Pseudomonas oleovorans Strain KBPF-004 Culture Supernatants Reduced Seed Transmission of Cucumber green mottle mosaic virus and Pepper mild mottle virus, and Remodeled Aggregation of 126 kDa and Subcellular Localization of Movement Protein of Pepper mild mottle virus

  • Efforts to control viral diseases in crop production include several types of physical or chemical treatments; antiviral extracts of a number of plants have also been examined to inhibit plant viral infection. However, treatments utilizing naturally selected microorganisms with activity against plant viruses are poorly documented. Here we report isolation of a soil inhabiting bacterium, Pseudomonas oleovorans strain KBPF-004 (developmental code KNF2016) which showed antiviral activity against mechanical transmission of tobamoviruses. Antiviral activity was also evaluated in seed transmission of two tobamoviruses, Pepper mild mottle virus (PMMoV) and Cucumber green mottle mosaic virus (CGMMV), by treatment of seed collected from infected pepper and watermelon, respectively. Pepper and watermelon seeds were treated with culture supernatant of P. oleovorans strain KBPF-004 or control strain ATCC 8062 before planting. Seeds germinated after treatment with water or ATCC 8062 yielded about 60% CGMMV or PMMoV positive plants, whereas < 20% of KBPF-004-treated seeds were virus-infected, a significantly reduced seed transmission rate. Furthermore, supernatant of P. oleovorans strain KBPF-004 remodeled aggregation of PMMoV 126 kDa protein and subcellular localization of movement protein in Nicotiana benthamiana, diminishing aggregation of the 126 kDa protein and essentially abolishing association of the movement protein with the microtubule network. In leaves agroinfiltrated with constructs expressing the coat protein (CP) of either PMMoV or CGMMV, less full-size CP was detected in the presence of supernatant of P. oleovorans strain KBPF-004. These changes may contribute to the antiviral effects of P. oleovorans strain KBPF-004.

  • Crown gall of stonefruit and nut trees is one of the very few plant diseases subject to efficient biological control. The disease is caused by the soil-inhabiting bacteria Agrobacterium tumefaciens and Agrobacterium rhizogenes and the original control organism was a non-pathogenic isolate of A. rhizogenes strain K84. Control is achieved by dipping planting material in a cell suspension of strain K84 which specifically inhibits pathogenic strains containing a nopaline Ti plasmid. Because the agrocin 84-encoding plasmid (pAgK84) is conjugative, it can be transmitted from the control strain to pathogenic strains which, as a result, become immune to agrocin 84 and cannot be controlled. To prevent this happening, the transfer genes on pAgK84 were located and then largely eliminated by recombinant DNA technology. The resulting construct, strain K1026, is transfer deficient but controls crown gall just as effectively as does strain K84. Field data from Spain confirm that pAgK84 can transfer to pathogenic recipients from strain K84 but not from strain K1026. The latter has been registered in Australia as a pesticide and is the first genetically engineered organism in the world to be released fro commercial use. It is recommended as a replacement for strain K84 to prevent a breakdown in the effectiveness of biological control of crown gall. Several reports indicate that both strains K84 and K1026 sometimes control crown gall pathogens that are resistant to agrocin 84. A possible reason for this is that both strains produce a second antibiotic called 434 which inhibits growth of nearly all isolates of A. rhizogenes, both pathogens and non-pathogens. Crown gall of grapevine is caused by another species, Agrobacterium vitis. It is resistant to agrocin 84 and cannot be controlled by strains K84 or K1026. It is different from other crown gall pathogens in several characteristics, including the fact that, although a rhizosphere coloniser, its also lives systemically in the vascular tissue of grapevine. Pathogen free propagating material can be obtained from tissue culture or, less surely, by heat therapy of dormant cuttings. A number of laboratories are searching for a biocontrol strain that will prevent, or at least delay, reinfection. A non-pathogenic A. vitis strain F/25 from South Africa looks very promising in this regard.

좀향유의 삽목 증식 및 시비에 따른 유묘의 생장 효과 (Cutting Propagation and Seedling Growth Effect According to Fertilizer Application of Elsholtzia minima Nakai)

  • 본 연구는 제주특산식물인 좀향유의 현지 내・외 보존을 위한 기초 연구로서 증식체계를 마련하고 전처리제 처리에 따른 발근 특성과 시비에 따른 유묘의 생장 효과를 등을 알아보았다. 온실과 밀폐상의 온습도 조사결과 평균 온도는 비슷하였으나 공중 습도는 온실보다 밀폐상이 높은 습도를 유지하였으며, 좀향유 삽목 후 온실은 10일, 밀폐상은 7일만에 최초 발근을 확인하여 밀폐상의 빠른 발근율을 보였으며 온실과 밀폐상에서의 생존율, 발근율, 뿌리수 등은 근소하게 밀폐상이 높게 조사되어 온실보다 밀폐상이 효과적이었다. 발근억제물질 제거를 위한 전처리제 처리는 AgNO3 77.5%를 제외한 다른 처리구에서 95% 이상의 높은 발근율로 조사되어 좀향유의 경우 발근억제 물질은 적은 것으로 판단된다. 유묘의 용기별 질소 시비량 및 Osmocote 시비시험 결과 시비량과 용기의 조건에 따라 생존율과 생장 등에 차이를 보였다. 배양판에서 완효성 비료인 Osmocote를 시비한 결과 생존율, 지상부, 지하부 생장 등이 가장 양호하여 좀향유의 유묘 증식을 위해서는 Osmocote 시비가 바람직 할 것으로 판단된다.

매장문화재 보존형 역사공원의 설계 양상 - 능곡선사유적공원, 안산신길역사공원, 용죽역사공원을 대상으로 - (A Design Aspects of Historic Parks Preserving Buried Cultural Heritages - In the Case of Neunggok Prehistoric Remains Park, Ansan Singil Historic Park, Yongjuk Historic Park -)

  • 본 연구는 매장문화재 보존형 역사공원의 설계 여건으로서 관련 법규와 제도를 고찰한 후, 비지정문화재를 보존조치한 능곡선사유적공원, 안산신길역사공원, 용죽역사공원에 대한 사례 연구를 진행하여 설계 양상을 도출하였다. 연구의 결과는 다음과 같다. 첫째, 역사공원 관련 법규에 따르면 유적지의 보존과 활용을 동시에 만족시켜야 하고, 역사관련시설공간, 광장, 휴게공간, 운동공간, 교육문화공간, 편익공간을 도입할 수 있다. 둘째, 매장문화재의 보존만 강조한 공간구성과 동선 체계에 의해서 유구보존공간과 기능적 공간들이 격리되고, 주변 토지이용을 고려한 이용행태를 수용하지 않아서 역사공원의 효용성이 적었다. 셋째, 건축적 방식의 노출 현지보존, 움집 재현, 복토 현지보존 후 철쭉이나 석재로 수혈주거지 위치를 표시하는 소극적 유구보존방식과 관람 위주의 활용으로 인하여 역사공원의 정체성이 약했다. 넷째, 이용자 접근을 막는 울타리가 유구의 체험을 방해하고, 노출된 유구의 상부를 보호하는 수직 구조물이 역사공원의 경관을 압도하였다. 다섯째, 매장문화재 정보를 전달하는 안내판에 전문용어 위주의 텍스트와 발굴 사진만 제시하여 유구 공간을 이해하기 어려웠다.

광물찌꺼기적치장의 안전관리를 위한 적정 모니터링 방안 연구 (Proper Monitoring Methods for Safety Management of Tailings Dam)

  • 광물찌꺼기적치장 안전 모니터링을 위한 적치장 안전에 영향을 미치는 요소를 분석하고 이러한 요소를 감시할 수 있는 계측장비를 검토하였다. 또한, 이러한 결과를 국내 광물찌꺼기 적치장(20개소, 18개 광산)에서 발생되는 결함에 대해 적용 가능한 계측장비를 선정하였다. 요소분석은 표면 침식, 파이핑 및 사면 불안정으로 구분하여 평가하였으며, 이를 다시 세분하여 표면 침식은 식생, 복토층 및 광물찌꺼기, 파이핑은 차수층, 우수배제시설 및 침출수, 사면불안정은 사면, 콘크리트 옹벽 및 석축으로 구분하여 결함의 지표 및 계측항목을 설정하였다. 계측은 주로 상시 및 변위계측이 가능한 CCTV의 활용이 높게 평가되었으며, 전기비저항탐사, 음향계측기, 다점온도모니터링, 구조물경사계, 강우량계 및 유량계 등의 활용도가 높은 것으로 평가되었다. 또한, 국내 적치장에서 발생되는 결함에 적용한 결과, 주로 파이핑과 관련된 차수층, 우수배제시설 및 침출수에서 결함이 주로 발견되어 이와 관련된 계측장비의 활용이 높은 것으로 평가되었다.

자동 채소 정식기 묘 취출 메커니즘 분석(I) (Analysis of Pick-up Mechanism for Automatic Transplanter( I ))

  • 농업현장에서는 각종 작물을 재배할 때, 작물 성장을 빠르고 효과적으로 하기 위하여 일정한 크기로 성장한 묘를 밭으로 옮겨 심게 되는데, 이러한 묘 정식작업을 수행할 때 채소 정식기가 사용된다. 본 채소 정식기는 인력에 의존하고 있는 채소 정식작업의 기계화를 촉진하기 위하여 2조식의 전자동 육묘트레이 공급 및 취출된 묘를 식부호퍼에 이송할 수 있도록 개발하였다. 또한 두 가지 취출방식에 따른 묘 결주율을 비교하여 판단하였다. 핑거타입 묘 취출 시험결과 256주 묘에서 묘를 뽑아내지 못하는 경우가 17주, 상토가 부서지는 경우가 15주 등 결주율이 13.7%로 나타났으며, 포크타입 묘 취출 시험결과 200주 묘에서 묘는 100% 뽑아내지만 이송 중 낙하되는 경우가 6개 등 결주율이 4%로 나타났다. 따라서 전자동 2조식 정식의 묘 취출장치로서 포크타입이 적당한 것으로 판단되었다.

국내 절화국화 재배농가의 재배와 수확 후 관리현황 분석 및 개선 방안 (Analysis and Improvement Plan of Cultivation and Postharvest Management Status of Cut Chrysanthemum Farms in Korea)

  • 본 연구는 국내에서 절화국화를 재배하고 있는 27개 농가를 선정하여 시설과 재배현황, 수확 후 관리 및 유통 실태를 조사하였다. 재배농가들의 60%는 PE필름 비닐하우스에서 토양 관비재배 방법으로 절화국화를 재배하고 있었으며, 전남과 부산은 스탠다드 국화를 주로 재배하고 있었다. 자가채취 또는 업체에서 구매한 삽수를 플러그 육묘하여 발근묘를 생산하고 있었으며, 전남지역에서는 주로 재배상에 직접 삽목하고 있었다. 절화국화를 수확한 후 전처리로 열탕처리 또는 수돗물로 물올림을 실시하는 농가가 66.6%로 나타났다. 예냉은 70.4% 농가에서 실시하지 않았으며, 예냉하는 농가들은 주로 $2{\sim}4^{\circ}C$에서 실시하고 있었다. 절화국화를 수확한 후 유통 물량을 조절하기 위해 70.4%의 농가에서 저장을 하고 있었으며, $2{\sim}4^{\circ}C$에서 48시간 이상 저장하고 있었다. 절화국화를 출하 전에 주로 개인별로 선별하고 있었으며, 일부 수출 농가에서만 공동선별을 하고 있었다. 유통에 있어서 모든 농가들은 국내로 출하하고 있었으며, 이들 농가 중 44.4%는 수출을 겸하고 있었다. 국내 시장으로 출하하는 경우, 63.0%의 농가에서 화훼경매장을 이용하고 있었다.

기상재해와 수도재배상의 대책 (Improvement in Rice Cultural Techniques Against Unfavorable Weather Condition)

  • The climatic impacts have been the environmental constraints with soil characteristics to achieve self sufficiency of food production in Korea. In this paper, the distribution and appearance of impacts and the changes in climatological status due to recent trend of early transplanting of rice are widely discussed to derive some countermeasures against the impacts, being focussed on cultural A long term analysis of the climatic impact appearances of the last 74 years showed that drought, strong wind, flood, cold spell and frost were the major impacts. Before 1970's, the drought damage was the greatest among the climatic impacts; however, the expansion and improvement of irrigation and drainage system markedly decreased the damage of drought and heavy rain. The appearance of cold damage became more frequent than before due to introduction of early transplanting for more thermophilic new varieties. Tongillines which were from Indica and Japonica crosses throw more attention to cold damage for high yields to secure high temperature in heading and ripening stages and lead weakness to cold and drought damage in early growth stage after transplanting. The plants became subject to heavy rain in ripening stage also. For the countermeasures against cold damage, the rational distribution of adequate varieties according to the regional climatic conditions and planting schedule should be imposed on the cultivation. A detoured water way to increase water temperature might be suggestable in the early growth stage. Heavy application of phosphate to boost rooting and tillering also would be a nutritional control method. In the heading and ripening stages, foliar application of phosphate and additional fertilization of silicate might be considerable way of nutritional control. Since the amount of solar radiation and air temperature in dry years were high, healthy plants for high yield could be obtained; therefere, the expansion of irrigation system and development of subsurface water should be performed as one of the national development projects. To minimize the damage of strong wind and rainfall, the rational distribution of varieties with different growing periods in the area where the damage occurred habitualy should be considered with installation of wind breaks. Not only vertical windbreaks but also a horizontal wind break using a net might be a possible way to decrease the white heads in rice field by dry wind. Finally, to establish the integrated countermeasures against the climatic impacts, the detailed interpretation on the regional climatic conditions should be conducted to understand distribution and frequency of the impacts. The expansion of observation net work for agricultural meteorology and development of analysis techniques for meteorological data must be conducted in future together with the development of the new cultural techniques.

제주지역의 기후변화에 따른 농가의 작목전환 실태 -한농대 졸업생 감귤 농가를 중심으로- (A study on the crop switching of farmers in Jeju Islands related to the climate changes - focused on the citrus farms of the graduates of the KNCAF -)

  • The aim of this study is to investigate to which degree farmers did the crop switching and cultivar renewal as a confrontational strategy to climate change, and which problems they had in that process, and then to provide the supporting plans for them. We conducted a questionnaire survey of 15 citrus farmers of the KNCAP graduates in Jeju Island. Most of the survey respondents agreed to the climate change of Jeju Island and the subtropical climate of its coastal area. The farmers have experienced irregular weather such as abnormal high temperature, frequent rain, and droughts, resulting in the harmful insects and new weeds attack. As the climate change strategies, they are adopting a greenhouse culture system, improving a soil drainage using reorganization of planting space, making a new pest management program, and trying to switch a crop to subtropical fruits. It is expected that 50% of the survey respondents have changed their crops or will do; and 73 % of them have changed cultivar or have a plan to do. Only a few farmers directly pointed to a reason for their efforts to change the crops or to renew the cultivars as the anti-climate change strategy, however, most farmers answered the reason was to increase profitability by meeting their consumers' tastes. Presently, it is not the anti-climate change strategy but increase of profitability by meeting the consumers' needs the reason why most of the survey respondents have changed their crops or renewed the cultivars, while a few of them switched their crops to a subtropical fruit trees due to climate change. On the crop switching, they had some difficulties such as a labor shortage, availability of land, operating costs and instable income. On the cultivar renewal, also, they encountered the lack of cultivating techniques for new cultivars and the dim future for the new market. In long-term perspective, Jeju's farmers need new information and educational programs about the effect of climate change on agriculture of Jeju, and cultivation techniques for new crops and new cultivars.

An Analysis of the Rice Situation in Nicaragua for Improving National Production.

  • Nicaragua is located in Central America, climatic conditions are considered tropical dry forest. Statistics reflex that in Nicaragua exits 24,000 rice farmers. National rice production only covers 73% of the national consumption. It exists two sowing system: irrigation and rainfed. Varieties used in both systems are mid-late maturity (120-135 days), there are 14 released varieties for irrigation, eight for rainfed, and eight landraces used in rainfed. The current breeding system (introduction of lines from Colombia) has increased the national production, however, has some limitation due to the lack of enough variability, reducing the proability of finding good genotypes and therefore the possibility of satisfying 100% of the demand. The purpose of this study was to analyze the problems that must be resolved in the short and long term to improve rice productivity in Nicaragua. In this paper we explain some proposal for an improvement plan. The selection of varieties with high adaptability to various cultivation environmental conditions it is necessary, also to thoroughly manage seed purity to supply certified seeds. In rice cultivation technology, it needs to improve seedling standing and weeding effect by improving soil leveling and water-saving cultivation technology. Also, proper fertilization and planting density must be established in irrigated and rain-fed areas. Furthermore, capacity must be strengthened by collecting and training with the most recent agricultural technology information, as well as by revitalizing the union rather than the individual farmer. It is necessary to develop varieties highly adaptable to the Nicaraguan cultivation environment, as well as to expand irrigation facilities and cultivation technology suitable for weather conditions in rain-fed areas. Last, it is necessary to maintain the consistency of agricultural policy for continuous and stable rice production in response to climate change events such as drought or intermittent heavy rain.
