Objective : The aim of this study was to investigate anti-inflammatory activity of SD-01 methanol extract in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Methods : Cytotoxic activity of SD-01 methanol extract on RAW 264.7 cells was measured using 5-(3-caroboxymeth-oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. The nitric oxide (NO) production was measured by Griess reagent system. And proinflammatory cytokines and $PGE_2$ were measured by ELISA method. The levels of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), $I{\kappa}$-B-alpha and nuclear NF-${\kappa}$ B p65 expression were detected by western blot. Results : Our results indicated that methanol extract of SD-01 significantly inhibited the LPS-induced NO, $PGE_2$ production and iNOS, COX-2 expression accompanied by an attenuation of TNF-$\alpha$, IL-$1\beta$, IL-6 and MCP-1 production in RAW 264.7 cells. Moreover, methanol extract of SD-01 treatment also blocked LPS-induced NF-kB activation. Conclusion : These findings indicate that methanol extract of SD-01 inhibits the production of pro-inflammatory mediators and cytokines via suppression of NF-${\kappa}$ B activation. Take together, these results indicate that methanol extract of SD-01 has the potential for use as an agent of anti-chronic inflammatory diseases.
우리는 한국생명공학연구원에서 제공한 소재를 탐색 후 ACE를 선별하였고 이 소재의 항염증 효능을 평가하였다. ACE는 세포독성이 없는 농도에서 LPS에 의한 iNOS의 발현과 NO의 생성을 유의적으로 억제하였다. ACE는 LPS에 의해 증가한 IKK, IκB, p65의 인산화와 IL-1β의 생성을 유의적으로 억제하였다. 이 논문은 ACE가 NO/iNOS 발현 및 NF-κB 신호 전달 경로의 억제를 통해 항염증 효능을 나타냄을 밝힌 최초의 논문으로서 의의를 가진다. 또한 이 결과를 바탕으로 ACE는 항염증 기능성식품 또는 의약품소재로써 활용가치가 높을 것으로 기대된다.
The present study describes the preliminary evaluation of the anti-inflammatory activities of Phellinus linteus (PL) and Phellinus linteus Grow in Germinated Brown Rice (BRPL). In order to effectively screen for anti-inflammatory agents, we first examined the extracts' inhibitory effects on the expression of pro-inflammatory cytokines activated with lipopolysaccharide. Moreover, we examined the inhibitory effects of the PL and BRPL extracts on pro-inflammatory factors such as NO, iNOS, $TNF-{\alpha}$ and $IFN-{\gamma}$ in murine macrophage RAW 264.7 cells. NO production and iNOS expression was significantly augmented in LPS treated cell, the production of NO and iNOS was greater in the BRPL than in the PL group. In addition, protein and mRNA levels of $TNF-{\alpha}$ and $IFN-{\gamma}$ in BRPL showed relatively more potent pro-inflammatory-activity inhibition compared to that of PL. These results suggest that BRPL may have significant effects on inflammatory factors, and may be a potential anti-inflammatory therapeutic materials.
Objectives : Forsythiae Fructus (Forsythia koreana Nakai) has been used anti-inflammatory, diuretics, antidote, and antibacterials in traditional herbal medicine. The present study is focused on the inhibitory effect of Forsythiae Fructus ethanol extract (FF-E) on the production of inflammatory mediators such as NO, iNOS and proinflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) in LPS-stimulated BV-2 cells, a mouse microglial cell line, and investigated the scavenging activity of FF-E. Methods : BV-2 cells were pre-incubated with FF-E for 30 min and then stimulated with LPS (1 ${\mu}g/m{\ell}$) at indicated times. Cell toxicity of GCF was determined by MTT assay. The levels of NO, PGE2 and cytokines were measured by Griess assay and ELISA. The mRNA and protein expressions of iNOS and cytokines were determined by RT-PCR and Western blotting. Free radical scavenging activity of GCF was determined by DPPH assay in tube test. Results : FF-E significantly inhibited the excessive production of NO, $PGE_2$, $TNF-{\alpha}$, and $IL-1{\beta}$ in LPS-stimulated BV-2 cells. In addition, FF-E attenuated the mRNA and protein expressions of iNOS, and proinflammatory cytokines. FF-E also significantly scavenged the DPPH free radicals in a dose-dependent manner. Conclusions : These results indicate that FF-E exhibits anti-inflammatory property by suppressing the transcription of inflammatory mediator genes, suggesting the anti-inflammatory property of FF-E may make it useful as a therapeutic agent for the treatment of human neurodegenerative diseases.
To search for immunoactive natural products exerting anti-inflammatory activity, we have evaluated the effects of the ethanol extracts of Rubus coreanus Miq. (ERC) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $Interferon-{\gamma}\;(IFN-{\gamma})$ production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased $IFN-{\gamma}\;and\;TNF-{\alpha}$ production. Consistent with these results, the protein level of inducible Nitric Oxide Synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by ethanol extracts of ERC in a dose-dependent manner. These results suggest that ERC may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.
Objective: In this study, we investigated the protective effect of rhododendron mucronulatum extract (RME) on allergic reactions and inflammation. Methods: The effect of RME was determined using ELISA and RT-PCR in RBL-2H3 mast cells and RAW 264.7 macrophage cells. We determined cell viability, β-hexosaminidase release, and the synthesis of IL-4 and TNF-α in RBL-2H3 cells. In addition, we determined NO from RAW 264.7 and the gene expression of IL-1β, iNOS, IL-6, TNF-α, and IL-10. Results: RME inhibited β-hexosaminidase release and synthesis of IL-4 and TNF-α in RBL-2H3 by the anti-DNP IgE plus DNP-HSA stimulation. In addition, RME inhibited the production of NO and the gene expression of IL-1β, iNOS, IL-6, and TNF-α in LPS-stimulated RAW 264.7 cells. Conclusion: From these results, we concluded that RME possesses anti-allergic activity and anti-inflammatory activity due to the inhibition of mast cells and macrophage function.
본 연구에서는 식물자원으로부터 생리활성을 보유한 새로운 기능성 소재를 찾고자 하였다. 이를 위해 수종의 중국 자생 식물을 분석하여 강한 항산화능을 보유한 3종(Malus hupehensis, Ophiorrhiza cantonensis, 그리고 Psychotria rubra)을 선별하고, 각 추출물의 항산화능과 항염증 생리활성을 분석하였다. 먼저 각 추출물의 항산화능을 DPPH radical 소거능을 통해 분석한 결과 모두 양성 대조군으로 사용한 ascorbic acid와 유사한 정도의 높은 활성을 보여 각 소재가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 $H_2O_2$ 및 LPS에 의해 유도된 ROS에 대한 각 추출물의 소거능을 분석한 결과에서도 농도의존적인 강한 소거능을 보였다. 뿐만 아니라 대표적인 항산화효소 중 하나로 항산화능 보유 천연물에 의해 발현이 유도되는 HO-1 및 그 전사 인자인 Nrf-2의 단백질 발현이 각 추출물의 처리에 의해 증가됨을 보였다. 한편 각 소재가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 모두 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이와 같은 각 소재의 NO 생성 및 iNOS 발현 억제 효과는 염증 상위신호 전달계인 NF-${\kappa}B$ 및 AP-1의 조절을 통해 일어날 가능성을 보였다. 이러한 결과를 통해 중국 자생 식물 3종의 높은 항산화능과 항염증 활성을 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.
본 연구에서는 LPS에 의해 활성화된 RAW264.7 대식세포에서의 NO 생성 및 iNOS와 HO-1 단백질 발현의 변화를 측정하여 개망초(Erigeron annuus L.) 꽃(EAF) methanol 추출물의 항염증 효과를 확인하였다. RAW264.7 대식세포에 LPS를 처리한 결과 NO의 함량이 11.48 ${\mu}m$ 수준으로 증가하였다. 그러나 EAF methanol 추출물(25, 50, 100, 200 ${\mu}g$/mL)을 처리하였을 때 NO의 함량이 12.82, 9.61, 6.83, 2.52 ${\mu}m$로 농도 의존적으로 감소하였다. 또한 EAF methanol 추출물은 NO의 생성에 관여하는 iNOS 단백질의 발현을 농도 의존적으로 저해하였으며 HO-1 단백질의 발현을 유도하였다. EAF methanol 추출물에 의한 HO-1 발현이 NO 생성에 미치는 영향에 대해 확인하기 위해 HO-1의 inhibitor인 ZnPP를 사용하였다. 실험결과 ZnPP를 처리하여 HO-1의 발현을 저해하였을 때 추출물에 의해 감소된 NO의 함량이 다시 증가되었다. 본 연구 결과, EAF methanol 추출물은 염증을 유발하는 중요 인자인 NO를 저해하였고, iNOS의 발현을 억제시켰으며 산화적 손상으로부터 세포 보호 방어 기작에 관여하는 HO-1의 발현 등 항염증에 우수한 효과를 보였으며 항염증 연구의 기초 자료로 활용될 것으로 예상된다.
iNOS(inducible nitric oxide synthase)와 TLR-4(toll-like receptor 4) 유전자는 모든 생물의 면역반응에 필요한 필수유전자로 알려져 있다. iNOS는 닭에서 19번 염색체에 위치하고 있으며, NO를 면역기관에서 생산되어 면역반응을 일으키는 것으로 알려져 있으며, TLR-4는 TLRs(toll-like receptors)중 하나로 알려져 있고, 그람 음성균의 LPS을 인식하여 초기 면역반응에 참여하는 것으로 알려져 있다. 이들의 유전적 변이는 살모넬라균에 감염되었을 때 맹장의 장내 균총에 영향을 주는 것으로 알려져 있다. 본 연구는 iNOS의 intron 24 영역 내 C14513T와 TLR-4의 exon 3 영역 내 G4409T 변이 지역을 대상으로 3품종(한국 재래닭, 코니쉬, 로드아일랜드 레드)을 이용하여 유전자형을 확인하고, 경제 형질 간의 연관성 분석을 실시하였다. 분석 결과, iNOS의 변이 지역(C14513T)은 한국 재래닭과 로드아일랜드 레드종에서 270일령 몸무게와 유의적인 연관성이 확인되었으며, TLR-4의 변이 지역(G4409T)은 경제 형질과의 연관성이 확인되지 않았다. 본 연구 결과는 면역 및 경제 형질 관련 표지인자 발굴을 위한 유용한 기초 자료로 활용될 수 있으며, 추가적인 연구를 통해 분자 육종을 위한 유전표지인자 개발에 활용이 가능할 것으로 사료된다.
Background: Celecoxib, a COX-2 specific inhibitor, has recently been used for the treatment of rheumatoid arthritis. However, the molecular and cellular mechanisms of celecoxib against RA inflammation remain to be defined. To elucidate the action mechanism of celecoxib on inflammatory cells, we investigated the effect of celecoxib on the production of two important mediators of inflammation, nitric oxide and PGE2 Methods: RAW 264.7 cells stimulated with LPS were preincubated with various concentrations of celecoxib (from $10^{-8}$ to $10^{-5}$ M) and $10{\mu}M$ hydrocortisone, respectively. The production of NO and PGE2, the end products of iNOS and COX-2 genes, were estimated in culture supernatants by Greiss method and EIA, respectively. The expression of iNOS gene, COX-2 gene, $NF-{\kappa}B$, and $I-{\kappa}B$ were determined by RT-PCR and western blot analysis. Results: Celecoxib and hydrocortisone inhibited the production of NO and PGE2 in dose dependent manner, when RAW 264.7 cells were stimulated with LPS. The expression of iNOS was also down-regulated by celecoxib and hydrocortisone. Interestingly, COX-2 gene differentially expressed according to the dose of celecoxib, a decrease with lower dose ($10^{-8}$ M) but an increase with higher dose ($10^{-5}$ M). $NF-{\kappa}B$ binding activity was decreased by lower dose of celecoxib, whereas was not affected by higher dose of it. The expression of $I-{\kappa}B$ was suppressed by higher dose of celecoxib. Conclusion: The celecoxib strongly suppressed the production of NO and PGE2 in LPS-stimulated RAW264.7 cells. The decrease of NO seems to be linked to the inhibition of iNOS by celecoxib. The lower and higher dose of celecoxib differentially regulated the COX-2 expression and $NF-{\kappa}B$ activity.
