Isolation of Ammonia Oxdizing Bacteria and their Characteristics (암모니아 산화세균의 분리와 그 특성)

  • This study was carried out far isolation and characterization of ammonia oxidizing bacteria (AOB) from aquacultural place and sludges of waste water collected in Pusan. One autotrophic AOB, Nitrosomonas sp. and 8 heterotrophic AOB (2 strains of Bacillus sp., 2 strains of Acinetobacter sp., Xanthomonas sp., Alcaligenes sp., Pseudomonas sp., Sphingobacterium sp.) were isolated. and identified. Variation of total nmmonia nitrogen (TAN) and $NO_2-N$ in mineral salt media containing 10mg/ $\ell$ of NHCl for 15 days in differents 9 strains was measured in order to examine the ablitity of ammonia oxidation. TAN was started to reduce after 4 days incubation and ca. 2 mg/$\ell$ of TAN was decreased after 15 days incubation by Nitrosomonas sp., At that time, $NO_2-N$ was produced to 0.023$\~$0.036 mg/$\ell$. Heterotrophic AOB showed the low ability of ammonia oxidation, 0.02$\~$0,04 mg/$\ell$ of TAN was decreased and $NO_2-N$ was produced to 0.01$\~$0.51 mg/$\ell$ after 15 days. When each strain of 8 heterotrophs was incubated in mimeral salt media containing 10 mg/$\ell$ $NH_4Cl$ and 50 mg/$\ell$ glucose, and 50 mg/$\ell$ $NH_4Cl$ and 5 g/$\ell$ glucose, the diminution of TAN was 1.12$\~$3.38 mg/$\ell$ and 1$\~$20 mg/$\ell$, respectively.

  • To remove dissolved $NH_{4}$$^{+}$ in the aquaculture system, one ammonia oxidizing bacterium, Nitrsomonas sp. PK1, was isolated from samples collected in many aquacultural place and sludges of waste water. The stationary phase of this atrain was reached after 9 days, and the maximum $NO_{2}$ production was shown from 3 days to 9 days. In the selective medium, 0.1% of glucose was the good carbon source for growth. However, the $NO_{2}$productivity was repressed by the addition of glucose to the medium. When $Zn^{++}$ ion was supplemented to the medium, growth and the $NO_{2}$ productivity was increased, 10mM of $ZnCl_{2}$ was the optimal concentration for growth and 1 mM of $ZnCl_{2}$ was the optimal concentration for the production of $NO_{2}$, respectively.

Effect of Probiotics on Water Quality in the Shrimp (Fenneropenaeus chinensis) Ponds (대하 (Fenneropenaeus chinensis) 양식장 사육수에 미치는 Probiotics의 효과)

  • Applications of probiotics to shrimp ponds were carried out to determine their effects on water quality. Fermented solutions consisting of Bacillus spp. and Nitrosomonas spp. were applied to a 4 ha shrimp (Fenneropenaeus chinensis) pond from July to September, 2000. In the pond treated with probiotics, daily variations of DO and pH, and concentrations of DIN and DIP were lower than those in the ponds without probiotic treatment. Concentration of phytoplankton was less variable and the number of species was more variable in the probiotic-treated pond than those in the control pond. Variation of bacterial numbers and the number of Vibrio spp. were lower in the treated pond than those in the control pond. It is confirmed that the probiotics can be used to improve water quality of the shrimp ponds.

Changes of Nitrifying Bacterial Populations in Anaerobic-Anoxic-Oxic Reactors (혐기-무산소-호기 반응조내 질화세균군의 변화)

  • This study was carried out to investigate the changes of nitrifying bacterial populations including Nitrosomonas sp. and Nitrobacter sp. in $A^2/O$ pilot plant with the configuration of anaerobic-anoxic-oxic reactors. The suspended nitrifying bacterial populations in mixed liquor and those of attached populations on granular carrier surface made by molded waste tire were analyzed by Fluorescent in situ Hybridization(FISH) method. The nitrification rate of a pilot plant showed the value of $1.97{\sim}2.98\;mg\;N/g$ MLVSS hr. The ratios of suspended ammonia oxidizer including Nitrosomonas sp. (NSO) to total bacteria in each reactor were oxic < anoxic < anaerobic. On the contrary, the ratios of suspended nitrite oxidizer including Nitrobacter sp. (NIT) were anaerobic < anoxic < oxic. The thickness, dry density and mass of the attached biomass on granular carriers were $180{\sim}188\;{\mu}m$, $38.5{\sim}43.9\;mg/cm^3$, $29.4{\sim}32.5\;mg/g$, respectively. Also, the ratios of attached nitrifier to total bacteria on granular carriers were similar regardless of ammonia/nitrite-oxidizer (NSO; 3.2%, NIT; 2.8%) and very low compared to those(NSO; $22.8{\sim}28.4%$, NIT; $17{\sim}26%$) of suspended nitrifier.

  • The effects of temperature on nitrification of enriched nitrifiers were investigated by using kinetics and thermodynamics method through the batch test. Aquaculture recirculating water, which was sampled at Chung Cheong Buk-Do Inland Fisheries Research Institute, was analized to observe the characteristics of nitrification. Temporal variation of ammonium, nitrite and nitrate concentration was measured at batch experiments. Activation energy was calculated using Arrhenius equation with the oxidation rates of specific ammonium or nitrite ion. These oxidation rates were measured at temperature range of $6-35^{\circ}C$ and ammonium concentration range of 0.2-1.8 mg/L. Two distinct activation energy of Nitrosomonas sp. at temperature $6-15^{\circ}C\;and\;15-35^{\circ}C$ was 93.1 and 25.0 KJ/mol, respectively. Nitrate accumulation was observed at temperature over $15^{\circ}C.$

Cloning and Nucleotide Sequence Analysis of the rpoH Gene from Methylovorus sp. Strain SS1 DSM11726 (Methylovorus sp. Strain SS1 DSM11726으로부터 rpoH 유전자의 클로닝과 염기서열 분석)

  • Using complementation of RpoH deficient E. coli strain A7448, the rpoH gene encoding heat shock sigma factor 32 (${\sigma}^{32}$) from Methylovorus sp. strain SS1 DSM11726 was cloned and sequenced. Sequence analysis of a stretch of 1,796-bp revealed existence of an open reading frame encoding a polypeptide of 284 amino acid (32,006 dalton). Deduced amino acid sequence of the Methylovorus sp. strain SS1 RpoH showed that 59.6%, 39.1% and 51.4% identities with those of Nitrosomonas europaea (${\beta}$-proteobacteria), Agrobacterium tumefaciens ($\alpha$-proteobacteria) and E. coli (${\gamma}$-proteobacteria). The expression level of the functional ortholog of RpoH of Methylovorus sp. strain SS1 was increased transiently after heat induction, further indicating that it functions as a heat shock sigma factor.

Effects of Probiotic Microbes on Growth Performance, Innate Immunity, and Pathogen Sensitivity in Cultured Olive Flounder (Probiotic 기능을 가진 미생물을 함유한 양어용 생균제가 넙치의 성장, 선천성면역 및 항병능에 미치는 영향)

  • Probiotic principles can be applied in aquaculture for the purpose of growth and immunity stimulation, disease prevention and eventually better production performance. This study was to assess effects of combinations of microbes containing two Bacillus sp., plus one Lactobacillus sp. as the basal preparation (BSL-LAB), and additional Nitrosomonas sp. (nitrifying bacteria consortium, NBS) in olive flounder (Paralichthys olivaceus). The effects examined were growth parameters, hematologic parameters, innate immunity and pathogen challenge test. Fish were assigned to 4 treatments as Control (no probiotics), Group A (Bacillius and Lactobacillus to culture water), Group B (Bacillius and Lactobacillus both in water and feed), Group C (same as Group B with additional Nitrosomonas in feed). Fish were allocated to the above 4 groups, each group being composed of triplicate 30 fish, for a 7-week feeding in the laboratory. Positive effects were observed both in growth and pathogen sensitivity with all three probiotic combinations. Such effects were attributed to improved innate immune functions. This result indicates that the tested probiotic microbes are beneficial to olive flounder aquaculture.

Change of Sludge Consortium in Response to Sequential Adaptation to Benzene, Toluene, and o-Xylene

  • Activated sludge was sequentially adapted to benzene, toluene, and o-xylene (BTX) to study the effects on the change of microbial community. Sludge adapted to BTX separately degraded each by various rates in the following order; toluene>o-xylene>benzene. Degradation rates were increased after exposure to repeated spikes of substrates. Eleven different kinds of sludge were prepared by the combination of BTX sequential adaptations. Clustering analyses (Jaccard, Dice, Pearson, and cosine product coefficient and dimensional analysis of MDS and PCA for DGGE patterns) revealed that acclimated sludge had different features from nonacclimated sludge and could be grouped together according to their prior treatment. Benzene- and xylene-adapted sludge communities showed similar profiles. The sludge profile was affected from the point of the final adaptation substrate regardless of the adaptation sequence followed. In the sludge adapted to 50 ppm toluene, Nitrosomonas sp. and bacterium were dominant, but these bands were not dominant in benzene and benzene after toluene adaptations. Instead, Flexibacter sp. was dominant in these cultures. Dechloromonas sp. was dominant in the culture adapted to 50 ppm benzene. Thauera sp. was the main band in the sludge adapted to 50 ppm xylene, but became vaguer as the xylene concentration was increased. Rather, Flexibacter sp. dominated in the sludge adapted to 100 ppm xylene, although not in the culture adapted to 250 ppm xylene. Two bacterial species dominated in the sludge adapted to 250 ppm xylene, and they also existed in the sludge adapted to 250 ppm xylene after toluene and benzene.

Changes of Microbial Community Depending on Different Dissolved Oxygen in Biological Nitrogen Removal Process (생물학적 질소제거 공정에서 용존산소변화에 따른 미생물의 군집변화)

  • PCR-DGGE method was applied to analyze changes of microbial community in simultaneous nitrification and denitrification (SND) bioreactor with various DO concentrations. In the analysis of eubacterial community, band profiles of DGGE were similar with 2 or 1 mg/L DO concentrations in the reactor. Experimental results led to 16 different bacteria being identified, including 5 dominant strains(3 strains of Uncultured Bacterium, 1 strains of Bacillus, 1 strains of Uncultured Bacteroidetes). DGGE results at 0.5 mg/L DO concentration led to 12 strains being identified, including 7 dominant strains(5 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). DGGE results at 0.1 mg/L DO concentration led to 11 strains being identified, including 3 dominant strains(1 strains of Uncultured Bacterium, 2 strains of Zoogloea sp.). In DGGE band profiles of $\beta$-AOB($\beta$-Ammonia Oxidizing Bacteria), only one band was observed. This band had 97% similarity with Nitrosomonas sp. done DNB Y20. This band was clearly observed at the 2, 1 and 0.5 mg/L DO concentrations, while the brightness of the band at 0.1 mg/L DO concentration was mostly dimmed. In DGGE band profiles of denitrification process, 5 bands(3 strains of Uncultured organism containing nirS, 2 strains of Uncultured organism containing nirK) were observed. Among those bands, the brightness of one band was gradually increased at the lower DO concentrations. This band has 86% identity with Uncultured organism clone eS1 cd1 nirS gene, partial cds. Based on this result, it could be concluded that Uncultured organism clone eS1 cd1 nirS gene, partial cds is a predominant microorganism in the denitrification process.

  • Ammonia oxidizing bacteria(AOB) oxidize ammonia to nitrite and are important microorganisms which control nitrification. Several environmental factors such as dissolved oxygen(DO), temperature and pH influence the growth of AOB. In this work, to assess the effect of DO concentration on AOB, four aerobic biofilm reactors packed with ceramic media were operated 1, 3, 5 and 7 mgDO/L, respectively. The optimal DO concentration with stable nitrification efficiency in aerobic biofilm reactor was above 5.0 mg/L. To assess the relationship between the DO concentration and the characteristics of AOB in aerobic biofiim reactor, DGGE and cloning based on PCR targeting 16S rRNA and amoA gene were performed. Additionally, INT-DHA activity test was proceeded to estimate the activity of AOB. As the results of DGCE and cloning, the community of AOB and the ratio of Nitrosomonas sp. changed little in spite of different nitrification efficiencies. INT-DHA activity test showed that the activity of AOB decreased as DO concentration decreased. It means that DO concentration does not affect the community of AOB, but the activity of AOB.
