The Protective Effects of Phenolic Constituents from Gastrodia elata on the Cytotoxicity Induced by KCI and Glutamate

  • Seven phenolic compounds (1-7) were isolated from the tubers of Gastrodia elata. Their structures were elucidated on the basis of MS and NMR spectral data. p-Ethoxymethyl phenyl-O-${\beta}$-D-glucoside (1) was proved to be a new compound, with N-(p-hydroxybenzyl)-adenosine (7) isolated from this plant for the first time. In this study, the protective effects of the six constituents (1-6) on PC12 cells against the cytotoxicity induced by KCI and glutamate were also investigated. The viability of the PC12 cells was significantly enhanced by pretreatment with the six phenolic constituents.

  • Six phenolic constituents, 2-methoxybenzyl benzoate (1), negletein (2), 2',3'-dihydroxy-4',6'­dimethoxydihydrochalcone (3), 5,6-dihydroxy-7-methoxy-dihydroflavone (4), astilbin (5), and quercitrin (6) were isolated from the methanol extract of the dried leaves of Desmos chinensis. Their structures were elucidated from spectral and chemical data. Of these constituents, compounds 2 ($IC_{50}$: 3.89 $\pm$ 0.39 $\mu$M) and 3 ($IC_{50}$: 9.77 $\pm$ 0.26 $\mu$M) exhibited potent inhibitory activity against nuclear factor of activated T cells (NFAT) transcription factor, and compound 1 ($IC_{50}$: 28.4 $\pm$ 2.62 $\mu$M) exhibited moderate inhibitory activity.

  • A chemosystematic study on euphorbiaceous plants in Korea has been performed by using phenolic constituents. The phenolic characteristics of subfamilies, genera and species were well distinguished from one another. Hydrolyzable tannins as constituents were considered to be a valuable taxonomic character in elucidating systematic relationships among the related taxa whereas flavonoids could be used in the classification of infraspecific taxa in this family. The phenolic fingerprints of each of the plants would be considered as a good tool to identify the species. In comparison with the morphological classification system, the chemical relationship supported the subfamilial system of Webster (1975) and the further division of Euphorbia sensu lato by Hurusawa (1954).

  • Hamamelis japonica (Hamamelidaceae), widely known as Japanese witch hazel, is a deciduous flowering shrub that produces compact clumps of yellow or orange-red flowers with long and thin petals. As a part of our ongoing search for phenolic constituents from this plant, eleven phenolic constituents including six flavonol glycosides, a chalcone glycoside, two coumaroyl flavonol glycosides and two galloylated compounds were isolated from the flowers. Their structures were elucidated as methyl gallate (1), myricitrin (2), hyperoside (3), isoquercitrin (4), quercitrin (5), spiraeoside (6), kaempferol 4'-O-β-glucopyranoside (7), chalcononaringenin 2'-O-β-glucopyranoside (8), trans-tiliroside (9), cis-tiliroside (10), and pentagalloyl-O-β-D-glucose (11), respectively. These structures of the compounds were identified on the basis of spectroscopic studies including the on-line LCNMR-MS and conventional NMR techniques. Particularly, directly coupled LC-NMR-MS afforded sufficient structural information rapidly to identify three flavonol glycosides (2 - 4) with the same molecular weight in an extract of Hamamelis japonica flowers without laborious fractionation and purification step. Cytotoxic effects of all the isolated phenolic compounds were evaluated on HCT116 human colon cancer cells, and pentagalloyl-O-β-D-glucose (11) was found to be significantly potent in inhibiting cancer cell growth.

Analysis of the essential oil composition of fresh Panax ginseng root and identification of novel phenylalkenal compounds

  • This study investigated the essential oil composition of fresh Panax ginseng root and identified novel compounds from ginseng oil. The oil was divided into five fractions (neutral, basic, phenolic, acidic, and aldehydic). In total, 149 constituents, including 29, 19, and 38 compounds in the basic, phenolic, and aldehydic fractions, respectively, were identified by gas chromatography (GC) and GC-mass spectrometry (MS). The primary constituents of the total ginseng volatile oil were α-humulene (13.91% as a peak area), bicyclogermacrene (13.59%), β-caryophyllene (8.24%), α-neoclovene (7.78%), and α- and β-panasinsenes (5.14% and 7.53%). The primary constituents of the basic fraction were 2-isopropyl-3-methoxypyrazine (35.51%), 3-sec-butyl-2-methoxy-5-methylpyrazine (31.54%), 2-isobutyl-3-methoxypyrazine (8.64%), and 2-methoxy-3-methylpyrazine (8.40%), whereas in the phenolic fraction, these were benzoic (25.40%), octanoic (11.57%), nonanoic (9.16%), propionic (6.35%), and decanoic acids (6.16%). The primary constituents of the aldehydic fraction were 4-(2-furyl)-3-buten-2-one (23.41%), benzaldehyde (10.18%), cis-2-heptanal (9.42%), 3-(α-furyl)-propenal (8.51%), and 2-phenyl-2-butenal (7.28%). Among these, the phenylalkenal compounds, including 2-phenyl-2-butenal, 2-methyl-3-phenyl-2-propenal, 5-methyl-2-phenyl-2-pentenal, 5-methyl-2-phenyl-2-hexenals, 2-phenyl-2-octenal, and 2-phenyl-2-nonenal, were newly identified in this study as ginseng volatile constituents. Furthermore, 2-phenyl-2-nonenal was identified as a plant-based volatile constituent for the first time in this study.

A Study on The Phenolic Content of Potatoes (감자에서의 페놀화합물에 대한 연구)

  • Two varieties, Lasoda and Sebago potatoes, were studied. Different cooking methods, conventional oven baking at $218^{\circ}C$ and microwave oven baking, have been used to compare the retention of the phenolic compounds. Peeled cortex samples of fresh and cooked potatoes were analyzed for total phenols, phenolic constituents, and moisture loss. Phenolic content was higher in fresh potatoes than in cooked potatoes. Laasoda had higher phenolic content (3. 63mg) than Sebago (1. 71mg). Potatoes with higher phenols (Lasoda) also had larger quantities of chlorogenic acid. There was a greater moisture loss in conventional oven baking potatoes than in microware oven.

  • Chemical investigation on a sample of the needles of Himalayan Taxus baccata has resulted in the isolation of several taxoids including taxol (1) 10-deacetyl-baccatin III (2) and 2-deacetoxytaxinine J (3) along with different lignans (6 and 7) and simple phenolics (8, 9, 10, 11 and 12). The occurrence of 4-(4'-hydroxyphenyl)-butane-2-one and 4-(4'-hydroxyphenyl)-trans-but 3-ene-2-one (8) in Taxus species is reported for the first time. The $^{13}C-NMR$ spectral data of two rearranged taxiod constituents, brevifoliol (4) and 13-decinnamoyltaxchinin B (5) are presented. The acid-catalyzed decomposition of taxol has been discussed. The synthesis of other two constituents, rhododendrol (10) and hibalactone (7) has been described.

  • For the investigation of medicinal resource in Phyllanthus ussuriensis, the studies were conducted to evaluate the pharmaco-constituents in Phyllanthus ussuriensis, which is used as folk medicine in China. From the hot water extract of leaves, three phenolic compounds were isolated and identified as phloroglucinol, gallic acid and rutin by physico-chemical properties and spectroscopic evidences$(IR,{\;}^1H-NMR,{\;}^{13}C-NMR{\;}and{\;}Mass,{\;}etc.)$.

Anti-platelet Effect of the Phenolic Constituents Isolated from the Leaves of Magnolia obovata

  • In the course of our work on anti-platelet constituents from plants, eight phenolic compounds, $({\pm})-syringaresinol$ (1), 4-hydroxybenzaldehyde (2), 4-hydroxybenzoic acid (3), vanillic acid (4), 4-hydroxy cinnamic acid (5), quercetin 3-O-rhamnoside (6), rutin (7), and quercetin $3-(2^G-rhamnosylrutinoside)$ (8) were isolated from the methanol extract of the leaves of Magnolia obovata. The compounds were identified based on the spectroscopic data. Compound 2, 3, 5, 6, 7 and 8 were isolated for the first time from genus Magnolia. 1 and 6 showed same order of inhibitory potencies as acetylsalicylic acid (ASA) to rat platelet aggregation induced by all the stimulators tested. The remaining six compounds showed only mild effects.

Phenolic Constituents of Boehmeria pannosa and α-Glucosidase Inhibitory Activity (왕모시풀의 페놀성 성분 및 항당뇨 활성)

  • Boehmeria pannosa (Urticaceae) is a perennial herb widely distributed in Korea. In this study, investigation on the constituents of B. pannosa was conducted by chromatographic techniques and spectroscopic analysis. As a result, nine compounds including eight phenolic compounds, 3-hydroxy-1-(4-hydroxyphenyl)propan-1-one (1), β-hydroxypropiovanillone (2), C-veratroylglycol (3), 3-hydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl)-propan-1-one (4) 2,3-dihydroxy-1-(4-hydroxy-3,5-dimethoxyphenyl) -1-propanone (5), p-coumaric acid (6), 4-hydroxybenzoic acid (7), vanillic acid (8) and one lignan, (-)-(7R,8S)-dihydrodehydrodiconiferyl alcohol (9) were isolated from the EtOAc-soluble fraction of B. pannosa. Among them, compounds 5, 6 and 9 inhibited α-glucosidase inhibitory activity more than 50% at the concentration of 100 μM.
