포도씨 추출물의 총 카테킨 함량과 전자공여능에 대한 마이크로웨이브 추출조건 최적화 (Optimization of Microwave-assisted Extraction Conditions for Total Catechin and Electron Donating Ability of Grape Seed Extracts)

  • 포도씨의 총 카테킨 함량과 추출물의 항산화성을 극대화하기 위한 마이크로웨이브 추출조건의 최적화를 시도하였다. 중심합성계획에 따라 추출조건(microwave power $0{\sim}120\;W$, 에탄올 농도 $0{\sim}100%$, 추출시간 $1{\sim}5\;min$)을 설계하고, 종속변수로서 추출물의 수율, 카테킨 함량 및 전자공여능을 회귀 분석함으로써 최적 추출조건을 예측하였다. 모든 회귀식의 $R^2$는 0.9 이상으로 1% 수준에서 유의성이 인정되었다. 총 카테킨 함량의 최대 추출 값은 434.16 mg%로 예측되었으며, 추출조건은 microwave power 122.76 W, 에탄올 농도 42.88%, 추출시간 4.67 min으로 나타났다. 추출물에 대한 세 가지 종속변수의 극대 값을 얻기 위한 추출조건 범위는 $75{\sim}150\;W$, $40{\sim}60%$$3.5{\sim}5.0\;min$이었다. 이상의 예측 값(총 추출수율 6.72%, 총 카테킨 함량 408.65mg%, 전자공여능 83.33%)은 실제 값과 유의적인 차이가 없었으며, 최적화된 마이크로웨이브 추출법(112.5 W, 50% 에탄올, 4.2분)은 현행추출법 (80% 에탄올, $60^{\circ}C$, 3시간, 150 rpm)에 비해 추출효율이 우수하였다.

건칠(乾漆) 추출물(Nexia)의 Quality Control을 위한 기시법 및 안정성 연구 (The Study on the Process and Quality Control of Rhus Verniciflua Stokes Extract (Nexia))

  • Rhus Verniciflua Stokes has been used for long history to treat blood stasis and cancer in Oriental Medicine. But urushiol, a constituent of Rhus Verniciflua Stokes has T-cell mediated allergic reaction, so it has made clinical limitation. Nexia is allergen removed extract of Rhus Verniciflua Stokes and its extraction process as well as quality control is needed to be standardized. The optimized extraction condition is as follow; temperature is $90{\sim}95^{\circ}C$, volume of slovent 10 times, extraction time >6 hours, pressure $, slovnet water or EtOH(60%), yield 8.1${\sim}$9.8(w/w). HPLC pattern and isolation study showed Nexia contain fisetin(>13.0%), fustin(>7.0%) and no urushiol.

  • This study was conducted to mass-produce brown stock optimized by using a high-pressure heating extractor and to use brown stock as a material for developing various products. For these purposes, we attempted to produce standardized brown stock by extracting brown stock using a high-pressure heating extractor and compared it with brown stock extracted by the traditional method in terms of general elements and mechanical and sensory characteristics. With regard to how to prepare optimal brown stock, the best brown stock was that extracted seven times repeatedly by the traditional method, but the method had a large economic loss in terms of material consumption and took a long time in extraction. Thus, considering time and labor, it was concluded that extraction at 120$^{\circ}C$ for 15 hours using a high-pressure heating extractor is the optimal extraction condition in terms of economic efficiency and quality. The results of this study are expected to be useful as a practical material for making brown stock production process more convenient, applying cooks' traditional cooking techniques to mass production, maintaining standardized superior quality and taste, and improving shelf life.

율무로부터 항치매성 Acetylcholinesterase 저해물질의 최적추출 조건 및 특성 (Optimal Extraction Condition and Characterization of Antidementia Acetylcholinesterase Inhibitor from Job's Tears (Coix lachrymajobi L.))

  • For the development of a new antidementia functional food or alternative drug using agricultural products, Job's tears (Coix lachrymajobi L.), which shows high acetylcholinesterase (AChE) inhibitory activity (55.1%) was selected and the extraction conditions of AChE inhibitor were optimized. AChE inhibitor of Job's tears was maximally extracted when it was treated with 60% methanol at $40^{\circ}C$ for 6 h. The AChE inhibitor of the methanol extracts was partially purified by systematic solvent extraction, thin layer chromatography, silica gel chromatography and reverse-phase HPLC and the partial purified AChE inhibitor with inhibitory activity ($IC_{50}$) of $0.608\;{\mu}g$ was obtained. The partial purified AChE inhibitor was soluble in methanol and hexane, and insoluble in water. Its maximum absorption spectra was 230 nm and also it was stable in the range of $30^{\circ}C$ and $70^{\circ}C$ and pH 4.0-8.0 for 1 h.

  • The ether extract(EE) of leaf tobacco contains diverse compounds such as lipid, resinoid, paraffin hydrocarbons and pigments. Although the correlation of EE concentertion with leaf tobacco has not been established clearly, it is known that in some concentration range, EE concentration of leaf tobacco exerts good influence on the quality of tobacco. Recently, The American Oil Chemist's Society introduced new method(Am 5-04; AOCS, 2009) to determine EE concentration by AT15 extractor. This method is based on extraction with organic solvent at relatively high temperature and pressure, which significantly reduces the extraction time and ensures safety. The aim of this study is to optimize analytical condition of AOCS method for analyzing EE concentration of leaf tobacco. When sample pre-drying time and extraction time of XT15 were set to 3 hour and 30 minute and 30 minute respectively, EE concentration obtained from AOAC method. Statistical analysis(T-test) showed that there is no difference(P>0.05) between EE concentrations from two methods.

Extraction of Optimal Operation Condition of QAM Envelope Tracking System using Combined Cost Function of Bandwidth and Efficiency

  • In this paper, we suggest a combined cost function to find out the optimal operation of an envelope tracking system, and evaluated its performance with Quadrature Amplitude Modulation (QAM) waveform, with which envelope tracking coefficients for the peak drain efficiency and the bandwidth of power amplifiers are determined. Based on the classical envelope tracking theory, the operation of the supply modulator, which is a key part of the envelope tracking process, is modeled and analyzed mathematically. Then characteristics of the modulator by setting envelope shaping function as a cubic polynomial and sweeping the coefficients of this function was analyzed. By sweeping the coefficients, efficiency and bandwidth at each condition with 64-QAM signal was used to obtain optimal point of the supply modulator. Compared to the conventional shaping functions, the optimized function showed the bandwidth reduction by 12.7 percent point while the efficiency was maintained.

양파껍질로부터 Flavonoid 물질의 추출조건 최적화 (Optimal Extraction Conditions of Flavonoids from Onion Peels via Response Surface Methodology)

  • 본 연구에서는 양파가공부산물인 양파껍질로부터 반응표면분석법을 이용하여 유효성분인 flavonoid 물질의 최적 추출조건을 설정하고자 하였다. 양파껍질의 일반성분은 수분 4.68%, 회분 6.67%, 조단백질 및 조지방이 5.03%, 0.71%이었고, 무기질은 Ca이 20.07 g/kg으로 가장 많은 함량을 차지하였고 K이 5.54 g/kg, Mg 2.17 g/kg의 순으로 함량이 많았다. 중심합성계획법에 의한 flavonoid 물질의 최적 추출조건은 다중회귀분석 및 능선분석 결과, 결정계수는 0.772이었으며, 적합결여검증(p>0.05)에서는 0.278, 전체적인 모형에서 유의성(p<0.05)을 보여 반응모형이 적합한 것으로 판단되었다. 따라서 최대점을 만족하는 각 변수의 조건은 추출용매의 농도는 70%, 추출온도 $40^{\circ}C$, 추출용매의 pH 5.3, 원료에 대한 추출용매의 비율은 1:63(w/v)이었다.

  • Effects of pretreatment and extraction conditions on total polyphenol yield from licorice root were investigated using statistical method. For pretreatment, heat treatment at $121^{\circ}C$ for 10 min was applied. Licorice root content in solvent (10, 20, and 30%) ethanol concentration (20, 40, and 60%) and reaction time (1, 2, and 3 h) were used as variables for extraction conditions. Two experiments, with heat treated and no treated licorice, were prepared with same experimental design. Box behnken design was employed and produced a total of 15 trials. Total polyphenol yield from licorice root was not affected by heat treatment. Among variables, licorice content in solvent showed most significant effect regardless of other variables (p<0.05). Finally, optimum conditions for the extraction of total polyphenol from licorice root was detected as following: 10% of licorice in solvent, 52% ethanol as solvent, 2 h of reaction time and non-heat treatment and the extraction yield from optimized condition was 17.6 mg/g licorice root.

반응표면분석법을 이용한 새싹보리 마이크로웨이브 추출공정의 최적화 (Optimization of microwave-assisted extraction process of Hordeum vulgare L. by response surface methodology)

  • 본 연구는 새싹보리를 이용하여 제품 개발에 적용할 수 있도록, 생리활성물질의 최적 추출구간을 설정하는 데에 목적이 있다. 에탄올 농도(0-100%), 마이크로웨이브 전력(60-300 W), 추출시간(4-20분)을 종속 변수로 설정한 후, 빠르고 추출 수율이 좋은 마이크로웨이브추출법을 이용하여 16구의 다른 추출 조건을 중심합성계획법에 따라 설정하여 새싹보리를 추출하였다. 이 후, 추출물의 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH 라디칼 소거능 활성을 측정하였다. 모든 회귀식의 $R^2$는 0.9 이상으로 5% 수준 이내에서 유의성이 인정되었다. 총 폴리페놀의 최적 추출조건은 에탄올 농도 58.94%, 마이크로웨이브 전력 209.04 W, 추출시간은 18.17분으로 나타났으며, 총 플라보노이드의 최적 추출 조건은 에탄올 농도 52.7%, 마이크로웨이브 전력 73.03 W, 추출시간은 5분으로 나타났다. DPPH 라디칼소거능 활성의 경우, 에탄올 농도 75.84%, 마이크로웨이브 전력 210.79 W, 추출시간 은 6.5분으로 나타났다. 조건에 따른 TPC, TFC 그리고 DPPH 라디칼 소거능 활성의 예측값은 각각 3.84 mg GAE/g, 3.00 mg RE/g 그리고 35.43%의 수치를 나타냈다. 최적 범위 내 임의의 점, 즉 에탄올 농도 40%, 마이크로웨이브 전력 120 W, 추출시간은 18분에서 실험값은 3.38 mg GAE/g, 2.64 mg RE/g, 그리고 37.94%를 나타냈으며 예측값과 실제 실험값은 유사한 값을 보였다.

황기의 추출조건 및 유효성분의 HPLC 분석법 평가 (Evaluation on Extraction Conditions and HPLC Analysis Method for Bioactive Compounds of Astragali Radix)

  • This study has been conducted to establish the optimal extraction process and HPLC analysis method for the determination of marker compounds as a part of the materials standardization for the development of health functional food materials from Astragali radix. Five extraction conditions including the shaking extraction at room temperature and the reflux extraction at $85^{\circ}C$ with 30%, 50% and 95% ethanol were evaluated. Reflux extraction with 50% ethanol showed the highest extraction yield as $27.27{\pm}2.27%$, while the extraction under reflux with 95% ethanol showed significantly the lowest yield of $10.55{\pm}0.24%$. The quantitative determination methods of calycosin-7-O-${\beta}$-D-glucoside and calycosin as marker compounds of Astragali radix extracts were optimized by HPLC analysis using a Thermo Hypersil column ($4.6{\times}250mm$, $5{\mu}m$) with the gradient elution of water and acetonitrile as the mobile phase at the flow rate of $0.8mLmin^{-1}$ and a detection wavelength of 230nm. The HPLC/UV method was applied successfully to the quantification of two marker compounds in Astragali radix extracts after validation of the method with the linearity, accuracy and precision. The contents of calycosin-7-O-${\beta}$-D-glucoside and calycosin in 50% ethanol extracts by reflux extraction were significantly higher as $1,700.3{\pm}30.4$ and $443.6{\pm}8.4{\mu}g-1$, respectively, comparing with those in other extracts. The results indicate that the reflux extraction with 50% ethanol at $85^{\circ}C$ is optimal for the extraction of Astragali radix, and the established HPLC method are very useful for the evaluation of marker compounds in Astragali radix extracts to develop the health functional material from Astragali radix.
