C57BL/6J Mice에서 스테비아(Stevia rebaudiana bertoni) 잎 추출물의 항비만 효과 (Effect of Stevia rebaudiana Bertoni Leaf Extract on Antiobesity in C57BL/6J Mice)

  • 미국의 영양사협회(32)에서는 설탕에서 얻은 열량을 전체 섭취량의 10-15%까지로 유지할 것을 권장하고 있고, WHO(33)에서는 전체 섭취 열량의 10% 이하로 권장하고 있다. 서구사회에서 뿐만 아니라 우리나라에서도 설탕 섭취량은 꾸준히 증가하고 있는데, 성인 1인 1일 당류 공급량은 1962년 4.8 g에서 1987년 41.9 g, 2003년 57.4 g으로 나타났으며(34), 2007년 한국인의 1일 평균 설탕 섭취량은 100 g, 총 섭취량의 20%로 권장량의 두 배 이상으로 점차 증가되고 있다(35). 그러나 본 실험에서 사용된 스테비아의 당도는 설탕의 200배 이상이나 칼로리는 설탕의 90분의 1에 불과하여 설탕의 대체제로서 가능성이 충분할 것으로 보여진다. 이에 본 연구는 스테비아 잎 추출물 보강이 고지방식이로 비만이 유도된 C57BL/6J mice에서 항비만 효과를 알아보고자 하였다. 실험동물은 정상 대조군(NC), 고지방 대조군(HC), 고지방식이+스테비아 잎 추출물 투여군(HLSV, 1 mL/kg/day), 고지방식이+스테비오사이드 투여군(HSS, 1 mL/kg/day) 4군으로 나누어 사육하였으며, 연구결과는 다음과 같다. 체중증가량은 스테비아 잎 추출물 투여군(HLSV)에서 고지방 대조군(HC)과 비교 시 유의적으로 감소하였으나, 스테비오사이드 투여군(HSS)은 고지방 대조군(HC)비교 시 감소하는 경향을 보였고 유의적 차이는 없었다. 부고환지방은 스테비아 잎 추출물 투여군(HLSV)과 스테비오사이드 투여군(HSS)에서 고지방 대조군(HC) 보다 유의적으로 감소하였고, 등지방은 스테비아 잎 추출물 투여군(HLSV)에서 유의적으로 감소하였다. 혈중 및 간중 중성지방은 스테비아 잎 추출물 투여군(HLSV)이 고지방 대조군(HC)보다 유의적으로 감소하였으며, 스테비오사이드 투여군(HSS)은 고지방 대조군(HC)에 비해 혈중 중성지방이 유의적으로 감소하였다. 혈중 카르니틴 중 NEC의 농도는 고지방 대조군(HC) 보다 스테비아 잎 추출물 투여군(HLSV)과 스테비오사이드 투여군(HSS)에서 유의적으로 증가하였으며 혈중 ASAC와 간중 acyl/free carnitine은 고지방 대조군(HC)과 비교 시 스테비아 잎 추출물 투여군(HLSV)과 스테비오사이드 투여군(HSS)에서 증가하는 경향을 보였다. 지질대사와 관련한 체내 mRAN 발현정도를 측정한 결과 ACS mRNA 발현 수준은 고지방대조군(HC)과 비교 시 스테비아 추출물 투여군(HLSV)와 스테비오사이드 투여군(HSS)에서 증가하는 경향을 보였으며 특히, 스테비오사이드 투여군(HSS)에서 유의적으로 증가하였다. PPAR${\alpha}$ mRNA발현 수준은 고지방대조군(HC)과 비교 시 스테비아 추출물 투여군(HLSV)와 스테비오사이드 투여군(HSS)에서 유의적으로 증가하였으며, CPT-I mRNA 발현 수준은 고지방대조군(HC)과 비교 시 스테비아 추출물 투여군(HLSV)이 유의적으로 증가하였다. ACC mRNA 발현 수준은 정상대조군(NC)과 비교 시 고지방대조군(HC)에서 유의적으로 증가하였고, 스테비아 추출물 투여군(HLSV)은 고지방 대조군(HC)과 비교 시 유의적으로 감소하였으나 스테비오사이드 투여군(HSS)은 차이가 없었다. 결과적으로 스테비아 잎 추출물은 스테비오사이드에 비하여 체내 지질대사개선에 더 효과적인 것으로 보였으며 이는 스테비아의 다양한 phytochemical에 의한 것으로 보여진다. 따라서 이러한 결과는 스테비아가 비만을 치료하거나 예방하는데 효과적으로 활용될 수 있는 가능성을 보여주었다.

Inhibitory Effects of a Korean Strain Gpf01 Identified as Pseudomonas fluorescens on Cucumber mosaic virus

  • An antiviral producing bacterial strain was isolated from a ginseng rhizosphere in Kangwon province of Republic of Korea. In order to identify the bacterial strain, microbiological, physiological and biochemical tests were performed, along with RAPD, 16S rRNA, 16S-23S rRNA ITS (intergenic spacer region) and DNA-DNA hybridization analyses. The bacterium was found to be a strain of Pseudomonas fluorescens, which was designated as Gpf01. The strain was grown in Muller-Hinton (MH) broth, and the culture supernatant obtained was filtered through a $0.45{\mu}l$ filter. It was further boiled at $100^{\circ}C$ and tested in two experiments for its ability to control a yellow strain of Cucumber mosaic virus (CMV-Y). In the first experiment, boiled culture filtrate (RCF) was treated on one half of the leaves of Chenopodium amaranticolor followed by CMV- Y inoculation on both halves. In the second experiment, BCF was treated on the lower leaves of Nicotiana tobacum var. Xanthi-nc, with the CMV-Y mechanically inoculated onto the upper untreated leaves. In the first experiment, BCF treatment was able to considerably reduce the number of viral lesion, and in the second experiment, plants treated with BCF showed no visible viral symptoms compared to the Muller-Hinton (MH) media treated controls 15 days post inoculation (dpi), and remained symptomless throughout the study period. Thus, Gpf01, identified as P. fluorescence, was able to produce an antiviral component in the culture filtrate, which was found to be heat stable, non-phytotoxic and effective in local as well as systemic hosts of CMV.

기술창업교육프로그램 품질이 참여자 만족과 추천의향에 미치는 영향 연구 (The Effects of the Quality of Technology Entrepreneurship Educating Program on Participant's Satisfaction and Referring Will)

  • 본 논문은 기술창업교육프로그램의 품질이 교육 참여자의 만족도와 타인에 추천의향에 미치는 영향도를 분석하였다. 이 분석의 목적은 기술창업교육프로그램의 방안으로 실행된 TEC(Technology Entrepreneurship & Commercialization: 미국 노스캐롤라이나주립대 개발 & 한국 기술사업화교육팀 보정) 프로그램의 유효성을 평가하고 이에 대한 개선방안을 도출해 성공창업을 견인하기 위해 국가적 차원에서 실시하고 있는 창업교육의 우수한 대안을 제시하는데 있다. 특히, 본 논문은 세부적 분석을 위해 PZB의 SERVQUAL 모형에 근거하여 TEC 교육 서비스의 품질평가 문항을 구축하고, TEC 교육이수자들에 대한 설문조사를 통해, 이것이 참여자의 만족도에 미치는 영향과, 만족도가 추천의향에 미치는 영향을 실증 분석하였다. 분석결과, 본 논문은 SERVQUAL 항목 중 유형성과 확신성 그리고 교육내용은 참여자의 만족도에 (+) 영향을 주는 것으로 나타났으며 교육참여자의 만족도는 추천의향과 (+) 관계가 존재함을 확인하였다. 이는 TEC 프로그램이 그 동안의 교육실시과정에서 우수한 학습환경의 조성과 신뢰 있는 교육내용에서 상당한 교육 만족도를 확보하고 있음을 입증하고 있다.

흑색 황산3가크롬을 이용한 태양열 흡열판 선택흡수막 도금기술 (Technology of selective absorber coatings on solar collectors using black chromium+3 sulfate acid on substrates)

  • One of the most important factors that have a large influence on performance of the solar water heater system is performance of the solar collector, more detailedly, coating technology on the surface of the solar collector, which can provide high solar absorptance and low emittance. The core of the coating technology is to coat solar selective surfaces. In this study, various performance experiments are carried out using $Cr_2(SO_4)_3{\cdot}15H_2O$ coating technology. Here, IGBT(Insulated Gate Bipolar Transistor) of 5000A-15V was used as the surface processing rectifier which can stably output power and also can control voltage and current. The plating solution mainly contains black chrome$^{+3}$ concentration, H-y Conductivity, N-u Complex, NF Additive and NC-2 Wetter. Before applying the black chrome coating on the copper plate, optimal conditions are provided by using various preprocessing methods such as removal of fat, activation, electrolytic polishing, nickel strike, copper sulfate plating and bright neckel plating, and then the automatic continuous coating experiment are performed according to plating time and cathode current density. In the experiment, after the removal of fat, chemical polishing, nickel strike and activation processes as the preprocessing methods, the black chrome coating was performed in a plate solution temperature of $28^{\circ}C$ and a cathode current density of $18A/cm^2$ for 90 seconds. The thickness of chrome and nickel on the coated plate is $0.389{\mu}m$, $159{\mu}m$ respectively. As a result of the coating experiment, it showed the most excellent performance having a high solar absorptance of 98% and a low emittance of $5{\pm}1%$ when the black chrome surface had a thickness of $0.398{\mu}m$.

Fruiting Body Extracts of Paecilomyces tenuipes Ameliorate Lipid and Antioxidant Metabolism in Rats Fed a High Fat-Cholesterol Diet

  • The present study examined the lipid lowering and antioxidant activity of Paecilomyces tenuipes. Eight week-old male Sprague-Dawley rats were fed one of the three diets, a reference diet without cholesterol addition (NC), a high fat (17 g/ 100 g)-high cholesterol (1 g/100 g) diet (HC) and a HC diet supplemented with 3% P. tenuipes (PT) for 30 days. Total lipid and total cholesterol were reduced significantly by 33 and 37%, respectively, in the PT diet group compared with controls. A similar reduction was found for low density lipoprotein (LDL) cholesterol levels, while plasma high density lipoprotein (HDL) cholesterol and triglyceride (TG) concentrations were not significantly different among groups. Hepatic total lipid and total cholesterol levels, but not hepatic TG levels, were significantly decreased in the PT group compared to the HC group. The administration of P. tenuipes increased the plasma total antioxidant potential and decreased the levels of lipid peroxidation. These results suggest that P. tenuipes exerts significant health benefits through the modulation of physiological functions including a variety of atherogenic lipid profiles and antioxidants in hypercholesterolemia.

박판제품의 블랭킹 및 피어싱과 굽힘 가공을 위한 순차이송용 공정 및 금형 설계와 가공자동화 시스템 (A Progressive Automated-Process Planning and Die Design and Working System for Blanking or Piercing and Bending of Sheet Metal Product)

  • This paper describes a research work of developing a computer-aided design and manufacturing of irregular shaped sheet metal product for blanking or piercing and bending operations. An approach to the system is based on the knowledge-based rules. Knowledge for the system is formulated from plasticity theories experimental results and the empirical knowledge of field experts, This system has been written in AutoLISp on the AutoCAD and in customer tool kit on the SmartCAM with a personal computer and is composed of nine modules which are input and shape treatment, flat pattern-layout, pro-processor module. Based on the knowledge-based rules, the system is designed by considering several factors, such as material and thickness of product complexities of blank geometry and punch profile sheet metal to give flat pattern and automatically account for the adjustment of bending allowances to match tooling requirements by checking dimensions and generating NC data automatically according to drawings of die-layout module. Results carried out in each module will provide efficiencies to the designer and the manufacturer of blanking or piercing and bending die in this field.

다축 RP 소프트웨어 기술을 이용한 스캐폴드 제조 장비 개발 (Development of Scaffold Fabrication System using Multi-axis RP Software Technique)

  • The scaffold serves as 3D substrate for the cells adhesion and mechanical support for the newly grown tissue by maintaining the 3D structure for the regeneration of tissue and organ. In this paper, we proposed integrated scaffold fabrication system using multi-axis rapid prototyping (RP) technology. It can fabricate various types of scaffolds: arbitrary sculptured shape, primitive shape, and tube shape scaffolds by layered dispensing biocompatible/ biodegradable polymer strands in designated patterns. In order to fabricate the 3D scaffold, we need to generate the plotting path way for the scaffold fabrication system. We design a data processing program - scaffold plotting software, which can convert the 3D STL file, primitive and tube model images into the NC code for the system. Finally, we fabricated the customized 3D scaffolds with high accuracy using the plotting software and the fabrication system.

스크류 유량계의 개발 및 정밀 성능평가에 관한 연구 (A Study on the Development and Precise Performance Test of a Screw Type Flowmeter)

  • This research introduces the efficient modeling and manufacturing method using reverse engineering combined with rotational simulation of a pair of screw and mill-turn machining realized a proposed NC program. Because previously developed model had insufficient flow measuring accuracy, we considered that matter in the aspect of method of modeling and performance test. For that reason we modified the modeling which could minimize a gab between male and female screw, and developed precise tester which consists of constant tank, flowmeter and load cell, etc., and then conducted the test for defining characteristic and accuracy of flowmeter and repeated same test 5-times. Consequently we could obtain satisfied measuring accuracy and reproducibility indicated in the catalog of master model. Hence we give our conclusion as to the validity of developing accurate screw type flowmeter using the proposed process such as reverse engineering, mill-turn machining and precise performance test.

무역학전공 관련 NCS의 이해와 활용에 관한 연구 (Understanding and Utilization of NCS on International Trade Major in University Education)

  • Recently Korea has stepped into a stage of sluggish development, and the unemployment of young people has become a major issue. Especially in 1998 IMF economic shock was influential in formulating government policy and social economic structure on employment. Restructuring now becomes everyday words and further restructuring is already under way. Owing to the growing economic crisis and resultant unemployment and initial appointment shall be considered conditional. Young job seekers are grappling with mastering their native language and unnecessary experiences. These unnecessary experiences are needless waste of time and money. Educational system in university may be affected by environmental changes in population of students and business crisis. Sometimes the departments made a very low rate of employment may be abolished or merged in university. The government will demand us in the work of reform by doing NCS. The government is now ask to teach, train and employ students on the basis of NCS especially in high-school, job training center, and college and university. NCS has advantages and disadvantages. NCS may reduce waste of time and money to make unnecessary experiences, provide standard course to prepare educational system. It will be a big help to adapt properly and make better employment system. The most negative element of the program is application to humanities and social sciences by the same standard with technology and engineering department in the university. Standardization of each educational course will not react quickly to rapidly changing situations in the future.

  • In CNC machining, a 3D(3-dimension) linear segment and a 2D(2-dimension) circular arc are general forms given by CAD/CAM system. Generally, the 2D circular arc machining is processed using dividing into some linear segments. A 3D circular arc also don't exist in the standard form of NC data. This paper present a algorithm and method for real-time machining of a circular arc(not only the 2D one, but also the 3D one). The 3D circular arc machining is based on the 2D circular arc machining. It only needs making a new coordinate system, converting given 3D points(a start point, a end point, and a center point of a 3D circular arc) into points of the new coordinate system, and processing a inverse transformation about a interpolated point. The proposed algorithm was implemented and simulated on PC system. It was confirmed to give a gcod result.
