Comparison of Relative Antioxidant Capacity and Antiperoxidation Activity of Traditional Medicines in Vitro

  • To investigate the antioxidant capacity of traditional Korean medicines, water extracts from 57 species were tested on their antioxidant activity using radical scavenging effects against ABTSㆍ+. Of which, Rhus javanica, Caesalpinia sappan, Rosa rugosa, Spatholobus suberectus, and Magnolia obovata showed strong antioxidant capacities at 10㎍ concentration. Therefore, antioxidant capacities of 5 traditional medicine extracts in the different concentration (1㎍, 5㎍, and 10㎍) were determined. The 5 traditional medicine extracts was detected in antioxidant capacity dose dependently. R. javanica was showed the highest antioxidant capacity, the antioxidant activity at 1 ㎍ of herbal extract being 0.85 mM TE. At the same time, the antiperoxidation effects of these 5 medicines were determined. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by S, suberectus, R. rugosa, and M. obovata. Extract of C. sappan was showed the highest inhibition against lipid peroxidation compared with 4 herbal extracts in vitro. These traditional medicines are a potent antioxidant capacity and antiperoxidation activity, further investigation into the in vivo antioxidant therapeutic potential for treatment of human disorders such as Alzheimer's disease.

항염증 효능을 가진 단미 한약의 2001년 이후 실험 연구 결과에 대한 고찰 (A Review of Experimental Study for Herbal Simplexes of Anti-inflammatory Effects Since 2001)

  • Objective : The purpose of this study is to survey the experimental studies for herbal simplexes of Anti-inflammatory Effects by reviewing oriental medicine journals since 2001. Methods : We collected 65 research papers on Oriental herbal simplexes medicines (herbs and Pharmacopunctures) of Anti-inflammatory effects in the internet site OASIS using the keyword 'Anti-inflammation'. Then we analyzed them according to published journals, years, and research methods, etc. Results : 65 papers in relation to Oriental herbal medicines of anti-inflammatory effects have been published in many kinds of journals and shows a tendency to increase recently. There are 60 cases of using herbal simplexes and 5 cases of Pharmacopuncture. Conclusions : This result will provide useful information of field of Oriental herbal medicine for the development and clinical use of anti-inflammatory related herb simplexes. In the future, we have to perform more studies such as clinical trial and research papers on developed design and herb-combined prescription study for anti-inflammatory effects.

한약 정보 표준화를 위한 의약품 식별 표준 (IDMP) 분석 및 고찰 (Review of Identification of Medicinal Products (IDMP) Standards for Standardization of Herbal Medicine Information)

  • Objectives : The purpose of this study was to apply informations related to herbal medicines to IDMP (Identification of Medicinal Products), an ISO standards related to medicinal products substances, for systematic collection of data through the integration of informations on distribution, manufacturing, and management of herbal medicines. Methods : By analyzing ISO 11238 and ISO/TS 19844, elements that can be used in the information model of herbal medicine were derived from the identification of medicinal products information model on substances. The labeling specified in the safety and quality control regulations for herbal medicines was mapped to the IDMP information model, and ginseng was applied as an example. Results : Herbal medicine corresponded to substance in IDMP. Among the five types of substances specified by IDMP, herbal medicines were expressed as structurally diverse. Scientific name was used as an invariant property of herbal medicine, and the substance level included information about source material and modification, and specifically included information about the scientific name, medicinal part, fraction, and processing. In addition, the specified substance level had information on the constituents, characteristic attributes, manufacturing, and grade of the herbal medicine. Conclusions : It is necessary to establish a code system for identifying herbal medicines. In order to apply the IDMP standards, research on the development of standard terms is required to express the characteristics of herbal medicines. In addition, information for identification of herbal medicines is also required, and information from production to consumption should be systematically accumulated and managed for actual application.

한약의 독물동태학적 특성 (The Toxicokinetic Characteristics of Korean Traditional Medicines)

  • Toxicokinetics of Korean Traditional Medicines(TKM) is the description of what rate TKM will enter the body and what happens to it once it is in the body in terms of toxicology. However, it is not easy to understand TKM toxicokinetics because of various factors such as a mixture of 2-30 kinds of herbal materials containing thousands of chemicals, and complex chemical properties. For these reasons, little is known about toxicokinetics of TKM. This study was aimed to characterize and review the absorption, distribution and metabolism of korean traditional medicines in a view of toxicokinetics. For this aim, some of korean traditional medicines were reviewed on a basis of drug-drug interaction, biotransformation and intestinal metabolisms by bacteria. As the factors affecting mainly on toxicokinetics of TKM, individual herbal material's degree of lipophilicity and metabolic rate, and decoction components according to different kinds of herbal materials were considered. Other factors such as intestinal pH and bacterial activity for metabolism affecting on TKM toxicokinetics, especially in small intestine. It would be a better way for improving the adverse or poor effects caused by TCM if the factors affecting on toxicokinetics of TKM is considered.

한약재 추출물에 대한 전혈응집억제 효능 검색 (Effect of Extracts for Herbal Medicines on the Inhibition of Whole Blood Aggregation)

  • 고미의 성질을 강하게 나타내는 한약재를 대상으로 맛과 효능과의 상관관계를 실험적으로 알아보기 위한 연구를 시도하였다. 본 연구에서는 콜라겐으로 유도된 인간전혈응집을 이용하여 고미약재 20품목에 대한 혈소판응집 억제능을 측정하였다. 이 중에 3종의 열수추출물과 2종의 70% 에탄올추출물이 p<0.001 수준에서 유의하게 전혈응집 억제 활성을 나타내었다. 특히, 전혈응집 억제능이 가장 뛰어난 후보소재로 황련이 선별되었으며 관능요원에 의한 관능평가에서 쓴맛이 가장 강한 한약재로 평가되었다. 이상의 실험결과는 전통의학이론을 바탕으로 한약재의 사용에 따른 효능을 실험적 근거로 제시할 수 있고 쓴맛이 약한 한약재보다 쓴맛이 강한 한약재가 혈액순환이 원활하게 하는 가능성의 경향이 있는 것으로 기대되었다.

황흑산(黃黑散)의 메티실린 내성 황색포도상구균에 대한 항균 활성과 옥사실린과의 시너지 효과 (The antimicrobial activity of Hwangheuk-san and synergy effect with oxacillin against methicillin-resistant Staphylococcus aureus)

  • Objectives : Hwangheuk-san is a complex prescription composed of oriental traditional medicine and has been reported for antioxidant, antimicrobial and anticancer effects in the recent study. Methicillin-resistant Staphylococcus aureus (MRSA) is one of important causes of fatal infectious diseases such as septicemia, endocarditis, toxic shock syndrome, pneumonia, skin and soft tissue infections (SSTIs). S. aureus is reported as being for a variety of human diseases and its epidemiological relevance is mainly due to their ability of becoming highly resistant to common antimicrobials such as tetracycline, penicillin, cphalosporin and aminoglycoside. The objective of this study is to determine the antimicrobial effect of Hwangheuk-san ethanol extracts (HHS) and synergistic effects with antibiotics oxacillin against MRSA.Methods : The antimicrobial activity of HHS was measured by the disc diffusion method, broth microdilution method and the checkerboard dilution test, time-kill curve assay was performed to investigate synergistic effects with antibiotics oxacillin against MRSA.Results : HHS showed antimicrobial activity against MRSA with a MIC value of 125 ㎍/㎖. In the checkerboard test, the interaction of HHS with antibiotics oxacillin produced almost synergy or partial synergy against MRSA. This study showed that HHS reduced the MICs of oxacillin tested, and a remarkable antibacterial effect of HHS, with membrane permeability enhancers.Conclusions : These results suggest that HHS has the antimicrobial effect and synergistic effects with antibiotics oxacillin against MRSA. This study thus can be a valuable source for the development of a new drug with low MRSA resistance.

부자 및 천오를 포함한 한약이 간 손상 수치에 미치는 영향 (The Consequences of Long-period Dosages of Herbal Medicine Including Boo-ja and Cheon-o on Liver Injury)

  • Objectives : Boo-ja and Cheon-o are not commonly prescribed, but are necessary for some clinical conditions, dispite the fact that some negative effects have been known to occur with these medicines. Hence, consequences for aspartate aminotransferase(AST), alanine aminotransferase(ALT) due to herbal medicines, including boo-ja and cheon-o are here reported. Methods : From January 2005 to July 2005, results were analyzed for 36 inpatients belonging to the sixth internal medicine department of Dongeui Oriental Medical Hospital who took herbal medicine, including boo-ja and cheon-o, over 20 days. The study is about the comparison and the investigation of LFT, and all results were taken upon their hospitalization and upon their departure. The standard of liver injury was used as a standard for their examinations. Results were as follows : No Change in normal limit : 28 of the 36 Increased AST. ALT from when they were hospitalized : 2 of 36 Normalized AST. ALT from when they were hospitalized with results higher than normal : 6 of 36 These results show no side-effects, therefore suggest that these herbal medicines, including boo-ja and cheon-o, do not cause adverse side-effects for patients of this kind if administered as was done in this study.

무동무언증과 인지기능장애를 주소로 하는 급성 백질뇌증 환자 치험 1례 (A Case of Acute Leukoencephalopathy Patient with Akinetic Mutism and Cognitive Dysfunctions)

  • This case report presents a 34-year-old female patient. Her chief complaints are akinetic mutism and cognitive dysfunctions. We have diagnosed her with leukoencephalopathy. She was treated with Korean traditional herbal medicines (Joojackwhan) for 33 days continuously. The effects of treatment were measured by the Korean Mini - Mental State Examination and Modified Bathel Index. After treatment, her chief complaints have been improved. Furthermore, the K-MMSE, MBI scores increased. From this case, we deduced that the Korean traditional herbal medicines (Joojackwhan) could be effective for cognitive dysfunctions and akinetic mutism caused by leukoencephalopathy.

한국약용식물의 최종당화산물 생성저해활성 검색 (III) (Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (III))

  • Enhanced formation and accumulation of advanced glycation end products (AGEs) have been implicated as a major pathogenesis process leading to diabetic complications, normal aging, atherosclerosis and Alzheimer's disease. In our ongoing project to discover novel treatments for diabetic complications from natural sources, we have investigated on the inhibitory activity of 67 ethanol extracts from 57 Korean herbal medicines against the formation of AGEs in vitro. Of these, 22 extracts were found to have a significant AGEs inhibitory activity ($IC_{50}$<50 ${\mu}g$/ml) compared with aminoguanidine ($IC_{50}$=75.98 ${\mu}g$/ml). Particularly, 6 extracts from 3 herbal medicines, Castanea crenata (flower, leaf, bark-twig), Acer tatarium subsp. ginnala (fruit) and Sapium japonicum (leaf, twig) showed (approximately 8-17 fold) stronger inhibitory activity than that of aminoguanidine.

급성 요추부 염좌로 입원했던 환자군의 독활탕 치료 효과에 대한 후향적 고찰 (A Retrospective Study of Acute Lumbar Sprain Patients on the Effect of Korean Medicine Treatment with Dokhwal-tang (Duhuo-tang))

  • Objectives This study investigated the effect of korean medicine treatment with Dokhwal-tang (Duhuo-tang) on Lumbal sprain patients by comparing with other herbal medicines. Methods 17 lumbar sprain patients of group treated with Dokhwal-tang (Duhuo-tang) were treated with Dokhwal-tang (Duhuo-tang), acupunture, cupping, oriental physical therapy. 13 lumbar sprain patients of group treated with other herbal medicines were treated with 9 prescriptions of herbal medicine, acupunture, cupping, oriental physical therapy. ODI (Oswestry diability index), VAS (Visual Analogue Scale) were checked to evaluate the effectiveness of the treatment. Results 1) ODI, VAS of patients treated with Dokhwal-tang (Duhuo-tang) were all improved and there were statistical significance (p<0.05). 2) VAS of group treated with Dokhwal-tang (Duhuo-tang) and that of group treated with other herbal medicines were all improved significantly. And treatment period of group treated with Dokhwal-tang (Duhuo-tang) was shorter than that of group treated with other herbal medicines. But there was no statistical significance between VAS of group treated with Dokhwal-tang (Duhuo-tang) and that of group treated with other herbal medicines. 3) In comparing medical history period on group treated with Dokhwal-tang (Duhuo-tang), ODI, VAS of hyper-acute phase group and acute phase group were improved but them of sub-acute phase group were less improved. Conclusions According to the study, korean complex therapy with Dokhwal-tang (Duhuo-tang) might improve acute lumbar sprain.
