Petrological and Geological Safety Diagnosis of Multi-storied Stone Pagoda in the Daewonsa Temple, Sancheong, Korea (대원사 다층석탑의 지질학적 및 암석학적 안전진단)

  • The multi-storied Daewonsa stone pagoda (Treasure No. 1112) in the Sancheong, Korea was studied on the basis of deterioration and geological safety diagnosis. The stone pagoda is composed mainly of granitic gneiss, partly fine-grained granitic gneiss, leucocratic gneiss, biotite granite and ceramics. Each rock of the pagoda is highly exfoliated and fractured along the edges. Some fractures in the main body and roof stones are treated by cement mortar. This pagoda is strongly covered with yellowish to reddish brown tarnish due to the amorphous precipitates of iron hydroxides. Dark grey crust by manganese hydroxides occur Partly, and some Part coated with white grey gypsum and calcite aggregates from the reaction of cement mortar and rain. As the main body, roof and upper part of the pagoda, the rocks are developed into the radial and linear cracks. Surface of this pagoda shows partly yellowish brown, blue and green patchs because of contamination by algae, lichen, moss and bracken. Besides, wall-rocks of the Daewonsa temple and rock aggregates in the Daewonsa valley are changed reddish brown color with the same as those of the pagoda color. It suggests that the rocks around the Daewonsa temple are highly in iron and manganese concentrations compared with the normal granitic gneiss which color change is natural phenomena owing to the oxidation reaction by rain or surface water with rocks. Therefore, for the attenuation of secondary contamination, whitening and reddishness, the possible conservation treatments are needed. Consisting rocks of the pagoda would be epoxy to reinforce the fracture systems for the structural stability on the basements.

Effect of Calmodulin on Ginseng Saponin-Induced $Ca^{2+}$-Activated $Cl^{-}$ Channel Activation in Xenopus laevis Oocytes

  • We previously demonstrated the ability of ginseng saponins (active ingredients of Panax ginseng) to enhance $Ca^{2+}$-activated $Cl^{-}$ current. The mechanism for this ginseng saponin-induced enhancement was proposed to be the release of $Ca^{2+}$ from $IP_{3}-sensitive$ intracellular stores through the activation of PTX-insensitive $G\alpha_{q/11}$ proteins and PLC pathway. Recent studies have shown that calmodulin (CaM) regulates $IP_{3}$ receptor-mediated $Ca^{2+}$ release in both $Ca^{2+}-dependent$ and -independent manner. In the present study, we have investigated the effects of CaM on ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current responses in Xenopus oocytes. Intraoocyte injection of CaM inhibited ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement, whereas co-injection of calmidazolium, a CaM antagonist, with CaM blocked CaM action. The inhibitory effect of CaM on ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement was dose- and time-dependent, with an $IC_{50} of 14.9\pm3.5 {\mu}M$. The inhibitory effect of CaM on saponin's activity was maximal after 6 h of intraoocyte injection of CaM, and after 48 h the activity of saponin recovered to control level. The half-recovery time was calculated to be $16.7\pm4.3 h$. Intraoocyte injection of CaM inhibited $Ca^{2+}$-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement and also attenuated $IP_{3}$-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement. $Ca^{2+}$/CaM kinase II inhibitor did not inhibit CaM-caused attenuation of ginseng saponin-induced $Ca^{2+}$-activated $Cl^{-}$ current enhancement. These results suggest that CaM regulates ginseng saponin effect on $Ca^{2+}$-activated $Cl^{-}$ current enhancement via $Ca^{2+}$-independent manner.

Suppression of reactive oxygen species generation as a part of antioxidative effect of plant extracts (식물추출물 항산화효능 기전의 일부로서의 활성산소 발생 억제 효과)

  • Chemical scavenging of reactive oxygen species (ROS) is considered a major mechanism of antioxidant effects, but preventing ROS generation can be more efficient in attenuating oxidative damage. In this study, the extracts of plants, Solanum lycopersicum, Ailanthus altissima, Equisetum arvense, and Oenothera biennis, were tested to determine whether their antioxidative effects are driven by the prevention of superoxide generation from mitochondria, a major ROS generator. While all the extracts efficiently attenuated the elevation of ROS levels in human fibroblasts and inflammation-induced mice, those of S. lycopersicum, A. altissima, and O. laciniata only suppressed mitochondrial ROS generation and reduced levels of lipofuscin and lipid peroxidation. Furthermore, the extracts of A. altissima and O. laciniata extended the lifespan of fruit flies. Our results suggest that plant extracts with anti-oxidative effects differ in their ability to prevent ROS generation, which may be associated with the attenuation of oxidative damage in cells and animal tissues.

Development of Safe Stove System using Sound Wave Fire Extinguisher (음파 소화기를 이용한 안전 스토브 시스템 개발)

  • In this paper, the architecture of a safe stove with an automatic fire suppression function using a sound wave fire extinguisher has been proposed and developed for the first time. A microcontroller connected to a fire sensor detects and suppresses a fire by driving a fire extinguisher. The sound wave fire extinguisher is composed of a speaker and collimator, and is driven by a driver module including an audio amplifier. The attenuation of the sound wave is reduced by preventing the sound diffusion with an enclosure surrounding a stove. The frequency of the sound wave is set to 50 Hz, and the sound pressure of 93 dBA is measured at the distance of 0.5 m. It takes maximum 8 and 15 seconds to suppress the flame from 7-cc and 14-cc flammable liquid, respectively, which corresponds to 24% and 42% of the natural extinguishing time. Since the proposed safe stove is non-toxic and leaves no residues over the conventional ones, it would combine with various home appliances to suppress early-stage fires and prevent fire expansion.

  • 산업혁명 이후 화석연료의 광범위한 사용, 삼림 벌채, 토지사용의 변화 등과 같은 인위적 활동은 대기 중 온실가스(GHGs, greenhouse gases) 농도를 지속적으로 증가시켜 지구의 기후위기를 유발하였다. 우리나라의 경우 최근 30년 사이 평균 온도가 1.4℃ 상승하였으며, 국제사회의 일원으로 책임을 다하기 위해 2016년 11월 3일 파리협정을 비준하였다. 이에 파리협정의 목표인 산업화 이전 대비 지구 평균온도 상승을 2℃ 아래, 가능한 1.5℃ 아래로 억제하기 위해 2050년까지 CO2 순배출량을 0으로 만들어야 하며, 이를 위해 다양한 정책 마련과 함께 경제 및 사회 전반에 걸쳐 많은 노력이 경주되고 있는 실정이다. 탄소중립을 달성하기 위해서는 첫 번째로 GHGs 배출을 줄이고, 두번째로 대기에서 CO2 포집을 촉진하기 위해 현재 가동되는 다양한 산업분야의 생산 시스템을 개혁하는 것이 가장 중요한 과제로 고려되고 있다. 그동안 지하수토양 관련 연구분야에서는 지속가능성(sustainability), 복원성(resilience), 녹색성장(green growth) 등과 같은 사회적 요구에 부응하여, 녹색정화(green remediation), 자연 저감(natural attenuation), 탄소포집저장(carbon capture and sequestration), 지열발전등의 기술이 초기단계로 개발이 되고 연구가 되어 왔다. 이러한 기존 연구들은 탄소중립2050의 달성을 위해 고도화되어야하며, 추가적으로 자연 및 인위기원 탄소배출 연구, 토양의 역할을 고려한 저탄소 토지이용 기술, 광물탄산화 등의 연구 및 기술개발이 필요하다고 판단된다. 본 논문에서는 탄소중립2050의 간단한 내용과 함께, 이를 달성하기 위한 지하수토양 분야의 혁신기술 및 선도연구를 소개하였다.

Anti-diabetic effects of aqueous and ethanol extract of Dendropanax morbifera Leveille in streptozotocin-induced diabetes model (Streptozotocin에 의해 유도된 당뇨모델동물에서 황칠나무 (Dendropanax morbifera Leveille)의 열수추출물과 에탄올추출물의 당뇨 질환 개선 효능)

  • Purpose: Dendropanax morifera Leveille (DML) exhibits diverse biological and pharmacological activities, including anti-oxidative effect, anti-cancer activity, hepatoprotection, immunological stimulation, and bone regeneration. As part of the identification for novel functions of DML, we investigated the therapeutic effects of DML on diabetes induced by streptozotocine (STZ) treatment. Methods: First, the four extracts including the water extract of leaf (DLW), the ethanol extract of leaf (DLE), the water extract of stem (DSW), and the ethanol extract of stem (DSE) were collected from the leaf and stem of DML using a hot water and ethanol solvent. Alterations in body weight, glucose concentration, insulin level, and pancreatic islet structure were investigated in diabetic mice after treatment with extracts of DML for 2 weeks. Results: Among four extracts, the highest level of total polyphenols and total flavonoids was detected in DLW, while the lowest level of these was measured in DSE. The radical scavenging activity was also higher in DLW than in the other three extracts at the concentration of $25-100{\mu}g/mL$, although this activity was maintained at a constant level in all groups at the concentration of $500{\mu}g/mL$. Based on the results of anti-oxidant activity, DLW and DLE were selected for examination of anti-diabetic effects in a diabetes model. Body weight was gradually decreased in all STZ treated groups compared with the No treated group. However, four STZ/DML treated groups maintained a high level of body weight during 7-14 days, while the STZ/vehicle treated group showed a gradual decrease of body weight during the same period. Also, a significant decrease or increase in the concentration of glucose and insulin in the blood of the diabetes model was detected in a subset of groups, although the highest increase was detected in the STZ/DLE-200 treated group. In addition, the histological structure of pancreatic islet was significantly recovered after treatment with DLW and DLE. Conclusion: These results suggest that DLW and DLE may contribute to attenuation of clinical symptoms of diabetes as well as prevent the destruction of pancreatic ${\beta}$-cells in STZ-induced diabetes mice.

Bulnesia Sarmienti Aqueous Extract Inhibits Inflammation in LPS-Stimulated RAW 264.7 Cells (RAW 264.7세포에서 lipopolysaccharide로 유발시킨 염증반응에 대한 Bulnesia sarmienti 열수추출물의 억제효과)

  • Bulnesia sarmienti (BS), a traditional South American herbal medicine native to Gran Chaco, has been used to treat various human ailments. We investigated the cytotoxic activities and the inhibitory effects of BS bark extract(0, 50, 100 and $200\;{\mu}g/\;mL$) on the production of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), cyclooxygenase (COX) and proinflammatory cytokines ($IL-1{\beta}$, IL-6 and $TNF-{\alpha}$) in the lipopolysaccharide (LPS) (100 ng/ml)-stimulated murine macrophage cell line RAW264.7. The levels of NO, COX, PGE2 production and proinflammatory cytokines ($IL-1{\beta}$, IL-6 and $TNF-{\alpha}$) were measured by ELISA kit. Cell viability, as measured by the MTT assay, showed that BS extract had no significant cytotoxicity in RAW264.7 cells. BS extract significantly inhibited the LPS-induced NO, $PGE_2$ and COX production accompanied by an attenuation of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ formation in macrophages. These results suggest that BS extract has potential as an herbal medicine for the treatment of inflammatory diseases.

The Extended Site Assessment Procedure Based on Knowledge of Biodegradability to Evaluate the Applicability of Intrinsic Remediation (자연내재복원기술(Intrinsic Remediation)적용을 위한 오염지역 평가과정 개발)

  • The remediation of contamiated sites using currently available remediation technologies requires long term treatment and huge costs, and it is uncertain to achieve the remediation goal to drop contamination level to either back-ground or health-based standards by using such technologies. Intrinsic remediation technology is the remediation technology that relies on the mechanisms of natural attenuation for the containment and elimination of contaminants in subsurface environments. Initial costs for the intrinsic remediation may be higher than conventional treatment technologies because the most comprehensive site assessment for intrinsic remediation is required. Total remediation cost, however may be the lowest among the presently employed technologies. The applicability of intrinsic remediation in the contaminated sites should be theroughly investigated to achieve the remedial goal of the technology. This paper provides the frame of the extended site assessment procedure based on knowledge of biodegradability to evaluate the applicability of intrinsic remediation. This site assessment procedure is composed of 5 steps such as preliminary site screening, assessment of the current knowledge of biodegradability, selecting the appropriate approach, analyzing the contaminant fate and transport and planning the monitoring schedule. In the step 1, followings are to be decided 1) whether to go on the the detailed assessment or not based on the rules of thumb concerning the biodegradability of organic compounds, 2) which protocol document is selected to follow for detailed site assessment according to the site characteristics, contaminants and the relative distance between the contamination and potential receptors. In the step 2, the database for biodegradability are searched and evaluated. In the step 3, the appropriate biodegradability pathways for the contaminated site is selected. In the step 4, the fate and transport of the contaminants at the site are analyzed through modeling. In the step 5, the monitoring schedule is planned according to the result of the modeling. Through this procedure, users may able to have the rational and systematic informations for the application of intrinsic remediation. Also the collected data and informations can be used as the basic to re-select the other remediation technology if it reaches a conclusion not to applicate intrinsic remediation technology at the site from the site assessment procedure.
