Diagnosis of Phythopthora sp. and Its Concentration by Potato Slices in Series Culture Soils.

  • Tomato soil pathogens(Phythopthora spp.) analyed high rates in series culture soil and existed in culture parts. To make a diagnosis of Phythopthora sp. and Its concentration, potato slices were manufactured to a round shape(2.5cm) or retangular form(1x4cm). and then, The potato slices dipped into diagnostic reagents with an antibiotic substance for 2∼4hours. Potato slices treated with a few reagents varied into 15cm depths in innoculated soils for 24hrs. Mycelium of the Phytophthora root rot fungus, Phythopthora capsici, were produced easily on potato slice. We collected many potato slice samples on diseased fields in various area. After storage of 24hrs in 20$^{\circ}C$ incubator, White mycelium of Phythopthora sp. formed on potato slice surface. Dilute concentrations of Phythopthora sp. was detected very low contents(1${\times}$10$^1$sporangia/g). But expressing Phythopthora root rots on potato slice did not developed larger lesions upon storage time in room temperature. These results suggest that the use of potato slice in a series of soil cultural system may still serve as efficient means of diagnosis of Phythopthora root rots in the absence of control measures.

Control of Phythophthora Blight of Pepper by grafting

  • Control effect of grafting with rootstocks on Phythophthora blight of pepper was evaluated. The 40∼45 day old seedlings of five pepper cultivars, Wangdaeback, Jinmi, Boosa, Samsungcho and Pochunngchun, as a scion were grafted with ten species of root stocks. There wasn't graft incompatibility in all scion-root stock combinations. After 21 days from inoculation with Phytopkhora capsici, incidence of Phythophthora blight of plant grafted with Kataguruma, R-Safe, R-16, YCM 334 and SCM 334 of rootstocks tested was decreased by 64.6∼100% compared to non-grafted plant whose disease severity was 3.7. However, plant grafted with Umsung native and Yeongdong native pepper was more sensitive to Phythophthora blight than non-grafed plant. Control effect by grafting was inversely proportional to virulence of inoculum and not significantly different among scion cultivars used. And resistant reaction of scion against Phytophthora blight was not affect that of scion-root stock plant in this study.

저온 생장성 Pseudomonas fluorescens M45와 MC07을 이용한 오이의 생육촉진과 모잘록병의 방제 (Enhancement of Plant Growth and Suppression of Damping-off of Cucumber by Low Temperature Growing Pseudomonas fluorescens Isolates)

  • Growth rates of the low temperature growing isolates, Pseudomonas fluorescens M45 and MC07, reached maximum stationary phase within 50 hrs at the low temperature, 4$^{\circ}C$. But an ordinary biocontrol agent P. putida Pf3 did not reach logarithmic growth phase until 80 hrs. The culture filtrates of M45 and MC07 significantly inhibited the mycelial growths of Pythium ultimum, Rhizoctonia solani and Phytophthora capsici. Detached cotyledons of cucumber grown on Murashige and Skoog agar medium were much enhanced in their growth, compared to those without the filtrates. Population densities of M45 and MC07 in the rhizosphere at 14$^{\circ}C$ were more stable than at 27$^{\circ}C$. When M45 and MC07 were treated into soil, the population density of MC07 continuously increased up to 9 days after treatment, and sustained the initial inoculum density up to 60 days. Cucumber damping-offs caused by P. ultimum and R. solani were significantly reduced by applying M45 as seed-inoculant and by soil treatment with MC07. The combined treatment of M45 and MC07 provided greater effect in reducing the disease incidence than that obtained by single treatments.

고추 재래종 선발계통의 주요 성분 함량 (Major Chemical Content in Selections from Korean Local Cultivars of Pepper)

  • 우수 재래 고추 품종인 풍각초에서 선발된 23계통과 칼미초에서 선발된 20계통 및 기타 대조품종들의 과실에 대하여 맛과 빛갈에 관여하는 주요 성분함량을 측정하였다. 칼미초에서 선발된 계통들은 풍각초에서 선말된 계통들 보다 전반적으로 신미성분(Capsaicin), 색소(Capsanthin) 및 당함량(sugsr content)이 더 높았다. 품질이 우수한 것으로 알려진 다복, 수비초 및 칠성초는 신미성분 함량은 중 정도이고 당함량은 높은 편이었다. 그러나 역병 저항성으로 도입된 PI201234는 신미성분 함량은 높으나 당함량이 낮았다.

Selection of KYC 3270, a Cellulolytic Myxobacteria of Sorangium cellulosum, against Several Phytopathogens and a Potential Biocontrol Agent against Gray Mold in Stored Fruit

  • During 2002-2008 in Korea, 455 extracts from myxobacteria consisting of 318 cellulolytic and 137 bacteriolytic myxobacteria were isolated, which were then screened for antifungal activity against the phytopathogens Botrytis cinerea, Colletotrichum acutatum, Penicillium sp., Pyricularia grisea, and Phytophthora capsici. 204 isolates had antifungal activity, causing both a clear zone due to blocked spore germination and inhibition of mycelial growth; most (199) were from cellulolytic (Sorangium cellulosum) and only five were from bacteriolytic myxobacteria. B. cinerea, the best controlled among the five tested pathogens, had a unique group of antifungal isolates of myxobacterial extracts compared to the other pathogens' groups. Among seventy-nine bioactive myxobacteria, four isolates, KYC 3130, KYC 3247, KYC 3248 and KYC 3270, were selected and all were cellulolytic. Liquid culture filtrates of these four myxobacteria were applied to tomato, cherry tomato, strawberry, and kiwi fruits 5 h before inoculation with gray mold conidia; then the treated fruits were placed in an airtight container and the experiment was repeated six to eight times. Incidence (%) of gray mold on fruit of the infected control treatment was 84-98%, whereas it was only 5-21% after the KYC 3270 treatment. After KYC 3270 treatment of the four fruits, mold control was 79-95%, which was highest among the filtrates and statistically the same as treatment with fludioxonil, a registered chemical against gray mold of stored fruits.

Potential of the Volatile-Producing Fungus Nodulisporium sp. CF016 for the Control of Postharvest Diseases of Apple

  • In vitro and in vivo mycofumigation effects of the volatileproducing fungus Nodulisporium sp. CF016 isolated from stem of Cinnamomum loureirii and the role of its volatile compounds were investigated against phytopathogenic fungi. The volatile compounds produced by Nodulisporium sp. CF016 inhibited and killed a wide range of plant and storage pathogens including to Pythium ultimum, Rhizoctonia solani, Fusarium oxysporum, Phytophthora capsici, Sclerotinia sclerotiorum, Colletotrichum coccodes, Magnaporthe oryzae, Alternaria panax, Botrytis cinerea and Penicillium expansum. Mycofumigation with wheat bran-rice hull cultures of Nodulisporium sp. CF016 showed in vivo antifungal activity against gray mold caused by B. cinerea and blue mold caused by P. expansum of apple. The most abundant volatile compound produced by Nodulisporium sp. CF016 was $\beta$-elemene followed by 1-methyl-1,4-cyclohexadiene, $\beta$-selinene and $\alpha$-selinene. Nodulisporium sp. CF016 could be an attractive mycofumigant in controlling postharvest diseases of various fruits including apple.

Functional Characterization of PR-1 Protein, β-1,3-Glucanase and Chitinase Genes During Defense Response to Biotic and Abiotic Stresses in Capsicum annuum

  • Spatial and temporal expression of pathogenesis-related (PR) gene and proteins has been recognized as inducible defense response in pepper plants. Gene expression and/or protein accumulation of PR-1, $\beta-1,3-glucanase$ and chitinase was predominantly found in pepper plants during the inoculations by Xanthomonas campestris pv. vesicatoria, Phytophthora capsici and Colletotrichum coccodes. PR-1 and chitinase genes were also induced in pepper plants in response to environmental stresses, such as high salinity and drought. PR-1 and chitinase gene expressions by biotic and abiotic stresses were regulated by their own promoter regions containing several stress-related cis-acting elements. Overexpression of pepper PR-1 or chitinase genes in heterogeneous transgenic plants showed enhanced disease resistance as well as environmental stress tolerances. In this review, we focused on the putative function of pepper PR-1, $\beta-1,3-glucanase$ and chitinase proteins and/or genes at the biochemical, molecular and cytological aspects.

Antifungal Activity of Paenibacillus kribbensis Strain T-9 Isolated from Soils against Several Plant Pathogenic Fungi

  • The bacterial strain T-9, which shows strong antifungal activity, is isolated from the soils of Samcheok, Gangwondo and identified as Paenibacillus kribbensis according to morphological and taxonomic characteristics and 16S rRNA gene sequence analysis. The P. kribbensis strain T-9 strongly inhibits the growth of various phytopathogenic fungi including Botrytis cinerea, Colletotricum acutatum, Fusarium oxysporum f. sp. radicis-lycopersici, Magnaporthe oryzae, Phytophthora capsici, Rhizoctonia solani, and Sclerotium cepivorum in vitro. Also, the P. kribbensis strain T-9 exhibited similar or better control effects to plant diseases than in fungicide treatment through in vivo assays. In the 2-year greenhouse experiments, P. kribbensis strain T-9 was highly effective against clubroot. In the 2-year field trials, the P. kribbensis strain T-9 was less effective than the fungicide, but reduced clubroot on Chinese cabbage when compared to the control. The above-described results indicate that the strain T-9 may have the potential as an antagonist to control various phytopathogenic fungi.

Distribution and Antifungal Activity of Endophytic Fungi in Different Growth Stages of Chili Pepper (Capsicum annuum L.) in Korea

  • This study investigated the distribution of endophytic fungi obtained from the surface sterilized tissues of leaves, stems and roots of chili pepper ($Capsicum$ $annuum$ L.) plants in seedling, flowering and fruiting stages in Korea and their antifungal activity. A total of 481 isolates were recovered and were identified using molecular techniques. Based on rDNA ITS gene sequence and phylogenetic analysis, 21 fungal genera were characterized, belonging to 16 Ascomycota and 5 Basidiomycota. $Penicillium$ in seedling stage, $Fusarium$ in flowering stage, $Colletotrichum$ followed by $Fusarium$, $Alternaria$ and $Xylaria$ in fruiting stage was predominant and $Alternaria$, $Cladosporium$ and $Fusarium$ were common in all growth stages. Among 481 endophytes 90 phenotypes were evaluated for the antimicrobial activity against three major pathogens ($Phytophthora$ $capsici$, $Colletotrichum$ $acutatum$ and $Fusarium$ $oxysporum$) of chili pepper. Among them 16 isolates inhibited the growth of at least one test microorganisms. Three strains showed a broad spectrum antifungal activity and displayed strong inhibition against chili pepper pathogenic fungi.

Isolation and Antifungal and Antioomycete Activity of Streptomyces scabiei Strain PK-A41, the Causal Agent of Common Scab Disease

  • The actinomycete strain PK-A41 was isolated from a soil sample from pepper fields in Ko-yang, Korea. The strain PK-A41 inhibited the mycelial growth of some plant pathogenic fungi and oomycete, Alternaria mali, Colletotrichum orbiculare, Fusarium oxysporum f.sp. lycopersici, Magnaporthe grisea, Rhizoctonia solani, and Phytophthora capsici. The presence of LL-diaminopi-melic acid in the cell wall extract and the nucleotide sequence of the 16S rDNA region of the strain PK-A41 was assigned to Streptomyces scabiei. Further morpho-logical, biochemical, and pathological analyses also confirmed the strain PK-A41 to be S. scabiei, which is pathogenic to potato tubers. The maximum antibiotic production of the strain PK-A41 was achieved when grown on the glycerol peptone broth (GPB) medium for 9 days.
