Foodborne illness caused by Noroviruses (NVs) is increasing rapidly in Korea. This study developed an effective detection protocol for NVs found in contaminated oysters and lettuce through an investigation using the major steps of virus particle separation, concentration and RT-PCR. As a surrogate model for NVs, the cultivable feline calicivirus (FCV) that belongs to the same Caliciviridae family was used. Instead of using a time-consuming ultracentrifugation method, efficient methods based on solvent extraction and PEG precipitation procedure were applied. Direct homogenization of a 25g sample of whole oyster and lettuce in 175mL PBS provided the simplicity that would be needed in the actual field of food product examination. The overnight PEG precipitation step at $4^{\circ}C$ was reduced to 3 h by placing the reaction tube in ice and by adjusting the PEG concentrations. The application of the use of chloroform and 0.2 ${\mu}m$ syringe filtration together showed a better detection efficiency than the use of chloroform alone in removing PCR inhibitors for both oyster and lettuce samples. Also, dilution of the extracted RNA solution before PCR provided increased sensitivity. The improved detection protocol developed in this study could be efficiently applied to detect FCV and most likely NVs from oysters and lettuce.
A comparison of analytical approaches for Levoglucosan ($C_6H_{10}O_5$, commonly formed from the pyrolysis of carbohydrates such as cellulose) and used for a molecular marker in biomass burning is made between the four different analytical systems. 1) Spectrothermography technique as the evaluation of thermograms of carbon using Elemental Carbon & Organic Carbon Analyzer, 2) mass spectrometry technique using Gas Chromatography/mass spectrometer (GC/MS), 3) Aerosol Mass Spectrometer (AMS) for the identification of the particle size distribution and chemical composition, and 4) two dimensional Gas Chromatography with Time of Flight mass spectrometry (GC${\times}$GC-TOFMS) for defining the signature of Levoglucosan in terms of chemical analytical process. First, a Spectrothermography, which is defined as the graphical representation of the carbon, can be measured as a function of temperature during the thermal separation process and spectrothermographic analysis. GC/MS can detect mass fragment ions of Levoglucosan characterized by its base peak at m/z 60, 73 in mass fragment-grams by methylation and m/z 217, 204 by trimethylsilylderivatives (TMS-derivatives). AMS can be used to analyze the base peak at m/z 60.021, 73.029 in mass fragment-grams with a multiple-peak Gaussian curve fit algorithm. In the analysis of TMS derivatives by GC${\times}$GC-TOFMS, it can detect m/z 73 as the base ion for the identification of Levoglucosan. It can also observe m/z 217 and 204 with existence of m/z 333. Although the ratios of m/z 217 and m/z 204 to the base ion (m/z 73) in the mass spectrum of GC${\times}$GC-TOFMS lower than those of GC/MS, Levoglucosan can be separated and characterized from D (-) +Ribose in the mixture of sugar compounds. At last, the environmental significance of Levoglucosan will be discussed with respect to the health effect to offer important opportunities for clinical and potential epidemiological research for reducing incidence of cardiovascular and respiratory diseases.

It is well known that volatile compounds including disinfection by-products as well as emissive dissolved gas in water can be removed effectively by air stripping. The micro-bubbles of flotation unit are so tiny as microns while the diameter of fine bubbles applied to air stripping is ranged from hundreds to thousands of micrometer. Therefore, the micro-bubbles in flotation can supply very wide specific surface area to transfer volatile matters through gas-liquid boundary. In addition, long emission time also can be gained to emit the volatile compound owing to the slow rise velocity of micro-bubbles in the flotation tank. There was a significant difference of the THMs species removal efficiency between air stripping and flotation experiments in this study. Moreover, the results of comparative experiments on the removal characteristics of THMs between air stripping and flotation revealed that the mass transfer coefficient, $K_La$ showed obvious differences. To overcome the limit of low removal efficiency of dissolved volatile compounds such as THMs in flotation process, the operation range of bubble volume concentration is required to higher than the operation condition of conventional particle separation.

This study was carried out to investigate the residue level of fluoroquinolones in hen's general eggs and specific eggs by microbiological assay method and high performance liquid chromatography (HPLC) method. HPLC separation was carried out by reversed phase chromatography on a Symmetry $C_{18}$ (250${\times}$4.6 mm, $5{\mu}m$ particle size) with a phase composed of distilled water (containing 0.4% triethylamine and phosphoric acid) : Methanol (780 : 220, v/v), pumped isocratically at a flow rate of 1.0ml/min. A fluorescence detector was utilized with an excitation wavelength of 278nm and an emission wavelength of 456nm. The calibration curves were linear $({\gamma}^2{\geq}0.999)$ over a concentration range of $0.025{\sim}0.4{\mu}g/ml$. Average recoveries of the five fluoroquinolones in whole eggs at fortified levels of $0.05{\sim}0.2{\mu}g/g$ were ranged mean $78.1{\sim}91.7%$ and low coefficient of variation was less than 10% for all analysed samples. The limits of detection and limits of quantification for whole eggs were $1.2{\sim}6.0ng/g$ and $2.3{\sim}9.1ng/g$, respectively. Only one hen's general eggfrom chicken farm in Incheon was detected with the residual fluoroquinolones (Microbiological assay method; 1 of 47 general eggs) ; the range of residual concentration enrofloxacin was 0.12ppm. Those in food stores were detected with the residual fluoroquinolones (Microbiological assay method; 4 of 88 general eggs) ; the ranges of residual concentration enrofloxacin were $0.15{\sim}2.2 ppm$, ciprofloxacin $0.01{\sim}0.06ppm$, and hen's specific eggs (40) in food stores were not detected. For the microbiological assay method of fluoroquinolones in hen's eggs, as the results of comparative analysis, the disc diffusion method with E coli may be a little highly detected for the residual fluoroquinolones.
This study was focused on iron removal from illite by L-ascorbic and oxalic acids. Iron has been shown as a secondary mineral such as iron oxides and hydroxides in illite ores. It is also known as a primary agent to degrade brightness index of the ores. Methods such as physical separation and chemical leaching with strong inorganic acids have been widely used to remove the iron from the ores. However, these methods are expensive and give rise to environmental problems. In this study, we examined an alternative method using solutions with different set of combination of sulfuric, hydrochloric, L-ascorbic, and oxalic acids. Compared to chemical treatments with only inorganic acids, our results demonstrate that an addition of L-ascorbic acid in inorganic acids results in decreasing both total concentrations of the inorganic acids and time for the treatments. The treatment with 0.15 M L-ascorbic acid and 0.25 M sulfuric acid in solution for 60 min significantly improved the brightness index from 42.4% to 74.4%. This improvement is similar to that of treatment with only 2.5 M sulfuric acid alone for 150 min. Mineralogical and chemical analyses were performed to compare the effect of acid leaching on illite powders. No obvious differences are observed in the mineralogical characteristics and particle size distributions of the samples. These results suggest that the treatment with the addition of L-ascorbic acid in sulfuric acid could effectively remove iron without modifying the physicochemical properties of illite under conditions used in this study.
The phase change of $(U_{1-x}Nd_x)_3O_8$ powder produced by oxidation of Nd-doped $UO_2$ pellet at $500^{\circ}C$ was investigated by high temperature oxidation heat treatment at $900{\sim}1500^{\circ}C$ under an air atmosphere. The XRD analysis results showed that the formation of $(U_{1-y}Nd_y)O_{2+z}$ phase and $U_3O_8$ phase from metastable $(U,Nd)_3O_8$ phase initiated at a temperature of $1000^{\circ}C$. The relative integrated intensity of $(U_{1-y}Nd_y)O_{2+z}$ phase to $U_3O_8$ phase increased with increasing of the oxidation temperature from 1100 to $1500^{\circ}C$. And also, it was found from the SEM observation that the particle size of $(U_{1-y}Nd_y)O_{2+z}$ phase increased with increasing of the oxidation temperature. However, electrone probe X-ray microanalyzer (EPMA) analysis results showed that Nd contents in $(U_{1-y}Nd_y)O_{2+z}$ phase decreased with increasing of the oxidation temperature. This behavior on the ground of XRD, SEM, and EPMA analysis data could be interpreted in terms of the transportation of U ions from $U_3O_8$ phase into $(U_{1-y}Nd_y)O_{2+z}$ phase through the interface of two phases during high temperature oxidation.

In the present study, pure RHO zeolite was hydrothermally synthesized by using 18-crown-6 ether as a structure directing agent(SDA), and the small gases adsorption was investigated. Synthesized RHO zeolite was a cube shape particle of which average edge length was around $1.2{\mu}m$ and composed of primary crystallites having a diameter of around 100 to 200 nm. RHO zeolite structure was stable under 3h calcination at $600^{\circ}C$. Water adsorption data announced that RHO zeolite has a specific surface area of 483.32 m2/g and its micropore diameter was about 4 A. Gas adsorption was studied in the pressure range of 50 to 500 kPa for $CO_2$, $N_2$, $O_2$ and $H_2$. It was evident that RHO zeolite showed a strong $CO_2$ adsorption behavior. Especially, RHO zeolite showed a transient $CO_2$ adsorption behavior. The 3h $CO_2$ up-take at 50 kPa and 500 kPa was 1.283 and 3.357 mmol/g, respectively. The $CO_2/H_2$ selectivity was around 16 at 500 kPa. Compared with gas adsorption data for some representative microporous adsorbents, it was certain that RHO zeolite is a beneficial adsorbent for $CO_2/H_2$ separation.
The deterioration and structural damage such as exfoliation, cracks, and separation of painted layer on the wall paintings of Maitreya Hall in Geumsan-sa temple have been accelerated since it was re-positioned to the original place after the dismantling from the building in 1993. The examination of which result and analysis described in this study, is a preliminary survey for establishing conservation plan of the wall paintings. It aimed at the understanding of the physical and chemical characteristics of the materials applied in the 1993 conservation. The research focused on the south walls which displayed the worst condition compared to other walls. Samples for the examination for the understanding of micro-structure, chemical composition, cristalisation, and particle distribution, were collected for finishing, middle, and consolidated layers of the walls between pillars and the ones between brackets. Those samples were collected from separated fragments of the walls. The sample analysis displayed that: 1. the 1993 conservation used the similar type of weathered soil as the original for the finishing layer, and such soil and sand for the middle layer; 2. those walls are composed of a group of mineral particles which are relatively equal in size and shape and in their distribution; 3. the mineral particles were cohered forming solid aggregate due to the application of acrylic resin for the reinforcement on the wall. The main composition of crystalisation on the first and the second reinforcement layers of the back walls were lime plaster ($CaSO_4{\cdot}2H_2O$). The overall examination confirmed that the priority of the future conservation treatment should be given to the removal of the first and the second layers of reinforcement and the treatment on the back walls which were partially consolidated.
In order to develop a new separation technology, supercritical fluid extraction process was used to produce high purity pigments and fatty acids from seaweed (Undaria pinnatifida). Supercritical carbon dioxide was used as a solvent and ethanol as an entrainer. The sample was treated by a frozen drier and experiments were conducted with a semi-batch flow system at various operating conditions (pressure range, $10.3\~17.2$ MPa; temperature range, $30\~45^{\circ}C$: particle size, $500\~1,000{\mu}m$ extraction time, 60 min). Characteristics of the recovered pigment (chlorophyll a) and fatty acids were determined by UV-spectrophotometry and gas chromatography, respectively. The highest extraction efficiency for fatty acids and pigments was achieved at 12.4 MPa, $35^{\circ}C$, $500{\mu}m$of seaweed size.
The average specific cake resistance, the most important indicator for cake filtration and solid-liquid separation, is measured by filtration experiment. But the exact value is difficult to measure because of the other influences such as sedimentation during filtration. This study, a little more stable method named filtration-permeation is proposed for measuring average specific cake resistance. The filtration-permeation is composed of permeation of particle eliminated water through pre-formed cake by filtration. Using 1 wt% calcium carbonate suspension, the filtration-permeation experiments were performed for 8 kinds of filter media at the conditions of 0.5 atm and 0.2 atm, 1 and 3 sheets of filter media. At each specific condition, three to five times filtration-permeation were accomplished. As a result, stable permeation speed is measured. According to this experimental result, the characteristics of permeation and the effect of sedimentation are analyzed with Ruth's equation. The one way analysis of variance (one way ANOVA) is applied to the average specific cake resistances of filtration and permeation obtained with the selected three kinds of filter media. The average specific cake resistances between 0.5 atm and 0.2 atm by filtration do not distinguished, but those by permeation is perfectly distinguished. The experimental results during permeation have a very narrow distribution than that measured during filtration. The analysis of filtration experiments, it was verified that the resistance of filter medium by traditional method is of no significance. Finally, the migration of small particles through the medium composed of fiber glass at low pressure was studied.
