Alterations of Hematological Parameters, Plasma Constituents and Antioxidant Responses in the Sablefish Anoplopoma fimbria Depending on Salinity (염분농도에 따른 치어기 은대구(Anoplopoma fimbria)의 혈액학적 성상, 혈장성분 및 항산화반응의 변화)

  • Juvenile Anoplopoma fimbria (mean length $15.6{\pm}1.4cm$, mean weight $68.7{\pm}4.3g$) were exposed to 4 months with the different levels of salinity [100 (35.0), 90 (31.5), 80 (28.0), 70 (24.5), 60 (21.0), 50 (17.5), and 40 (14.0) % (psu)] for 4 months. Hematological parameters such as red blood cell (RBC) counts, hematocrit (Ht), and hemoglobin (Hb) concentrations were substantially decreased under salinities of 50% psu or lower. Of the measured inorganic plasma constituents, magnesium was notably decreased, whereas there was no effect on calcium. Among organic plasma components, glucose and cholesterol were significantly increased, and total protein was decreased. Among enzyme plasma components, glutamic oxalate transaminase (GOT), glutamic pyruvate transaminase (GPT), and alkaline phosphatase (ALP) were significantly increased under salinities of 50% psu or lower. Antioxidant responses such as glutathione S-transferase (GST) and glutathione (GSH) were significantly decreased at salinities of 50% psu or lower. The results of this study indicate that salinity affects the blood parameters, plasma constituents, and antioxidant responses of A. fimbria.

The Changes of NO, nNOS, Norepinephrine by Acupucture at LU7, HT5, PC6 Acupoints in Rats (수삼음경의 락혈 침자가 백서의 혈위 조직내 nNOS. NO와 조직 및 혈장 Norepinephrine의 변화에 미치는 영향)

  • Objectives : A previous study demonstrated that the connecting points of three yang meridians attenuated changes of nNOS, and Norepinephrine(NE) in rats. The current study investigated the changes in nNOS, NO and NE upon the needle insertion at varying depths at the connecting point of three yin meridians of the hand. Methods : Needles were inserted into rats, on both left and right sides of the connecting point, including the LU7, HT5 and PC6 acupoints which are three yin meridians of the hand. After insertion, needles were retained for five minutes. Each acupuncture groups were treated acupuncture at each acupoint and at the depths of superficial, middle and deep layer. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessel was extracted to examine the changes in the expression of nNOS, NO and NE. Results : Compared with the superficial layer group, nNOS production significantly decreased in the middle and deep layer at LU7 acupoint group and in the deep layer at HT5, PC6 acupoint group. The tissue NE decreased in the deep layer on PC6 acupoint and the plasma NE increased at the middle layer at LU7 acupoint group but decreased at the deep layer on at LU7 acupoint group. Conclusions : Acupuncture at connecting points of three yin meridians of the hand can regulate the activities of nNOS, and NE.

Hematological Characteristics of Olive Flounder (Paralichthys olivaceus) in Culture Farm Influenced by Heated Effluent Water from a Power Plant in Summer (여름철 온배수 수역에 인접한 양식장 넙치(Paralichthys olivaceus)의 혈액 성상)

  • Effect of heated water from a power plant in summer on hematological characteristics of flounder (Paralichthys olivaceus) were investigated. Hematocrit level fluctuated between 15.7 and 16.3% in control farm, but it significantly increased to 17.7% in the high-WT farm. The corresponding values for hemoglobin concentration were 9.9~18.0 $g/㎗$ for the control farm and 6.2~19.4 $g/㎗$ for the exposed farm. The plasma cortisol remained between 0.6 and 2.8 ng/$ml$ in the former but significantly decreased 317.1 to 3.3 ng/$ml$ in the later. The plasma glucose also decreased from 36.5~46.0 to 23.5~36.5 mg/$dl$. Plasma osmolality decreased from 551.5~597.0 to 391.0~466.5 mOsm/kg.

Effect of Coicis Semen on Starvation Stress in Mice (억이인이 생쥐의 기아 Stress에 미치는 영향)

  • Objective : In 2001, the rate of obesity in Korea reached 30.6%. There are many therapeutic ways to reduce body weight, such as low and very low calorie diet, exercise therapy, behavior modification therapy, etc. However, in many cases the patients feel stress under obesity treatment because of starvation. This study was aimed to evaluate the anti-starvation stress effect of Coicis Semen on mice. Methods : First, the mice were divided into 6 groups : Normal (group with no starvation), Control (administrated normal saline 6 times before starting 36 hours starvation), and Samples A, B, C, and D (administrated 0.25, 0.5, 1.0, 3.0 g/kg Coicis Semen respectively 6 times before starting 36 hours starvation). Then the plasma corticosterone level and rectal temperature were measured. The norepinephrine, dopamine, DOPAC (dihydroxy-phenylacetic acid), 5-HT (5-hydroxytryptamine) and 5- HIAA (5-hydroxy-indole-acetic acid) in the hypothalamus were measured by the HPLC method. Result : I. The rectal temperature in Sample group D showed a significant difference (P<0.05) compared with the Control group. 2. The DOPAC in Sample groups A, C and D showed the significant difference (P<0.05) compared with the Control group. Conclusion : It might be recognized that Coicis Semen has an anti-starvation stress effect.

Effects of Acupuncture at the Sea Point on the Changes of Plasma and Tissue Levels of NO, nNOS, Norepinephrine in Rats (12경맥(經脈)의 오수혈중(五輸穴中) 합혈(合穴) 침자(鍼刺)가 백서(白鼠)의 혈위(穴位) 조직내 NO, nNOS와 조직 및 혈장 Norepinephrine의 변화에 미치는 영향(影響))

  • Objectives : This study is to observe the changes in the expression of neurotransmitters, such as NO, nNOS, and NE, upon the needle insertion to the sea points, which is one of the five transport points. Methods : Needles were inserted into rats, on both left and right sides of all sea points, including the LU5, PC3, HT3, LI11, TE10, SI8, SP9, LR8, KI10, ST36, GB34, and BL40, which are the sea points of five transport points for 12 meridian vessels. After insertion, needles were retained for five minutes. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessels were extracted to examine the changes in the expression of NO, nNOS and NE. Results : In terms of the effect in NO production, there was a significant decrease only in the LU5 point, whereas there was a significant increase in the TE10 point alone. In terms of the expression of nNOS within tissues, none of the experimental groups showed significant changes based on the results of immunohistochemistry and western blotting. Regarding the formation of norepinephrine within tissues, the HT3, SP9, and KI10 point showed a significant decrease, while the PC3 and LR8 point showed a significant increase. Production of plasma norepinephrine was significantly increased at the TE10, SP9, LR8, GB34, and BL40 point. Conclusions : The effect of needles applied at the sea points of five transport points of 12 meridian vessels on the functions of NO, nNOS, and NE could be observed, and it is considered that the effect of needle stimulation on nervous system disorders could be studied through additional researches based on this one.

Development of Modeling System for Assessing Essential Amino Acid Requirements Using Surgically Modified Rainbow Trout (외과적 수술에 의한 송어의 혈장 아미노산 농도 측정을 이용한 아미노산 요구량 설정 모델 개발에 관한 기초연구)

  • A new technique combining forced-feeding and dorsal aorta cannulation was developed to monitor concentration of nutritions in the blood circulation and their metabolites in rainbow trout. To study the effect of dorsal aorta cannulation on stress, 30 rainbow trout (523$\pm$5.4 g; Mean$\pm$SD) were divided into 6 groups of 5 individuals each. A group was anesthetized and blood samples were taken at 0, 3, 6, 12, 24 or 48 h after dorsal aorta cannulation. Hematocrit peaked at 6 h and returned to 0 values by 12 h after dorsal aorta cannulation. Plasma cortisol and glucose concentrations also peaked at 6 h and returned to 0 values by 48 h after dorsal aorta cannulation. Based on the plasma cortisol and glucose concentrations, the rainbow trout recovered from the operation of dorsal aorta cannulation within 48 h. To compare the patterns of plasma free amino acid concentrations after force-feeding in the fish with dorsal aorta cannulation, 5 dorsal aorta cannulated individuals (511$\pm$6.2 g) were kept in a cage. After 48 h starvation, they were anesthetized and blood samples were taken at 0, 4, 8, 12, 24, 36 or 48 h after forced-feeding. The concentration of all plasma free amino acids, except isoleucine, leucine, phenylalanine, and tryptophan, also peaked at 4 h and returned to 0 values by 24 h after feeding. The combined technique allows forced-feeding and repeated sampling of blood in rainbow trout with minimum stress.

The inhibition of Hypertension-related Response by $17\beta$-estradiol and the Increase of $17\beta$-estradiol Activity by Electrical Stimulation ($17\beta$-estradiol의 고혈압 유도반응 억제와 인체적용 전기자극의 $17\beta$-estradiol 활성 증가)

  • Purpose: $17\beta$-estradiol is the most active endogenous estrogen, which is related to favorable changes in the plasma lipid profile, to relaxation of the coronary vessels, and to a decrease in platelet aggregation and vascular smooth muscle cell migration. However, although the beneficial effect of estrogens on plasma lipoproteins (ie, lowering low-density lipoprotein and increasing high-density lipoprotein cholesterol) contributes to cardiovascular protection, it does not fully account for the protective effect, particularly in the application of physical therapy, including low frequency electrical stimulation. Methods: The aim of this study was to demonstrate the inhibition of stressors, such as endothelin-1 (ET-1), serotonin (5-hydroxytryptamine, 5-HT), prostaglandin $F2\alpha$ ($PGF2\alpha$), and a protein kinase C (PKC) activator 12-deoxyphorbol 13-isobutyrate (DPB), induced isometric tension by $17\beta$-estradiol in vascular smooth muscle strips, respectively. In addition, the effects of low frequency electrical stimulation at the meridian points (CV-3, -4, Ki-12, SP-6, LR-3, BL-25, -28, -32, -52) on the indirect antihypertensive effect were examined by monitoring the changes in the serum $17\beta$-estradiol concentration in healthy volunteers. Results: Isometric tension analysis showed that the responses of inhibited tension by $17\beta$-estradiol were similar to the same stressors in rat aortic smooth muscle strips. Furthermore, although the continued amplitude modulation (AM) type of electrical stimulation was not increased significantly by electrical stimulation, the current of the frequency modulation (FM) type of low frequency electrical stimulation increased the serum $17\beta$-estradiol concentration in normal volunteers. Conclusion: These results, in part, suggest that $17\beta$-estradiol has the capacity to supress stressor-induced muscle tension, and electrical stimulation, particularly current of the FM type, has a modulatory effect on the sex steroid hormones, particularly $17\beta$-estradiol, in healthy volunteers.

Changes in metabolic rate and hematological parameters of black rockfish (Sebastes schlegeli) in relation to temperature and hypoxia (수온과 저산소에 따른 조피볼락(Sebastes schlegeli)의 호흡대사와 혈액성상의 변화)

  • Experiments were performed to investigate changes in metabolic rate (MO2), critical oxygen saturation (Scrit) and hematological parameters of black rockfish, Sebastes schlegeli exposed to hypoxia at 15, 20 and 25℃. The MO2 was measured at an interval of 10 min using intermittent-flow respirometry. The normoxic standard metabolic rate (SMR) was 116.5±5.5, 188.6±4.2 and 237.4±6.8 mg O2/kg/hr, and Scrit was 22.1±1.2, 30.6±1.5 and 41.9±1.4% air saturation at 15, 20 and 25℃, respectively. Q10 values were 2.62 between 15 and 20℃, 1.58 between 20 and 25℃, and 2.04 over the full temperature range. In the investigation of blood (hematocrit and hemoglobin) and biochemical parameters (plasma cortisol, glucose, electrolyte and osmolality), the rockfish were subjected to Scrit for each temperature during 4 hr. All of hematological parameters of the rockfish exposed to hypoxic water were significantly higher than those of normoxic control. Moreover, blood and biochemical parameters of the rockfish maintained to normoxic water showed the tendency of increase with temperature, and were significantly higher at 25C. As a result of this experiment, it was found that physiological stress due to hypoxia increased at high temperature.

Correlation between Metabolic Syndrome and Plasma Homocysteine Level in Patients with Silent Lacunar Infarction (무증상성 열공성 뇌경색 환자에서 호모시스테인과 대사증후군의 상관관계에 대한 연구)

  • Objectives : This clinical study was done to examine metabolic syndrome (MS) and plasma homocysteine (HCY) level in patients with silent lacunar infarction (SLI) and in normal controls. Methods : A total of 154 patients, who were over 20 years of age and visited the stroke prevention check-up center of a university hospital from December 2006 to December 2010, were examined by brain CT or brain MRI, and classified into two groups. We compared the components of MS and HCY levels between patients with SLI (n=74) and normal controls (n=80). Modified ATP III definition was used for diagnosis of MS while Korean standard for waist circumference was used. Results : Prevalence of MS was significantly higher in the SLI group than the normal group. HCY was also significantly higher in the SLI group than the noraml, and the odds ratio (OR) for SLI, comparing high HCY level (${\geq}10{\mu}mol/L$) with low HCY level ($<10{\mu}mol/L$), was 3.64 (95% confidence interval (CI); 1.81-7.29, p<.0001). However, there was no correlation between MS and HCY in the SLI group. Prevalence of diabetes and hypertension (HT) was higher in the SLI group than the normal group, but there was no significant difference in blood lipids level between the SLI and normal groups. Of note, HT itself was enough to be an independent risk factor for SLI (OR; 4.58, 95% CI; 1.91-11.01, p=0.001). Body mass index, waist circumference, waist-hip ratio and visceral fat area were significantly higher in the SLI than normal group, and visceral fat area was enough to be an independent risk factor for SLI (OR; 2.41, 95% CI; 1.04-5.59, p=0.040). Conclusions : SLI is shown to have significant correlation with HCY and prevalence of MS, however there is no relationship between HCY and prevalence of MS in patients with SLI.

Comparison of Blood Physiology in Juvenile Black Seabream (Acanthopagrus schlegeli) Reared in Converted Freshwater from Seawater and Seawater from Freshwater (해수사육에서 담수사육으로, 담수사육에서 해수사육으로 전환된 감성돔 치어, Acanthopagrus schlegeli의 혈액생리학적 비교)

  • Comparison of blood properties in juvenile black seabream (Acanthopans schlegeli) between transfer from seawater to freshwater (sFW) and transfer from freshwater to seawater (fSW) were investigated for 60 days. Plasma cortisol levels in sFW were significantly increased from 34.2 $\pm$ 28.6 ng/mL at the beginning to 365.6 $\pm$ 136.0 ng/mL at 3 hours, and decreased to the beginning level at 24 hours. However, in fSW, no significant differences in the cortisol levels were recognized throughout experimental period. No significant difference was found in the glucose levels between sFW and fSW. The transfer from seawater to freshwater of juvenile black seabream resulted in reduced $Na^{+}\;and\; Cl^{-}$ concentrations for 24 hours (decreasing from 165.3 $\pm$ 2.5, 146.0$\pm$ 2.2 to 118.3 $\pm$ 12.3, 78.0$\pm$ 7.0 mEq/L, respectively), but these were completely recovered the beginning levels at 30 days. Total protein and AST showed no significant differences between the two rearing conditions, while ALT was markedly elevated at 3 hour in sFW and at 24 hours in fSW. Ht in sFW was increased from 18.5 $\pm$ $0.6\%$ at the beginning to 25.3 $\pm$ $4.0\%$ at 12 hours, and was decreased to the beginning level at 24 hours. Ht, RBC and Hb in fSW were also significantly higher at 12 hours, but recovered to their initial levels at 24 hours. All fish were dead until 50 days in sFW while survival rate in fSW was $85\%$ at the end of experiment.
