오디 당침출액의 HepG2 세포에서 H2O2로 야기된 산화적 스트레스 보호 효과 (Protective effects of mulberry (Morus alba) sugar extracts on hydrogen peroxide-induced oxidative stress in HepG2 cell)

  • 오디 당침출액(MSE)의 산화적 스트레스 개선 효과를 확인하기 위하여 HepG2 세포에 $H_2O_2$로 산화적 스트레스를 유도시킨 다음, MSE의 보호효과를 확인하였다. MSE를 40일간 저장하여 DPPH radical scavensing을 통해 DPPH radical 소거능이 유의적으로 좋았던 저장 40일용 MSE를 선택하여 세포 실험에 적용하였다. HepG2 세포에 $500{\mu}M$ $H_2O_2$를 처리하여 산화적 스트레스를 유발시키고, MSE를 처리하여 세포 생존율을 확인한 결과, MSE 처리로 인한 세포 생존율이 유의적으로 증가하였고, ROS 생성과 과산화물에 대한 지표로 측정된 MDA 농도도 MSE 처리로 인해 효과적으로 억제되었다. 또한, $H_2O_2$ 처리로 감소된 SOD 및 CAT 활성이 MSE 처리로 인해 유의적으로 높아졌으며, $H_2O_2$를 처리로 인한 세포핵의 apoptosis body가 MSE 처리로 인해 감소함을 확인하였으며, 이는 caspase-3 활성 MSE가 억제시킴으로 인해 세포를 보호하고 있음을 확인하였다. 이상의 결과로부터 오디 당침출액은 산화적 스트레스로부터 야기되는 세포독성과 apoptosis로부터 세포 보호 효과를 확인함에 따라 향후 노화와 관련된 다양한 연구소재의 기초 자료 및 질병 예방 소재로의 가능성을 확인하였다.

Effects of a Mixture of Eugenol, Thymol and Malate on Growth Performance, Beef Quality and Liver Function in Hanwoo Finishing Steers Fed a High-Concentrate Diet

  • Thirty six Hanwoo steers (average BW, $564.5\;{\pm}\;25.7\;kg$; average age, approximately 23 months) were used to evaluate the effects of a mixture of eugenol (14%), thymol (1%) and malate (85%) on growth performance, beef quality and liver function in Hanwoo finishing steers fed a high-concentrate diet. A feeding trial was conducted for 7 months in National Agricultural Cooperative Federation farm located in Anseong, Korea. Steers were assigned randomly to one of three treatments: control (without additive supplementation), treatment 1 (0.05% additive of concentrate), treatment 2 (0.1% additive of concentrate). The results of this study showed that initial and final BW averaged 564 and 755 kg, respectively, and BW gain was significantly higher (P<0.05) for steers fed the additive mixture than for those fed no mixture (0.78 and 0.79 vs. 0.69 kg/d, respectively). Serum aspartate aminotransferase in the T2 treatment was decreased during the 24 to 31 months of age. Although supplementation of additives resulted in no substantial effect on carcass characteristics, it had a potential effect to improve feed efficiency and AST concentration in Hanwoo finishing steers fed a high-concentrate diet. In conclusion, a mixture of eugenol, thymol and malate has shown promise in improving feed efficiency and liver function in the finishing phase of Hanwoo steers.

임신 여성 근로자의 제대혈 중 망간 농도 추정 (Estimation of Manganese Levels in Cord Blood of Pregnant Workers)

  • Objectives: This study aims to analyze manganese (Mn) concentrations in maternal and cord bloods at delivery and to estimate the Mn exposure risk for fetuses whose mothers were occupationally exposed to Mn. Materials and Methods: Forty-six pairs of maternal and cord blood samples were collected at delivery from mothers who were occupationally unexposed to Mn. Mn concentrations of blood were analyzed by graphite furnace atomic absorption spectrometer. Mn exposure levels for fetuses of female workers were estimated by simulating two working exposure scenarios. Results: The geometric mean concentration of Mn in maternal and cord blood were $27.0(1.34){\mu}g/L$, $46.6(1.25){\mu}g/L$, respectively. Transfer ratios of Mn from maternal to cord blood were $1.81{\pm}0.62$, which indicated that the Mn concentrations in cord blood were higher than those in maternal blood. Mn concentrations in cord blood for the worse or general scenarios were estimated to $22.3-1,881{\mu}g/L$ and $1.59-308{\mu}g/L$, respectively. The probabilities of exceeding $74{\mu}g/L$, which was adopted as a reference level reported in a previous study, were 95% and 44% for the two scenarios, respectively. Conclusions: Comparable levels of Mn exposure in maternal or cord blood to those in this study have shown various health effects in previous studies. This suggests that Mn exposure levels in mothers and fetuese in Korea need to be monitored and managed. In addition, female workers who are occupationally exposed to Mn should be protected from the exposure since their fetuses can be exposed to Mn at risky levels during their pregnancy.

High frequency direct plant regeneration from leaf, internode, and root segments of Eastern Cottonwood (Populus deltoides)

  • Simple, reproducible, high frequency, improved plant regeneration protocol in Eastern Cottonwood (Populus deltoides) clones, WIMCO199 and L34, has been reported. Initially, aseptic cultures established from axillary buds of nodal segments from mature plus trees on MS liquid medium supplemented with $0.25mg\;1^{-1}$ KIN and $0.25mg\;1^{-1}$ IAA. Nodal and internodal segments were found to be extra-prolific over shoot apices during course of aseptic culture establishment, while $0.25mg\;1^{-1}$ KIN concentration played a stimulatory role in high frequency plant regeneration. Diverse explants, such as various leaf segments, internodes, and roots from in vitro raised cultures, were employed. Direct plant regeneration was at high frequency of 92% in internodes, 88% in leaf segments, and 43% in root segments. This led to the formation of multiple shoot clusters on established culture media with rapid proliferation rates. Many-fold enhanced shoot elongation and growth of the clusters could be achieved on liquid MS medium supplemented with borosilicate glass beads, which offer physical support for proliferating shoots leading to faster growth in comparison to semi-solid agar or direct liquid medium. SEM examination of initial cultures confirmed direct plant regeneration events without intervening calli. In vitro regenerated plants induced roots on half-strength MS medium with $0.15mg\;1^{-1}$ IAA. Rooted 5- to 6-week-old in vitro regenerated plants were transferred into a transgenic greenhouse in pots containing 1:1 mixture of vermicompost and soil at $27{\pm}2^{\circ}C$ for hardening and acclimatization. 14- to 15-week-old well-established hardened plants were transplanted to the field and grown to maturity. The mature in vitro raised poplar trees exhibited a high survival rate of 85%; 4-year-old healthy trees attained an average height of 8 m and an average trunk diameter of 25 cm and have performed well under field conditions. The regeneration protocol presented here will be very useful for undertaking genetic manipulation, providing a value addition to Eastern Cottonwood propagation in future.

Effects of reducing inclusion rate of roughages by changing roughage sources and concentrate types on intake, growth, rumen fermentation characteristics, and blood parameters of Hanwoo growing cattle (Bos Taurus coreanae)

  • Objective: Reducing roughage feeding without negatively affecting rumen health is of interest in ruminant nutrition. We investigated the effects of roughage sources and concentrate types on growth performance, ruminal fermentation, and blood metabolite levels in growing cattle. Methods: In this 24-week trial, 24 Hanwoo cattle ($224{\pm}24.7kg$) were fed similar nitrous and energy levels of total mixed ration formulated using two kinds of roughage (timothy hay and ryegrass straw) and two types of concentrate mixes (high starch [HS] and high fiber [HF]). The treatments were arranged in a $2{\times}2$ factorial, consisting of 32% timothy-68% HS, 24% timothy-76% HF, 24% ryegrass-76% HS, and 17% ryegrass-83% HF. Daily feed intakes were measured. Every four weeks, blood were sampled, and body weight was measured before morning feeding. Every eight weeks, rumen fluid was collected using a stomach tube over five consecutive days. Results: The mean dry matter intake (7.33 kg) and average daily gain (1,033 g) did not differ among treatments. However, significant interactions between roughage source and concentrate type were observed for the rumen and blood parameters (p<0.05). Total volatile fatty acid concentration was highest (p<0.05) in timothy-HF-fed calves. With ryegrass as the roughage source, decreasing the roughage inclusion rate increased the molar proportion of propionate and decreased the acetate-to-propionate ratio; the opposite was observed with timothy as the roughage source. Similarly, the effects of concentrate types on plasma total protein, alanine transaminase, Ca, inorganic P, total cholesterol, triglycerides, and creatinine concentrations differed with roughage source (p<0.05). Conclusion: Decreasing the dietary roughage inclusion rate by replacing forage neutral detergent fiber with that from non-roughage fiber source might be a feasible feeding practice in growing cattle. A combination of low-quality roughage with a high fiber concentrate might be economically beneficial.

유류오염지역의 지하수 수질특성 및 동절기 토양경작법의 온도보전을 위한 현장사례 연구 (The Characteristics of Groundwater and a Field Test for Thermal Insulation of Landfarming of Petroleum Contaminated Soil in Winter Season)

  • The objectives of this study were to identify the characteristics of groundwater in the petroleum contaminated site and to evaluate the applicability of house-type landfarm facilities heated with briquette stoves in winter season. The six monitoring wells were installed at the site where pH, dissolved oxygen, and temperature were all measured. Also groundwater contaminants, benzene, toluene, ethylbenzene, xylene and total petroleum hydrocarbon, were analyzed twice. House-type two landfarm facilities ($12m{\times}40m{\times}4.8m$) each installed with four briquette stoves were constructed. During four rounds treatment process, VOCs, moisture, temperature were monitored and soil contaminants were analyzed. The pH was 6.37 and considered subacid and DO was measured to be 3.12 mg/L. The temperature of groundwater was measured to be $9.48^{\circ}C$. The groundwater contaminants were detected only in the monitoring wells within the contaminated area or close to it showing that the groundwater contaminated area was similar to the soil contaminated area. During the landfarm process, 73.3% of VOCs concentration in interior gas was decreased and moisture was lowered from 17.7% to 13.4%. In the morning, at 8:00 am, the temperature was decreased showing soil ($5.5^{\circ}C$) > interior ($4.8^{\circ}C$) > exterior ($3.5^{\circ}C$). In the afternoon, at 2:00 pm, the temperature was soil ($8.6^{\circ}C$) < interior ($9.9^{\circ}C$) < exterior ($11.5^{\circ}C$) with solar radiation. The temperature difference between interior and exterior was $0.7^{\circ}C$ in the morning, but it was $1.6^{\circ}C$ in the afternoon. A total of 130 days were taken for four round landfarm processes. Each process was completed within 33 days showing 80% of cleanup efficiency ($1^{st}$ order dissipation rate(k) = 0.1771).

Metabolizable energy requirement for maintenance estimated by regression analysis of body weight gain or metabolizable energy intake in growing pigs

  • Objective: Feed energy required for pigs is first prioritized to meet maintenance costs. Additional energy intake in excess of the energy requirement for maintenance is retained as protein and fat in the body, leading to weight gain. The objective of this study was to estimate the metabolizable energy requirements for maintenance ($ME_m$) by regressing body weight (BW) gain against metabolizable energy intake (MEI) in growing pigs. Methods: Thirty-six growing pigs ($26.3{\pm}1.7kg$) were allotted to 1 of 6 treatments with 6 replicates per treatment in a randomized complete block design. Treatments were 6 feeding levels which were calculated as 50%, 60%, 70%, 80%, 90%, or 100% of the estimated ad libitum MEI ($2,400kJ/kg\;BW^{0.60}\;d$). All pigs were individually housed in metabolism crates for 30 d and weighed every 5 d. Moreover, each pig from each treatment was placed in the open-circuit respiration chambers to measure heat production (HP) and energy retained as protein ($RE_p$) and fat ($RE_f$) every 5 d. Serum biochemical parameters of pigs were analyzed at the end of the experiment. Results: The average daily gain (ADG) and HP as well as the $RE_p$ and $RE_f$ linearly increased with increasing feed intake (p<0.010). ${\beta}$-hydroxybutyrate concentration of serum tended to increase with increasing feed intake (p = 0.080). The regression equations of MEI on ADG were MEI, $kJ/kg\;BW^{0.60}\;d=1.88{\times}ADG$, g/d+782 ($R^2=0.86$) and $ME_m$ was estimated at $782kJ/kg\;BW^{0.60}\;d$. Protein retention of growing pigs would be positive while REf would be negative at this feeding level via regression equations of $RE_p$ and $RE_f$ on MEI. Conclusion: The $ME_m$ was estimated at $782kJ/kg\;BW^{0.60}\;d$ in current experiment. Furthermore, growing pigs will deposit protein and oxidize fat if provided feed at the estimated maintenance level.

Two Crystal Structures of $Tl^+$ and $Zn^{2+}$ Exchanged Zeolite A, $Tl_{12-2x}Zn_x-A$ (x=4.3 and 3.25)

  • The structures of $Tl_{12-2x}Zn_x-A$ (x = 4.3 and 3.25), vacuum dehydrated zeolite A with all $Na^+$ ions replaced by $Tl^+$ and $Zn^{2+}$ as indicated, have been determined by single-crystal X-ray diffraction techniques in cubic space group Pm3m at 21(1) $^{\circ}C$ (a=12.100(2) ${\AA}$ for $Tl_{3.4}Zn_{4.3}-A$ and a=12.092(2) ${\AA}$ for $Tl_{5.5}Zn_{3.25}-A$). The crystals of $Tl_{3.4}Zn_{4.3}-A$ and $Tl_{5.5}Zn_{3.25}-A$ were prepared by flow method using exchange solutions in which mole ratios of $TlNO_3$,/TEX> and $Zn(NO_3)_2$ were 1:50 and 1:1, respectively, with total concentration of 0.05 M. The structures of the dehydrated $Tl_{3.4}Zn_{4.3}-A$ and $Tl_{5.5}Zn_{3.25}-A$ were refined to yield the final error indices $R_1$ = 0.075 and $R_2$ = 0.075 with 236 reflections, and $R_1$ = 0.057 and $R_2$ = 0.064 with 202 reflections, respectively, for which I > 3$\sigma$(I). Both structures indicate that Zn(II) ions are coordinated by three framework oxygens: the Zn(II) to O(3) distances are 2.08(1) ${\AA}$ for $Tl_{3.4}Zn_{4.3}-A$ and 2.07(1) ${\AA}$ for $Tl_{5.5}Zn_{3.25}-A$, respectively. In each structure, the angle subtended at Zn(II), O(3)-Zn(II)-O(3) is 119.9(3)$^{\circ}$ for $Tl_{3.4}Zn_{4.3}-A$, and 120.0(3)$^{\circ}$ for $Tl_{5.5}Zn_{3.25}-A$, respectively, close to the idealized trigonal-planar value. Zn(II) ions prefer to 6-ring sites. $Tl^+$ ions do not have any preference to a particular site but occupy simultaneously both at the 6-ring sites and 8-ring sites.

  • 가스크로마토그래피-질량분석기 (GC/MS)를 이용하여 뇨 중의 cholesterol, lanosterol, desmosterol 의 동시분석법을 개발하였다. 뇨 시료는 ${\beta}$-glucuronidase/arylsulfatase를 첨가하여 효소가수분해한 후 에틸아세테이트-헥산 (2:3, v/v)으로 추출하였으며 추출한 잔사를 MSTFA/TMSI/TMCS (100:2:5 v/v/v)로 유도체화한 후 GC/MS의 selective ion monitoring mode에서 분석하였다. 개발된 분석방법은 검량선에서 좋은 직선성 ($r^2=0.998{\sim}0.999$)과 회수율 (80.0%~113%)을 보였으며 5-200 ng/mL의 농도범위에서 15% 이내의 정확도와 정밀도를 보였다. 개발된 분석법은 고지혈증 유도 쥐에 pravastatin을 7일동안 70과 250 mg/kg/day로 경구투여한 후 채취한 뇨 시료를 분석하는데 적용하였다. 측정 결과 뇨 중 스테롤의 농도가 약물 투여한 쥐에서 대조군에서 보다 유의성 있게 낮아 약물효과를 관찰할 수 있었다. 이 결과로부터 개발된 방법을 뇨 중 스테롤의 농도를 정량함으로서 스타틴약물의 효과를 측정하는데 적용할 수 있음을 확인하였다.

비파괴 연속 생체중 측정장치의 개발 및 이에 의한 상추의 생장반응 분석 l. 양액의 이온 농도 및 명ㆍ암 처리가 생장에 미치는 영향 (Analysis of Growth Response by Non - destructive, Continuous Measurement of Fresh Weight in Leaf Lettuce 1. Effect of Nutrient Solution and Light Condition on the Growth of Leaf Lettuce)

  • 환경변화에 따른 작물생체의 반응을 근속하고 정확히 해석하기 위하여, 로드셀과 마이크로컴퓨터를 이용하여 수경조건에서 생체중을 비파괴적으로 연속측정할 수 있는 시스템을 개발 하였다. 또한 이 시스템을 이용하여 양액의 이온농도 및 명암 조건에 따른 상추의 생장반응을 조사 하였던 바, 그 결과를 요약하면 다음과 같다. 가. 작물의 생체중을 비파단 연속적으로 측정할 수 있는 이 장치의 측정오차는 측정범위 0-2000g에서 0.05%인 土1.0g 이하로 나타났다. 나. 로드셀에서 출력된 지시중량은 수면위의 생체중량과 잘 일치하였다. 다. 상추의 생체중은 정식후 18일에 정식 당시에 비해서 44배 정도 증가하였고, 상대생장율은 정식후 5일까지는 미약하였으나 이후에 급격히 높아져 정식후 13일에 최대에 달하여 29.6% day$^{-1}$ 이었다. 정식후 18일간의 상대생장율은 10.7-29.6% day$^{-1}$ 범위에 있었다. 라. 양액의 무기 이온 농도에 따른 상추의 생장량은 EC 1.4-2.2mS/cm 에서 가장 높았으며, EC 0.7 이하나 2.8 이상에서는 떨어졌는데, 특히 지하부의 생장량은 2.8 이상의 고농도에서 현저히 낮았다. 마. 양액의 이온 농도에 따른 생체중량의 증가속도는 정식후 7일까지는 큰 차이가 없었으나 이후 차이가 나기 시작하여 정식후 20일에는 양액의 이온농도에 따라 큰 차이가 있었다. 사. 암흑 처리후 12시간 까지는 정상적으로 생체중이 증가 하였으며, 이후 78시간까지도 완만한 증가 양상을 나타내었으나 그 이후에는 감소하는 경향이었다. 아. 명조건에서 암조건으로 환경이 변화함에 따라 생체중량은 급격히 증가하였고 반대의 조건에서는 일시적인 생체중 감소가 있었는데, 이와 같은 양상은 생육단계, 광조사 시각 및 시간을 달리하 여도 같은 경향이었다.음은 물론 식물안정화를 위해 도입되는 식물의 생육 개선에 효과가 있음을 확인할 수 있었다.서 기후인자와 인간영향을 어떻게 분리해 낼 것인지에 대한 문제점이 남는다. 예를 들어 화분분석의 경우 1차적인 환경변화에 따른 식생조성변화를 반영한다. 인간이 활발한 농경활동을 하게 되면서 주변식생을 제거하게 되고, 제거된 나대지에는 재배작물과 잡초들이 자라게 된다. 그리고 이들 화분은 주변 소택지에 퇴적물과 함께 퇴적되어 화분분석 결과 벼과(科)(Gramineae)와 문화지표수종이라 부르는 여뀌속(屬)(Persicaria), 국화과(科)(Composite), 쑥속(屬)(Artemisia), 쐐기풀속(屬)(Urtica) 등의 화분에 있어 확연한 조성변화를 보여준다. 그러나 화분을 이용한 분석은 토탄층이나 유기질 퇴적물이라는 한정된 토양에서만 분석이 가능하며, 조성변화에 있어서 역시 기후와 인간활동의 영향 모두를 반영하기 때문에 이를 구분해 주는 명확한 기준이 필요하다. 따라서 본 연구에서는 이러한 다양한 대리자료 중 고고학 유적지에 적용 가능한 동식물 화석 및 미화석, 지리, 지질학적 자료에 어떠한 것들을 적용할 수 있는지를 살피고, 몇 가지 대리자료를 중심으로 선사인의 농경활동에 대해 살펴보고자 한다.avascular injection of the iodinated contrast materials.석 시기중 성공적인 착상성공율은 56%(71/126)였다..8H2O를 사용함으로써 host-guest적인 반응을 유도시키는데 있어 물의 가장 실용적이고 효과적인 수열용매임도 알았다.
