Glow-light tetra, Hemigrammus erythrozonus is a teleost belonging to Characidae. The natural habitate of this fish is the wild in the Essequibo river, Guyana and South America. The oogenesis of glow-light tetra was investigated by light microscopy to compare with those of other families. A pair of ovary was located between swim bladder and intestines. The ovary was of white color and ellipsoidal shape with the major axis 11 mm and the minor axis 4 mm. Cytoplasm of oogonia was basophilic and many nucleoli were located at inside of nuclear membrane. In primary oocytes, yolk vesicles were distributed only in the marginal area and egg envelope was not formed on the outside of an egg. In secondary oocytes, the egg envelope was formed and yolk vesicles in the cytoplasm were increased than the earlier stage. The amount of basophilic substance was decreased. In case of matured egg, thickness of egg envelope and size of egg were increased, basophilic substance was distributed in only around the egg envelope. The yolk vesicles were changed to yolk mass in accordance with development. In conclusion, the oogenesis of glow-light tetra, Hemigrammus erythrozonus was characterized by the increase in cell size, the formation and accumulation of yolk, and the decrease of basophilia a in the cytoplasm. The oogenesis of glow-light tetra seems to share common patterns in Characidae, teleost and have a similar pattern with other teleost.
Early cleavage is a reliable prognostic tool for successful embryo transfer in assisted reproduction because early cleaved embryo show better pregnancy rate after transfer. There for, preparation of good embryo recipient is important factor to optimize efficiency of pig cloning. The present study was performed to evaluate the effect of early cleavage on the in vivo development of cloned embryos and to analyze breed, parity and estrous synchrony to optimize recipient for pig cloning. In vitro matured porcine oocytes derived from local slaughterhouse and fibroblasts derived from miniature pig fetuses were used for somatic cell nuclear transfer (SCNT). Reconstructed embryos were transferred to recipient pigs on the same day of SCNT or after 1~2 days of in vitro culture for selecting early cleaved embryos. Breed, parity and date of standing estrous of recipients were recorded for analysis. After 25~35 days after embryo transfer pregnancy was diagnosed using ultrasonography, and pregnant recipients were monitored till delivery. Between purebred and crossbred, no significant difference was founded in both pregnancy and delivery rates. However, early cleaved embryos showed significantly higher pregnancy (46.2%) and delivery (12.8%) rates compared to non-selectively transferred group (24.8% and 4.5%, respectively). The results also showed that the recipients showing standing estrous on the same day of SCNT and less than 4 parities were most suitable for pig cloning.

For the artificial induction of the sexual maturation of Anguilla japonica, salmon pituitary extract (SPE) is continuously injected into females, and the eggs obtained from artificial sexual maturation are artificially fertilized with sperms and hatched. However, repeated injection of SPE in the abdominal cavity causes tremendous stress in females, which may prevent their complete sexual maturation and reduce the immune system function, ultimately resulting in death. In addition, the poor quality of the ovulated eggs can reduce the hatching and survival rate of larvae. In the present study, sexual maturation of females was induced by inserting an osmotic (OS) pump containing hormone analogs known to effectively induce sexual maturation into the abdominal cavity of female eels, and the effect of the OS pump on the induced sexual maturation was investigated. Our study results showed that the gonadosomatic index (GSI) was significantly higher in the fish subjected to SPE injection than those subjected to human chorionic gonadotropin (hCG), gonadotropin-releasing hormone analog (GnRHa), and methyl testosterone (MT) injections, either separately or in combination. In addition, a histological analysis showed that the oocytes in the SPE OS pump groups were more mature (entered the nuclear shift stage) than those in the other groups. These results suggest that an osmotic pump containing hormone analogs can be used to induce sexual maturation in female A. japonica artificially.
This study is written to bring the proposal forward for the direction of south korean naval force. The political situation and the circumstance of the world, especially in the area of Pacific Ocean, are changing very rapidly. North Korea has been always the conventional existing intimidator for South Korea since the 6·25-War. Additionally, the strengthening movements of the national defense, which is easily noticed from China and Russia, is also an other part of intimidating countries against South Korea. Those three mentioned countries are continually developing the asymmetrical warfare systems, for example a strategic nuclear weapon. Since the Obama Administration, the Asia-Pacific Rebalancing-Strategy has been changed as an East Asian foreign policy. Nowadays, Trump Administration renamed the 'United States Pacific-Command' to 'United States Indo-Pacific Command'. The purpose of this is not only letting India to participate in american alliance, but also reducing an economic burden, which is often mentioned in USA. West Germany was located in the very similar geopolitical position during the Cold War just like South Korea these days. And that's why the strategy of West German Navy is worthy of notice for south korean naval force to decide its suitable strategy. Most of all, the two most important things to refer to this study are the plan to expand naval air force and the realistic political stand for us to take it. In conclusion, I laid an emphasis on maintenance of 'green-water-navy'. instead of selecting the strategy as a 'blue-water-navy'. The reason I would like to say, is that south korean navy is not available to hold the unnecessary war potential, just like Aircraft-Carrier. However, this is not meaning to let the expansion of naval force carelessly. We must search the best solution in order to maintain the firm peace within the situation. To fulfill this concept, it is mostly very important to maintain the stream of laying down a keel of destroyers, submarines and air-defense-missile, as well as the hight-tech software system, taking a survey of 4th industrial revolution. Research and development for the best solution of future aircraft by south korean navy is likewise necessary. Besides, we must also set the international diplomatic flexibly. As well as maintaining the relationship with US Forces, it is also very important to improve the relationship with other potential allied nation.
It is an important issue to develop quality assessing system for biofuel for the purpose of accelerating the mass production of biofuel. It is particularly challenging to conduct testing method in the mass production of bioethanol while meeting quality specifications such as ASTM (American Society for Testing & Materials) D4806-10. In order to address this challenge, this paper proposes on-line spectroscopic quality assesment system based on Near Infrared spectrum and Partial Least Squares method in Chemometrics. As a result of testing a number of preprocessing methods and Partial Least Squares, it was found out that Savitzky-Golay method showed the best performance in terms of spectrum correction, noise reduction, and model maintenance. The proposed system allows us to assess multiple quality components continuously using spectroscopic facilities with the cheap cost. Since the value of R2 is more than 0.99, it is possible to replace the laboratory analysis.
The purpose of this study was to investigate the anti-proliferative and apoptotic effects of acid extract of Gracilaria verrucosa (AEG) on RC-58T/h/SA#4 primary human prostate cancer cells. AEG significantly decreased the cell viability of prostate cancer cells in a dose-dependent manner. AEG also showed relatively low cytotoxicity on normal cell (RWPE-1). The morphology of prostate cancer cells treated with AEG was distorted to shrunken cell masses. In addition, it was revealed that AEG induced cell death as evidenced by increased formation of apoptotic body and nuclear condensation. Furthermore, AEG clearly modulated the down regulation of Bcl-2 (anti-apoptotic)/Bax (pro-apoptotic) family and activated caspase-3 as an effector caspase in a dose-dependent manner. AEG inhibited cell proliferation induced by environmental hormones as a bisphenol A in a dose-dependent manner. These results indicate that AEG act as anti-proliferative effects as a potential therapeutic agent on primary human prostate cancer cells.
In aquatic invertebrates, particularly marine gastropods, organotin compounds induce irreversible sexual abnormality in females, which is termed imposex, at very low concentrations. Organotin compounds are agonists for nuclear receptors such as RXRs and $PPAR{\gamma}$. However, the imposex phenomenon has not been reported to act as an antagonist on estrogen receptors in other species, including vertebrates and invertebrates. In order to gain insights into the antagonistic activity of organotin compounds on estrogen receptors (ERs), we examined the inhibitive effect of these compounds on estradiol-dependent ${\beta}$-galactosidase activity using the yeast two-hybrid detection system consisting of a combination of the human estrogen receptor ($hER{\beta}$) ligand-binding domain and the co-activator steroid receptor co-activator-1 (SRC1). Tributyltin-hydroxide (TBT-OH) and triphenyltin-chlorine (TPT-Cl) exhibited an inhibitive effect on $E_2$-dependent transcriptional activity, similar to antagonistic chemicals such as 4-hydroxytamoxifen (OHT) or ICI 182,780, at a very low concentration of $10^{-14}$ M TBT or $10^{-10}$ M TPT, respectively. The yeast growth and transcriptional activity with transcriptional factor GAL4 did not exhibit any effect at the tested concentration of TBT or TPT. Moreover, the yeast two-hybrid system using the interaction between p53 and the T antigen of SV40 large did not describe any effect at the tested concentration of OHT or ICI 182,780. However, the interaction between p53 and T antigen was inhibited at a TBT or TPT concentration of $10^{-9}$ M, respectively. These results indicate that TBT and TPT act as inhibitors of ER-dependent reporter gene transcriptional activation and of the interaction between $hER{\beta}$ LBD and the co-activator SRC1 in the yeast two-hybrid system. Consequently, our data could partly explain the occurrence of organotin compound-induced imposex on the endocrine system of mammals, including humans.

This study was performed to determine the anticancer effects of cultivated Orostachys japonicus (COJ) and wild Orostachys japonicus (WOJ) on primary human prostate cancer cells (RC-58T/h/SA#4 cells). The morphology of cells treated with COJ and WOJ was distorted to shrunken cell masses. In addition, cell death induced by COJ and WOJ was associated with increased population of cells in sub-G1 phase as well as the formation of apoptotic bodies and nuclear condensation. COD and WOJ markedly reduced the number of viable prostate cancer cells in a dose-dependent manner, and cell numbers were lower than control cells. COJ and WOJ also inhibited increases in cell proliferation induced by environmental hormones such as dioxin and bisphenol A in charcoal-treated FBS (cFBS) medium. COJ and WOJ methanol extracts at the tested concentrations (150, 300, and 600 ${\mu}g/mL$) also dose-dependently inhibited cell proliferation induced by environmental hormones. These results indicate that COJ and WOJ exert anticancer effects on primary human prostate cancer cells.

This study aimed to investigate glucose uptake mechanisms and metabolic mechanisms for absorbed glucose in HepG2 cells treated with Acanthopanax senticosus water extract (ASW). A colorimetric assay kit was used to measure polyphenol content, glucokinase (GK) activity, glucose uptake, glucose consumption in cell culture medium, and glycogen content. RT-PCR and western blotting were performed to examine changes in the expression levels of glucose transporter 2 (GLUT2), hepatocyte nuclear factor $1{\alpha}$ ($HNF-1{\alpha}$), phosphatidylinositol 3-kinase (PI3k), protein kinase B (Akt), phospho-AMP-activated protein kinase (AMPK), phosphoenolpyruvate carboxykinase, GK, and glycogen synthase kinase $3{\beta}$ ($GSK3{\beta}$). Increased glucose uptake upon ASW treatment was confirmed to result from increased expression of $HNF-1{\alpha}$, which is one of the transcription factors acting on the GLUT2 promoter. From the measurements of GK activity, we observed that ASW had an effect on glucose phosphorylation, and we also confirmed that increased AMPK phosphorylation promoted glycolysis and suppressed gluconeogenesis. We confirmed that the increase in glycogen upon ASW treatment was induced by activation of Akt by PI3k, followed by phosphorylation of $GSK3{\beta}$. This study demonstrates that ASW activates glucose metabolic mechanisms in liver cells and is therefore a potential candidate to alleviate diabetes.

Matrix metalloproteinase 1 (MMP-1), a calcium-dependent zinccontaining collagenase, is involved in the initial degradation of native fibrillar collagen. Tissue necrosis factor-alpha (TNFα) is a pro-inflammatory cytokine that is rapidly produced by dermal fibroblasts, monocytes/macrophages, and keratinocytes and regulates inflammation and damaged-tissue remodeling. MMP-1 is induced by TNFα and plays a critical role in tissue remodeling and skin aging processes. However, the regulation of the MMP1 gene by TNFα is not fully understood. We aimed to find additional cis-acting elements involved in the regulation of TNFα-induced MMP1 gene transcription in addition to the nuclear factor-kappa B (NF-κB) and activator protein 1 (AP1) sites. Assessments of the 5'-regulatory region of the MMP1 gene, using a series of deletion constructs, revealed the requirement of the early growth response protein 1 (EGR-1)-binding sequence (EBS) in the proximal region for proper transcription by TNFα. Ectopic expression of EGR-1, a zinc-finger transcription factor that binds to G-C rich sequences, stimulated MMP1 promoter activity. The silencing of EGR-1 by RNA interference reduced TNFα-induced MMP-1 expression. EGR-1 directly binds to the proximal region and transactivates the MMP1 gene promoter. Mutation of the EBS within the MMP1 promoter abolished EGR-1-mediated MMP-1 promoter activation. These data suggest that EGR-1 is required for TNFα-induced MMP1 transcriptional activation. In addition, we found that all three MAPKs, ERK1/2, JNK, and p38 kinase, mediate TNFα-induced MMP-1 expression via EGR-1 upregulation. These results suggest that EGR-1 may represent a good target for the development of pharmaceutical agents to reduce inflammation-induced MMP-1 expression.
