Objective : The purpose of this study was to investigate the effects of Bee Venom on the lipopolysaccharide(LPS), sodium nitroprusside(SNP), hydrogen peroxide$(H_2O_2)$-induced expression inducilble nitric oxide synthetase(iNOS), tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) and nuclear factor kappa B(NF-kB) in RAW 264.7 cells, a murine macrophage cell line. Method : The expressions of expression iNOS and TNF-${\alpha}$ were determined by western blotting with corresponding antibodies. The expressions of expression NF-kB was assayed by EMSA method. Results : 1. The 0.5, 1 and $5{\mu}g/mg$ of bee venom on LPS-induced expression of iNOS, the $5{\mu}g/mg$ of bee venom on SNP-induced expression of iNOS and the $1{\mu}g/mg$ of bee venom on $H_2O_2$-induced expression of iNOS compared with control were inhibited significantly. 2. The 0.5, 1 and $5{\mu}g/mg$ of bee venom inhibited significantly LPS and $H_2O_2$-induced expression of TNF-${\alpha}$ compared with control, respectively. The $0.5{\mu}g/mg$ of bee venom increased significantly SNP-induced expression of TNF-${\alpha}$ compared with control. 3. The $5{\mu}g/mg$ of bee venom on LPS-induced expression of NF-kB, the $0.5{\mu}g/mg$ of bee venom on SNP-induced expression of NF-kB and the 0.5, $5{\mu}g/mg$ of bee venom on $H_2O_2$-induced expression of NF-kB were inhibited significantly compared with control, respectively.
Korean bennudagrass collections showed diverse genetic variations in their morphology, growth habit, and cytological aspects. Chromosome number and nuclear DNA content of the bennudagrasses indicated a ploidy level ranging from triploid (2n=3x) to hexaploid (2n=6x). In this study, we investigated the different responses of antioxidant enzymes (superoxide dismutase, catalase, peroxidase, ascorbate peroxidase) and cell membrane stability of those bennudagrass cytotypes to lower temperature and shorter day length, which meets a dormant induction in Korea. All the antioxidant enzymes were found to be higher during dormant stage, while the heme-containing catalase which converts hydrogen peroxide ($H_2O_2$) to water and oxygen molecules was activated before dormant initiation in the three cytotypes except for hexaploid bennudagrass. The triploid and tetraploid which exhibited relatively finer leaves and a rapid establishment speed were found to show increased activities of superoxide dismutase and peroxidase enzyme. The malondialdehyde(MDA) which is a product of lipid peroxidation in the cell membrane damaged by the hydroxyl radical was increased in all cytotypes as temperature declined, and tri- and tetraploids which had more protective antioxidant enzymes demonstrated a significantly lower MDA production. Similarly electrolyte leakage was higher in penta- and hexaploidy, seemingly more damage to cell membrane when low temperature was implemented. Results indicated that antioxidant responses of different cytotypes were genetically specific, which needs to be investigated the relevance with the low temperature tolerance in the bermudagrass further at the molecular level.

In this study, the anti-oxidative, anti-inflammatory, anti-melanogenic activities of Endlicheria anomala (Nees) Mez methanol extract (EAME) were evaluated by use of in vitro assays and cell culture model systems. The results revealed that EAME scavenges various radicals such as 1,1-diphenyl-2-picryl hydrazyl hydrogen peroxide induced reactive oxygen species, and lipopolysaccharide induced nitric oxide. Furthermore, EAME induced the expression of anti-oxidative enzymes such as heme oxygenase 1, thioredoxin reductase 1, NAD(P)H dehydrogenase 1, and their upstream transcription factor, nuclear factor-E2-related factor 2. Moreover, EAME inhibited in vitro DOPA oxidation and 3-isobutyl-1-methylxanthine induced melanogenesis in B16F10 cells. Its anti-melanogenic activity will have originated from the inhibition of tyrosinase enzyme activity and melanogenesis related protein expression. Taken together, these results provide the important new insight that E. anomala possesses various biological activities such as anti-oxidative, anti-inflammatory, and anti-melanogenic. Therefore, it might be utilized as a promising material in the fields of nutraceuticals and cosmetics.

Computational Fluid Dynamics (CFD) analysis has been performed to estimate the air temperature inside an Auxiliary Feed Water (AFW) Motor-Driven (MD) pump room for the case where there is loss of Heating, Ventilation, and Air-Conditioning (HVAC). A transient calculation for the closed pump room without cooling by any HVAC system shows that the volume-averaged air temperature reaches around $60^{\circ}C$ for a transient period of 8.0 h. From previous studies, the external air and surface boundary temperatures are assumed to increase slowly starting from an initial temperature of $35^{\circ}C$. For the cases where the door is opened at 2, 4, and 6 h after the initiation of HVAC failure, the average air temperature promptly drops by about $4^{\circ}C$ when the door is opened and then slowly increases. The current calculations based on the CFD technique predict the rate of increase of air temperature to be lower than that determined by previous conservative calculations on the basis of a lumped model.

Strontium (90Sr) and nickel (59Ni) have been considered as key radionuclides in the safety assessment of radioactive waste disposal. Through various efforts to impede the migration of radioactive nuclides underground, it has been established that some minerals generated from the corrosion of the waste containers have a positive chemical interaction with these radionuclides. Among these minerals we selected mackinawite (FeS), an iron and sulfur compound, and performed a sorption experiment for the Sr and Ni in FeS under anoxic and alkaline conditions by reflecting deep underground environments. The effects of pH on sorption were likewise investigated in the pH range of 8 ~ 12. As a result, it was found that strontium failed to exhibit a good sorption capacity in a weak alkaline range, while nickel showed a noticeably higher sorption affinity over the entire experimental pH range. Moreover, we determined that as the pH increased in the solution, the distribution coefficients (Kd) were increased for both nuclides, which reflects when an alkalinity increses, the surface of the mineral charges much negatively by detaching the hydrogen or cations on the mineral surface. Thus, it can be concluded that the cationic nuclides of Sr and Ni can attach easily to the mineral under strong alkalinity.

Malus melliana (Hand.-Mazz.) Rehder (M. melliana) is a Chinese plant that belongs to the Rosaceae family. There have been no previous reports regarding its bioactivity. In this study, the anti-oxidative and anti-inflammatory activities of M. melliana ethanol extract (MMEE) were evaluated using a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity assay, reactive oxygen species (ROS) scavenging activity assay, nitric oxide (NO) inhibitory activity assay, and the analysis of related protein expressions through Western blot hybridization. MMEE showed potent scavenging activity against DPPH, similar to ascorbic acid, a well-known anti-oxidative agent, which was used as a positive control. MMEE also inhibited hydrogen peroxide-induced ROS in RAW 264.7 cells. Moreover, MMEE induced the expression of an anti-oxidative enzyme, heme oxygenase 1, and its upstream transcription factor, nuclear factor E2-related factor-2, in a dose-dependent manner. On the other hand, MMEE was associated with a reduction in NO production, which was induced by the lipopolysaccharide treatment of RAW 264.7 cells. The expression of inducible nitric oxide synthase, which is the upstream regulator of NO production, was also inhibited. Taken together, these results suggest that MMEE has anti-oxidative and anti-inflammatory properties, thus appearing to be a potential anti-oxidant and anti-inflammatory agent. The further identification of active compounds that confer the biological activities of MMEE may be necessary.
Peroxisome proliferator-activated receptors (PPARs) are members of nuclear receptors and their activation induces regulation of fatty acid storage and glucose metabolism. Therefore, the $PPAR\gamma$ is a major target for the treatment of type 2 diabetes mellitus. In order to generate pharmacophore model, 1080 known agonists database was constructed and a training set was selected. The Hypo7, selected from 10 hypotheses, contains four features: three hydrogen-bond acceptors (HBA) and one general hydrophobic (HY). This pharmacophore model was validated by using 862 test set compounds with a correlation coefficient of 0.903 between actual and estimated activity. Secondly, CatScramble method was used to verify the model. Hence, the validated Hypo7 was utilized for searching new lead compounds over 238,819 and 54,620 chemical structures in NCI and Maybridge database, respectively. Then the leads were selected by screening based on the pharmacophore model, predictive activity, and Lipinski's rules. Candidates were obtained and subsequently the binding affinities to $PPAR\gamma$ were investigated by the molecular docking simulations. Finally the best two compounds were presented and would be useful to treat type 2 diabetes.

Objective : To identify the effects of the water extract of Liriope platyphylla tuber (Liriopis tuber, LT) on the activation of astocytes, we investigated the regulatory effects of LT extract on H2O2-induced oxidative damage in C6 rat astrocytes. Methods : LT extract was extracted with boiling water. C6 cell line were treated with LT extract at 1, 2, and 3 mg/㎖ or without for 30 min and then stimulated with H2O2 at 5 ㎛ for 24 hr. The cell viability was measured by MTT assay. The expression of glial fibrillary acidic protein (GFAP), signal transducer and activator of transcription 3 (STAT3), phospho-STAT3 (pSTAT3), cyclooxygenase (COX-2), Nuclear factor-κB (NF-κB), superoxide dismutase 2 (SOD2), heme oxygenase-1 (HO-1), catalase, Akt, phospho-Akt (p-Akt) phosphoinositide 3-kinases (PI3K), and protein kinase C alpha (PKCα) proteins were determined by Western blot, respectively. GFAP expression was also observed with immunocytochemistry under a fluorescence microscope. Results : LT extract induced cell proliferation in H2O2-stimulated C6 cells. LT extract significantly inhibited the expression of GFAP, NF-κB and COX-2 and increased the expression of HO-1 and the phosphorylation of STAT3 in H2O2-stimulated C6 cells. LT extract also significantly increased the phosphorylation of Akt and decreased the expression of PKCα in a dose-dependent manner in H2O2-stimulated C6 cells. Conclusions : LT extract can regulate H2O2-induced activation of astrocytes through inhibiting the expression of NF-κB, COX-2 and regulating Akt / HO-1, STAT3 or PKCα signaling pathway.
Objectives: Paeonia japonica has been widely used for gynecopathy and analgesic effects in Korean Traditional Medicine. The aim of the present study is to determine the antioxidant effect of Paeonia japonica extracts(PJE) by using mouse embryonic fibroblast cells(MEF cells). Methods: We evaluated Radical Scavenging Activity of PJE by the DPPH assay. Protective effect of the PJE on the hydrogen peroxide($H_2O_2$) induced oxidative damage of MEF cells was analyzed by the MTT assay. The Morphological changes of MEF cells induced by P. japonica, $H_2O_2$ and P. japonica+$H_2O_2$ was evaluated by DAPI staining. And effect of PJE on the rate of apoptosis in MEF cells was measured using flow cytometry with Annexin V-FITC and PI double staining. Results: We observed that PJE contain significant DPPH radical scavenging activity. Cell viability of oxidative damaged cells treated with various concentrations of $H_2O_2$ was increased by treatment with PJE. Flow cytometric analysis of the cells treated with $H_2O_2$ in the absence or presence of PJE showed that the crumbled G1 peak was accumulated by the treatment with $H_2O_2$ alone, but restored by addition of PJE. Portion of cells that undergo apoptosis mediated by oxidative stress was decreased by treatment of PJE. The nuclear fragmentation occurred in the oxidative damaged MEF cells was also decreased by PJE treatment. Conclusions: Taken together, our results suggest that PJE exhibits significant antioxidant activity and functions to inhibit cell death mediated by oxidative damage induced apoptotic pathways.

Hydrazine based reductive dissolution applied on magnetite oxide was investigated. Dissolution of Fe(II) and Fe(III) from magnetite takes place either by protonation, surface complexation, or reduction. Solution containing hydrazine and sulfuric acid provides hydrogen to break bonds between Fe and oxygen by protonation and electrons for the reduction of insoluble Fe(III) to soluble Fe(II) in acidic solution of pH 3. In terms of dissolution rate, numerous transition metal ions were examined and Cu(II) ion was found to be the most effective to speed up the dissolution. During the cycle of Cu(I) ions to Cu(II) ions, the released electron promoted the reduction of Fe(III) and Cu(II) ions returned to Cu(I) ion due to the oxidation of hydrazine. In the experimental results, the addition of a very low amount of cupric ion (about 0.5 mM) to the solution increased the dissolution rate about 40% on average and up to 70% for certain specific conditions. It is confirmed that even though the coordination structure of copper ions with hydrazine is not clear, the $Cu(II)/H^+/N_2H_4$ system is acceptable regarding the dissolution performance as a decontamination reagent.
