Change of population density of tobacco whitefly (Bemisia tabaci, Aleyrodidae, Hemiptera) by RNAi (RNAi에 의한 담배가루이(Bemisia tabaci, 가루이과, 노린재목)의 개체군 밀도변화)

  • Ninety genes randomly selected from tobacco whitefly (Bemisia tabaci) cDNA library was studied for selecting target gene in order to control of tobacco whitefly using TRV-VIGS vector (tobacco rattle virus-virus induced gene silencing vector) with RNAi. First of all, the occurrence of B. tabaci adult according to agro-infiltration of TRV was no significant difference. And that of TRV inserted tobacco whitefly cDNA showed a significant difference in each sample. P CV and N CV sample were more than 80% could be confirmed in 5 samples, for example, wh11, wh36, wh46, wh50 and wh71. Lastly, the occurrence of nymph and egg also showed a significant difference in each sample. That could be confirmed in 11 samples, for example, wh01, wh09, wh10, wh15, wh16, wh23, wh24, wh48, wh64 and wh66. In case of wh46, wh50 and wh71 sample could be confirmed that occurrence of B. tabaci adult was many, but occurrence of B. tabaci nymph and egg was a little. So sample showed a physioecological good effect to control of whitefly need to be investigated variation of gene expression in whitefly body using qRT-PCR through individual test.

Seasonal Development and Control of Parthenolecanium corni in Blueberry Shrubs (블루베리에 발생하는 말채나무공깍지벌레의 계절적 발육과 방제)

  • Seasonal development of Parthenolecanium corni was observed from 4 Jun. 2019 (nymph) - 25 Jun. 2020 (1st Gen. nymph) in blueberry shrub in Sacheon city, Gyeongsangnam-do. To investigate their development, more than 5 twigs sprouted in 2018 were taken from the farm nearly weekly basis. The development of each scale were examined under the stereomicroscope and chemical control was conducted in the blueberry shrub with available three insecticides. The results on the development period and Centigrade Degree-Days accumulation (DDC) obtained are as follows: egg-laying period (peak): 12 -26 May 2020 (24 May)(DDC, 110.0-188.5 (173.6)); egg-hatching period (peak): 9 - 23 June 2020 (19 June)(DDC, 325.2-480.8(435.6); egg period: 26 days; nymph movement from overwintered adult to new leaves 16-25 June 2020 (DDC, 410,5-500.4); nymph movement from leaf to twig (peak) to become adult: 4-18 Feb. 2020 (8 Feb.). Eggs no. /adult (range): 956.8 ± 73.4 (13 - 3497). Size (mm) of egg, 0.29 ± 0.020(L), 0.15 ± 0.013(W); of egg-hatched nymph, 0.35 ± 0.018(L), 0.18 ± 0.007(W), 0.09 ± 0.007(eye distance); and of adult, 4.30 ± 0.893(L), 2.64 ± 0.520(W). The egg-hatched nymphs from the overwintered adult moved to the backside leaf of new shoot in which they found about 95% until leaf is falling by early February in next year. They overwintered as 2nd instar and occurred univoltine. For the control of the 1st instar crawler, three insecticides treated on 16 and 30 July at the registered dose for Ceroplastes japonicus. Acetamiprid 8WP showed 96.9% mortality at 21 days after 1st treatment.

Biological Characteristics of Lycorma delicatula and the Control Effects of Some Insecticides (꽃매미(Lycorma delicatula)의 생태 특성 및 약제 살충 효과)

  • Biological Characteristics of Lycorma delicatula and insecticidal activity of some insecticides against nymphs of L. delicatula was investigated. Nymph of L. delicatula had 4 instars, and color of body was black. There were white spots on the body of 1st-3rd nymph. Upper body became red at 4th nymph. Adult forewings were brownish, and had black spots. Color of hind wing were red. The egg mass was covered with a yellowish brown secretion. The adult of L. delicatula emerged once a year. Among test insecticides, deltamethrin 1% EC and fenitrothion 50% EC showed very quick and strong insecticidal activity against the 2nd-3rd nymphs of L. delicatula. Imidacloprid 4% SL and clothianidin 8% SC showed 100% insecticidal activity at 24h after treatment. Thiacoprid 10% SC revealed the weakest insecticidal activity among the insecticides tested.

Ectoparasites: immature Japanese hard ticks (Ixodes nipponensis; Acari: Ixodidae) on Korean lizards

  • Although lizards are important hosts for hard ticks (Ixodidae), very few studies have been conducted in South Korea. To determine whether or not hard ticks can infest lizards endemic to South Korea, we examined 77 lizards of four species (Eremias argus, Sincella vandenburghi, Takydromus amurensis, and Takydromus wolteri) that were collected at 22 different sites between April and October 2010. We confirmed that all four lizard species can be infested by Ixodes nipponensis larvae or nymphs. Of the 62 E. argus examined, we found an average of 12.5 larvae on two lizards and an average of one nymph on one lizard. We found seven nymphs on one S. vandenburghi. We found an average of two nymphs on two of the five T. amurensis and an average of one nymph on four of the nine T. wolteri. Ixodes nipponensis larvae and nymphs were found most frequently on the foreleg axillae (87.8%), followed by the forelegs (7.3%), the eyelids (2.4%), and the ears (2.4%) of the lizards. To the best of our knowledge, this is the first report of I. nipponensis infestations of lizards endemic to South Korea.

Characterization of Chl a Fluorescence of Hydrophytes under Cadmium Stress (카드뮴 스트레스에 대한 수생식물 5종의 엽록소형광 반응)

  • The effects of $Cd^{2+}$ ions on the Chl a fluorescence of 5 hydrophytes (e.g. Lemna, Salvinia, Ricciocarp, Nymph, Typha plants) were investigated in order to select $Cd^{2+}$-sensitive plant species and to get informations on physiological responses of plants to $Cd^{2+}$ stress. Lemna plants were most sensitive to cadmium stress, while Nymph plants were tolerant. However, in all $Cd^{2+}$-treated plants, Fv/Fm, the maximum photochemical efficiency of PS II, decreased in proportion to the increase of $Cd^{2+}$ concentration and treatment time. The Chl a fluorescence transient O-J-I-P was also considerably affected by $Cd^{2+}$ ions; the fluorescence yield decreased consid- erably in steps J, I and P in $Cd^{2+}$ treated plants, although it followed a typical polyphasic rise in non-treated plants. In Lemna plants, the functional parameters, ABS/CS, TRo/CS와 ETo/CS and RC/CS, decreased in proportion to the increase of $Cd^{2+}$ concentration, while N, Mo and Kn increased. The structural parameters, ${\Phi}po$, ${\Phi}po$/($1-{\Phi}po$), Plabs, SFlabs, Kp and RC/ABS, also decreased according to the increase of $Cd^{2+}$ concentration. Consequently, Lemna plants will be useful as a experimental model system to investigate responses of plants. And several functional or structural parameters could be applied to determine quantitatively the physiological states of plants under stresses.

Effect of Cooking Oil and Yolk Mixture on Control of Bemisia tabaci on Fruit Vegetables (난황유를 이용한 과채류 발생 담배가루이 방제)

  • This study was conducted to develop an organic control method of tobacco whitefly (Bemisia tabaci) by using cooking oil and yolk mixture (COY). Mortality rate against B. tabaci nymph on paprika leaf was 94.5% by COY treatment and it was not significantly different from that by pyridaben treatment. Mortality rate against B. tabaci nymph on tomato leaf was 41.4% by COY treatment and it is lower than that by neem extract. But mortality rates of B. tabaci adult by COY and neem extract were no significant differences. Oviposition of B. tabaci were decrease respectively 75.8% and 52.7% by COY on paprika and tomato leaves. In paprika and tomato greenhouses, damaged by B. tabaci, COY was sprayed twice and resulted in respectively 100% and 38% control values.

Acaricidal and antibacterial toxicities of Valeriana officinalis oils obtained by steam distillation extraction (수증기 증류 추출법에 의해 얻어진 미국산 길초근 정유의 살비효과 및 항균효과)

  • The chemical compositions of the essential oil of Valeriana officinalis roots obtained by steam distillation method were analyzed by GC-MS. The 16 constituents were identified in the V. officinalis oil, and the most abundant compounds were patchouli alcohol (18.69%) and ${\beta}$-gurjunene (15.26%). Acaricidal effects of the V. officinalis oil were evaluated against Tyrophagus putrescentiae, Haemaphysalis longicornis larva and H. longicornis nymph by contact bioassay. The $LD_{50}$ values against T. putrescentiae, H. longicornis larva and H. longicornis nymph were 28.01, 178.26 and $207.98{\mu}g/cm^2$, respectively. Agar disc diffusion bioassay showed the antibacterial activity of the V. officinalis oil against foodborne pathogens, especially L. monocytogenes. These results showed that the essential oil of V. officinalis roots derived from USA has a potential for development as acaricide and antimicrobial.

Reciprocal effect of ethyl formate and phosphine gas on two quarantine pests, Tetranychus urticae(Acari: Tetranychidae) and Myzus persicae(Hemiptera: Aphididae)

  • Fumigation of fruits and vegetables during quarantine and pre-shipment(QPS) treatment should be effective with a shorter fumigation time to minimize phytotoxicity. In this research study, a shorter fumigation time, 2 hours exposure which is shorter than that of the current commercial fumigation procedures using a lower dose of ethyl formate (EF) mixed with phosphine (PH3) on strawberry was investigated. The reciprocal effect between EF and PH3 against nymphs and adult Myzus persicae (Sulzer) and Tetranychus urticae (Koch) was evaluated. In addition, L(Ct)50 and L(Ct)99 of EF only and EF mixed with PH3 were analyzed at 5℃ and 20℃. The synergistic ratio (SR) of L(Ct)50 and L(Ct)99 for the nymph and adult stages of M. persicae were >1.0, which indicated a synergistic effect between EF and PH3. However, the SR values of L(Ct)50 and L(Ct)99 of the nymph and adult stages of T. urticae were ≤1.0 indicating that there was no synergistic effect between the two fumigants against T. urticae. Our results showed that the reciprocal effect between EF and PH3 has different effects on M. persicae and T. urticae. This could be attributed to the biological and physical differences between the class Arachnida and Insecta. The synergistic effect between EF and PH3 against M. persicae within a shorter exposure period and without phytotoxicity on fruits and vegetables will significantly benefit the horticultural industry.

A Rare Tropical Stonefly Brahmana flavomarginata (Plecoptera: Perlidae: Acroneuriinae) from Vietnam

  • The perlid stonefly Brahmana flavomarginata Wu, originally described from China from the male adult, is described using reared male and female adults and nymphs from Vietnam. The male adult of B. flavomarginata is distinguished from its congeners by the blackish-brown body color and distinct head marking, hemitergal triangular chitin plates, and a median semicircular smooth subgenital plate (hammer) on the abdominal sternum IX. The female adult has a large and round subgenital plate which extends to the posterior margin of the abdominal sternum X. The nymph can be distinguished by the relatively small compound eyes and the body covered by many long stout setae and short golden brown hair-like setae. This species is known in southwestern China (Yunnan Province) and northern Vietnam (Lao Cai Province, Cao Bang Province).

  • From December 1992 to February 1993, all vagrants and children admitted to public welfare facilities supported by the Government of Republic of Korea were examined for the presence of nit, nymph or adult of head louse. Of 36,055 persons examined, the number of positive was 7,393 (20.5%). Intensive control measures are needed to control this ectoparasitic infestation.
