Simultaneous Analysis of the Coloring Compounds in Indigo, Phellodendron bark, and Madder Dye Using HPLC-DAD-MS

  • Indigotin, indirubin, berberine, palmatine, alizarin, and purpurin are major pigments of indigo plant, Phellodendron bark, and madder. The six pigments were examined using the HPLC-DAD-MS instrument for the purpose of the simultaneous detection of the pigments in a single sample run. The HPLC-DAD-MS method examined the individual pigment solutions in DMSO, a solution containing 6 pigments, and the DMSO extract of the silk dyed with a dye solution of 5 pigments excluding indirubin. The retention times of the HPLC chromatograms, ${\lambda}_{max}$ of the uv-vis absorption bands in the DAD analyses, and the molecular ions detected for the compound peaks in the MSD analyses were consistent throughout the analyses of individual pigment solutions, mixed pigment solutions, and dye extracted from silk dyeing. The developed instrumental method of the simultaneous detection of six pigments can identify dye in an exhumed textile if the textile is dyed using any one (or multiple) pigments of indigo, Phellodendron bark, or madder plant.

Protoberberine Alkaloids and their Reversal Activity of P-gp Expressed Multidrug Resistance (MDR) from the Rhizome of Coptis japonica Makino

  • Six protoberberine alkaloids were isolated from the chloroform layer of the rhizome of Coptis japonica Makino (Ranunculaceae). The structures of the isolated compounds were determined to be 6-([1,3]dioxolo[4,5-g]isoquinoline-5-carbonyl)-2,3-dimethoxy-benzoic acid methyl ester (1), oxyberberine (2), 8-oxo-epiberberine (3), 8-oxocoptisine (4), berberine (5) and palmatine (6) by physicochemical and spectroscopic methods. The compound 3 (8-oxo-epiberberine) was first isolated from natural sources. The compounds were tested for cytotoxicity against five tumor cell lines in vitro by SRB method, and also tested for the MDR reversal activities. Compound 4 was of significant P-gp MDR inhibition activity with ED50 value $0.018\;{\mu}g/mL$ in MES-SA/DX5 cell and $0.0005\;{\mu}g/mL$ in HCT15 cell, respectively.

Acetylcholinesterase Inhibitors from the Aerial Parts of Corydalis speciosa

  • In a bioassay-guided search for acetylcholinesterase inhibitors from Korean natural resources, four isoquinoline alkaloids, corynoxidine (1), protopine (2), palmatine (3), and berberine (4) have been isolated from the methanolic extract of the aerial parts of Corydalis speciosa. Structures of these compounds were elucidated on the basis of spectroscopic techniques. These compounds inhibited acetylcholinesterase activity in a dose-dependent manner, and the $IC_50$ values of compounds 1-4 were 89.0, 16.1, 5.8, and 3.3 $\mu$ M, respectively.

Determination of Coptidis Rhizoma Alkaloids in Preparations by Spectrophotometric Method (흡광도측정법에 의한 제제 중 황련 알칼로이드의 정량)

  • The Coptidis rhizoma is known for containing protoberberine alkaloids. Berberine, coptisine and palmatine are the major constituents of protoberberine alkaloids. The alkaloids were isolated and determined by forming complex compounds from Coptidis rhizoma in preparation I(Sam-Hwang-Sa-Sim-Tang) and II(Hwang-Ryen-Tang). For the determination of these alkaloids, a new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex compound ion. The absorbance of alkaloidal complex compounds in l.2-dichloroethane solution was measured at 625 nm. Calibration curve for the alkaloids isolated from Coptidis rhizoma was linear over the concentration range of 0.2-0.3 mg/ml. The method was proved to be rapid, simple and reliable for the isolation and the determination of the alkaloids in Coptidis rhizoma preparation I and II.

Chemical Constituents from the Bark of Phellodendron amurense and Their Cytotoxic Effects on HL-60 Human Leukemia Cells

  • Phellodendri Cortex, phellodendron bark, has been used as a stomachic for intestinal function control and as an antimicro and anti-inflammatory agent. In this phytochemical study, eight compounds, berberine (1), palmatine (2), syringin (3), (+)-syringaresinol di-O-${\beta}$-D-glucopyranoside (4), salvadoraside (5), citrusin B (6), osmanthuside H (7), and kelampayoside A (8), were isolated from the bark of Phellodendron amurense. Their structures were elucidated by comparing spectroscopic data with reported values. Compounds 1 - 8 were evaluated for cytotoxic activity against HL-60 human promyelocytic leukemia cells in vitro. Among them, compounds 1 and 2 reduced the viability of HL-60 cells significantly, with $IC_{50}$ values of 26.0 and $18.5{\mu}M$, respectively.

  • The quality control of natural products is principal key to guarantee the Good Manufacturing Practices (GMP) and Good Clinical Practices (GCP) for the functional food, pharmaceuticals and cosmeceuticals in the industry. In this study, we examined the quantitative analysis of berberine as marker substance of Coptidis Rhizoma by high performance liquid chromatography-photodiode array detector (HPLC-DAD). The HPLC method was validated and met all the requirements for the quality control analysis recommended by FDA and ICH. The berberine was separated on a Xterra $C_{18}$ column ($5{\mu}m$, $4.6{\times}250mm$) using mobile phase consisting of distilled water and acetonitrile with $KH_2PO_4$ (3.4 g) and $Na_2SO_4$ (1.7 g). Calibration curve of berberine has been estimated (y = 42293.47x-41589 with the correlation coefficient 0.9999). The amount of berberine was calculated as 4.25%. And berberastine, palmatine, columbamine, jatrorrhizine, epiberberine, berberine and coptisine in the Coptidis Rhizoma were identified by high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-ESI-MS) method.

Assessment of the Quality Variance among Commercial Hwang-Ryun-Hae-Dok-Tang by Simultaneous Analysis of Characterizing Compounds (황련해독탕제제의 지표성분 동시 분석을 통한 품질비교)

  • A study was performed to assess the quality equivalence of commercially available Hwang-Ryun-Hae-Dok-Tang granules from various vendors in terms of the contents of characterizing compounds. 'Hwang-Ryun-Hae-Dok-Tang'(or 'Huanglian Jiedu Tang') is the complex medicinal herbal product which is composed of four different herbal drugs; Scutellariae Radix, Coptidis Rhizoma, Phellodendri Cortex and Gardeniae Fructus. Using a high performance liquid chromatography and direct analysis in real time mass spectrometer (DART-MS), five characterizing compounds such as baicalin, baicalein, berberine, palmatine, and geniposide were analyzed to compare the variance of the contents between the products from four different vendors. Although five characterizing compounds were all successfully detected from all the products, the contents of each compound were shown to have significant difference among vendors. For real time monitoring of characterizing compounds in medicinal herbal products, DART-MS can be successfully adopted as a powerful analytical tool.

Effect of Hwangryunhaedok-tang on Blood Pressure and Renal Functions in Spontaneously Hypertensive Rats (황연해독탕(黃連解毒湯)이 자발적(自發的) 고혈압(高血壓) 백서(白鼠)의 혈압(血壓) 신장(腎臟) 기능(機能)에 미치는 영향(影響))

  • The present study designed to investigate whether hwangryunhaedok-tang show an anti-hypertensive effect and elucidate its possible mechanism in spontaneously hypertensive rats. The systolic blood pressures (SBP) were significantly decreased as an oral administration of hwangryunhaedok-tang compared with their control group. The urine volume was significantly increased by administration of hwangryunhaedok-tang but urinary sodium (UNaV), potassium (UKV), chloride excretion (UCIV) were not remarkably affected. The urinary creatinine excretion rate (UcrV) was significantly increased in rats administered with hwangryunhaedok-tang in association with increase of creatinine clearance (Ccr). The urine osmolality (Uosmol) was significantly decreased in SHR administered with hwangryunhaedok-tang without being changed in solute-free water reabsorption ( TcH20). The expressions of Aquaporin 2 (AQP-2). 3 and ${\alpha}\;1$, ${\beta}\;1$ subunits of Na.K-ATPase were determined by Western blot analysis to assess the role of these proteins in association with changes of renal functions in SHR administered with hwangryunhaedok-tang. The expression of AQP-2 and 3 protein was significantly down-regulated in the kidney of SHR administered with hwangryunhaedok-tang compared with those in control rats without being altered expression of ${\alpha}\;1$, ${\beta}\;1$ subunits of Na,K-ATPase. In the in vitro assay, Angiotensin converting enzyme (ACE) was inhibited by hwangryunhaedok-tang in a dose-dependent manner. Berberine and/or palmatine, which are well known as a main components of hwangryunhaedok-tang, also have an ACE inhibitory effects in a dose-dependent manner. Taken together, these results suggest that hwangryunhaedok-tang lowered blood pressure through the increase of diuresis caused by down-regulation of water channels and the inhibition of Angiotensin converting enzyme.

Determination of isoquinoline alkaloids by UPLC-ESI-Q-TOF MS: Application to Chelidonium majus L.

  • In this study, we set up an analytical method that can be used for rapid and accurate determination of representative isoquinoline alkaloids in medicinal plants using UPLC-ESI-Q-TOF MS (ultra pressure liquid chromatography-electrospray ionization-quadrupole-time-of-flight mass spectrometry). The compounds were eluted on a C18 column with 0.1 % formic acid and acetonitrile, and separated with good resolution within 13 min. Each of the separated components was characterized by precursor ions (generated by ESI-Q-TOF) and fragment ions (produced by collision-induced dissociation, CID), which were used as a reliable database. We also performed method validation: analytes showed excellent linearity ($R^2$, 0.9971-0.9996), LOD (5-25 ng/mL), LOQ (17-82 ng/mL), accuracy (91.6-97.4 %) as well as intra- and inter-day precisions (RSD, 1.8-3.2 %). In the analysis of Chelidonium majus L., magnoflorine, coptisine, sanguinarine, berberine and palmatine were detected by matching retention times and characteristic fragment ion patterns of reference standards. We also confirmed that, among the quantified components, coptisine was present in the highest quantity. Furthermore, alkaloid profiling was carried out by analyzing the fragment ion patterns corresponding to peaks of unknown components. In this manner, protopine, chelidonine, stylopine, dihydroberberine, canadine, and nitidine were tentatively identified. We also proposed the molecular structure of the fragment ions that appear in the mass spectrum. Therefore, we concluded that our suggested method for the determination of major isoquinoline alkaloids by UPLC-Q-TOF can be useful not only for quality control, but also for rapid and accurate investigation of phytochemical constituents of medicinal plants.

Review of Pharmacological Effects of Coptidis Rhizoma and its Bioactive Compounds (황련(黃連)과 구성 생화합물의 약리작용에 대한 고찰)

  • Objectives: Coptidis Rhizoma is one of the most popular and multi-purpose herbs in traditional medicine. The purpose of this study was to contribute to research and applications of Coptidis Rhizoma in clinic, by analyzing and reviewing international studies on its chemical compositions and pharmacological effects. Methods: This study analyzed 344 articles published from 2000 to 2010 in PubMed, Refworks, Riss, and KTKP. The search keywords were "Coptis chinensis", "Coptis japonica", "Coptidis Rhizoma", "huanglian" and "huanglian in Chinese". From them, we selected 114 articles which met our inclusion criteria. Results: This study reviewed 114 articles on Coptidis Rhizoma and its active components in terms of 'Active components', 'Experimental studies', 'Clinical studies', 'Industrial use' and 'Side Effects/Toxicity'. Conclusions: The active components of Coptidis Rhizoma are berberine, coptisine, epiberberine, palmatine, jateorrhizine, magnoflorine, worenine, etc. It is reported that Coptidis Rhizoma and its active components have anti-inflammatory, antibacterial, antitumor, and antioxidant activity, and cardiovascular, hepatoprotective, antidiabetic, neuroprotective, gastrointestinal, pain relieving, discharge phlegm and metrocyte proliferation effects. Moreover, we found that Coptidis Rhizoma can be used for bath preparation, cosmetic products and as a natural antimicrobial substance.
