Observation of Growth Inhibition of Elsinoe fawcettii on Satsuma Mandarin Leaves Pre-treated with Rhizobacterial Strains by a Scanning Electron Microscope (식물근권세균을 처리한 감귤 잎에서 주사전자현미경을 통한 감귤 더뎅이병균의 생장 억제 관찰)

  • Elsinoe fawcettii causing citrus scab was suppressed by rhizobacterial strains such as Burkholderia gladioli MRL408-3, TRH423-3 and Pseudomonas fluorescens THJ609-3, TRH415-2 having antifungal activity. The leaf surface of Satsuma mandarin, which was pre-treated with the rhizobacterial strains, was observed by a scanning electron microscope (SEM) after inoculation with E. fawcettii. The number of lesions was reduced on the leaves pre-treated with the rhizobacterial strains compared to those of untreated leaves. Especially, the lesions numbers was apparently reduced on the leaves pre-treated with B. gladioli MRL408-3. The observation by SEM revealed that not only the germination rate but also the length of germ tube of the pathogen were decreased on the rhizobacterial strains pre-treated leaves. These inhibition of the fungal growth was more strongly expressed on the leaves pre-treated with commercial fungicide imibenconazole, by which the lesions was rarely found on the leaves. Based on these results, it was suggested that rhizobacterial strains may inhibit the germination and growth of the E. fawcettii on the surface of citrus leaves, resulting in decrease of disease severity.

Gray Mold of Nephrolepis Caused by Botrytis cinerea (Botrytis cinerea에 의한 네프로레피스 잿빛곰팡이병)

  • In February of 2000-2001, the gray mold disease occurred on nephrolepis (Nephrolepis sp.) grown in a flower nursery farm in Suwen, Korea. Typical symptoms were water-soaked brown or blackish lesions on terminal leaf blades. Severely infected leaves were entirely blighted with grayish fungal mycelia formed on the surface. Conidia of the fungus in mass were hyaline or gray, 1-celled, mostly ellipsoid or ovoid and $13.5{\sim}16.9{\times}6.8{\sim}9.2{\mu}m$ in size. Conidiophores were formed on PDA with $8.7{\sim}11.1{\mu}m$ in width. The sclerotia were readily formed within 2 or 3 days on PDA. In addition, the Biolog database gave the causal fungus a high similarity to Botrytis cinerea (78%) with a match probability of 100%. Pathogenicity of the causal organism was proved according to Koch's postulate. The causal organism was identified as Eotrytis cinerea based on its mycological characteristics and utilization of carbon sources with Biolog system as supporting data. This is the first report of gray mold of nephrolepis caused by Botrytis cinerea in Korea.

Occurrence of Freesia Basal Rot Caused by Sclerotium sp. (Sclerotium sp.에 의한 프리지아 균핵병 발생)

  • Basal rot of freesia caused by a Sclerotium sp. occurred at Incheon areas. Incidence of the disease reached up to 45% and averaged 17.0% in the fields. Typical symptoms consisted of sheath dry and leaf blight due to rots on basal leaves. The causal fungus was identified as Sclerotium sp. based on following mycological characteristics. The fungus formed sclerotia on cultural media and plant tissues, but did not produce asexual spores. On cultural medium, aerial mycelia of the fungus changed color from white to clay with cultural age and smelled musty odor. Numerous irregular and elliptical black microsclerotia of the fungus were formed on potato dextrose agar (PDA) after 5 days of incubation at $25^{\circ}C$ and sized $115{\sim}200{\times}95{\sim}150 (av. 145{\sim}126.5){\mu}m$. The fungus grew at $10{\sim}32^{\circ}C$ and $pH 4.0{\sim}8.5$. However, the optimal temperature and pH for mycelial growth of the fungus were $24^{\circ}C$ and 5.5 respectively. The isolate showed present pathogenicity to not only freesia but gladiolus in the pathogenicity test, and the symptoms were similar to those observed in the fields. Basal rot of freesia caused by Sclerotium sp. is firstly reported in Korea.

  • It is worth verifying the effectiveness of data integration between data with different features. This study investigated whether the data integration affects the accuracy of deep neural network (DNN), and which integration method shows the best improvement. This study used two different public datasets. One public dataset was taken in an actual farm in India. And another was taken in a laboratory environment in Korea. Leaf images were selected from two different public datasets to have five classes which includes normal and four different types of plant diseases. DNN used pre-trained VGG16 as a feature extractor and multi-layer perceptron as a classifier. Data were integrated into three different ways to be used for the training process. DNN was trained in a supervised manner via the integrated data. The trained DNN was evaluated by using a test dataset taken in an actual farm. DNN shows the best accuracy for the test dataset when DNN was first trained by images taken in the laboratory environment and then trained by images taken in the actual farm. The results show that data integration between plant images taken in a different environment helps improve the performance of deep neural networks. And the results also confirmed that independent use of plant images taken in different environments during the training process is more effective in improving the performance of DNN.

Effect of Light Receiving rate on Growth and Quality of Ginseng Cultivated in Plastic House

  • Ginseng is a shade-plant cultivated using shading facilities. However, at too low light levels, root growth is poor, and at high light levels, the destruction of chlorophyll reduces the photosynthesis efficiency due to leaf burn and early fall leaves. The ginseng has a lightsaturation point of 12,000~15,000 lux when grown at 15 to 20℃ and 9,500 lux at 25℃. This study was conducted to select the optimal light intensity of 3-year-old ginseng grown in blue-white film plastic house. The seeds were planted in the blue-white film plastic house with different light receiving rate (March 17, 2020). Between April and September, the average air temperature in the house was 20.4-20.7℃. Average soil temperature was 18.3℃-18.5℃. The chemical properties of the test soil was as follows. The pH level was 7.0-7.4, EC was 0.5-0.6 dS/m, OM was at the levels of 33.6-37.7 g/kg, P2O5 was 513.0-590.8 mg/kg, slightly higher than the allowable 400 mg/kg. The amount of light intensity, illuminance, and solar radiation in the blue-white film house was increased as the light-receiving rate increased and the amount of light intensity was found to be 9-14% compared to the open field, 8-13% illuminance and 9-14% solar irradiation respectively. The photosynthesis rate was the lowest at 3.1 µmolCO2/m2/s in the 9% light blue-white plastic house and 4.2 and 4.0 µmolCO2/m2/s in the 12% and 14% light blue-white plastic house, respectively. These results generally indicate that the photosynthesis of plants increases with the amount of light, but the ginseng has a lower light saturation point at high temperatures, and the higher the amount of light, the lower the photosynthetic efficiency. The SPAD (chlorophyll content) value decreased as the increase of light-receiving rate, and was the highest at 32.7 in 9% light blue-white plastic house. Ginseng germination started on April 11 and took 13-15 days to germinate. The overall germination rate was 82.9-85.8%. The plant height and length of stem were long in the 9% light-receiving plastic house. The diameter of stem was thick in the 12-14% light-receiving plastic house. In the 12% and 14% light-receiving plastic house, the length and diameter of taproot was long and thick, so the fresh weight of root per plant was 20 g or more, which was heavier than 16.9 g of the 9% light-receiving plastic house. The disease incidence (Alternaria blight, Gray mold and Damping-off etc.) rate were 0.9-2.7%. The incidence of Sclerotinia rot disease was 7.5-8.4%, and root rot was 0-20.0%. The incidence ratio of rusty root ginseng was 34.4-38.7% level, which was an increase from the previous year's 15% level.

Controlling Effect of Agricultural Organic Materials on Phytophthora Blight and Anthracnose in Red Pepper (고추 역병과 탄저병에 대한 친환경유기농자재의 방제 효과)

  • A total of 20 agricultural organic materials including inorganic compounds, plant oils and plant extracts were used in the study for assessing the control efficacy on pepper diseases. Among inorganic compounds, only copper hydroxide showed inhibitory effect on both Phytophthora capsici causing Phytophthora blight and Colletotrichum acutatum causing anthracnose. Phosphorous acid inhibited the growth of P. capsici on PDA, and Sulfur/quicklime had it on that of C. acutatum. Plant essential oil, rosemary oil, and rapeseed oil among plant oils and plant extract of Japanese apricot/ginkgo nut inhibited the mycelial growth of the two pathogens. In the screening using pepper plant seedlings, the control efficacy on Phytophthora blight in 6-leaf stage of seedling was superior to that in 4-leaf stage of seedling. A protective effect on Phytophthora blight was displayed by copper hydroxide, sulfur/quicklime, water soluble calcium, phosphorous acid, plant essential oil, and cloves extract. When C. acutatum was inoculated by the non-wound method, copper hydroxide and rapeseed oil showed excellent protective activities with control values of 91.3% and 82.6%, respectively. However, copper hydroxide did not show any activity, when C. acutatum was inoculated after wounding pepper fruits. All organic materials never showed the curative effect on Phytophthora blight and anthracnose in pepper seedling assay and fruit assay.

Occurrence of Brown Patch on Kentucky Bluegrass Caused by Rhizoctonia solani AG-1 IB (Rhizoctonia solani AG-1 IB에 의한 Kentucky Bluegrass 갈색잎마름병 발생)

  • Brown patch caused by Rhizoctonia solani AG-1 IB occurred on Kentucky bluegrass during late May through early October 2010 at golf course in Gyeongbuk Province, Korea. Disease symptoms on the turfgrass for spring season were leaf blights dying from the leaf tip, which appeared patches of brown color in the field. However, it appeared patches of dark brown color or gray brown color in fall. The fungus (B-7 isolate) of brown patch was isolated from the diseased leaf tissue and cultured on potato-dextrose agar (PDA) for identification. The young hyphae had acute angular branching and few septa and mature hyphal branches showed about 90-degree angles and development of monilioid cells, which were morphologically identical to Rhizoctonia solani AG-1 IB reported previously. DNA sequences of ribosomal RNA gene (internal transcribed spacer) of the fungus were homologous with similarity of 99% to those of Rhizoctonia solani AG-1 IB isolates in GenBank database, confirming the identity of the causal agent of the disease. Pathogenicity of the fungus was also confirmed on the creeping bentgrass and Kentucky bluegrass by Koch's postulates. This is the first report of brown patch on Kentucky bluegrass caused by Rhizoctonia solani AG-1 IB in Korea.

Yield Loss Assessment and Determination of Control Thresholds for Powdery Mildew of Eggplant (Solanum melongena) (시설 가지 흰가루병 발생정도에 따른 수량변화와 경제적 방제수준 설정)

  • This study was carried out to develop the economic thresholds for powdery mildew on eggplant. To investigate the relationship between powdery mildew incidence degree and yield, experimental plots with ten treatments as the initial disease degree were established. Disease severity exhibited negative and significant correlation with fruit weight and number of fruit. There existed close correlation between rate of diseased leaf area and yields in the greenhouse (control with fungicide Y = -36.5X+2938.5 $R^2=0.91$ $r=-0.913^*$, no fungicide: Y = -29.57X+2574.4 $R^2=0.73$ $r=-0.858^*$). There existed close correlation between rate of diseased leaf area and yield loss in the greenhouse (Y = 1.27X-2.4 $R^2=0.88$ $r=0.91^*$). The economic thresholds for powdery mildew on eggplant was below 14.7% rate of diseased leaf area per plant in the greenhouse.

Introduction of VP6 Gene into Potato Plant by Agrobacterium-mediated Transformation and Analysis of VP6 Expression in Transgenic Potatoes (Rotavirus VP6 유전자의 감자식물체내로의 도입과 형질전환체의 발현분석)

  • A VP6 fragments was subcloned with BamHI in the binary pMBP-1 vector under Califlower Mosaic Virus (CaMV) 355 promoter and neomycin phosphotransferase II (npt II) gene. The recombinant binary vector was mobilized into Agrobacterium-tumefaciens LBA4404 by the freeze-thaw method and potato (Solanum tubensum L. cv Desiree) was transformed by modified leaf-disc cocultivation. Shoots were induced on MS medium with 0.01 mg/L NAA, 0.1 mg/L GA$_3$, 2.0 mg/L Zeatin, 100.0 mg/L kanamycin, 500.0 mg/L carbenicillin. In order to identify the copy number of VP6 into potato plant, total genomic DNA was isolated from transgenic potato and analysed by Southern blotting. Genomic DNA and total mRNA analysis demonstrated the incorporation of the foreign gene into the potato genome, as well as their transcription.

Combined Treatment with Gamma Irradiation and Sodium Dichloroisocyanurate to Control Leaf Blight and to Preserve Cut Lilies (절화 백합 잎마름병 방제 및 품질 보존을 위한 감마선 150 Gy와 Sodium Dichloroisocyanurate 병용처리)

  • For the purpose of practical application of gamma irradiation for export lily without the ionic energy damage, 150 Gy gamma ray was suggested as the safe level on cut lilies, the oriental cultivar Siberia (white) and Sorbonne (pink). The combined treatments of 150 Gy gamma ray and sodium dichloroisocyanurate (NaDCC) at 0, 40, 100, and $200{\mu}g/l$, were tried to control lily leaf blight caused by Botrytis elliptica. Incidences of petal blight on the two oriental lilies at control were 93%-95%, whereas those at the combined treatments of 150 Gy and $100{\mu}g/l$ or higher of NaDCC were 76%-83%. However, severity of petal blight was not reduced by the combined treatments. Gamma irradiation at 150 Gy did not change the cut lily quality such as improve cut lily longevity of vase-life compare with the control. In addition, fresh weight, chlorophyll contents, and the color of pink petals of Sorbonne were not changed by 150 Gy of gamma irradiation. Therefore, we recommend the combined treatment of 150 Gy gamma irradiation and $100{\mu}g/l$ NaDCC for the standard operating procedure on cut lily for export quarantine to control leaf blight.
