Diversity and Saline Resistance of Endophytic Fungi Associated with Pinus thunbergii in Coastal Shelterbelts of Korea

  • The Black Pine, Pinus thunbergii, is widely distributed along the eastern coast of Korea and its importance as a shelterbelt was highlighted after tsunamis in Indonesia and Japan. The root endophytic diversity of P. thunbergii was investigated in three coastal regions; Goseong, Uljin, and Busan. Fungi were isolated from the root tips, and growth rates of pure cultures were measured and compared between PDA with and without 3% NaCl to determine their saline resistance. A total of 259 isolates were divided into 136 morphotypes, of which internal transcribed spacer region sequences identified 58 species. Representatives of each major fungi phylum were present: 44 Ascomycota, 8 Zygomycota, and 6 Basidiomycota. Eighteen species exhibited saline resistance, many of which were Penicillium and Trichoderma species. Shoreline habitats harbored higher saline-tolerant endophytic diversity compared with inland sites. This investigation indicates that endophytes of P. thunbergii living closer to the coast may have higher resistance to salinity and potentially have specific relationships with P. thunbergii.

Effects of Temperature and Humidity on Fungal Occurrence in Dried Red Pepper during Storage

  • Dried red peppers are frequently contaminated with mycotoxins during storage. To determine the effect of storage environments on fungal occurrence and subsequent mycotoxin accumulation in dried red peppers, we monitored red pepper powder and whole fruit samples for fungal occurrence under various temperatures and relative humidity (RH) conditions during 340 days. Fungal occurrences fluctuated in both pepper forms throughout the storage but they were higher in pepper powder than whole one, higher under low temperatures (-20℃, 0℃, or 4℃) than others (10℃, 25℃, or 30℃), and higher under RH 93% than RH 51% and 69% in both peppers. The samples exhibiting high fungal occurrences were associated mainly with dominant species such as Aspergillussydowii, Penicillium solitum, P. roqueforti, P. polonicum, or P. chrysogenum. Mycotoxigenic species, including A. flavus, A. ochraceus, A. westerdijkiae, A. tubingensis, and P. citrinum, were also detected throughout the samples. Although mycotoxins were not detected in the samples, mycotoxigenic potential of A. flavus, A. ochraceus, and A. westerdijkiae isolates were confirmed. These results show that low temperatures (-20℃, 0℃, or 4℃) and/or high surrounding RH (>93%) are not safe environments for storage of dried red peppers as fungal growth can occur under these conditions.

Biological Control of Soil-borne Diseases with Antagonistic Bacteria

  • Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.

국내 귀리의Fusarium속 균의 다양성 및 독소 화학형 (Identification and Chemotype Profiling of Fusarium Species in Korean Oat)

  • 본 연구에서는 귀리에서 독성곰팡이 발생현황을 평가하기 위해 2017년 2018년 4월부터 6월까지 귀리 재배지 강진, 정읍을 정기적으로 모니터링 하였다. 총 745개의 곰팡이 균주를 형태학적 방법으로 분리한 후 마커 유전자 염기서열을 분석하여 동정하였다. 분리된 곰팡이의 약 92%가 Fusarium속 균주였고, Penicillium속(5.9%)과 Aspergillus속(2.1%) 순으로 분리되었다. Fusarium속 균주의 대부분이 F. asiaticum (83.1%)이었고, F. incarnatum (5.4%), F. proliferatum (3.5%), F. fujikuroi (2.8%), F. tricinctum species complex (FTSC) 11 (1.5%), F. graminearum (1.0%) 순으로 많이 분리되었다. F. asiaticum의 약 97%가 nivalenol 독소화학형이 있었고, 3-acetyl deoxynivalenol (3.2%) 독소화학형과 15-acetyl deoxynivalenol (0.4%) 독소화학형도 발견되었다. 선발된 Fusarium 균들의 병원성 실험결과 F. asiaticum이 실험에 사용된 모든 식물체에서 다양한 범위의 병원성을 나타내었다. 병증이 있는 종실에서 분리된 F. graminearum and FTSC 11 균주들은 쌀귀리에서 강한 병원성을 나타내었다. 또한 FTSC 11 균주를 제외한 모든 균주들이 쌀배지에서 nivalenol (0.2-7.6 ㎍/g), deoxynivalenol (0.03-6.1 ㎍/g), zearalenone (0.1-27.0 ㎍/g)을 생성하였다. 본 연구는 국내에서는 처음 보고되는 F. asiaticum에 의한 귀리의 붉은 곰팡이병 사례이다. 본 연구의 결과는 밀, 보리, 벼와 마찬가지로 F. asiaticum이국내귀리에서우점종임을보여준다.

전통 대두발효식품 중에 존재하는 Ochratoxin A 생성균주의 특성 (Characteristics of the Ochratoxin A Producing Fungi in Traditionally Fermented Korean Soybean Foodstuffs)

  • 국내에서 재래적인 방법에 의하여, 생산 시판되고 있는 전통 발효식품인 메주(12종), 된장(28종) 및 간장(28종)을 수거하여 이들로부터 OA를 생성하는 fungi를 분리 및 동정하였고, 동정된 fungi 중 OA를 가장 많이 생성하는 4 isolates을 선택하여 이들의 생육과 OA 생성 특성을 조사하였다. 각 시료에서 불리해낸 222 fungi 중 OA를 생성하는 것은 39 isolates로, 곰팡이의 OA 생성률이 17.7%(39/222)이며, 이들 중 가장 많은 OA를 생성하는 4 Fungi는 Penicillium spp., Phialotubus microsporus, Eupenicillium lapidosum 및 Paecilomyces variotti이었다. 이들 균주는 $a_{w}$ 0.99에서 가장 잘 자랐고 $a_{w}$ 0.85에서 OA의 생성이 중지되었다. 또한 $30^{\circ}C$에서 억제되었다. Toxin 생성 최적 pH는 Paecilomyces variotti을 제외하고는 6.5이었으며 pH에 의해서 toxin 생성은 $a_{w}$와 달리 영향을 크게 받지 않았다.

훈증방제 처리한 참나무시들음병 감염목의 사상균 조사 (Investigation of Fungi in Pesticide Fumigated Oak Wilt-Diseased Logs)

  • 한국에서 발병하는 참나무시들음병은 암브로시아 곤충인 Platypus koryoensis.에 의해 전반되는 Raffaelea quercus-mongolicae 균류에 의해 발생한다. 이 병의 전파를 방지하기 위해 참나무시들음병 감염목을 벌채 후 비닐막을 씌워 훈증하고 벌채장소에 3년 이상 방치시키고 있다. 본 연구는 이런 훈증처리된 감염목에 존재하는 균류에 대한 정보를 얻고자 수행하였다. 천안 태조산 지역 두 곳에서 훈증처리된 감염목을 채집하고 진균을 분리하여 99개 균주를 얻었다. 분리 균주를 형태적 특성과 translation elongation factor 1-alpha 유전자와 ITS rDNA region 염기서열에 의거한 분자적 방법으로 동정한 결과 Hypocrea, Trichoderma, Penicillium 속에 속하는 종들이 확인되었다. 이중 Trichoderma 속에 속하는 종이 주요 균류이었다. 조사결과 병원균인 R. quercus-mongolicae와 매개충인 P. koryoensis는 검출되지 않았다. 본 연구결과는 훈증 처리한 참나무시들음병 감염목이 3년 이상 되면 더 이상의 피해를 유발할 위험요소가 없다는 과학적 증거를 제시한다.

김치 분리 유산균의 치즈 곰팡이 항진균 활성 (Antifungal activity against cheese fungi by lactic acid bacteria isolated from kimchi)

  • 본 연구는 치즈 숙성 중 오염원인 곰팡이균 제거기술 개발의 일환으로 수행하였다. 치즈 및 치즈 숙성실에서 자생하는 곰팡이를 분리하고, 지역별 김치로부터 유산균을 분리하여 치즈 곰팡이에 대한 항진균 활성을 측정한 결과는 다음과 같다. 치즈와 치즈 숙성실에 분리된 곰팡이는 총 8종으로Penicillium과 Cladosporium으로 동정되었다. 지역별 김치로부터 분리된 유산균은 총 44종이고 이 중 항진균 활성을 보인 유산균 22종을 동정한 결과 Lactobacillus와 Pediococcus로 확인되었다. 동정된 김치 분리 유산균 22종 중 항진균 활성도가 높은 유산균 6종(L. sakei subsp. ALJ011, L. sakei subsp. ALI033, L. sakei subsp. ALGy039, P. pentosaceus ALJ015, P. pentosaceus ALJ024, P. pentosaceus ALJ026)을 선발하여, 치즈 곰팡이균 8종에 대하여 항진균 활성을 측정한 결과 전주지역 김치에서 분리한 유산균인 P. pentosaceus ALJ015, P. pentosaceus ALJ024과 임실지역 김치에서 분리한 유산균인 L. sakei subsp. ALI033 균주가 치즈 곰팡이에 대한 항진균 활성이 높게 나타났다. 치즈 곰팡이에 대한 유산균 배양액의 최소저해 농도를 측정해 본 결과, 생육저지환으로 알아본 항균활성 실험과 동일하게 분리 곰팡이균 8종에 대하여L. sakei subsp. ALI033 유산균의 항진균 활성이 가장 크게 나타났다.

저장 땅콩에서 분리된 Aspergillus flavus의 다양성 및 독소생성능 (Diversity and Mycotoxin Production of Aspergillus flavus in Stored Peanut)

  • 땅콩의 독성곰팡이 및 곰팡이독소 오염현황을 조사하기 위해 고창 지역의 저장고에서 땅콩을 수집하였다. 수집된 땅콩은 Fusarium (17.2±28.0%), Penicillium (12.4±28.0%), Aspergillus (8.0±7.6%) 속 곰팡이로 주로 오염되어 있었으며, 그 외에 Talaromyces, Rhizopus, Rhizoctonia, Trichocladium, Clonostachys, Mucor, Chaetomium, Trametes, Epicoccum, Humicola 속 곰팡이가 검출되었다. 땅콩시료에서 아플라톡신은 검출되지 않았으나, 29점의A. flavus 균주가 분리, 동정되었다. 그 중 17점의 균주를 선발하여 potato dextrose agar (PDA) 배지 상에서 아플라톡신 생성능을 분석한 결과 0.61-187.82 ㎍/kg의 농도범위에서 모두 아플라톡신을 생성하였다. 이들 17 균주는 아플라톡신 B1, B2도 생성하였으며, 일부(5 균주)는 G1 또는 G2를 생성하였다. 이 연구는 국내 저장 땅콩에서A. flavus 오염률 및 독소생성능에 대한 첫 보고이다.

식품제조공장 내 공중부유미생물 오염도와 오염진균동정 (Identification and Concentration of Airborne Microbes in Food Manufacturing Plants)

  • To evaluate the indoor air quality of food manufacturing plants, the presence of viable bacteria and fungi was assessed in the indoor air of the facilities at which 9 food items were manufactured. Air samples were collected from the general zone, low clean zone and clean zone of each factory with an air sampler, in combination with plate counts agar using for bacteria, and dichloran-glycerol agar for fungi. The samples were incubated at $25^{\circ}C$ for 4 to 7 days. After culture, the colony forming units (CFU) on each plate were counted and corrected with a positive hole conversion table. The average concentration of bacteria was $2.2{\times}10^3\;CFU/m^3$ in the general zone, $1.2{\times}10^3\;CFU/m^3$ in the low clean zone and $7.3{\times}10^2\;CFU/m^3$ in the clean zone. The average concentration of fungal microbes was $2.5{\times}10^3\;CFU/m^3$ in the general zone, $2.6{\times}10^3\;CFU/m^3$ in the low clean zone, and $2.0{\times}10^2\;CFU/m^3$ in the clean zone. No meaningful differences were detected between the general zone and the low clean zone, but the clean zone had significantly lower concentrations than the other zones. Additionally, the identification of the fungi was performed according to morphological method using a giant culture and slide culture. The fungi were identified as belonging to 18 genera, and the genera Cladosporium(33%), Penicillium(29%) and Aspergillus(26%), predominated. Aspergillus isolates were identified to species level, and A. ochraceus, a mycotoxigenic species, was identified. As part of the effort to control the quality of the indoor air of food manufacturing plants, our results show that continued studies are clearly warranted.

Biocontrol Activity of Bacillus amyloliquefaciens CNU114001 against Fungal Plant Diseases

  • A total of 62 bacterial isolates were obtained from Gomsohang mud flat, Mohang mud flat, and Jeju Island, Republic of Korea. Among them, the isolate CNU114001 showed significant antagonistic activity against pathogenic fungi by dual culture method. The isolate CNU114001 was identified as Bacillus amyloliquefaciens by morphological observation and molecular data analysis, including 16SrDNA and gyraseA (gyrA) gene sequences. Antifungal substances of the isolate were extracted and purified by silica gel column chromatography, thin layer chromatography, and high performance liquid chromatography. The heat and UV ray stable compound was identified as iturin, a lipopeptide (LP). The isolate CNU114001 showed broad spectrum activity against 12 phytopathogenic fungi by dual culture method. The semi purified compound significantly inhibits the mycelial growth of pathogenic fungi (Alternaria panax, Botrytis cinera, Colletotrichum orbiculare, Penicillium digitatum, Pyricularia grisea and Sclerotinia sclerotiorum) at 200 ppm concentration. Spore germ tube elongation of Botrytis cinerea was inhibited by culture filtrate of the isolate. Crude antifungal substance showed antagonistic activity against cucumber scleotiorum rot in laboratory, and showed antagonistic activity against tomato gray mold, cucumber, and pumpkin powdery mildew in greenhouse condition.
