Levels and Subsequent Growth in Cattle

  • To determine the effects of short-term dietary supplementation of branched-chain amino acids (BCAAs) during nursing (from 3 to 28 days of age) on plasma BCAA levels and subsequent growths in cattle, 12 nursing male Holstein calves, randomly assigned to control and treatment groups (n = 6 in each group), orally received a daily supplement of essential BCAAs (2 g/kg body weight/day; 1:1:1 of valine, leucine and isoleucine) or not. The plasma BCAA levels increased linearly after the administration. During the treatment period, average daily gain (ADG) was lower in the treatment group (0.43${\pm}$0.07 kg/day) than the controls (0.71${\pm}$0.07 kg/day, p<0.05). However, at 2 months of age, ADG was significantly higher in the BCAA-treated group (1.16${\pm}$0.26 kg/day vs. 0.51${\pm}$0.06 kg/day, p<0.05). Furthermore, at age 8, 9 and 10 month, ADG in the treated group (1.35${\pm}$0.23, 1.46${\pm}$0.07 and 1.60${\pm}$0.16 kg/day, respectively) showed a linear increase and was significantly higher than that in the control group (0.88${\pm}$0.14, 0.70${\pm}$0.21 and 1.11${\pm}$0.11 kg/kg, respectively, p<0.05). Overall, ADG was 15.6% higher in the treatment group (1.26${\pm}$0.05 kg vs. 1.09${\pm}$0.04 kg; p<0.05). The final body weight at slaughter was 14.8% higher in the treatment group (759.5${\pm}$17.7 kg vs. 661.7${\pm}$21.2 kg, p<0.01). Thus, the supplementation of BCAAs during nursing improves ADG and carcass weight in cattle and is a useful husbandry technique for beef cattle.

  • Novel, effective methods to control and prevent spoilage and contamination by pathogenic microorganisms in meat and meat products are in constant demand. Non-thermal pasteurization is an ideal method for the preservation of meat and meat products because it does not use heat during the pasteurization process. Atmospheric pressure cold plasma (APCP) is a new technology for the non-thermal pasteurization of meat and meat products. Several recent studies have shown that APCP treatment reduces the number of pathogenic microorganisms in meat and meat products. Furthermore, APCP treatment can be used to generate nitrite, which is an essential component of the curing process. Here, we introduce the effectiveness of APCP treatment as a pasteurization method and/or curing process for use in the meat and meat product processing industry.

췌장 콜레스테롤 에스터레이즈 저해제인 초두구 추출물의 혈중 콜레스테롤 저해효과 (Plasma Cholesterol-Lowering Effects of Alpiniae katsumadaii Extract as an Inhibiter of Pancreatic Cholesterol Esterase Activity)

  • 췌장에서 분비되는 cholesterol esterase(CEase)와 소장의 acyl coenzyme A: cholesterol acyltransferase(ACAT)는 흡수된 콜레스테롤을 다시 ester화하는데 관여한다. 한방재료 중 건위, 진토 및 수렴지사작용 등을 가지는 초두구(Alpiniae Katusumadaii Semen)의 에탄올 추출물이 in vitro에서 cholesterol esterase활성에 대하여 강한 저해작용을 보였으며 에탄올추출물의 용매분획 중 클로로포름분획이 다른 분획들, 즉 에틸 아세테이트분획, 부탄올분획 및 물 분획보다 강하였다. 초두구의 클로로포름분획이 고 콜레스테롤식이를 섭취시킨 rat에서 혈중 콜레스테롤치를 저하시키는 효과가 있는지 측정한 실험에서 chloroform분획을 100 mg/kg 및 150 mg/kg 섭취시켰을 경우 각각 총cholesterol 함량이 11.8% 및 20.8% 감소하였으며 HDL-cholesterol 함량 및 동맥경화지수는 정상군에 가깝게 회복된 것으로 나타났다. 또한 초두구 chloroform분획의 독성검사 결과 mouse에서 $LD_{50}$은 1,625mg/kg으로 산출되었다.
