Effects of Growth Regulators on Adventitious Root Growth and Eleutherosides and Chlorogenic Acid Accumulation in Air Lift Bioreactor Cultures of Eleutherococcus koreanum (생물반응기 배양에서 생장조절제에 따른 섬오갈피 부정근 증식 및 Eleutheroside와 Chlorogenic Acid 생산)

  • The influence of different growth regulators on biomass of adventitious roots and secondary metabolites accumulation in bioreactor cultures of Eleutherococcus koreanum was studied. The maximum growth of adventitious roots was observed in the presence of $2.0\;\cal{mg/L}$ IBA (6.7 g DW/L). However the productivity of eleutheroside E was not significantly different among IBA levels. High level of thidiazuron (TDZ) efficiently increased both root growth and secondary metabolites production. Especially when $0.1\;\cal{mg/L}$ TDZ was combined with $3.0\;\cal{mg/L}$ IBA, the highest eleutheroside E accumulation ($3,327{\mu}g/g $DW) was observed. When the same TDZ level was treated in combination with $5.0\;\cal{mg/L}$ IBA, both chlorogrnic acid and eleutheroside B were accumulated to maximum level. In contrast, zeatin remarkably suppressed the accumulation of both eleutheroside B and chlorogenic acid.

In Vitro Propagation of Commonly Used Medicinal Trees in Korea

  • Forest medicinal resources, which constitute one of the non-timber forest products, have been regarded as healthy and highly valued products. To meet the increasing demand of the medicinal resources, it is necessary to improve the propagation methods of medicinal plants. In vitro propagation not only allows an opportunity for propagating plants in large numbers but also allows for enhancing the quality and quantity of the desired functional component of a plant by altering the growth factors, such as medium, carbon source, and plant growth regulators influence plant. There have been several studies of in vitro propagation methods, such as axillary bud culture, shooting, and embryogenesis, on Kalopanax septemlobus, Eleutherococcus sessiliflorus, Hovenia dulcis, and Schisandra chinensis in Korea between from 2000 through 2010. Furthermore, there have been attempts to proliferate callus and plantlets for producing useful natural compounds by using bioreactors. Here, we provide an account of the in vitro propagation methods of medicinal trees in Korea based on a review of several micropropagation studies.

Functional and Proteomic Analyses Reveal That ScpBXv Is Involved in Bacterial Growth, Virulence, and Biofilm Formation in Xanthomonas campestris pv. vesicatoria

  • Segregation and condensation protein B (ScpB) is essential for replication and segregation in living organisms. Here, we reported the functions of ScpBXv (ScpB-like protein in Xanthomonas campestris pv. vesicatoria) using phenotypic and proteomic analyses. Growth of $Xcv{\Delta}scpBXv$ (ScpBXv knockout mutant) was reduced under both slow and fast growth conditions in rich medium, but comparable to this of the wild-type in plant-mimic conditions. Interestingly, the mutant was significantly less virulent than the wild-type in tomato, indicating that ScpBXv is involved in virulence. To investigate ScpBXv-associated mechanisms, comparative proteomic analyses were carried out and the abundance of 187 proteins was altered. Among them, diverse transcriptional regulators involved in biofilm formation and virulence were abundant in the wild-type. We further showed that biofilm formation of $Xcv{\Delta}scpBXv$ was reduced. This study provides new insights into the functions of ScpBXv in bacterial replication and biofilm formation, which may contribute to the virulence of Xcv.

Effects of Different Natural Extracts and Plant Growth Regulators on Plant Regeneration and Callus Induction from Pseudobulbs Explants through in vitro Seed Germination of Endangered Orchid Bulbophyllum auricomum Lindl.   • Bulbophyllum auricomum Lindl. is a rare orchid and has flowers with an attractive fragrance. The present study investigated the tissue culture method for micropropagation. Capsules derived from artificial self-pollination were obtained for the best seed germination in MS basal medium. Plant growth regulators (1.0 mg·L-1 of BAP and 2.0 mg·L-1 of NAA) were affected by callus induction from subcultured pseudobulb explants. For the callus subculture, different natural plant extracts were tested in 11 treatment media. Among them, MS medium with 150 mL·L-1 of coconut water was generally effective in fresh weight (1.75 ± 0.08) and (3.01 ± 0.20) of callus proliferation and PLBs induction at 1 and 2 months, respectively, followed by an MS combination of 30 g·L-1 of banana and 20 g·L-1 of potato extract. The results of a comparative study of different MS mediums containing plant growth regulators with a natural extract combination and MS medium supplemented with natural extract only showed that MS medium supplemented with a combination of natural extracts (150 mL·L-1 of coconut water) and plant growth regulators (2.0 mg·L-1 of BAP and 1.0 mg·L-1 of NAA) obtained the highest shoot regeneration (3.37 ± 0.17) and (6.41 ± 0.68) after 1 month and 2 months of culturing, respectively.

Production of Giant Hyssop Oil by Plant Tissue Culture (식물조직배양에 의한 배초향유의 생산)

  • Callus was derived from the seedlings of Agastache rugosa(Labiatae). The growth rate of callus and the production of essential oil were studied with the variation of culturing conditions. 2, 4-D 2ppm in the medium was more effective for the production of essential oil than NAA 2ppm. The growth rate of callus and the production of essential oil were inhibited by the illumination of the light. The essential oils from Agastache rugosa and the callus cultivated on the medium containing 2, 4-D 2 ppm and kinetin 0.2 ppm were analysed by TLC, gas chromatography and mass spectrometry. These two oils showed different compositions. The main component of the plant oil, methyl chavicol was not contained in the callus oil.

Micropropagation from Corm Apical Meristems Culture of Freesia refrecta Hybrida (정단 분열 조직배양에 의한 후리지아의 미세번식)

  • Corm apical meristem cultures of thirteen glasshouse freesia cultivars were tested to investigate the possibility of micropropagation using MS basal medium supplemented with 2,4-D, NAA(0.1, 0.5, 1.0mg/L, respectively) and BA (0.5∼2.0mg/L). The majority of the tested cultivars could be induced callus and shoot buds in all culture condition. The combinations of NAA and BA appeared superior to that of 2,4-D and BA depending on cultivars for callus induction and shoot formation. Among the cultivars, 'Golden Yellow' showed the highest regeneration capacity on MS media with 0.5mg/L NAA and 1.0 mg/L BA. The highest percentage of regeneration and the greatest number of shoot from calli were obtained through successive subculture on MS medium supplimented with 0.5mg/L BA. In that condition, more than 60 % shoot regeneration and average of 25.1 shoots per explant was achieved. Transformed shoots on half-strength MS medium without plant growth regulators rooted easily.

In vitro seed germination and callus formation on flower bud of Korean mistletoe ( Viscum album L. var. cololatum [Kom.] Ohwi) (겨우살이 종자 발아 및 화아 배양에 의한 캘러스 형성)

  • Effects of growth regulators and culture conditions on seed germination, haustorium development, and callus formation of Korean mistletoe (Viscum album var. coloratum (Kom.) Ohwi) were described. Histological examination showed that seed of V. album contained one or two zygotic embryos with rod shape, and actively dividing cells were mainly distributed in radicle region rather than cotyledon of zygotic embryo. The most significant factor for seed germination and haustorium development of V. album was the requirement of the light. Various growth regulators examined in this study failed to substitute the effect of the light on seed germination. The frequency of callus formation was highest at 27.3% when flower buds were cultured onto B5 medium containing $0.1\;mgl^{-1}$ IAA. Explants from other organs were recalcitrant in forming calluses. Culture conditions described in this study could be applied for production of useful metabolites and multiplication of V. album in future.

Regeneration of Adventitious Shoots from Populus deltoides Bartr. (미류나무 엽육조직에서 식물체 재분화)

  • Adventitious organs (shoots and roots) from mature leaves of Populus deltoides (Eastern cottonwood) clones were induced on WPM (Woody Plant Medium) containing fructose as a carborn source and different combinations of either BA (Benzyl aminopurin) or TDZ (Thidiazuron) with NAA (Naphthaleneacetic acid). The morphologies of adventitious shoots were different depending on the kinds of growth regulators and the length of shoots induced on the medium containing BA were generally longer than that of TDZ. BA 0.5 mg/L and TDZ 0.01 mg/L combined with either 0.05 or 0.1 mg/L NAA showed the highest values in the frequency of regeneration (63% to 100%) the number of shoots per explant and in the organogenic sites per explant among tested three clones (A, B and C). Although there was a minor clonal difference in the development of adventitious shoots from leaf segments, their responses to growth regulators were very similar.

Effects of Plant Growth Regulators, Medium Salt Strength and Nitrogen Ratio on Cell Culture of Gymmema sylvestre (식물생장조절물질, 무기물 농도 및 질소원 비율이 Gymmma sylvestre 세포 배양에 미치는 영향)

  • This study was carried out to investigate the effects of plant growth regulators, medium salt strength and nitrogen ratio on cell culture of Gymnema sylvestre. Cell growth was inhibited by 2,4-D higher than 1.0 mg L$^{-1}$, but not by kinetin lower than 0.5 mg L$^{-1}$. Maximal cell growth was obtained at 1.0 mg L$^{-1}$ 2,4-D and 0.1 mg L$^{-1}$ kinetin. Cell growth was greatest at 1x MS medium but high strength of MS medium inhibited cell growth due to low water potential in the medium. In $NH_4^+:NO_3^-$ ratio of 0:60 (i.e. 0.0 mM $^NH_4^+$ and 60.0 mM $NO_3^-$), cells growth was highest but cells were smaller and whiter compared with those in other $NH_4^+:NO_3^-$ ratio. Reduced cell growth was observed with continuous culture. These results suggested that optimal cell culture of G. sylvestre could be achieved with 1x MS medium with 20:40 ratio of $NH_4^+:NO_3^-$ supplemented with 1.0 mg L$^{-1}$ 2,4-D and 0.1 mg L$^{-1}$ kinetin.

Effects of Growth Regulators on Shoot Regeneration and Polysaccharide Production of Orostachys japonicus Berger

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. O. japonicus was cultured in media containing various growth regulators and carbon sources. The highest regeneration rate was achieved in 1.0 and $3.0\;mg\;l\;^{-1}$ of 2,4-D concentration, while the lowest was obtained in $10.0\;mg\;l\;^{-1}$ 2,4-D concentration. When different carbone sources were added in the culture medium, plant growth was high in 3% sucrose treatment. The micropropagated shoots were successfully acclimatized in artificial soils and produced comparable amont of polysaccharide compred to parent cultivated plants.
