Explosion Hazard Assessment of Pharmaceutical Raw Materials Powders (원료의약품 분진의 폭발 위험성 평가)

  • Hazard risk of explosion on pharmaceutical raw materials dust in pharmaceutical industry often exists when it is handled or processed in the industrial sites, and explosion accident is caused by this. In this study, the dust explosion characteristics of the three pharmaceutical raw materials samples were measured. The main explosion characteristics are as follows: $P_{max}$, MIE and MIT of loxoprofen acid having $5.31^{\circ}C$ of median diameter are obtained 8.4 bar, 1 mJ < MIE < 3 mJ and $550^{\circ}C$. $P_{max}$, MIE and MIT of camphorsulfonate having $95.63^{\circ}C$ of median diameter are obtained 7.9 bar, 30 mJ < MIE < 100 mJ and $510^{\circ}C$. $P_{max}$, MIE and MIT of rifampicine having $26.48^{\circ}C$ of median diameter are obtained 7.9 bar and 1 mJ < MIE < 3 mJ and $470^{\circ}C$. The deflagration index ($K_{st}$) and the explosion index (EI) were obtained by using these data. The explosion hazard assessment of pharmaceutical raw materials dust was compared and examined. As a result, the explosion hazard assessment according to deflagration index and explosion index were the explosion class with St 2 and the explosion hazard rating of severe for loxoprofen acid & rifampicine and St 1 and strong for clopidogrel camphorsulfonate, respectively.

  • The identification step of raw drug materials is an indispensible procedure in the GMP manufacturing process within the pharmaceutical industry. However, wet chemistry methods for identification of drug materials, used by the various Pharmacopeia are time-consuming and expensive steps. In this paper, near-infrared spectroscopy (NIRS) has been developed for identifying eleven drug substances including calcium pantothenate, cefaclor, cefoperazone, cephradine, dextromethorphan, ehtambutol, nicotinamide, pyrozinamide, tramadol, vitamin C, and vitamin E. Also the aim of ths work is to consturct a new algorithm for calibration model using soft independence modeling of class analogy (SIMCA) with Malinowskis Indicator Function (IND), which is used for finding the number of principal components of each class of the SIMACA model. The use of NIR technique with pattern recognition to qualify raw materials can make it possible to monitor process in real time as well as to control all procedures in the pharmaceutical industry. As the result, the samples identified of 183 different batches from 11 different compounds were separated clearly by SIMCA with 2nd derivative spectra in the NIR region of 1100∼2400 nm.

Anti-inflammatory effect of Crypsinus hastatus biorenovation extract (고란초 생물 전환 추출물의 항염증 효과)

  • Biorenovation is a method for converting materials using the enzyme properties of microorganisms. Natural products converted by that method increase physiological activity or reduce cytotoxicity. In this study, we investigated the anti-inflammatory activity of crypsinus hastatus prothallium (CH) and biorenovated CH prothallium (CHB) using RAW 264.7 cells stimulated with lipopolysaccharide (LPS). CHB inhibited the production of nitric oxide, prostaglandin E2 and cytokines (interleukin-6, interleukin-1β, tumor necrosis factor-α) compared to CH at a concentration of 50-200 ㎍/mL. In addition, CHB concentration of 200 ㎍/mL inhibited the expression of inducible nitric oxide synthase and cyclooxygenase-2 protein by LPS stimulation to the level of the untreated control group. These results indicate that CHB could be a novel anti-inflammatory agent for cosmetic and pharmaceutical ingredients.It also suggests that the application of biorenovation has potential usefulness in developing anti-inflammatory materials.   • Atenolol and related compounds found in raw materials and commercial products were analyzed by reversed-phase high-performance liquid chromatography. A mixed solution of phosphate buffer (3.4 g/l, pH 3.0), tetrahydrofurane and methanol (800:20:180, v/v/v) including sodium octanesulfonate (1 g/l) and tetrabutylammonium-hydrogensulfate (0.4 g/l) was used as mobile phase at the flow rate of 0.25 ml/min. Detection was carried out at UV 226 nm. Atenolol related compounds, such as bis ether, tertiary amine and blocker acid were identified by comparing the retention time of the standard. The within-day and between-day precisions of the separated compounds were less than 1.2% and 3.4%, respectively. The contents of related compounds of the tested samples were under the limit prescribed in the European Pharmacopoeia. The pattern of the related compounds showed that atenolol raw materials and products could be classified in three different groups, indicating that the materials originated from different source or treated in different way.

  • Pinus densiflora, the Korean Red Pine, is the predominant tree species of the cool, temperate forests of northeast Asia, occurring in pure stands across Korea, Japan, and parts of northern China and Russia. Pinus densiflora leaves, pollen, and bark have been widely used for traditional medicine, or edible purposes. However, pine cones contain many bioactive phytochemicals, but they are rarely used as natural raw materials. This study was conducted to evaluated the anti-inflammatory effect of pine cone extracts and its possbility of natural sources were evaluated. Pine cones were extracted with 80% methanol, concentrated and then partitioned with ethyl acetate, and the organic layer was used as a sample. The Pine cone Ethyl acetate Fraction (PEF) showed no toxicity to RAW 264.7 cells at a concentration of less than $50{\mu}g/ml$. PEF inhibited the production of nitric oxide (NO) in RAW 264.7 cells treated with lipopolysaccharide (LPS). Also, It suppressed the expression of cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS) and transcription of nuclear factor-kappa B (NF-${\kappa}B$). These results suggest that pine cones can be used as an effective natural material for anti-inflammatory agent.

  • Houttuynia cordata Thunb. grown in wet earth in China, Japan and Korea, has often been cited in medical literature for its medicinal effects. In this paper, the pharmaceutical effects of Houttuynia cordata Thunb., featured in the East Asian literature were studied. It was revealed that it has a wide variety of uses, including remedial treat-ments for pneumonia, antidote, inflammation, syphilis, abscess, paralysis and gynecological diseases. Its application methods also are numerous: drinking as tea or squeezed juice, application to wounds, wet dressing, chewing raw roots and mixing with other plants, liquor and food materials. The major pharmaceutical effects are as follows: antibiosis, immunity enhancement, urination, pain relieving, vasodilator and cough lozenge.

The Application of Ion Chromatographic Method for Bioavailability and Stability Test of Iron Preparations

  • Postabsorptive serum iron level was determined after oral administration of the compounds to human. In serum and whole blood, $Fe^{3+}$ was measured by ion chromatography (IC) using a pyridine-2,6-dicarboxylic acid (PDCA) as an eluent. The serum sample solutions were pretreated with I N HCI and 50% TCA. The whole blood sample solutions were treated with 3 N HCI for 30 min at $125^{\circ}C$. The limit of detection (LOD) of the IC technique is $0.2 {\mu}M$ for$Fe^{2+}$and 0.1 $\mu$M for $Fe^{3+}$. The area under concentration (AUC) can be obtained by the above analytical condition. In addition, to compare the stability of $Fe^{2+}$ to that of $Fe^{3+}$ in pharamaceutical preparations, accelerated stability test was carried out. After storing the samples under $40^{\circ}C$, 75%RH in light-resistant container for various time intervals, the contents of iron of different valencies were determined separately by the IC technique and the change and/or the interchange of among those iron species in preparations was investigated. Iron raw materials are stable, but $Fe^{2+}$ in$Fe^{3+}$ source materials was slightly converted to $Fe^{3+}$ by oxidation. $Fe^{2+}$ in$Fe^{3+}$ source raw materials and $Fe^{3+}$ in $Fe^{2+}$ raw materials are determined as impurities. Therefore, IC technique is found to be an appropriate method for comparative evaluation of dissimilar bioavailability of $Fe^{2+}$ and $Fe^{3+}$, stability of $Fe^{2+}$ and $Fe^{3+}$ raw materials and preparations.

  • Thalictrum rochebrunianum var. grandisepalum (TRG) is a Korean endemic plant, and it is widely used for edible, medicinal, landscape materials. In this study, we examined the protein and mRNA expression levels of MITF, tyrosinase, TRP-1 and TRP-2 by TRG extract (TRGE) in IBMX-treated melanocytes to evaluate the possibility of using TRG as a whitening material. IBMX were reported as melanin synthesis enhancers. It could increase intracellular melanin synthesis by activation of the microphthalmia-associated transcription factor (MITF) signaling pathway. TRGE did not show cytotoxicity at concentrations below $100{\mu}g/ml$ in B16F10 cells. TREG dose-dependently inhibited protein and mRNA levels of MITF, tyrosinase, TRP-1 and TRP-2. Therefore, we suggest that TRGE is an important natural resource for cosmetic raw materials for whitening function.

Enhancement of the Anti-inflammatory Activities of Aralia continentalis Kitagawa Extracts Fermented by Lactobacillus plantarum (추출용액에 따른 유산균 발효 땅두릅의 항염증 효과)

  • We investigated the anti-inflammatory activities of various organic solvent extracts with and without Lactobacillus plantarum fermentation of Aralia continentalis Kitagawa which has hypotensive effects in addition to excitatory effects on the central nervous system. It has been used to treat arthritis, colds, neuralgia, rheumatism, and itchy skin. Our extracts were tested for their anti-inflammatory potential on NO production and the expression of inflammatory factors in lipopolysaccharide-stimulated RAW264.7 macrophages. Extracts with and without L. plantarum fermentation were prepared using water, ethanol, hexane, ethyl acetate, and butanol. The RAW264.7 cells were tested for toxicity and the anti-inflammatory activity of each extract was determined at a concentration with no toxicity to the cells. The extracts used in this study significantly inhibited both the production of NO and the mRNA expression of COX-2 and iNOS, the major inflammatory factors. The production of inflammation-related cytokines $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ was also significantly reduced. These results suggest that the extracts involving fermentation by L. plantarum can inhibit cytokines by controlling the expression of inflammatory cytokine genes. It is considered that the water, ethanol, and butanol extracts after fermentation with L. plantarum could be useful as functional natural materials with anti-inflammatory effects.

Assessment of Fertilizer Efficiency of Pharmaceutical Byproduct and Cosmetic Industry Wastewater Sludge as Raw Materials of Compost (제약업종 부산물 및 화장품 제조업 폐수처리오니의 비효검정)

  • Pharmaceutical byproduct sludge and cosmetic industry wastewater sludge can be used as a raw material of compost. Effects of three types of pharmaceutical byproduct sludge and one type of cosmetic industry wastewater sludge on soil properties and red pepper growth were investigated in a field based concrete pot ($2{\times}2m$). These sludges and pig manure ($5Mg\;ha^{-1}$, dry basis) were incorporated into the upper of loam soil 30 days prior to transplanting red pepper. Changes in soil properties and contents of heavy metals and toxic organic compounds in soil and plant were measured. And also plant growth measurement and bioassay of soil phytotoxicity were included. Contents of heavy metals were increased in the soils treated with the sludges. Plant growth in the sludge treatments were mostly inferior to that of NPK treatment, especially in early stage. Content of N in plant was lower in all sludge treatments at early and middle growth stages, and it was especially caused by characteristics and concentration of nitrogen and organic matter of sludges. Total yield of red pepper was highest in the NPK treatment and followed by pharmaceutical sludge 3, pig manure, pharmaceutical sludge 1, and pharmaceutical sludge 2, and the yield of cosmetic sludge treatment was considerably lower than others. HEM and PAHs contents in soil of cosmetic sludge treatment were $4.80mg\;kg^{-1}$ and $2,263.2{\mu}g\;kg^{-1}$, respectively. Root elongation of lettuce exposed to the water extract of soil treated with cosmetic sludge was about 20% of that found in the test with soil extract of non fertilization treatment. At present, raw materials of compost were authorized according to the contents of organic matter, heavy metals and product processing. Toxic organic compounds analysis and bioassay would be helpful for authorization and assessment of suitability of raw materials of compost.
