Determination of trace actinide (Am, Pu, Th, U) using alpha spectrometry and neutron activation analysis (알파분광법과 중성자방사화분석법에 의한 극미량의 악티늄계원소 (Am, Pu, Th, U)분석연구)

  • Determination of actinides in the environmental sample requires separation of each element. This procedure is tedious and time consuming. And also, the detection limits of some nuclides using alpha spectrometry are rather higher. To overcome the lower detection limit and complicated separation procedure, a simple analytical technique for the determination of actinide isotopes in the environmental samples was developed and applied to IAEA and NIST reference sediment samples. For the separation of actinides from matrix, anion exchange resin and TRU-spec extraction chromatography resin were used and chemical yields were obtained using natural uranium, thorium, $^{242}Pu$ and $^{243}Am$ tracers. For overcoming the higher detection limits of U and Th in alpha spectrometry, neutron activation analysis was applied. Using combined method, the detection limit was increased about 10 times. The activity values of each isotope were consistent with the reference values reported by IAEA and NIST.

  • In this study, film type radiation sensor tips are fabricated for remote sensing of X or g-ray with inorganic scintillators and plastic optical fiber. The visible range of light from the inorganic scintillator that is generated by X and g-ray is guided by the plastic optical fiber and is measured by optical detector and power-meter. It is expected that the fiber-optic radiation sensor which is possible to be developed based on this study is used for remote, fast and exact sensing of X or g-ray because of its characteristics such as very small size, light weight and no interference to electromagnetic fields.

  • This study was conducted, during the period of 20-30th, July in 1981, to survey measurement methods in thyroid uptake rate test in Seoul city. The results were summarized as follows: 1. For the great part of nuclear medcine department, a mount of radioiodine($^{131}I$) administrated to the patients was $50-100{\mu}Ci$ in thyroid uptake rate test. 2. Distribution of scintillation, counter with crystal size of $1\frac{1}{2}inch$ was 43%, 3inch(22%), 2.5inch(14%) and $2\frac{1}{2}inch$ was 7% in RAI uptake rate test. 3. When RAI uptake rate test was performed, distribution of collimator in use was flat field type collimator(78%) in general and cylindrical type collimator was 22%. 4. High voltage applied to the P-M tube was $900{\sim}1000V$(50%) and most units provided $3{\sim}15%$ of the window range for the $^{131}I$ peak $\gamma-ray$ energy. 5. Distribution on the use of neck phantom for measurements standard solution was 57% and distribution of b filter in use for room background counts and extrathyroidal tissue was 43% and 50%. 6. The distance between the counter and the source was 25cm(58%) in measuring radioactivity of standard solution, thyroid tissue and background radioactivity count. 7. The early uptake measurements(2, 4, 6 hours) are done after administration of the radioiodine dose and also 24-hour and 48-hour uptake measurements are done in routine test.

Protective effect of phloroglucinol against gamma radiation-induced oxidative stress in hair follicles (방사선으로 인한 산화적 손상에서 phloroglucinol의 모낭 보호 효과)

  • When exposed to gamma-rays, hair follicular cells immediately go through apoptosis, which hampers their rapid differentiation essential for the regeneration of hair. Phloroglucinol (PG) is a phenolic compound of Ecklonia cava, brown algae abundant in Jeju island, Korea. Containing plentiful polyphenols, PG is known for its instructive effects by inhibiting apoptosis, scavenging oxygen radicals, and protecting cells against oxidative stress. In this study, we demonstrate that PG rescues radiosensitive hair follicular cells from gamma radiation-induced apoptosis and DNA damage. To identify protective capacity of PG on hair follicles, we irradiated with 8.5 Gy (1.5 Gy/min) of gamma-rays to the whole body of C57BL/6 mice at day 6 after depilation with or without PG. In mice exposed to radiation, the expression of proapoptotic molecule p53 was downregulated in the skin of PG treated group. On immunohistochemical observation of the skin, PG inhibited the immunoreactivity of p53 and cleaved caspase-3. PG treatment protected hair follicular cells from cell death due to gamma-radiation. Our results suggest that PG presents radioprotective effects by inhibiting apoptosis of radiosensitive hair follicular cells and can protect hair follicular cells from gamma-ray induced damage.

Inhibition of Apoptosis by Elaeocarpus sylvestris in Mice Following Whole-body Exposure to Ionizing Radiation: Implications for Radioprotectors

  • Elaeocarpus sylvestris var. ellipticus (E.S.), which contains 1, 2, 3, 4, 6-penta-O-galloyl-beta-D-glucose (PGG), is reported to have the ability to scavenge oxygen radicals, thereby protecting rat neuronal cells from oxidative damage. The potential of an E.S. extract, which contains a rich PGG, to protect radiosensitive lymphocytes and intestinal crypt cells from radiation injury induced by a single whole-body irradiation (WBI) in vivo was investigated. Our results demonstrated that in immune cells, E.S. treatment decreased the percent of tail DNA, a parameter of DNA damage, compared with levels in untreated, irradiated controls. Furthermore, apoptosis was significantly decreased in lymphocytes and intestinal crypt cells of E.S.-treated mice compared with irradiated controls. These results suggest that the E.S. extract can strengthen the radioresistance of radiosensitive lymphocytes and crypt cells by preventing apoptosis. Therefore, it was concluded that E.S. extract has the radioprotective effects in vivo through an inhibition of apoptosis.

Anti-inflammatory Effects of Enzymatic Extract from Ecklonia cava on TPA-induced Ear Skin Edema

  • Anti-inflammatory potential of the enzymatic extract prepared by Kojizyme (ECK), a component of brown seaweeds Ecklonia cava (Alariaceae, Phaeophyta) in vivo was investigated. For the application of mouse ear edema model, 12-O-tetradecanoylphorbol acetate (TPA) was used, a topical inducer of a long-lasting inflammatory response. Our results demonstrated that ECK inhibited ear edema when topically applied to mouse ear skin. In histological evaluation, the inhibition activity of ECK on TPA-induced inflammation is similar to that of dexamethasone, although less strong. In addition, the mRNA expression levels of IL-$1{\beta}$, IFN-$\gamma$, TNF-$\alpha$, and cyclooxygenase-2 (COX2) and the immunoreactivity to inducible nitric oxide synthase (iNOS) and COX2 expressed mainly in inflammatory cells were down-regulated by ECK. These results indicate that ECK has anti-inflammatory effects through the inhibition of Th1 cytokines and 2 inducers of inflammation in TPA-induced ear skin edema.

Technical Status of Environmental Radiation Monitoring using a UAV and Its Field Application to the Aerial Survey (무인기를 이용한 광역부지 환경방사선측정 기술 현황 및 현장 적용 연구)

  • According to lessons learned from an accident of Fukushima Daiichi nuclear power plant, it is advisable to make a comprehensive radiation survey by the accident phase for efficient response and risk management using diverse survey platforms. This study focuses on the technical status of environmental radiation monitoring using a UAV (Unmanned aerial vehicle) and the performance test of developed aerial survey system based on two detectors with an high energy resolution through the field application to contaminated areas. Finally, the performance of aerial survey at diverse flight heights was successfully achieved by introducing the correction factor to represent the results into ambient dose rate at 1m above the ground.

  • Effective multiplication factor has been calculated for one PWR fresh fuel assembly immersed in a spent fuel storage vault on the basis of the neutron transport theory. A numerical calculation has been carried out by means of Sn approximation. The method employed in this study is that the energy domain is broken into 16 groups, the angular variable is divided into four discrete direction, i.e., $S_4$, and the spatial variable which is divided into fine meshes at the interface between different materials is discretized into 27 mesh points. The calculated $K_{eff}$ value of 0.6145 seems to be far small in comparison with the value obtained by other author for an infinite array of fuel assemblies.

Etoposide Induces Mitochondrial Dysfunction and Cellular Senescence in Primary Cultured Rat Astrocytes

  • Brain aging is an inevitable process characterized by structural and functional changes and is a major risk factor for neurodegenerative diseases. Most brain aging studies are focused on neurons and less on astrocytes which are the most abundant cells in the brain known to be in charge of various functions including the maintenance of brain physical formation, ion homeostasis, and secretion of various extracellular matrix proteins. Altered mitochondrial dynamics, defective mitophagy or mitochondrial damages are causative factors of mitochondrial dysfunction, which is linked to age-related disorders. Etoposide is an anti-cancer reagent which can induce DNA stress and cellular senescence of cancer cell lines. In this study, we investigated whether etoposide induces senescence and functional alterations in cultured rat astrocytes. Senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) activity was used as a cellular senescence marker. The results indicated that etoposide-treated astrocytes showed cellular senescence phenotypes including increased SA-${\beta}$-gal-positive cells number, increased nuclear size and increased senescence-associated secretory phenotypes (SASP) such as IL-6. We also observed a decreased expression of cell cycle markers, including PhosphoHistone H3/Histone H3 and CDK2, and dysregulation of cellular functions based on wound-healing, neuronal protection, and phagocytosis assays. Finally, mitochondrial dysfunction was noted through the determination of mitochondrial membrane potential using tetramethylrhodamine methyl ester (TMRM) and the measurement of mitochondrial oxygen consumption rate (OCR). These data suggest that etoposide can induce cellular senescence and mitochondrial dysfunction in astrocytes which may have implications in brain aging and neurodegenerative conditions.

The bactericidal effect of an atmospheric-pressure plasma jet on Porphyromonas gingivalis biofilms on sandblasted and acid-etched titanium discs

  • Purpose: Direct application of atmospheric-pressure plasma jets (APPJs) has been established as an effective method of microbial decontamination. This study aimed to investigate the bactericidal effect of direct application of an APPJ using helium gas (He-APPJ) on Porphyromonas gingivalis biofilms on sandblasted and acid-etched (SLA) titanium discs. Methods: On the SLA discs covered by P. gingivalis biofilms, an APPJ with helium (He) as a discharge gas was applied at 3 different time intervals (0, 3, and 5 minutes). To evaluate the effect of the plasma itself, the He gas-only group was used as the control group. The bactericidal effect of the He-APPJ was determined by the number of colony-forming units. Bacterial viability was observed by confocal laser scanning microscopy (CLSM), and bacterial morphology was examined by scanning electron microscopy (SEM). Results: As the plasma treatment time increased, the amount of P. gingivalis decreased, and the difference was statistically significant. In the SEM images, compared to the control group, the bacterial biofilm structure on SLA discs treated by the He-APPJ for more than 3 minutes was destroyed. In addition, the CLSM images showed consistent results. Even in sites distant from the area of direct He-APPJ exposure, decontamination effects were observed in both SEM and CLSM images. Conclusions: He-APPJ application was effective in removing P. gingivalis biofilm on SLA titanium discs in an in vitro experiment.
