Genome-wide association study for the interaction between BMR and BMI in obese Korean women including overweight

  • BACKGROUND/OBJECTIVES: This is the first study to identify common genetic factors associated with the basal metabolic rate (BMR) and body mass index (BMI) in obese Korean women including overweight. This will be a basic study for future research of obese gene-BMR interaction. SUBJECTS/METHODS: The experimental design was 2 by 2 with variables of BMR and BMI. A genome-wide association study (GWAS) of single nucleotide polymorphisms (SNPs) was conducted in the overweight and obesity (BMI > $23kg/m^2$) compared to the normality, and in women with low BMR (< 1426.3 kcal/day) compared to high BMR. A total of 140 SNPs reached formal genome-wide statistical significance in this study (P < $1{\times}10^{-4}$). Surveys to estimate energy intake using 24-h recall method for three days and questionnaires for family history, a medical examination, and physical activities were conducted. RESULTS: We found that two NRG3 gene SNPs in the 10q23.1 chromosomal region were highly associated with BMR (rs10786764; $P=8.0{\times}10^{-7}$, rs1040675; $2.3{\times}10^{-6}$) and BMI (rs10786764; $P=2.5{\times}10^{-5}$, rs10786764; $6.57{\times}10^{-5}$). The other genes related to BMI (HSD52, TMA16, MARCH1, NRG1, NRXN3, and STK4) yielded P < $10{\times}10^{-4}$. Five new loci associated with BMR and BMI, including NRG3, OR8U8, BCL2L2-PABPN1, PABPN1, and SLC22A17 were identified in obese Korean women (P < $1{\times}10^{-4}$). In the questionnaire investigation, significant differences were found in the number of starvation periods per week, family history of stomach cancer, coffee intake, and trial of weight control in each group. CONCLUSION: We discovered several common BMR- and BMI-related genes using GWAS. Although most of these newly established loci were not previously associated with obesity, they may provide new insights into body weight regulation. Our findings of five common genes associated with BMR and BMI in Koreans will serve as a reference for replication and validation of future studies on the metabolic rate.

Factors Predictive of Voice Therapy Outcome in Patients with Unilateral Vocal Fold Paralysis (일측성 성대마비 환자에서 음성치료 효과를 예측할 수 있는 인자)

  • Background and Objectives : Unilateral vocal fold paralysis is generally treated using injection laryngoplasty or voice therapy. However, the decision of treatment method is dependent on clinician's preference and hospital facilities without specific criteria. The purpose of the study was to examine factors predictive of voice therapy outcome in patients with unilateral vocal fold paralysis. Materials and Method : 38 patients diagnosed as unilateral vocal fold paralysis, aged from 24 to 81 years and undergone voice therapy more than 1 month were included. After 3 to 12 (mean 5.1) sessions of voice therapy, subjects had divided into responder group (RG, 28 patients) and non-responder group (NRG, 10 patients) according to G scale change. Paramters of perceptual assessment, acoustic and aerodynamic measure, and videostroboscopy were compared between two groups, and factors predictive of voice therapy result were analyzed. Results : RG patients showed significantly reduced rough, breathy, asthenic voice after voice therapy. Change of MPT and MFR was more substantial in RG than in NRG. By videostroboscopy, RG patients showed significantly more mucosal wave symmetry, glottal closure, reduced glottal gap index during the closed phase of phonation, while NRG patients showed more occurrences of abnomal supraglottic activities during phonation (p < 0.05). Poor outcome of voice therapy significantly associated with increased asthenic scale, short MPT, and less glottal closure (p=0.02). In addition, 90% of patients with MPT more than 5 seconds were in RG, whereas 56% of patients with MPT less than 5 secondes were in RG. Conclusion : Voice therapy is useful for large proportion of patients with unilateral vocal fold paralysis as an initial treatment method. However, patients with large asthenia scale, large glottic gap or MPT less than 5 seconds tend to have poor voice therapy outcome, and early injection laryngoplasty maybe recommended for these patients.

  • Embryonic stem (ES) cell-derived cardiomyocytes (ESCMs) must be specifically purified in order to prevent teratoma formation, and this confusing issue has hampered their clinical application. We therefore investigated a technique to generate pure labeled ESCMs for possible use in cardiac repair. We generated transgenic ES cell lines expressing enhanced green fluorescent protein (EGFP) under the transcriptional control of the $\alpha$-cardiac myosin heavy chain ($\alpha$-MHC) promoter. Differentiated EGFP-positive ES cells displayed characteristics of CMs. Furthermore, neuregulin-1 (NRG-1) upregulated the expression of the cardiac-restricted transcription factors Nkx2.5 and GATA-4, as well as differentiated CM factors ($\alpha$-MHC, $\beta$-MHC). Immunohistochemistry demonstrated that NRG-1 increased expression of cardiac-specific troponin T in the beating foci of the embryoid bodies. This work revealed a potential method for specifically labeling and enriching ESCMs by combining genetically-engineered ES cell clones and exogenous growth factor treatment.

  • Development of oligodendrocytes and the generation of myelin internodes within the spinal cord depends on regional signals derived from the notochord and axonally derived signals. Neuregulin (NRG)-1, localized in the floor plate as well as in motor and sensory neurons, is necessary for normal oligodendrocyte development. Oligodendrocytes respond to NRGs by activating members of the erbB receptor tyrosine kinase family. Here, we show that erbB2 is not necessary for the early stages of oligodendrocyte precursor development, but is essential for proligodendroblasts to differentiate into galactosylcerebroside-positive (GalC+) oligodendrocytes. In the presence of erbB2, oligodendrocyte development is normal. In the absence of erbB2 (erbB2-/-), however, oligodendrocyte development is halted at the proligodendroblast stage with a >10-fold reduction in the number of GalC+ oligodendrocytes. ErbB2 appears to function in the transition of proligodendroblast to oligodendrocyte by transducing a terminal differentiation signal, since there is no evidence of increased oligodendrocyte death in the absence of erbB2. Furthermore, known survival signals for oligodendrocytes increase oligodendrocyte numbers in the presence of erbB2, but fail to do so in the absence of erbB2. Of the erbB2-/- oligodendrocytes that do differentiate, all fail to ensheath neurites. These data suggest that erbB2 is required for the terminal differentiation of oligodendrocytes and for development of myelin.

  • Background: Arteriovenous malformations (AVMs) are rare diseases comprising abnormally dilated arteries and veins with an absence of a capillary network. Since these diseases are intractable after diagnosis, various treatment strategies have been examined, with continuous efforts to identify target genes. Here, we report relevant new target genes selected via gene microarray. Methods: Endothelial cells were isolated from samples collected from three patients with AVM and three healthy individuals, followed by microarray analysis. Additionally, quantitative PCR was performed to select genes highly relevant to AVM. Results: In the vascular endothelial cells derived from the tissues of patients with AVM, the expression of ANGPT1, ANGPT2, DLL4, IL6, NRG1, TGFBR1, and VEGFA was typically higher compared to those derived from normal tissues. Conclusion: Seven candidate genes were selected to analyze the pathophysiological mechanism of AVM. These results may aid in future directions of diagnosis and treatment.

  • The purpose of this study was to investigate blood pressure and its related factors of middle-aged women in Korea. For the collection of data, 410 women aged between 45-54 were selected through stratified sampling techniques at urban and rural areas in Korea. They were interviewed from Oct. 15 through Nov. 14 by use of questionare. Descriptive statistical methods, multiple stepwise regression methods and multiple discriminant analysis by S.P.S.S. were used to analyze the data. The obtained result are summarized as follows. 1. Systolic B.P was 119.2 mmHg. mean pressure, Diastolic B.P. was 77.7 mmHg. mean pressure. 2. Systolic B.P. was related with the area and the ownership of house; it was accepted (P<05) 3. The most important factor affecting the B.P of middle-aged women were weight It was related with systolic pressure(0.176) and diastolic pressure(0.157) That is, this study showed that for the middle-aged women(45-54 yrs.), health education about weight control which can prevent ascending B.P should be demanded.

  • To detect Single nucleotide polymorphisms (SNP) markers associated with Bovine viral diarrhea virus (BVDV) and Bovine respiratory syncytial virus (BRSV) S/P ratio in Korean Holstein dairy cattle, Genome-wide association study (GWAS) was performed using Illumina BovineSNP50 Beadchip. The number of phenotype data and genotype data were 107, and 294. respectively. Phenotype data were collected for four periods (0 week, 1 week, 4 week, 24 week) after having vaccinated (0 week no vaccinated period). A total of 36,257 SNPs was remained after quality control had been done by PLINK. The result of GWAS showed 6 SNP markers (BTB-01704243, BTB-01594395, ARS-BFGL-NGS-118070, ARS-BFGL-NGS-111365, BTA-65410-no-rs, Hapmap38331-BTA-61256) under BVDV and 4 SNP markers (ARS-BFGL-NGS-109861, Hapmap53701-rs29017064, ARS-BFGL-NGS-71055, BTA-11232-no-rs) under BRSV. And also, 10 candidate genes found through 10 SNP markers (TBX18, CEP162, PAFAH1B1, METTL16, BRCA1, RND2, POLK, ENSBTAG00000051724, ADAM18, NRG3).

Effects of the Cultivated Areas on Antioxidant Compounds and Activities of Proso Millet (Panicum miliaceum L.) (재배지역이 기장(Panicum miliaceum L.)의 항산화성분 및 활성에 미치는 영향)

  • Effects of antioxidant compounds and antioxidant activities of proso millet cultivated in different areas were determined. The cultivated areas were Milyang (plain area of interior), Wonju (mountainous territory) and Sinan (coastal area), and cultivated varieties were Hwanggeumgijang (HGG), Byeoruk-gijang (BRG), Norangchal-gijang (NRG), Bulgeun-gijang (BGG), Whin-gijang (WG). The highest total polyphenol contents of methanolic extracts were 2.54 and 2.65 mg/g in BGG and BRG produced in Sinan, respectively. The highest total flavonoid content were 2.66 and 2.59 mg/g in BGG and BRG produced in Sinan, respectively. The highest total tannin contents were 1.87 and 1.42 mg/g in BRG and NRG produced in Sinan. The DPPH radical-scavenging activity was the highest value of BGG (8.54 mg TE/g) and BRG (8.53 mg TE/g) produced in Sinan. The ABTS radical-scavenging activity was the highest value of BGG and BRG of 19.48 and 19.29 mg TE/g in the grains produced in Sinan, respectively. Generally, there was difference in antioxidant compound contents on the methanolic extracts of proso millet between the cultivated areas and varieties.

Analysis of Microbiological Hazards From Working Gloves Used in the Processing of Shucked Pacific Oyster Crassostrea gigas (참굴(Crassostrea gigas) 탈각 공정에서 작업자 장갑에 의해 발생하는 미생물학적 위해요소 분석)

  • This study assessed the microbiological hazards of gloves worn during the shell shucking process of the oyster Crassostrea gigas, and we suggest an in situ method for minimizing microbial contamination. The study consisted of two groups, one in which the working gloves were periodically replaced (PRG) with new gloves, and another in which the gloves were not replaced (NRG). In the PRG group, gloves were replaced every 2 h during 8 h of processing. Food pathogenic bacteria Escherichia coli, Salmonella species, and Listeria monocytogenes were not found in any samples, including gloves and shucked oysters. However, Staphylococcus aureus (SA) was detected in some samples, and the contamination levels were correlated with the working time and the regular replacement of gloves. SA was not detected on gloves or oysters of the PRG group. However, it was detected in the range of <$15CFU/15cm^2$ to $2.9{\times}10^2CFU/15cm^2$ on gloves after 6 h of continuous work, and from <$15CFU/15cm^2$ to $2.23{\times}10^2CFU/15cm^2$ on oysters after 8 h. These results indicate that the SA contamination in shucked oysters originated from the working gloves, and that replacement of working gloves every 2-4 h will minimize SA contamination in oyster products.

Antioxidant Compounds and Antioxidant Activities of Different Varieties of Foxtail Millet and Proso Millet according to Cultivation Time (품종 및 재배시기에 따른 조와 기장의 항산화성분 및 항산화활성)

  • Effects of cultivation time on the antioxidant contents and activities of foxtail millet and proso millet were determined. The cultivation times were early-season (ES), normal-season (NS), and late-season (LS), and the cultivated varieties were Hwanggeum-cho (HGC), Cheongcha-cho (CCC), Samdamae (SDM), Gyeongkwan 1 (GK 1), Gyeongkwan 2 (GK 2), Ganghae-cho (GHC), Hwanggeum-gijang (HGG), Manhongchal-gijang (MHCG), Byeoruk-gijang (BRG), Norangchal-gijang (NRG), and Whin-gijang (WG). The total polyphenol, flavonoid, tannin content, and DPPH and ABTS radical scavenging activities of the methanolic extracts of foxtail millet and proso millet showed significant differences according to cultivation variety and time. The highest total polyphenol content of foxtail millet was 29.33 mg of gallic acid equivalent (GAE)/g of sample in GK 1 at LS, whereas that of proso millet was 23.48 mg of GAE/g of sample in BRG at LS. The highest total flavonoid content of foxtail millet was 2.12 mg of catechin equivalent (CE)/g of sample in CCC at ES, whereas that of proso millet was 4.49 mg of CE/g of sample in BRG at LS. The highest total tannin content of foxtail millet was 14.07 mg of tannic acid equivalent (TAE)/g of sample in SDM at LS, whereas that of proso millet was 15.59 mg of TAE/g of sample in BRG at LS. The highest DPPH radical scavenging activity of foxtail millet was 7.71 mg of TE/g of sample in CCC at NS, whereas that of proso millet was 12.66 mg of TE/g of sample in BRG at LS. The highest ABTS radical scavenging activity of foxtail millet was 8.05 mg of TE/g of sample in GK 1 at LS, whereas that of proso millet was 34.46 mg of TE/g of sample in BRG at LS. Generally, HGC, GK 1, and GHC had more beneficial effects at LS than ES or NS, whereas CCC, SDM, and GK 2 had more beneficial effects at NS than ES or LS, and proso millet had more beneficial effects at LS than ES or NS.
