섬유 폐수 활성 슬러지에서 분리한 Comamonas testosteroni의 생물학적 페놀 분해 (Biodegradation of Phenol by Comamonas testosteroni DWB-1-8 Isolated from the Activated Sludge of Textile Wastewater)

  • 산업화 이후 다양한 화학물질의 생산과 이용은 우리 삶의 질을 높이는데 기여하였지만 그로 인한 대량의 폐기물 방출은 필연적이며 환경오염은 날이 갈수록 심각해지고 있다. 환경오염에 의한 화학물질의 노출은 인간의 건강과 생태계에 악영향을 주고 있다. 우리의 삶에 영향을 줄 수 있는 오염된 환경을 정화하는 것은 매우 중요한 문제이다. 광범위한 석유화학제품의 사용으로 인해 독성 난분해성 방향족화합물이 토양, 지하수, 폐수 등에서 빈번하게 검출되고 있다. 특히 합성수지, 합성섬유, 염료, 살충제, 방부제 등의 원료로 사용되는 페놀은 주요 오염물질이며, 호흡곤란, 두통, 구토, 돌연변이, 발암 등을 일으킬 수 있다. 본 연구에서는 페놀분해균주인 DWB-1-8을 섬유폐수에서 분리, 동정하였으며 16S rRNA유전자 염기서열분석결과 Comamonas testosteroni로 동정 되었다. 본 실험 균주의 최적 생장 및 최적페놀분해 배양 조건은 0.7% K2HPO4, 0.6% NaH2PO4, 0.1% NH4NO3, 0.015% MgSO4·7H2O, 0.001% FeSO4·7H2O, 초기 pH 7 및 30℃ 였으며, 다른 독성 화합물인 벤젠, 톨루엔 혹은 크실렌(BTX)을 유일탄소원으로 이용하여 생장할 수 있었다.

사염화탄소로 섬유화가 유도된 흰쥐 간에서 털부처꽃 뿌리 추출물의 항산화 및 섬유화저해 활성 (Antioxidant and Anti-fibrotic Properties of Root Extract of Lythrumsalicaria L. in $CCL_4$-Induced Liver Fibrosis Rat Model)

  • Fifty percent ethanol extract of Lythrum salicaria Linne root (LSR) was tested in vitro on antioxidant activity, and furthermore was investigated on antioxidative and fibrosis protecting activities in $CCL_4$-induced liver fibrosis rat model. Ratio of hepatic GSH/GSSG (reduced glutathione/oxidized glutathione) as bio-parameter of antioxidant level in $CCL_4$ plus LSR-treated rats for 6 weeks significantly increased from 2.8- to 5.7-fold than that of $CCL_4$-treated rats at p < 0.05. Thiobarbituric acid reactive substances (TBARS) contents in $CCL_4$ plus LSR-treated rats ranged from 1.57- to 2.19-fold of normal rats and were lower than those in $CCL_4$ plus silymarin-treated rats ($1.78{\sim}2.46$-fold of normal rats) (p < 0.05). Amounts of hydroxyproline of liver tissue showing the content of total collagen, a parameter of fibrosis, in $CCL_4$ plus LSR-administrated rat livers were $4.9{\sim}8.8{\mu}g$/mg ($-47{\sim}-71%$, compared with that in $CCL_4$-treated rat livers ($16.6{\mu}g$/mg tissue), which were significantly lower than those in $CCL_4$ plus silymarin-administrated rats being $8.4{\sim}11.7{\mu}g$/mg ($-30{\sim}-50%$). This collagen reducing effect of liver tissue in $CCL_4$ plus LSR-treated rats was supported by histological observation using microscopy assay. From the results, we conclude that the root of L. salicaria have efficient antioxidant potential and effective antifibrotic activities.

Physostigmine과 procyclidine으로 구성된 복합예방제의 유기인제 해독효능 (Efficacy of Physostigmine and Procyclidine as Combinational Prophylactics of Organophosphate Poisoning)

  • Antidotal efficacy of combinational prophylactics composed of physostigmine plus procyclidine, alone or in combination with antidotes such as atropine plus 2-pralidoxime or atropine plus HI-6, was evaluated in rats. Physostigmine (0.1 mg/kg) plus procyclidine (3 mg/kg), pretreated subcutaneously 30 min prior to subcutaneous exposure to organophosphates of militarily importance, exerted protection ratios of 7.2, 6.5, 4.0, 2.9 and 8.0 fold for tabun, saris, soman, cyclosarin and V-agent, respectively. In comparison, low effects (1.7 fold for soman and 1.3 fold for cyclosarin) were achieved with the traditional antidotes atropine (17.4 mg/kg) plus 2-pralidoxime (30 mg/kg) administered intramuscularly immediately after organophosphate, in contrast to high effects (5.5 fold for soman and 160.0 fold for cyclosarin) with atropine (17.4 mg/kg) plus HI-6 (125 mg/kg), although the protection ratio markedly decreased when treatment of antidotes was delayed. Note- worthy, the combinational prophylactics markedly potentiated the effects of antidotes to higher than 5.0 fold in all cases. In addition, the combinational prophylactics fully prevented the seizures and excitotoxic brain injuries induced by a high dose (100 mg/kg, 1.3 LD$_{50}$) of soman. Taken together, it is suggested that the prophylactics composed of physostigmine and procyclidine, in combination with posttreatment antidotes, could be a promising regimen for the prevention of lethality, seizures and brain injuries induced by organophosphates possessing diverse properties.s.

밤 발효사료 제조과정에서 미생물 첨가수준이 영양소 함량과 반추위 내 발효특성에 미치는 영향 (Effects of Inoculant Application Level on Chemical Compositions of Fermented Chestnut Meal and Its Rumen Fermentation Indices)

  • This study aimed to estimate the effect of inoculant application level on chemical composition and bacterial count of fermented chestnut meal (FCM), and its rumen fermentation characteristics. The inoculant contained Lactobacillus acidophilus ($1.2{\times}10^{10}cfu/g$), Bacillus subtilis ($2.1{\times}10^{10}cfu/g$), and Saccharomyces cerevisiae ($2.3{\times}10^{10}cfu/g$). The chestnut meal mixed with molasses, double distilled water, and inoculant at 1 kg, 3 g, 480 mL, and 20 mL ratio for the basal chestnut meal diet. The double distilled water from basal chestnut meal diet was substituted with bacterial inoculant at a level of 0 (Control), 20 (Medium), and 40 mL (High) in the experimental diets. The mixed experimental diets were incubated at $39^{\circ}C$ for 7, 14, and 21 days, respectively. On 7 days of FCM incubation, the contents of crude protein (CP) (quadratic, P=0.043) and neutral detergent fiber (quadratic, P=0.071) decreased by increases of inoculant application levels, whereas bacterial count (quadratic, P=0.065) and rumen $NH_3-N$ (linear, P=0.063) increased. By increases of inoculant application levels on 14 days of FCM incubation, the increases were found on dry matter (DM) (quadratic, P=0.085), CP (quadratic, P=0.059), acid detergent fiber (quadratic, P=0.056), in vitro DM digestibility (linear, P=0.002), rumen total volatile fatty acid (VFA) (linear, P=0.057), and rumen iso-butyrate (linear, P=0.054). However, the decreases were found on bacterial count (linear, P=0.002), propionate (linear, P=0.099), and butyrate (quadratic, P=0.082). On 21 days of FCM incubation, in vitro DM digestibility (linear, P=0.002) and total VFA (linear, P=0.001) increased by increases of inoculant application levels, whereas the contents of CP (quadratic, P=0.034) and neutral detergent fiber (quadratic, P=0.047) decreased. These results indicate that the FCM with a medium level of inoculant application and 14 of fermentation had beneficial effects by increasing DM digestibility and rumen total VFA content, without altering bacterial count.

  • 시간지연이 존재하는 시스템의 제어기 설계에 많이 사용되는 Smith Predictor는 플랜트와 모델이 완전히 일치해야만 특성방정식에서 시간지연을 제거 할 수 있기 때문에 실제 공정에서는 적용하기가 매우 어렵다. 본 논문에서는 Smith Predictor의 모델을 플랜트와 일치한 식이 아닌 시간지연을 포함한 2차식 형태로 제안하여 큰 시간지연에서의 응답특성을 향상 시켰다. PI 제어기의 적분상수의 범위를 제안하였다.

근관세척액의 항균효과에 관한 연구 (A STUDY ON THE ANTIMICROBIAL EFFECT OF IRRIGATION SOLUTIONS)

  • The purpose of this study was to investigate antimicrobial effect of several irrigation solutions on 7 anaerobes and 2 aerobes, which are found frequently in infected root canals. The antimicrobial effects of normal saline, 3% $H_2O_2$, 0.5% & 3.5% NaOCl, 10% & 50% citric acid and mixed solutions of 3% $H_2O_2$ plus 3.5% NaOCl were compared. No. 80 paper points dipped in bacterial broth were soaked in each irrigation solutions and moved into thioglycolate broth, subcultured in agar plate for bacterial growth. The results were as follows: 1. Normal saline had no antimicrobial effect. 2. Mixed solutions of 3% $H_2O_2$ plus 3.5% NaOCl, 10% citric acid had relatively weak antimicrobial effect. 3.3% $H_2O_2$, 50% citric acid, 0.5% NaOCl showed relatively strong antimicrobial effect. 4. 3.5% NaOCl had the strongest antimicrobial effect among used 7 irrigation solutions.

Dicor와 수종 전장도재간의 전단결합강도 (THE SHEAR BOND STRENGTH BETWEEN DICOR AND SEVERAL VENEERING PORCELAINS)

  • Dicor has not been prescribed routinely, in spite of many advantages, because of esthetic limitations by excessive translucency and external shading. In an attempt to solve these problems, the technique of veneering Dicor by aluminous poreclain has been used and recently Dicor Plus system was developed. The purpose of this study was to evaluate the compatibility between Dicor and several veneering porcelains by measuring the shear bond strength and observing the failure mode and interface appearance with SEM. Total 55 Dicor disks(10.0mm diam. X 3.0mm thickness) were fabricated by lost wax technique and divided into five groups of 11. Veneering porcelains such as Dicor Plus, Vitadur Alpha, Vitadur N, Vivodent, and Ceramco II were built up over the center of the treated Dicor surface using paper tube(5.0mm diam. X 4.0mm height) and fired according to the manufacturesr’instructions. A representative sample from each group was completely embedded in epoxy resin and crosssectioned, and remaining 50 samples were embedded in epoxy resin with the bonded area perpendicular to table base. The shear bond strengths were measured by applying the shear load parallel to Dicor surface close to the bonded area. Failure modes and interface appearances were observed using SEM at 15 and 1000 magnification respectively. The obtained results were as follows : 1. The mean shear bond strengths showed Dicor-Dicor Plus(10.53 MPa); Dicor-Vitadur Alpha(8.84 MPa); Dicor-Vitadur N(7.37 MPa); Dicor-Vivodent(4.28 MPa); Dicor-Ceramco II(0.89 MPa). 2. The shear bond strength of Dicor-CeramcoII was significantly decreased compared with Dicor-Dicor Plus(p<0.01), but had no significant difference compared with Dicor-Vivodent(p>0.01). 3. The shear bond strengths of Dicor-Vitadur Alpha and Dicor-Vitadur N were not significantly different compared with Dicor-Dicor Plus(p>0.01). 4. SEM examination of bond failure modes revealed that Dicor-Dicor plus, Dicor-Vitadur Alpha, Dicor-Vitadur N exhibited cohesive failure within Dicor and Dicor-Vivodent exhibited adhesive failure. And Dicor-Ceramco III exhibited adhesive failure and cohesive failure within CeramcoIII together. 5. SEM examination of interfaces revealed that Dicor-Dicor Plus exhibited the most tight contact and Dicor-Vitadur Alpha, Dicor-Vitadur N exhibited acceptible contacts. But Vivodent exhibited discontinuous gap and Ceramco II exhibited large continuous gap.

Trimethyltin 유도성 인지기능 저하 동물 모델에 대한 들기름의 개선효과 (Amelioration of Trimethyltin-induced Cognitive Impairment in ICR Mice by Perilla Oil)

  • 본 연구에서는 들기름의 TMT 유도성 인지 기능 상실에 대한 개선 효과와 함께 가정에서 식용유로서 사용량이 많은 대두유와 그 효과를 비교 연구하였다. 실험에서 들기름과 대두유를 섭취한 마우스를 TMT로 인지 기능 손상을 유발하여 Y-maze test와 Morris water maze test 한 결과, 공간 인지 기능 및 학습능력 개선에 대해 대두유는 효과가 미비하였으나 들기름은 대조군과 유의적인 차이가 없는 정도의 유의적 개선 효과를 보였다. 동물 실험 후 mouse로부터 적출된 뇌 조직을 대상으로 AChE 활성, MDA 함량, SOD 활성 측정 및 산화된 glutathione 측정한 결과, 대두유는 TMT 단독 처리군의 경우와 유사한 반면 들기름은 TMT에 의해 손상을 입은 mice의 뇌 조직에서 AChE의 활성과 MDA 생성 및 GSH의 산화를 억제시킬 뿐만 아니라 SOD 활성을 유의성 있게 증가시킨 것으로 나타났다. 이러한 결과는 들기름에 존재하는 풍부한 생리 활성 물질로서 ${\omega}$-3계 지방산, 페놀화합물 그리고 비타민 E 등에 의한 것으로 사료된다. 결국 들기름은 상대적으로 우수한 인지 기능 개선 효과를 나타냈고, 이는 건강기능 식품으로서 고부가가치 소재로 활용될 수 있는 산업적 활용 가능성을 나타내는 것으로 판단된다.

Influence of Phytase and Xylanase Supplementation on Growth Performance and Nutrient Utilisation of Broilers Offered Wheat-based Diets

  • Individual and combined supplementation of phosphorus-adequate, wheat-based broiler diets with exogenous phytase and xylanase was evaluated in three experiments. The effects of the enzyme combination in lysine-eficient diets containing wheat and sorghum were more pronounced than those of the individual feed enzymes. The inclusion of phytase plus xylanase improved (p<0.05) weight gains (7.3%) and feed efficiency (7.0%) of broilers (7-28 days post-hatch) and apparent metabolisable energy (AME) by 0.76 MJ/kg DM. Phytase plus xylanase increased (p<0.05) the overall, apparent ileal digestibility of amino acids by 4.5% (0.781 to 0.816); this was greater than the responses to either phytase (3.6%; 0.781 to 0.809) or xylanase (0.7%; 0.781 to 0.784). Absolute increases in amino acid digestibility with the combination exceeded the sum of the individual increases generated by phytase and xylanase for alanine, aspartic acid, glutamic acid, glycine, histidine, isoleucine, phenylalanine, threonine, tyrosine and valine. These synergistic responses may have resulted from phytase and xylanase having complementary modes of action for enhancing amino acid digestibilities and/or facilitating substrate access. The two remaining experiments were almost identical except wheat used in Experiment 2 had a higher phytate concentration and a lower estimated AME content than wheat used in Experiment 3. Individually, phytase and xylanase were generally more effective in Experiment 2, which probably reflects the higher dietary substrate levels present. Phytase plus xylanase increased (p<0.05) gains (15.4%) and feed efficiency (7.0%) of broiler chicks from 4-24 days post-hatch in Experiment 2; whereas, in Experiment 3, the combination increased (p<0.05) growth to a lesser extent (5.6%) and had no effect on feed efficiency. This difference in performance responses appeared to be 'rotein driven'as the combination increased (p<0.05) nitrogen retention in Experiment 2 but not in Experiment 3; whereas phytase plus xylanase significantly increased AME in both experiments. In Experiments 2 and 3 the combined inclusion levels of phytase and xylanase were lower that the individual additions, which demonstrates the benefits of simultaneously including phytase and xylanase in wheat-based poultry diets.

  • 등량(等量)의 사료(飼料)로서 급식회수(給食回數)를 달리 했을때 면양(緬羊)의 증체(增體), 영양소(營養素)의 소화율(消化率), 열량이용성(熱量利用性), 체조직(體組織)의 화학성분(化學成分)에 미치는 영향(影響)을 연구(硏究)하기 위하여 32두(頭)의 면양(緬羊)으로 두개(個)의 실험(實驗)을 하였든바 그 성적(成績)은 다음과 같다. (1) 영양소(營養素)의 소화율(消化率) 및 ME가(價)에 있어서 1 일(日) 1회(回) 8급식구(給食區), 1 일(日) 8회급식구(回給食區), 1 일(日) 1회급식(回給食)+7회첨수구(回添水區)사이에 하등(何等)의 차이(差異)가 발견(發見)되지 않았다. (2) 매대사체중당(每代射體重當) 열발생량(熱發生量)은 1 일(日) 8회구(回區)가 1회구(回區)보다 훨씬 적었다. (3) 1 일(日) 8회구(回區)보다 증체량(增體量)이 더 많았다. (4) 1 일(日) 8회구(回區)의 경우 체단백질(體蛋白質), 지방(脂肪), 에너지의 증가(增加)가 1회구(回區)보다 훨씬 왕성하였다. (5) 증체(增體)된 부분(部分)의 체조성(體組成) 변화(變化)를 보면 1회구(回區)의 경우에 훨씬 많은 량(量)의 지방(脂肪), 단백질(蛋白質), 광물질(鑛物質)이 비축되었다. (6) 급식회수(給食回數)가 위에서 본바와 같은 영향을 일으키는 생리적기전(生理的機轉)의 일부(一部)가 설명(說明)되었다.
