The Experimental study on the Immuno-regultory effect of Notopterygii Rhizoma Extract on OVA-induced asthma in mice (강활추출물이 OVA-induced Asthma Mouse Model의 면역조절에 미치는 영향)

  • Objectives : The aim of this study was to investigate the asthma-suppressive and immuno-regulatory effect of Notopterygii Rhizoma(NR) extract on OVA(ovalbumin)-induced asthma in mice. Methods : C57BL/6 mice out of all the experimental groups, except the Normal group and the NRI group, were sensitized and challenged with OVA. C57BL/6 mice were exposed to OVA three times a week for 12 weeks and analyzed by flow cytometer, ELISA, H&E stain. Results : The concentrations of IL-4, IL-5, IL-13, IgE in serum of the OVA-NRII group decreased significantly compared with those of the OVA-Control group. The number of $Gr-1^+/CD11b^+$, $CCR3^+$, $CD3^+/CD19^+$, $CD3e^+/CD69^+$cells in the OVA-NRI group decreased significantly compared with those of the OVA-Control group. The collagen accumulation in the lung sections of the OVA-NRII group decredased signi- ficantly compared with that of the OVA-Control group. Conclusions : These results suggest that Notopterygii Rhizoma(NR) would be a effective candidate for herbal-originated anti-asthmatic drug. However, this drug should be further studied for characterization of the accurate action and underlying mechanism using variant disease model in the future.

Anti-thrombotic Activities of Hot-water Extracts Prepared from Various Parts of Nelumbo nucifera Gaertner (다양한 부위의 연(Lotus) 열수 추출물의 항혈전 활성)

  • To investigate the anti-thrombotic activities of the lotus (Nelumbo nucifera Gaertner), various hot-water extracts were prepared from the leaf, pod of seed (PS), seed, embryo of seed (ES), root, and the node of root (NR) of the lotus. The highest extraction ratio was found in the NR (20.3%), followed by the seed, root, leaf, ES, and PS. These extracts had pH and acidity levels ranging from 5.6~6.5 and 0.06~0.20%, respectively. The seed extract showed 70% brix, whereas the leaf and PS extracts showed less than 0.1% brix. The highest contents of total polyphenol (179.7 mg/g), total flavonoids (161.4 mg/g), and reducing sugar (161.4 mg/g) were observed in the leaf extract, and the highest total sugar content (873.0 mg/g) in the seed extract. Anti-coagulation assay of the extracts of NR, leaf, and PS showed strong activities. In particular, at a concentration of 5 mg/ml, the PS extract had 15-fold extended thrombin, prothrombin, and activated partial thromboplastin times. However, only the ES extract showed activities inhibitory to platelet aggregation, with treatment with 0.25 mg/ml of ES extract decreasing platelet aggregation to 25.1%, a reduction comparable to that effected by aspirin. The extracts other than the seed extract showed no hemolysis activities against human RBC at treatments of up to 1 mg/ml. These results suggest that the NR, PS, seed, and ES, all byproducts of the lotus agriculture industry, have high potential as novel sources of anti-thrombotic agent.

Syntheses and Spectroscopic properties of Palladium(II) Complexes with Bidentate Aminophosphine of N,N-Dialkyl-N'-diphenylphosphinodiaminoethane

  • Several new palladium(II) complexes of aminophosphines, $(Pd(L)X_2)$, (L = $Ph_2PNH$ ++ $NR_2$; R = $CH_3(L_1)$, $C_2H_5(L_2)$ : X = Cl, Br, I, and NCS) that contain two different donor atoms of nitrogen and phosphorus as ${\pi}$-electron acceptor, were synthesized and characterized by conductivity measurement, ir, and UV/Vis-spectra. For the dithiocyanatopalladium(II) complexes with aminophosphines, it was confirmed that the thiocyanate group trans to phosphorus is coordinated as Pd-NCS mode and the one trans to nitrogen as Pd-SCN mode, and the aminophosphines form six-membered chelate ring. The spectra of dihalogenopalladium(II) complexes with aminophosphines show that the band maxima are shifted to the short wavelengths as the concentration is decreased.

Biochemical and cultural characteristics of mineral-solubilizing Acinetobacter sp. DDP346 (미네랄 가용화능을 갖는 Acinetobacter sp. DDP346의 생화학적 및 배양학적 특성)

  • In this study, to select strains suitable as microbial agent from among rhizosphere microorganisms present in rhizosphere soil and roots, the mineral solubilization ability, antifungal activity against 10 types of plant pathogenic fungi, and plant growth-promoting activity of rhizosphere microorganisms were evaluated. As a result, DDP346 was selected because it has solubilization ability of phosphoric acid, calcium carbonate, silicon, and zinc; nitrogen fixing ability; production ability of siderophore, indole-3-acetic acid, and aminocyclopropane-1-carboxylate deaminase; and antifungal activity against seven types of plant pathogenic fungi. DDP346 showed a 99.9% homology with Acinetobacter pittii DSM 21653 (NR_117621.1); phylogenetic analysis also revealed a close relationship with Acinetobacter pittii based on the 16S rRNA base sequence. The growth conditions of DDP346 were identified as temperatures in the range of 10-40 ℃, pH in the range of 5-11, and salt concentrations in the range of 0-5%. In addition, a negative correlation coefficient (r2 = -0.913, p <0.01) was shown between pH change and the solubilized phosphoric acid content of Acinetobacter sp. DDP346, and this is assumed to be due to the organic acid generated during culture. Consequently, through the evaluation of its mineral solubilization ability, antifungal activity against plant pathogenic fungi, and plant growth-promoting activity, the potential for the utilization of Acinetobacter sp. DDP346 as a multi-purpose microbial agent is presented.

Genes expression by using cDNA Microarray in Whallak-tang (활락탕(活絡湯)의 cDNA Microarray를 이용한 유전자 발현에 미치는 영향)

  • Objective This study was undertaken to determine the effect of Whallak-tang on expression of CD/cytokine Genes. Methods The expression of CD/Cytokine Genes were examined by cDNA microarray using the human mast cell line(HMC-1). Results The expression of ATP5F1, FLJ20671, unknown, KIAA0342, OAS2, unknown genes were increased in $200{\sim}300%$ range. The expression of unknown, MDS006, IFITM1, MRPL3, ZNF207, FTH1, FBP1, NRGN, NR1H2, KIAA0747 genes were decreased in $0{\sim}33%$ range. Conclusion These results would provide important basic data on the possibility of the clinical treatment of Whallak-tang in musculoskeletal disease.

Effects of $NO_3^-$ Gradients on Nitrogen Fixation, Nitrate Reduction and Ureide Content of Soybean (대두의 공소개정, 공산환원 및 Ureide함량에 미치는$NO_3^-$의 영향)

  • Soybean, inoculated with effective Rhizobium japonicum 110, were grown by sand culture with nutrient solution containing either of 0, 1, 3, 10 or 30mM NO3-/l, and analyzed growth characteristics, NR activity, N2-fixation activity, and changes of ureide contents during the growing period. The amount of nodule formation decreased abruptly by nitrate treatment, the maximum nodule dry weight was 1.59, 1.05, 0.78, 0.09 and 0.008 g plant-1, respectively for each treatment on the 98th day. Specfic activity of N2-fixation showed the maximum rates of 140, 101, 37, 5 and 2.2 nM dw.mg-1.hr-1, respectively for each treatment in the earlier growth period. The maximum acetylene reduction activity on the 98th day after sowing was 81.5, 35.3, 14.3, 0.1 and 0.0045 $\mu$M C2H4 plant-1.hr-1, respectively for 0, 1, 3, 10 and 30 mM of NO3- gradients. Nitrate reduction activity increased along with nitrate gradients, and decreased abruptly with age. Relative abundance of ureides in plant organs was high in reproductive growth, and showed the maximum value in fully symbiotic dependent plant. Relative abundance of ureides in stem is a useful indication for the evaluation of nitrogen fixation in nodules of symbiotic plant.

Physiological and Molecular Characterization of NAD(P)H-Nitroreductase from Stenotrophomonas sp. OK-5 (Stenotrophomonas sp. OK-5에서 분리한 NAD(P)H-Nitroreductase의 생리학적 및 분자생학적 특성 연구)

  • Stenotrophomonas sp. OK-5 capable of degrading TNT has been found to have three nitroreductase fractions designated as NTR fractions I, II, and III. NTR in a previous study. This study was attempted to reveal physiological and molecular characteristics of NTR fractions I, II, and III in strain OK-5. Several chemicals (e.g., EDTA, NaCl, dithiothreitol, $\beta$-mercaptoethanol) were tested for their effect on enzyme activity of NTRs, demonstrating that enzyme activities of NTR fractions I, II, and III from OK-5 were inhibited in the presence of $\beta$-mercaptoethanol. Substrate specificity test showed that NTR fractions I, II, and III all have over 70% enzyme activities for nitrobenzene or RDX as a substrate. N-terminal amino acid sequence of NTR fraction I from Stenotrophomonas sp. OK-5 was $^1MSDLLNADAVVQLFRTARDS^20$ and exhibited 70% sequence homology with that of NTR from Xanthomonas campestris. NTR I gene from Stenotrophomonas sp. OK-5 (SmOK5nrI) shared extensive sequence homology in deduced amino acid sequence of PCR product with NTRs from Xanthomonas campestris (81 %), X. axonopodis (75%), Streptomyces avermitilis(30%), whereas they had low homology with that from P. putida KT2440 (pnrB) (16%).

Influence of Yeoldahanso-tang on the Hypoxic Damage of Cultured Cerebral Neurons from mouse and SK-N-MC cells (열다한소탕(熱多寒少湯)이 저산소성(低酸素性) 대뇌신경세포(大腦神經細胞) 손상에 미치는 영향(影響))

  • To elucidate the neuroprotective effect of Yeoldahanso-tang(YHT) on nerve cells damaged by hypoxia, the cytotoxic effects of exposure to hypoxia were determined by XTT(SODIUM3,3'-{I-[(PHENYLAMINO) CARBONYL]-3,4-TETRAZOLIUM}- BIS (4-METHOXY-6-NITRO) BENZENE SULFONIC ACID HYDRATE), NR(Neutral red), MTT(3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) and SRB(Sulforhodamin B) asssay. The activity of catalase and SOD(Superoxide dismutase) was measured by spectrophometry, and $TNF-{\alpha}$(Tumor cell necrosis $fector-{\alpha}$) and PKC(Protein kinase C) activity was measured after exposure to hypoxia and treatment of YHTWE. Also the neuroprotective effect of YHTWE was researched for the elucidatioion of neuroprotective mechanism. The results were as follows; 1. Hypoxia decreased cell viability measured by XTT, NR assay when cultured cerebral neurons were exposed to 95% N2/5% CO2 for $2{\sim}26$ minutes in these cultures and YHTWE inhibited the decrease of cell viability. 2. H2O2 treatment decreased cell viability measured by MTT, and SRB assay when cultured cerebral neurons were exposed to 1-80 ${\mu}M$ for 6 hours, but YHTWE inhibited the decrease of cell viability. 3. Hypoxia decreased catalase and SOD activity, and also $TNF-{\alpha}$ and PKC activity in these cultured cerebral neurons, but YHTWE inhibited the decrease of the catalase and SOD activity in these cultures. 4. Hypoxia triggered the apoptosis via caspase activation and internucleosomal DNA fragmentation. Also hypoxia stimulate the release of cytochrome c forom mitochondria. YHTWE inhibited the apoptosis via caspase activation induced by hypoxia. From these results, it can be suggested that brain ischemia model induced hypoxia showed neurotoxicity on cultured mouse cerebral neurons, and the YHTWE has the neuroprotective effect in blocking the neurotoxicity induced by hypoxia in cultured mouse cerebral neurons.

Inhibitory Effects of Changchuldoin-tanggamibang on Collagen Induced Arthritis in DBA/1J Mouse (창출도인탕가미방(蒼朮桃仁湯加味方)이 DBA/1J 생쥐의 collagen 유발 관절염 억제에 미치는 영향)

  • Objectives: This study was carried out to find the effects of Changchuldoin-tanggamibang (hereinafter referred to CDIT) on the inhibition of arthritis induced by collagen on DBA/1J mouse. Methods: The experimental mice were divided into four groups: normal group (Nr), control group (CIA-CT), methotrexate group (CIA-MTX), and Changchuldoin-tanggamibang group (CIA-CDIT). Cytotoxicity, hepatotoxicity, arthritis index, value of immunocytes in draining lymph node and paw joint, and rheumatoid factor (IgG, IgM) in serum were measured in vivo. Results: 1. Cytotoxicity against hFCs was not shown in any concentration. 2. Hepatotoxicity was low in the CDIT-treated group compared with the MTX group. 3. The arthritis index decreased significantly. 4. In total cell counts of DLN and paw joint, the cells in DLN increased significantly while there was a significant decrease in paw joint. 5. In lymph nodes, CD19+, CD3+, CD4+, CD8+, CD3+/CD8+, CD3+/CD69+, CD4+/CD25+, CD3+/CD49b+, and CD4+/CD44+ cells increased significantly, while B220+/CD23+, and CD11c+/MHCII+ cells decreased significantly. 6. In joints, CD3+, CD4+, CD4+/CD25+, and CD11b+/Gr-1+ cells decreased significantly. 7. The level of IgG decreased and the level of IgM significantly decreased compared with the control. 8. Anti-collagen II in serum decreased compared with the control. 9. Around the joint of the CDIT group, infiltration of inflammation, synovial hyperplasia, invasion of cytokine, of cartilage, deposition of collagen and synovial injury decreased compared with the control in histopathologic observation (HE, MT staining). Conclusions: Comparison of the results for this study showed that CDIT had immunomodulatory effects. We expect that CDIT could be used as a effective drug for not only rheumatoid arthritis but also another auto-immune diseases. Therefore, we have to survey continuously, looking for effective substances and mechanisms in the future.

In vitro Conidial Germination and Mycelial Growth of Fusarium oxysporum f. sp. fragariae Coordinated by Hydrogen Peroxideand Nitric Oxide-signalling

  • Chemicals related to hydrogen peroxide ($H_2O_2$) and nitric oxide (NO) generations were exogenously applied to Fusarium oxysporum f. sp. fragariae (Fof) causing Fusarium wilt disease in strawberry plants, and regulations of in vitro conidial germination and mycelial growth of the fungus by the chemical treatments were evaluated. $H_2O_2$ drastically reduced the conidial germination of Fof in a dose-dependent manner, and treatment with 3-amino-1,2,4-triazole (3-AT) catalase inhibitor also led to dose-dependent inhibition of conidial germination but relatively moderately. Gradual decreases in mycelial growth of Fof were found by high concentrations of $H_2O_2$, whilst exogenous 3-AT slightly increased the mycelial growth. Increasing sodium nitroprusside (SNP) NO donor, $N^G$-nitro-l-arginine methyl ester (L-NAME) NO synthase (NOS)-inhibitor and tungstate nitrate reductase (NR) inhibitor led to dose-dependent reductions in conidial germination of Fof in quite different levels. SNP conversely increased the mycelial growth but increasing L-NAME moderately decreased the mycelial growth. Tungstate strongly enhanced mycelial growth. Differentially regulated in vitro mycelial growths of Fof were demonstrated by SNP, L-NAME and tungstate with or without $H_2O_2$ supplement. Superoxide anion production was also regulated during the mycelial growth of Fof by nitric oxide. These results show that $H_2O_2$ and NO-associated enzymes can be suggested as fungal growth regulators of Fof as well as eco-friendly disease-managing agents in strawberry production fields.
