Packagng of Fresh Curled Lettuce and Cucumber by Using Low Density Polyethylene Films Impregnated with Antimicrobial Agents (항균소재를 함유시킨 저밀도폴리에틸렌 필름에 의한 상추와 오이의 포장)

  • Low density polyethlene(LDPE0 films of 50${\mu}{\textrm}{m}$ thickness were fabricated with addition of antimicrobial agents of Rheum palmatum extract, Coptis chinensis extract, sorbic acid and Ag-substitude inorganic zirconium matrix in 1% concentration. The films were compared in physical properties, tested in antimicrobial activity against some selected microorganisms on the agar plate medium and then applied for packaging fresh curled lettuce and cucumber to preserve their qualities. The films with Rheum palmatum extract, Coptis chinensis extract, and Ag-substituted inorganic zirconium matrix did not show any antimicrobial activity on the disk test against Escherichia coli, Staphylococcus aureus, Leuconostoc mesenteroides, Saccharomyces cerevisiae, Aspergillus niger, Aspergillus oryzae, Penicilluium chrysogenum, while film with sorbic acid did against E. coli, S. aureus and L. mesenteroides. The added antimicrobial agents changed the color and light transmittance of the films, but did not affect their mechanical tensile strength, heat shrinkage and wettability. For the packaged curled lettuce and cucumber stored at 5$^{\circ}C$ and 1$0^{\circ}C$, all the LDPE films impregnated with antimicrobial agents showed the reduced growth of total aerobic bacteria in the vegetables compared with control film without any additive until it reached the level around 108/g. They did not give any negative effect on other quality attributes during storage.

  • Industrial strain Improvement is concerned with developing or modifying microorga-nisms used In production of commercially important fermentation products. The aim is to reduce the production cost by improving productivity of a strain and manipulating specific cilarafteristic such as the ability to utilize cheaper raw materials or resist bacteriophages. The traditional empiri-cal approach to strain improvement is mutation combined with selection and breeding techniques. It is still used by us to improve the productivity of organisms in amino acids. organic acids andenzymes production. The breeding of high L-lysine-producing strain Au112 is one of the outstanding examples of this approach. It is it homoserine auxotroph with AEC, TA double metabolicanalogue resistant markers. The yield reaches 100g/1. Resides, the citric acid-producing organism Aspergillus nuger, Co827, its productivity reches the advanced level in the world, is also the result of a series mutations expecially with Co Y-radiation. The thermostable a-amylaseroducing strain A 4041 is the third example. By combining physical and chemical multations. the strain ,A 4041becomes an asporogenous, catabolite derepressed mutant with rifamycin resistant and methionine, arginine auxotroph markers. The a-amylase activity reaches 200 units/ml. The fourth successful example of mutation in strain improvement is the glucoamylase-producing strain Aspergillus nigerSP56 its enzyme activity is 20,000 units/ml, 4 times of that of the parental strain UV_11. Recently recombinant DNA approach Provides a worth while alternative strategy to Industrial strain improve-ment. This technique had been used by us to increase the thermostable a-amylase production and on some genetic researches.

Optimization of Fermentation Medium for Glycyrrhizin Biotransformation to Monoglucuronyl-glycyrrhetinic Acid by Plackett-Burman and Box-Behnken Design

  • Plackett-Burman (PB) and Box-Behnken (BB) experimental designs were used to optimize fermentation variables for the biotransformation of glycyrrhizin (GL) to monoglucuronyl-glycyrrhetinic acid (MGGA). The PB design was first used to screen the important factors among the medium variables. The steepest ascent method was used to approach the optimum range for each of these factors. The BB design was finally used to analyze the response surfaces of the screened factors for further optimization. The optimized conditions for this system were 0.7 g/L $MgSO_4{\cdot}7H_2O$, 1.19 g/L GL, and cultivation for six days. The biotransformation of GL to MGGA could reach up to 35.72%, which is a good result for this kind of transformation.

  • GOD electrochemically immobilized in PPy/GOD complex have an effect on redox properties of the complex. In the cyclicvoltammetry, GOD shows the redox reaction at the potential below -0.6Y vs. Ag/AgCI. That leads to new peaks in the cyclicvoltammograms in additional to typical PPy peaks. The pH of electrolyte solution during potential swing decreased to 4.4, and then increased to 10. That suggests the redox of GOD for the cycling. As the concentration of GOD was increased, the anodic wave of the new peaks was strong as much as increased. GOD obstructs the diffusion of electrolyte anion because of its net chain. Insulating property of GOD is cause that it made the faradic impedance of complex large in charge transfer. It suggests that increase of the concentration of GOD be against electrochemical coupling. Therefore, the concentration of GOD and electrochemical coupling should be dealt with each other. The apparent Michaelis-lenten constant ( K\`$_{M}$ ) was determined by 30.7 mmol d $m^{-3}$ fur the PPy/GOD complex. The value is of the same order of magnitude as that for soluble glucose oxidase from Aspergillus Niger.r.

  • A new series of ethyl 2-(4-substitutedbenzylidene)-5-(3'-(ethoxycarbonyl)biphenyl-4-yl)-7-methyl-3-oxo-3,5-dihydro-2H-thiazolo[3,2-a]pyrimidine-6-carboxylate derivatives ($\mathbf{8a-j}$) were synthesized. The newly synthesized compounds were characterized by $\mathbf{IR}$, $^1\mathbf{H}$ $\mathbf{NMR}$, $^{13}\mathbf{C}$ $\mathbf{NMR}$, $\mathbf{LCMS}$ $\mathbf{mass}$ and $\mathbf{C}$, $\mathbf{H}$, $\mathbf{N}$ analyses. All newly synthesized compounds were screened for their In vitro antioxidant activity (Scavenging of hydrogen peroxide, Scavenging of nitric oxide radical, and Lipid peroxidation inhibitory activity), antibacterial (Escheria coli, Pseudonmonas aeruginosa (gram-negative bacteria), Bacillus subtillis, Staphylococcus aureus (gram-positive bacteria)) and antifungal (Candida albicans Aspergillus niger) studies.

Aspergillus Associated with Meju, a Fermented Soybean Starting Material for Traditional Soy Sauce and Soybean Paste in Korea

  • Aspergillus is an important fungal genus used for the fermentation of Asian foods; this genus is referred to as koji mold in Japan and China. A. oryzae, A. sojae, and A. tamari are used in the production of miso and shoyu in Japan, but a comprehensive taxonomic study of Aspergillus isolated from Meju, a fermented soybean starting material for traditional soy sauce and soybean paste in Korea, has not been conducted. In this study, various Aspergillus species were isolated during a study of the mycobiota of Meju, and the aspergilli were identified based on phenotypic characteristics and sequencing of the ${\beta}$-tubulin gene. Most strains of Aspergillus were found to belong to the following sections: Aspergillus (n = 220), Flavi (n = 213), and Nigri (n = 54). The most commonly identified species were A. oryzae (n = 183), A. pseudoglaucus (Eurotium repens) (n = 81), A. chevalieri (E. chevalieri) (n = 62), A. montevidensis (E. amstelodami) (n = 34), A. niger (n = 21), A. tamari (n = 15), A. ruber (E. rubrum) (n = 15), A. proliferans (n = 14), and A. luchuensis (n = 14); 25 species were identified from 533 Aspergillus strains. Aspergillus strains were mainly found during the high temperature fermentation period in the later steps of Meju fermentation.

Antifungal Activities of Copper(II) with Biosensitive Macrocyclic Schiff Base Ligands Derived from 4-Aminoantipyrine Derivatives

  • Novel copper(II) complexes have been synthesized from the macrocyclic Schiff bases derived from Knoevenagel condensed ${\beta}$-ketoanilides (obtained by the condensation of acetoacetanilide and substituted benzaldehydes), 4-aminoantipyrine and ophenylene diamine. The structural features have been determined from their analytical and spectral data. All the Cu(II) complexes exhibit square planar geometry. Their high molar conductance values support their 1 : 2 electrolytic nature. The magnetic moment data provide evidence for the monomeric nature of the complexes. The X-band ESR spectra of the |$CuL^1$|$(OAc)_2$ in DMSO solution at 300 and 77 K were recorded and their salient features are reported. The in vitro biological screening effects of the investigated compounds were tested against the bacterial species Staphylococcus aureus, Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris and Pseudomonas aeruginosa and fungal species Aspergillus niger, Rhizopus stolonifer, Aspergillus flavus, Rhizoctonia bataicola and Candida albicans by well diffusion method. A comparative study of inhibition values of the Schiff bases and their complexes indicate that complexes exhibit higher antimicrobial activity than the Schiff bases. Copper ions proved to be essential for the growth-inhibitor effect. The extent of inhibition appeared to be strongly dependent on the initial cell density and on the growth medium.

Anti-inflammatory Activity of the Ethanol Extract from Magnoliae Flos on PAR2-mediated Edema (신이 에탄올 추출물의 PAR2-유발 부종에 대한 항염증 활성)

  • The flower of Magnolia denudata Desrousseaux(Magnoliaceae) has long been used for treatment of nasal disorder in Korea. The physiological activity of the Magnoliae Flos ethanol extract (MFX) was investigated. MFX showed antimicrobial activity. At doses of 100 and 200 mg/kg, MFX showed significant inhibition on both change in paw volume and vascular permeability. MFX (100 mg/kg) significantly inhibited PAR2 agonist-induced myeloperoxidase (MPO) activity in paw tissue. These results indicate that MFX has anti-inflammatory activity in PAR2-mediated paw edema.

Studies on the industrialization of the Korean KockJa.(I) - It's Isolation and physiological characteristics of Mold from Kock Ja. (한국국자(Kock Ja)의 발효생산력에 관한연구 (제 1보) - 국자중 함유사상균의 분리와 기성상)

  • Especially, we mainly dealt on the isolation of mold in the sample of the Korean products, Kock Ja. The kinds of the isolated strain are such as these, Rhizopus, Mucor, Aspergillus oryzae sp., aspergillus niger sp., Penicillum and Flungi Imperfecti. The action of the starch saccharification of isolated strain and the order of liquefying action are follows: The saccharification power was R-l>R-2>M-2> Kock Ja>M-1>O-2>N-1>O-4 The liquefying power was R-1, R-2>0-2>0-4>M-2, Kock Ja>M-1>N-1 We compared the pH's saccharification curve of each kind of strain with Kock Ja. The most suitable pH value of R-1, R-2 was the closest to pH 4. 0, close value with Kock Ja. The Rhizopus species on the saccharification action of each kind of strain in regard to raw wheat starch was stronger than other kinds of strain. We think that to generalize the above result, the Rhizopus species consists of an important strain of this Kock Ja, and is an important factor for the saccharification action of Kock Ja and the existence of Mucor species as well.

Phytochemical Screening, Isolation, Characterization of Bioactive and Biological Activity of Bungkang, (Syzygium polyanthum) Root-bark Essential Oil

  • Bungkang (Syzygium polyanthum) is a medium to tall plant which produces medicinal root-bark, the plant is normally found along inland river bank and produces small white flowers and fruits. Essential oils are among the most interesting components of the plant extracts consisting mostly of monoterpenoid or sesquiterpenoids. They are used as therapeutic agents in ethno, conventional, and complementary alternative medicines. Investigation and evaluation of the essential oil of Syzygium polyanthum as well as the antibacterial, antioxidant and antifungal activity was ascertained. The experiment was performed. 100 chemical constituents were obtained and two pure compound was isolated as Eugenol (1) and Farnesol (2). Significant growth inhibition of Staphylococcus aureus, (ATCCⓒ25923) Klebsiellia pneumonia (ATCCⓒ19155), Salmonella typhi (ATCCⓒ14028) and Escherichia coli (ATCC©25922) and the fungal strains Aspergillus flavin, Aspergillus niger, Candida, tropicalis, and Fusarium oxysporium was observed from the essential oil at concentration of 500 ㎍/mL. Antioxidant potential was observed to be strong of 18.42 ㎍/mL when compared to the control of 15.23 ㎍/mL. The result indicated that the oil obtained from root-bark of Syzygium polyanthum can be considered as an agent for antioxidant, antibacterial and antifungal in pharmaceutical food and cosmetic industries trails.
