백작약 추출물이 항산화활성, LDL 산화 억제 및 혈전용해에 미치는 영향 (Preventive Effects of Peony Root Extracts on Oxidative Stress, Thrombosis and Atherosclerosis)

  • Objectives: There is currently increased interest in the identification of natural antioxidant compounds derived from various plants. Peony Root (PR) is used worldwide for the treatment of many types of cardiovascular disease including atherosclerosis and hypertension. It has been used in Korean traditional medicine for the treatment of glycosuria, hypertension and cancer. However, to date, no studies concerning the antioxidant properties of PR have been conducted. Therefore, this study was conducted to evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species and anti-thrombosis effect in response to treatment with PR using various screening methods including biological and non-biological oxidants. Methods: In this study, the antioxidant activity of extract from PR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on collagen-induced platelet aggregation. Results: The PR extracts were found to have a potent scavenging activity of oxidative stress [DPPH, superoxide anion, hydroxyl radical, nitric oxide and peroxynitrite, etc.] as well as an inhibitory effect on LDL oxidation and on platelet aggregation. Conclusions: The PR extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.

Antioxidant & Antibacterial effects of Artichoke (Cynara Scolymus L.) leaf by various extract solvents

  • Antioxidant is an important role to protect the human body against damage by reactive oxygen species. However, the excessive intake of such antioxidant is known to cause a serious poisonous influence on one's liver, lungs and circulating system. Therefore, it is necessary to develop a safe natural antioxidant. For the purpose of developing natural antioxidant and antibacterial, the antioxidant activity and antibacterial effects of various extract solvents from Artichoke (Cynara Scolymus L.) leaf were determined. In this study, the extracts of Artichoke leaf dried from solvent extraction were examined by means of DPPH free radical scavenging activity and ABTS free radical scavenging activity. The effect of free radical scavenging compared with $\alpha$-tocopherol and L-ascorbic acid. In Artichoke leaf extract, evaluated by using DPPH and ABTS showed that the highest antioxidant activities were found to be in methanol extracts from DPPH radical ($IC_{50}$: $20.06{\mu}g\;mL^{-1}$), ABTS radical ($IC_{50}$: $16.01{\mu}g\;mL^{-1}$) and followed by ethanol > methyl chloride > ethyl acetate > n-Hexane. By using disc diffusion method, the antibacterial activity showed that the Artichoke leaf extract was found to be most effective against all of the tested organisms and the methyl chloride extract showed the most significant antibacterial effect against all of tests among 5 solvents extract, followed by ethyl acetate > n-Hexane > ethanol > methanol. As a result, optimal in antioxidant activity for Artichoke (Cynara Scolymus L.) leaf is methanol extract and for antibacterial effect is Methyl Chloride extract.

Hepatoma 세포주에서 H2O2 처리에 의한 Cu/Zn SOD의 발현 (Expression of Cu/Zn SOD according to H2O2 in Hepatoma cell line)

  • 생체는 산소를 소비하는 대사 과정 중에 초산화물(superoxide, $O_{2}$), 과산화수소($H_2O_2$), 수산 라디칼(OH)과 같은 다양한 활성산소(reactive oxygen)들을 생성하게 되며, 그 중에서도 hydrogen peroxide ($H_2O_2$)는 biological membrane을 자유롭게 통과하며, 세포내에서 hydroxyl radical 등의 반응성이 큰 활성 산소종(reactive oxygen species, ROS)을 발생시키는 작용을 하는 강력한 산화제이다. 세포를 계대 배양 (5, 15, 25, 35 passage)하여 $H_2O_2$를 농도별(100 ${\mu}M$, 500 ${\mu}M$, 1 mM, 5 mM)로 처리하고, 또한 $H_2O_2$의 처리 시간(30 분, 1 시간)을 변화시킴으로써, Hepatoma 세포주에서 $H_2O_2$ 처리에 의한 Cu/Zn SOD의 발현을 Northern blot을 통하여 다음과 같이 분석하였다. 1)Hepatoma 세포주에서 시간별, 농도별로 산화제를 처리 했을 때 각각의 경우에서 발현양의 차이는 적었지만, 오랜 시간동안 고농도의 산화제에 노출시켰을 때 항산화 능력이 증가한다는 것을 확인할 수 있었다. 2)계대배양을 증가시키는 것은 노화가 진행된다는 것을 의미하므로, 산화제를 처리했을 때 25 passage에서 35 passage 단계에서 항산화 효소의 발현 정도가 급격히 감소되는 것으로 미루어 보아 이 단계에서 노화가 진행되었음을 추측할 수 있었다. 3)동일한 시간과 농도로 처리했을 때 각각의 passage의 발현 level에서 보이는 양상과는 다르게 35 passage에서는 500${\mu}M$이상의 농도를 1 시간동안 노출시켰을 경우에 Cu/Zn SOD가 거의 발현되지 않았으며, 30 분 동안 노출시켰을 때에는 500 ${\mu}M$의 농도까지 방어할 수 있는 능력을 가진 것으로 보인다.

금불초 꽃 추출물의 항균 및 항산화 활성 (Antibacterial and Antioxidative Activities of Inula britannica Flower Extract)

  • 본 연구에서는 금불초 꽃 추출물의 피부 상재균에 대한 항균작용과 항산화, tyroinase 저해 효과에 관한 조사를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, P. acnes에 대한 ethyl acetate 분획의 MIC는 0.25 %로 나타났으며, methyl paraben과 비슷한 활성을 나타냈고 quercetin보다는 큰 항균활성을 나타내었다. 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 ethyl acetate 분획에서 $8.55{\mu}g$/mL로 측정되었다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 금불초 꽃 추출물의 총 항산화능은 ethyl acetate 분획 $0.24{\mu}g$/mL로, 50 % EtOH extract와 aglycone 분획 보다 ethyl acetate 분획에서 가장 큰 활성을 나타내었다. 금불초 꽃 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였다. 금불초 꽃 추출물의 경우 $5{\sim}100{\mu}g$/mL의 농도에서 광용혈을 억제하였다. 특히 당을 제거시킨 플라보노이드 aglycone 분획은 $25{\mu}g$/mL 농도에서 $\tau_{50}$이 164.15 min으로 매우 큰 세포보호 효과를 나타내었다. 미백 효과측정으로는 각각 tyrosinase의 활성 저해 효과를 측정하였다. Tyrosinase의 활성 저해 효과($IC_{50}$)는 ethyl acetate 분획에서 $87.03{\mu}g$/mL 로 나타났다. 이상의 결과들은 금불초 꽃 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, 금불초 꽃 성분 분석, ethyl acetate 분획의 tyrosinase 저해활성 그리고 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성을 확인하였다.

레스베라트롤의 세포보호 및 항산화 효과 (Cellular Protective Effects and Antioxidative Activity of Resveratrol)

  • 본 연구에서는, 레스베라트롤의 산화적 손상대한 세포 보호 효과와 항산화능을 조사하였다. 레스베라트롤의 Free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성은 ($FSC_{50}$) $103{\mu}M$로 나타났다. 루미놀화학발광법을 이용하여 $Fe^{3+}-EDTA/H_2O_2$계에서 레스베라트롤의 활성산소(Reactive oxygen species, ROS) 소거활성은($OSC_{50}$)을 평가하였다. 레스베라트롤은 $0.042{\mu}M$로 나타나 비교물질로 사용한 L-ascorbic acid ($0.405{\mu}M$)보다 9.6배 우수한 활성산소 소거활성을 보였다. 또한 레스베라트롤은 지용성 항산화제로 알려진 (+)-${\alpha}$-tocopherol보다 우수한 세포보호 활성을 나타내었다. 세포독성 조사에서 레스베라트롤은 $50{\mu}M$에서 세포독성을 나타내지 않았다. HaCaT 세포에 $800mJ/cm^2$의 UVB 또는 $30{\mu}M$의 rose-bengal을 처리하였을 때, 레스베라트롤은 농도의존적으로 세포의 산화적 손상에 대항하여 세포를 보호하였다. 그러나 10 mM의 $H_2O_2$를 처리한 경우에, 레스베라트롤은 세포보호 효과를 나타내지 않았다. 이들의 결과들은 레스베라트롤이 항산화능 및 세포보호 효과를 통해서 피부 노화를 예방하고 개선하는데 있어 이용 가능함을 시사한다.

여뀌 추출물의 항산화 및 항노화에 관한 연구 (Antioxidative and Antiaging Effects of Persicaria hydropiper L. Extracts)

  • 본 연구에서는 여뀌 추출물의 항산화, 미백, 항주름 효과를 통한 항노화 활성을 조사하였다. 여뀌 추출물의 free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 분획물 중 ethyl acetate 분획에서 $5.23\;{\mu}g/mL$로 가장 큰 활성을 나타내었다. Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 여뀌 추출물의 총 항산화능은 분획물 중 ethyl acetate 분획($0.40\;{\mu}g/mL$)이 큰 활성을 나타내었다. 여뀌 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였다. 여뀌 추출물의 경우 $1\;{\sim}\;10\;{\mu}g/mL$의 농도에서 광용혈을 억제하였다. 미백 및 주름억제 효과측정으로는 각각 tyrosinase 및 elastase의 활성 저해 효과를 측정하였다. Tyrosinase의 활성 저해 효과($IC_{50}$)는 aglycone 분획($8.90\;{\mu}g/mL$) 에서, elastase의 활성 저해 효과($IC_{50}$)도 aglycone 분획($2.37\;{\mu}g/mL$)에서 가장 큰 저해활성을 나타났다. 여뀌 추출물중 ethyl acetate 분획과 aglycone 분획에 대한 실험 결과로부터 항산화, 항노화 화장품 원료로서의 가능성을 확인하였다.

Cytoprotective Constituents of Alpinia katsumadai Seeds Against Glutamate-Induced Oxidative Injury in HT22 Cells

  • Glutamate-induced oxidative injury contributes to neuronal degeneration in many central nervous system (CNS) diseases, such as epilepsy and ischemia. Bioassay-guided fractionation of the MeOH extract of the seeds of Alpinia katsumadai Hayata (Zingiberaceae) furnished three phenolic compounds, alpinetin (1), pinocembrin (2), and (+)-catechin (3). Compounds 2 and 3 showed the potent neuroprotective effects on glutamate-induced neurotoxicity and reactive oxygen species (ROS) generation in the mouse hippocampal HT22 cells. In addition, Compounds 2 and 3 showed significant DPPH free radical scavenging effect. These results suggest that compounds 2 and 3 could be the effective candidates for the treatment of ROS-related neurological diseases.

Biological Activities and Partial Characterization of Beauveria bassiana Mycelium

  • Some biological activities of Beauveria bassiana were studied to elucidate pharmacological function of B. bassiana-infected larva of the silkworm. The mycelium consisted mainly of carbohydrate (65.8%), followed by protein (15.9%) and fat (8.3%). Glucose (68.8%), mannose (7.1%), and galactose (6.1%) were major components in carbohydrates. Ten amino acids including glutamine, threonine, valine, aspartic acid, alanine, leucine, serine, glycine, arginine, and isoleucine were found in protein as major amino acids. Various extracts were prepared from the freeze-dried mycelium of B. bassiana by systemic extraction and their biological activities were investigated. Among tested fractions, the hot-water extract (HW) contributed significantly to the anti-coagulant activity, anti-complementary activity, and stimulation of intestinal immune system. The methanol extract (ME) increased acetylcholinesterase (AChE) inhibition activity and reactive oxygen species (ROS) scavenging activity.
