Isolation and Characterization of Ammonia and Nitrite Nitrogen Oxidizing Strains (암모니아 및 아질산성 질소 산화균주의 분리 및 특성)

  • Characteristics and oxidizing ability of both $NH_4-N$ and$NO^2$-N were examined for the strains isolated from wastewater treatment facilities and from natural systems by using Winogradsky columns. In case of $NH_4$-N, the most efficient strain was Nitrosomonas KB1 isolated from wastewater treatment facility of K corporation and in case of $NO_2$-N, it was Nitrobacter KB2 from the same site as Nitrosomonas KB1. For Nitrosomonas KB1, 91% of $NH_4$-N was oxidized after 4 days of cultivation. Optimal growth temperature and initial pH of Nitrosomonas KB1 were $28^{\circ}C$ and 7, respectively. In comparison to oxidizing rates with changing initial concentration of $NH_4$-N, the ammonia oxidizing rate was increased up to 6.7 mg/day for the initial $NO_2$-N concentrations for the region lower than 100 mg $NH_4-N/L$, but it was gradually reduced for the region higher than 100 mg $NH_4-N/L$. For Nitrobacter KB2 90% of $NO_2$-N was removed after culturing for 4 days. Optimal growth temperature and initial pH of Nitrobacter KB2 was $28^{\circ}C$ and 7, respectively. And the nitrite oxidizing rate was increased in proportion to the initial concentrations of $NO_2$-N up to 200 mg/$\ell$, and it was maintained almost 4.2 mg/day irrespective of initial $NO_2$-N higher than 200 mg/L.

Presence and Growth of Ammonia-oxidizing Bacteria in Anaerobic Ammonium Oxidation Enrichment (아나목스 농후배양에서 암모니아 산화균의 자생 특성)

  • Anaerobic ammonium oxidation (AMX) is a cost-efficient biological nitrogen removal process. The coexistence of ammonia-oxidizing bacteria (AOB) in an AMX reactor is an interesting research topic as a nitrogen-related bacterial consortium. In this study, a sequencing batch reactor for AMX (AMX-SBR) was operated with a conventional activated sludge. The AOB in an AMX bioreactor were identified and quantified using terminal restriction fragment length polymorphism (T-RFLP) and real-time qPCR. A T-RFLP assay based on the ammonia monooxygenase subunit A (amoA) gene sequences showed the presence of Nitrosomonas europaea-like AOB in the AMX-SBR. A phylogenetic tree based on the sequenced amoA gene showed that AOB were affiliated with the Nitrosomonas europaea/mobilis cluster. Throughout the enrichment period, the AOB population was stable with predominant Nitrosomonas europaea-like AOB. Two OTUs of amoA_SBR_JJY_20 (FJ577843) and amoA_SBR_JJY_9 (FJ577849) are similar to the clones from AMX-related environments. Real-time qPCR was used to quantify AOB populations over time. Interestingly, the exponential growth of AOB populations was observed during the substrate inhibition of the AMX bacteria. The specific growth rate of AOB under anaerobic conditions was only 0.111 d-1. The growth property of Nitrosomonas europaea-like AOB may provide fundamental information about the metabolic relationship between the AMX bacteria and AOB.

  • To remove dissolved $NH_{4}$$^{+}$ in the aquaculture system, one ammonia oxidizing bacterium, Nitrsomonas sp. PK1, was isolated from samples collected in many aquacultural place and sludges of waste water. The stationary phase of this atrain was reached after 9 days, and the maximum $NO_{2}$ production was shown from 3 days to 9 days. In the selective medium, 0.1% of glucose was the good carbon source for growth. However, the $NO_{2}$productivity was repressed by the addition of glucose to the medium. When $Zn^{++}$ ion was supplemented to the medium, growth and the $NO_{2}$ productivity was increased, 10mM of $ZnCl_{2}$ was the optimal concentration for growth and 1 mM of $ZnCl_{2}$ was the optimal concentration for the production of $NO_{2}$, respectively.

Comparative analysis of core and pan-genomes of order Nitrosomonadales (Nitrosomonadales 목의 핵심유전체(core genome)와 범유전체(pan-genome)의 비교유전체학적 연구)

  • All known genomes (N=10) in the order Nitrosomonadales were analyzed to contain 9,808 and 908 gene clusters in their pan-genome and core genome, respectively. Analyses with reference genomes belonging to other orders in Betaproteobacteria revealed that sizes of pan-genome and core genome were dependent on the number of genomes compared and the differences of genomes within a group. The sizes of pan-genomes of the genera Nitrosomonas and Nitrosospira were 7,180 and 4,586 and core genomes, 1,092 and 1,600, respectively, which implied that similarity of genomes in Nitrosospira were higher than Nitrosomonas. The genomes of Nitrosomonas contributed mostly to the size of the pan-genome and core genomes of Nitrosomonadales. COG analysis of gene clusters showed that the J (translation, ribosomal structure and biogenesis) category occupied the biggest proportions (9.7-21.0%) among COG categories in core genomes and its proportion increased in the group which genetic distances among members were high. The unclassified category (-) occupied very high proportions (34-51%) in pan-genomes. Ninety seven gene clusters existed only in Nitrosomonadales and not in reference genomes. The gene clusters contained ammonia monooxygenase (amoA and amoB) and -related genes (amoE and amoD) which were typical genes characterizing the order Nitrosomonadales while they contained significant amount (16-45%) of unclassified genes. Thus, these exclusively-conserved gene clusters might play an important role to reveal genetic specificity of the order Nitrosomonadales.

Rapid Determination of the Maximum Specific Growth Rates of Nitrogen Oxidizing Bacteria by Fed-Batch Experiments (Fed-Batch 실험장치(實驗裝置)를 이용한 질산화(窒酸化) 미생물(微生物)들의 최대(最大) 성장율(成長率)의 결정(決定)에 관한 실험적(實驗的) 연구(硏究))

  • Nitrification reaction consists of two reactions: nitritification which oxidizes ammonia nitrogen to nitrite nitrogen and nitratification which oxidizes nitrite nitrogen to nitrate nitrogen. Each reaction is carried out by Nitrosomonas and Nitrobacter, respectively. The effective maximum growth rates for both bacteria have to be determined to design aeration tank whenever the aeration tanks have to nitrify ammonia nitrogen in influent. And these values are very important to use mathematical models such as IAWPRC model to simulate nitrification in activated sludge. There are several methods to determine these valves, however, the Fed-Batch experiments can determine these values within 72 hours. In this study, the mathematical equations and experimental procedures for Fed-Batch test are presented. Also, the experimental data and reported values are compared. The estimated mean values of maximum specific growth rates for Nitrosomonas and Nitrobacter are $0.5010day^{-1}$ and $0.6704day^{-1}$, respectively.

Effect of Probiotics on Water Quality in the Shrimp (Fenneropenaeus chinensis) Ponds (대하 (Fenneropenaeus chinensis) 양식장 사육수에 미치는 Probiotics의 효과)

  • Applications of probiotics to shrimp ponds were carried out to determine their effects on water quality. Fermented solutions consisting of Bacillus spp. and Nitrosomonas spp. were applied to a 4 ha shrimp (Fenneropenaeus chinensis) pond from July to September, 2000. In the pond treated with probiotics, daily variations of DO and pH, and concentrations of DIN and DIP were lower than those in the ponds without probiotic treatment. Concentration of phytoplankton was less variable and the number of species was more variable in the probiotic-treated pond than those in the control pond. Variation of bacterial numbers and the number of Vibrio spp. were lower in the treated pond than those in the control pond. It is confirmed that the probiotics can be used to improve water quality of the shrimp ponds.

Effect of Copper toxicant on Suspended and Attached Growth Nitrifying Bacteria (부유 및 부착성장 질화균에 미치는 구리 독성의 영향)

  • The effect of toxicant on the inhibition of nitrification was investigated, using concentrated nitrifying bacteria of both attached and suspended growth. This nitrifying organism was originally obtained from the activated sludge of sewage treatment plant and cultivated for more than three months. The object of this experiment is to determine the effect of the specific surface area and the growth condition of nitrifying bacteria on toxicity of heavy metal. The results of this study were as follows. The specific surface area of both attached and suspended growth of nitrifying organism was proven to be a major factors in determining the inhibition of nitrification of heavy metal such as $Cu^{++}$ion. When the condition of attachment and detachment was compared in an experiment using attached growth nitrifier, the effect on toxicant was 1.12 times less in attached condition than in detached condition for Nitrosomonas, and 1.09 times less for Nitrobacter. In case of suspended growth nitrifier, the effect on toxicant was 1.46 times less in non-ground condition than in ground condition for Nitrosomonas, and 1.35 times less for Nitrobacter. Also, similar results were obtained in a set of experiments, without adding nitrite to the substrate. In an experiment that compared attached condition using attached growth nitrifier with detached condition using attached growth nitrifier, the effect on toxicant was 1.83 times less in attached condition than in detached one for Nitrosomonas, and 1.78 times less for Nitrobacter. In case of suspended growth nitrifier, the effect on toxicant was 1.27 times less in non-ground condition than in ground condition for Nitrosomonas, and 1.32 times less for Nitrobacter.

Isolation of Ammonia Oxdizing Bacteria and their Characteristics (암모니아 산화세균의 분리와 그 특성)

  • This study was carried out far isolation and characterization of ammonia oxidizing bacteria (AOB) from aquacultural place and sludges of waste water collected in Pusan. One autotrophic AOB, Nitrosomonas sp. and 8 heterotrophic AOB (2 strains of Bacillus sp., 2 strains of Acinetobacter sp., Xanthomonas sp., Alcaligenes sp., Pseudomonas sp., Sphingobacterium sp.) were isolated. and identified. Variation of total nmmonia nitrogen (TAN) and $NO_2-N$ in mineral salt media containing 10mg/ $\ell$ of NHCl for 15 days in differents 9 strains was measured in order to examine the ablitity of ammonia oxidation. TAN was started to reduce after 4 days incubation and ca. 2 mg/$\ell$ of TAN was decreased after 15 days incubation by Nitrosomonas sp., At that time, $NO_2-N$ was produced to 0.023$\~$0.036 mg/$\ell$. Heterotrophic AOB showed the low ability of ammonia oxidation, 0.02$\~$0,04 mg/$\ell$ of TAN was decreased and $NO_2-N$ was produced to 0.01$\~$0.51 mg/$\ell$ after 15 days. When each strain of 8 heterotrophs was incubated in mimeral salt media containing 10 mg/$\ell$ $NH_4Cl$ and 50 mg/$\ell$ glucose, and 50 mg/$\ell$ $NH_4Cl$ and 5 g/$\ell$ glucose, the diminution of TAN was 1.12$\~$3.38 mg/$\ell$ and 1$\~$20 mg/$\ell$, respectively.

Rapid Detection of Ammonia-oxidizing Bacteria in Activated Sludge Based on 16S-rRNA Gene by Using PCR and Fluorometry

  • To detect whole ammonia-oxidizing bacteria in the activated sludge, group-specific primers targeting the 16S-rRNA gene of ammonia-oxidizing bacteria were used. The electrophoresis pattern of the PCR products seemed to produce a single band of approximately 1.0 k bp for the bacteria in activated sludge and Nitrosomonas europaea. No band was observed for nitrite-oxidizer Nitrobacter winogradskyi and heterotrophs such as Pseudomonas putida. Then direct measurement of the PCR product was made by fluorometry using the reagent Hoechist 33258, so that the fluorescent intensity was in proportional to the cell number of the sample up to 240. Total time required for the test was about 4 h including DNA extraction. The DNA fragments produced were cloned and their sequences showed high similarity to those of Nitrosomonas spp. This study showed the feasibility to detect ammonia-oxidizing bacteria and to esti-mate their population rapidly for the control of the nitrogen elimination process.
