Phellinus linteus KACC 93057P, '한경상황버섯'인공재배 자실체 페놀추출물의 항산화 활성 및 화학적 동정 (Chemical identification and antioxidant activity of phenolic compounds extracted from the fruiting body of 'Hankyong Sanghwang', Phellinus linteus KACC 93057P)

  • 참나무 단목원목으로 재배한 P. linteus KACC93057P(PLHS)2년산 자실체 60%에탄올 추출물의 총 페놀함량은 $19.05{\pm}0.32mg\;GAE/g$로 다른 비교구 버섯 종에 비하여 4배에서 10배이상의 높게 나타났다. $125{\mu}g/mL$농도에서 DPPH와 ABTS 라디칼 소거능은 62%와 100%를 나타냈으며 FRAP 환원력은 $0.7{\mu}MFeSO_4/g$으로 비교 구 버섯종 보다 2에서 10배이상의 유효활성이 검출 되었다. PLHS 자실체 ethyl acetate분배 추출물의 HPLC분석에서 styrylpyrone 계열의 화합물 davallialactone, hispidin, hypholomine B와 phenylpropanoid 계열의 화합물인 caffeic acid가 동정 되었다.

  • 본 연구에서는 버섯으로부터 새로운 항비만 치료(예방) 효과를 갖는 물질을 추출, 정제하여 그 특성을 규명하고, 이를 기능성식품 개발의 소재로 용용하고자 lipase 저해 활성이 높은 상황버섯(Phellinus linteus)의 lipase 저해 물질을 각종 chromatography로 정제 하였다. 먼저 상황버섯(Phellinus linteus) 자실체 추출물에 대한 용매 계통분획을 실시하여 저해 활성이 가장 높은 에틸아세테이트 추출물을 얻었고 이를 3차례의 sillicas gel column chromatography와 분취용 HPLC를 실시하여 $IC_{50}$값이 175 ng의 lipase 저해활성을 가진 연한 연두색 분말의 정제된 lipase 저해 물질을 얻었다. 정제한 lipase 저해물질을 일련의 NMR spectrometry를 실시한 결과 분자량이 523.06 Da으로 추정되었고 225.1 nm에서 최대 흡수 파장을 가진 물질이었다.

  • We have examined the effect of water extract of Phellinus linteus, a raw material of Korean traditional herbal medicine, on the induction of HL-60 cell differentiation. The proliferation of HL-60 cell was inhibited dose-dependently by treatment with various doses of P. linteus extract. It also caused a significant change in NBT reduction (7.5 times). The expression of CD11b and CD14 was increased in the cells treated with the extract, especially in those arrested at G0/G1 stage, which suggested that some components in P. Linteus extract induced HL-60 cell differentiation to granulocytic and monocyte lineages. Moreover, the expression levels of $p21^{WAF1/CIP}$ and $p27^{KIP}$ were up-regulated during HL-60 cell differentiation induced by P. Linteus extract. These results together suggest that P. Linteus extract contains potential HL-60 cell differentiation agents.

Development of Species-specific Primers for Rapid Detection of Phellinus linteus and P. baumii

  • Genus Phellinus taxonomically belongs to Aphyllophorales and some species of this genus have been used as a medicinal ingredients and Indian folk medicines. Especially, P. linteus and morphological-related species are well-known medicinal fungi that have various biological activities such as humoral and cell-mediated, anti-mutagenic, and anti-cancer activities. However, little is known about the rapid detection for complex Phellinus species. Therefore, this study was carried out to develop specific primers for the rapid detection of P. linteus and other related species. Designing the species-specific primers was done based on internal transcribed spacer sequence data. Each primer set detected specifically P. linteus (PL2/PL5R) and P. baumii (PB1/PB4R). These primer sets could be useful for the rapid detection of specific-species among unidentified Phellinus species. Moreover, restriction fragment length polymorphism analysis of the ITS region with HaeIII was also useful for clarifying the relationship between each 5 Phellinus species.

Immunostimulating Activity of Polysaccharides from Mycelia of Phellinus linteus Grown under Different Culture Conditions

  • Polysaccharides were extracted from mycelia of Phellinus linteus grown under different culture conditions. The in vitro immunostimulating activity was measured by plaque-forming cell (PFC) assay. The activity of the polysaccharides was different from that of mycelia from which was extracted. The number of PFC's ranged from 40 to 600 depending on the media. When P. linteus was cultured on a medium with mannose or starch as a sole carbon source, the fungus produced polysaccharide with the highest activity of 960 PFC. Activity was therefore increased by $50%$ compared with polysaccharide which was extracted from mycelia grown on medium with glucose. pH had little effect on the change in activity. All polysaccharides on media with different pH stimulated about 600 PFC. These results suggest that activity could be increased by polysaccharide modification through changes in physiological conditions.

Antioxidant Effects of Cheonggukjang Containing Phellinus linteus Extract

  • This study was carried out to examine the antioxidant effects of cheonggukjang combined with Phellinus linteus extract. The electron-donating activity (EDA) of cheonggukjang containing 0.3% P. linteus extract (0.3% CPLE) was higher than that of cheonggukjang only. EDA of the ethanol extract from cheonggukjang was higher than that of the water extract. The water and the ethanol extracts showed strong antioxidant activity with regard to peroxide value. However, the ethanol extract showed a higher peroxide value than the water extract. The nitrite scavenging activity of the ethanol extract was greater than that of the water extract, crresponding to the EDA and peroxide values for each extract. Therefore, the antioxidant effects were enhanced by adding 0.3% of extract from P. linteus in manufacturing cheonggukjang. It is suggested that P. linteus extract could be put into practice as an effective antioxidant agent.

  • Phellinus linteus is known as a medicinal mushroom, which has the pharmaceutical activity on tumors and inflammatory diseases in traditional Oriental medicine. However, despite extensive pharmacological studies on P. linteus, the liquor of fermentation using mycelium of P. linteus(LFMP) has not been investigated. In the present study, it was examined the effect of the evaporating extract from LFMP(E-LFMP) on the expression of inflammatory proteins and the generation of reactive oxygen species in human hepatocarcinoma HepG2 cells. E-LFMP inhibited acetaldehyde-induced morphological change in HepG2 cells. Also, E-LFMP inhibits expression of inflammatory proteins including cyclooxygenase(COX)-1 and COX-2 through suppressing nuclear translocation of nuclear factor κB(NF-κB) and degradation of inhibitory κBα(IκBα). In addition, E-LFMP inhibits generation of reactive oxygen species(ROS) by hydrogen peroxide(H₂O₂) in HepG2 cells. These results suggest that LFMP has the pharmaceutical, especially anti-inflammatory, activity similar to P. linteus mushroom.

상황버섯의 기술개발 동향과 시장분석 (Phellinus linteus; Market and Technology Trends Analysis)

  • 담자균류의 버섯은 대부분 영양생장단계의 균사체와 포자를 형성하는 생식생장단계인 자실체로 나누어진다. 그러나 자연산 상황버섯은 자연계에서의 분포수가 극히 적어 그 자실체를 얻기 어려우며, 이러한 희귀성으로 인해 매우 고가로 거래되고 있다. 1990년대 중반 상황버섯은 균사체 배양기술과 인공재배법이 확립된 후 농가에서 대량생산되기 시작하였으며, 이에 따라 상황버섯의 유효성분을 이용한 기능성 식품 및 의약품의 개발이 활발하였다. 그러나 인공재배법에 의한 상황버섯 추출물은 자연산 추출물의 항암 및 효능면에서 차이를 나타내고 있어, 자연산 자실체와 같은 수준에서의 약효를 나타내기 위한 인공배양기술과 고농도 유효성분을 획득하기 위한 추출법이 핵심기술로 대두되었고, 인공배양된 상황버섯과 자연산의 다각도적인 약효 비교에 관한 연구가 아직까지 진행되고 있다. 따라서 상황버섯의 유효성분이 높게 함유된 저렴한 추출물이 생산될 수 있는 기반 기술들이 기능성 음료용수, 식품 및 의약품으로 개발에 중요한 핵심기술로 떠오르고 있다. 국내 상황버섯 기능성 식품의 시장은 상황버섯의 효능에 대한 인지 증대, 웰빙열풍을 의한 건강 음료에 대한 선호도 증대 등으로 인해 향후에도 10% 이상의 높은 성장률을 보일 것으로 예상된다. 현재는 중소기업들이 주로 활동하고 있어 마케팅 및 유통의 한계로 인해 시장이 크게 확대되고 있지 않으나, 사업매력도가 긍정적으로 평가되고 있어 대기업에서도 상황버섯을 이용한 기능성 식품 시장에의 진출을 모색하고 있다.

Antimutagenicity of Phellinus linteus in Salmonella typhimurium

  • The mutagenicities and antimutagenicities of butanol (PL I) and water (PL II) extracts from the filtrate of the cultured broth of Phellinus linteus were examined using the Ames/Salmonella test. No mutagenic activity of PL I and PL II was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PL I and PL II showed inhibitory effects on the mutagenic activities induced by the directly-acting mutagens, 4-nitro-o-phenylenediamine (NPD) using the tester strain TA98 and sodium azide (NaN3) using the tester strain TA 100 in the absence of S9 mix. PLI and PL II also showed inhibitory effects on the mutagenicities of the indirectly-acting mutagens, 2-aminofluorene (2-AF) using the tester strain TA98 and benzo[a]pyrene (B[a]P) using the tester strain TA 100 in the presence of S9. These results suggest that P. linteus has an antimutagenic activity and may play a role in the prevention of cancer.

An Antioxidant Hispidin from the Mycelial Cultures of Phellinus linteus

  • In the course of screening for reactive oxygen species scavengers from natural products, an antioxidant was isolated from the mycelial culture broth of Phellinus linteus and identified as hispidin. The hispidin content was reached its maximum level at 12 days after onset of inoculation. About 2.5 mg/mL of hispidin was produced by P linteus in a yeast-malt medium (pH 5.8, $25^{\circ}C$). Hispidin inhibited 22.6 and 56.8% of the super oxide anion radical, 79.4 and 95.3% of the hydroxyl radical, and 28.1 and 85.5% of the DPPH radical at 0.1 and 1.0 mM, respectively. The positive control ${\alpha}-tocopherol$ scavenged 25.6 and 60.3%, 74.6 and 96.3%, and 32.7 and 77.5% of each radical, respectively, at the same concentrations. However, hispidin showed no significant activity on the hydrogen peroxide radical.
