Effect of Anti-Browning Agent Application on the Improvement of Quality Maintenance for Minimally Processed Pear Slices (갈변 방지제 처리가 최소 가공한 배 절편의 저장성 향상에 미치는 영향)

  • This study was conducted to increase the keeping quality of the minimally processed pear slices during the cold storage. Korean pears, 'Shin-go (Niitaka)', 'Chu-hwang' and 'Won-hwang’ were immersed in 1% ascorbic acid, 0.1% calcium propionate, 1% citric acid, 0.2% N-acetyl cysteine and 0.5 M 4-hexylresorcinol solution for 3 three minutes and then stored at $1{\pm}0.5^{\circ}C$ for 10 days. We have also examined into the firmness and the color difference of pears slices as affected by the application of the anti-browning agents. The firmness of pears slices which that were immersed in 0.1% calcium propionate and 0.2% N-acetyl cysteine solution were not significant at did not differ significantly after 10 days after of cold storage. However, the ${\Delta}E^*$ values of 'Shin-go' slices which that were treated with 0.1% calcium propionate and 0.2% N-acetyl cysteine solution and stored for 10 days decreased by 3.18 and 3.83, when compared with that in of the control, which decreased by 6.36. The ${\Delta}E^*$ values of 'Chu-hwang' slices which that were treated with 0.2% N-acetyl cysteine and 0.1% calcium propionate solution had the slight difference by differed by only 2.09 and 2.14, when compared with that in of the control, which differed by 3.04. The ${\Delta}E^*$ values of 'Won-hwang' slices which that were treated with 1% citric acid and 0.5 M 4-hexylresorcinol were 4.49 and 5.83, respectively, while that in of the control decreased by 8.95. It was assumed that the anti-browning agent treatment had the different activities among varieties of the pear varieties slices, however, application of 0.2% N-acetyl cysteine and 0.1% calcium propionate application had greater the higher antibrowning activity.

Volatile Flavor Components in Various Varieties of Pear (Pyrus pyrifolia N.) (배의 품종별 휘발성 향기성분)

  • Volatile flavor components in three varieties (shingo(niitaka), mansamgil (okusankichi) and chuwhang pears) of Pear (Pyrus pyrifolia N.) were extracted for 24 hours with pentane-diethylether (1 : 1, v/v) using the LLEP (liquid-liquid extraction & perforation). Neutral fraction was separated from the extract and then analyzed by GC-FID and GC/MS equipped with a fused silica capillary column (Carbowax 20M, HP). Individual components were identified by mass spectrometry and their retention indices. The totals of 52, 47 and 22 volatiles were identified in shingo, mansamgil and chuwhang pears, respectively. Ethyl acetate, propyl acetate, hexanal, 1-hexanol, ethyl butanoate, ethyl-3-hydroxy butanoate, ethyl-2-hydroxy propanoate were the main components in each samples, though there were several differeces in composition of volatile compounds. Total contents of volatile components isolated in shingo, mansamgil and chuwhang pears were 6.972, 2.776 and 2.653 mg/kg of pears.

Effect of 1-Methylcyclopropene Treatment on the Fruit Quality of Pear c.v. "Hwasan" during the Storage at Ambient Temperature (1-MCP 처리가 '화산' 배의 상온 저장 중 품질에 미치는 영향)

  • We investigated the effects of 1-methylcyclopropene (1-MCP) treatment (0, 0.5, and 1.5 ppm) for 16 hours, followed by storage at ambient temperature, on the qualities of pears (Pyrus pyrifolia c.v. "Hwasan"). Color differences, as measured using the Hunter L, a, and b values, showed the greatest decreases before storage, and 8 weeks after storage, when fruit was exposed to 1.5 ppm 1-MCP. The sensory qualities of c.v. "Hwasan" pears were good until 4 weeks of storage after treatment with 0 ppm 1-MCP, until 6 weeks when 0.5 ppm 1-MCP was employed, and until 7 weeks when 1.5 ppm 1-MCP treatment was performed. Fruit and storage quality in terms of all of hardness, sugar content, PPO activity, respiration amount, and ethylene production increased in the 8 weeks after application of 1.5 ppm 1-MCP. Consequently, it seems that 1-MCP treatment, especially at 1.5 ppm, before storage of pears at ambient temperature, effectively improves the quality of pears of c.v. "Hwasan".

Species, Damage and Population Density of Pseudococcidae Injuring Pear Fruits (배를 가해하는 가루깍지벌레류와 가해양상 및 밀도변동)

  • This study investigated species, damage to fruits and population density of Pseudococcidae on pear in Naju, Chonnam province from 1990 to 1991. Three species, Planococcus kraunhiae, Pseudococcus comstocki and Crisicoccus matsumotoi, were identified. Of them P. kraunhiae was most abundant as 80.6%. Rate of fruit damages was greatest in var. Poongsoo as 39.7% but locations of field did not affect the rates. Degree of damages on Singo(late var.), Poongsoo (midseason var.) and Haengsoo(early var.) was 21.2, 18.2 and 13.3%, respectively. Population density of scales within paper bag of fruits began to increase from mid-June and showed the first at mid-July, the second at mid-August and third peak at early October. Rate of fruit damages was 48% at the first peak and 50.6% at 7 days after the second peak. Density increased from early October again but migration was not active presumably because of cold weather.

Studies on Pear Phylloxera (Aphanostigma iakusuiense Kishida) (1) On the Control Methods (콩가루벌레 (Aphanostigma iakusuiense)에 관한 연구 (I) 방제시험을 중심으로)

  • Life history and control methods of Aphanostigma iakusuiense Kishida were studied during April to November, 1974, in the Naju pear orchard area, Cholla Namdo. The results are summarized as follows: 1. More than $60\%$ of wintering eggs were found at the height of 1.5 to 2.0m from the soil surface, with approximately $10\%$ at 0.5 to 1.5m and 1.5 to 2.5m respectively. 2. There was little difference in the size of egg masses and hatching ratio $(12.7-13.7\%)$ between the varieties of host trees, Mansamkil and Kooksoo. 3. Hatching was not observed until April 6, but hatches of $12.8\%$ on April 13 and $90.6\%$ on May 4 were found. 4. During the hatching season in early spring, 8 chemicals were applied to test effectiveness of control. C8514, Metasystox, Acar 338, and Parathion were more effective than either Sevin or Malix for control of this insect. 5. Highest populations of pear phylloxera ocurred from August 20 to September 20 at which time levels of eggs and larvae were simillar in abundance. 6. In tests for control of this insect after in paper bags, DDVP, Loxon, C8514, Sevin, and Binapacryl were more effective than either EPN or Otran. 7. Predacious mite, Protolaelape pygmaeus Miller, and several species of lady beetles (Coccinellidae) preying on this insect as natural enemies were collected. The predation ratio of mites varied according to region: that is 0.77 to $18.6\%$ in Bia Orchard; $7.88-48.7\%$ in Noan Orchard and almost none in Youngsanoo Orchard. 8. The varietal difference in fruit damage by this insect was in the order of Choseng Chuck, Kooksoo, followed by Sin Ko, Mansamkil, Keumchon Choo, and Changsimrang.

MARYBLYT Study for Potential Spread and Prediction of Future Infection Risk of Fire Blight on Blossom of Singo Pear in Korea (우리나라 신고배 화상병 꽃감염 확산 가능성 및 미래 감염위험 예측을 위한 MARYBLYT 연구)

  • Since fire blight (Erwinia amylovora) firstly broke out at mid-Korea in 2015, it is necessary to investigate potential spread of the invasive pathogen. To speculate environmental factors of fireblight epidemic based on disease triangle, a fire blight predicting program, MARYBLYT, was run with the measured meteorological data in 2014-2017 and the projecting future data under RCP8.5 scenario for 2020-2100. After calculating blossom period of Singo pear from phenology, MARYBLYT was run for blossom blight during the blossom period. MARYBLYT warned "Infection" blossom blight in 2014-15 at Anseong and Cheonan as well as Pyungtak and Asan. In addition, it warned "Infection" in 2016-17 at Naju. More than 80% of Korean areas were covered "Infection" or "High", therefore Korea was suitable for fire blight recently. Blossom blight for 2020-2100 was predicted to be highly fluctuate depending on the year. For 80 years of the future, 20 years were serious with "Infection" covered more than 50% of areas in Korea, whereas 8 years were not serious covered less than 10%. By comparisons between 50% and 10% of the year, temperature and amount of precipitation were significantly different. The results of this study are informative for policy makers to manage the alien pathogen.

Study on Evaluation of Carbon Emission and Sequestration in Pear Orchard (배 재배지 단위의 탄소 배출량 및 흡수량 평가 연구)

  • Objective of this study was to evaluate the carbon budget on 40 years old pear orchard at Naju. For carbon budget assessment, we measured the soil respiration, net ecosystem productivity of herbs, pear biomass and net ecosystem exchange. In 2015, pear orchard released about $25.6ton\;CO_2\;ha^{-1}$ by soil respiration. And $27.9ton\;CO_2\;ha^{-1}$ was sequestrated by biomass growth. Also about $12.6ton\;CO_2\;ha^{-1}$ was stored at pruning branches and about $5.2ton\;CO_2\;ha^{-1}$ for photosynthesis of herbs. As a result, 25.6 ton of $CO_2$ per ha is annually released to atmosphere. At the same time about 45.7 ton of $CO_2$ was sequestrated from atmosphere. When it sum up the amount of $CO_2$ release and sequestration, approximately $20.1ton\;CO_2\;ha^{-1}$ was sequestrated by pear orchard in 2015, and it showed no significant differences with net ecosystem exchanges ($17.8ton\;CO_2\;ha^{-1}\;yr^{-1}$) by eddy covariance method with the same period. Continuous research using various techniques will help the understanding of $CO_2$ dynamics in agroecosystem and it can be able to present a new methodology for assessment of carbon budget in woody crop field. Futhermore, it is expected that the this study can be used as the basic data to be recognized as a carbon sink.

Relationship of Resistance to Benzimidazole Fungicides with Mutation of β-Tubulin Gene in Venturia nashicola (Benzimidazole계 살균제에 대한 배 검은별무늬병균 Venturia nashicola 의 저항성과 β-Tubulin 유전자 돌연변이와의 관계)

  • Pear scab caused by Venturia nashicola has been reported as an important disease of pear resulting in lowering the quality of pear fruits. In this study, it was conducted to investigate the relationship between resistance of V. nashicola and mutation of ${\beta}$-tubulin gene and the fungicide resistance in field isolate group in benzimidazole fungicides. Responce of V. nashicola to carbendazim could be classified into 3 groups as sensitive that does not grow at all on PDA amended with $0.16{\mu}g/ml$ of carbendazim, low resistance that could not grow in $4.0{\mu}g/ml$ medium, and high resistance that can grow even at $100{\mu}g/ml$. Thirty isolates of V. nashicola collected from 3 regions as Wonju, Naju, and Okcheon were highly resistant to carbendazim. Analysis of the nucleotide sequence of ${\beta}$-tubulin gene of V. nashicola showed that there was no difference in the nucleotide sequence between the sensitive and the low-resistant isolate, but GAG at codon 198 (glutamic acid) was replaced with GCG (alanine) in the high-resistant isolate. Among 10 isolates obtained from the Okcheon, 5 isolates showed the substitution of glycine for glutamic acid, which were resistant to carbendazim, but more sensitive to the mixture of carbendazim and diethofencarb than others. Through these results, all isolates of V. nashicola isolated in pear orchard were found to be resistant to benzimidazoles. Also, mutants E198A and E198G at ${\beta}$-tubulin were found to be important mechanisms of V. nashicola resistance against benzimidazole fungicides.

The Study on Carbon Budget Assessment in Pear Orchard (배 재배지의 탄소수지 산정에 관한 연구)

  • This study was conducted to find out the methodology of carbon budget assessment among soil, atmosphere and plant. Soil respiration, net ecosystem productivity of herbs and net ecosystem productivity of woody plants have been measured in 30 years old pear orchard at Naju. Closed Dynamic Chamber (CDC) method was used to measure soil respiration and net ecosystem productivity of herbs. Net ecosystem productivity of woody plant (pear) was determined by eddy covariance method using the EddyPro (5.2.1) program. As for soil respiration, $429.1mgCO_2m^{-2}h^{-1}$ was released to atmosphere and sensitivity of soil temperature ($Q_{10}$) was 2.3. In case of herbs, respiration was superior to photosynthesis during measurement period. From 20 to 24 Jun 2015, the sum of absorbed and released $CO_2$ by herb's photosynthesis and respiration was $156.1mgCO_2m^{-2}h^{-1}$. Woody plants showed the $680.1mgCO_2m^{-2}h^{-1}$ of absorption by photosynthesis. In a farm scale, the sum of soil respiration, and net ecosystem productivity of herbs and woody plants was $0.04tonCO_2ha^{-1}$ during the measurement period, and it showed that pear orchard act as a $CO_2$ sink. This study using various approaches is expected to present a methodology for evaluating the carbon budget of perennial woody crop plantations.

Production and Developmental Pattern of Embryogenic Callus in Oenanthe javanica ($B_{L.}$) DC. (미나리 체세포 배발생 캘러스의 획득과 발달 형태)

  • This experiment was canted out to obtain embryogenic callus and to understand developmental mechanism of somatic embryogenesis in Oenanthe javanica ($B_{L.}$) DC. experiments included the examination of explant source and media for embryogenic callus production and the observation of developmental pattern of embryogenic cells and non-embryogenic cells. Embryogenic calli were formed on zygotic pro-embryos together with their endosperms when they were cultured on Ms media containing 1.0mg/L 2,4-D. Embryogenic calli were also formed on the intact surface in vitro grown stem or petiole segmentsafrer 6-8 weeks of culture, whereas non-embryogenic calli were formed on cut surfaces of the stem and petiole after 2 weeks of culture. Non-embryogenic calli were rhizogenic in suspension and solid media culture.
