Salmonella typhimurium LPS Confers Its Resistance to Antibacterial Agents of Baicalin of Scutellaria baicalensis George and Novobiocin: Complementation of the rfaE Gene Required for ADP-L-glycero-D-manno-heptose Biosynthesis of Lipopolysaccharide

  • The antibacterial mechanism of enterobacter Salmonella typhimurium was studied. The rfa (Waa) gene cluster of S. typhimurium encodes the core oligosaccharide biosynthesis of lipopolysaccharide (LPS). Among the rfa gene cluster, we recently cloned the rfaE gene, which is involved in ADP-L-glycero-D-manno-heptose biosynthesis. The rfaE mutant synthesizes heptose-deficient LPS, which consists of only lipid A and 3-deoxy-D-manno-octulosonic acid (KDO), thus making an incomplete LPS and a rough phenotype mutant. S. typhimurium deep-rough mutants with the heptose region of the inner core show a reduced growth rate, sensitivity to high temperature, and hypersensitivity to hydrophobic antibiotics such as baicalin isolated from the medicinal herb of Scutellaria baicalensis Georgi. Thus, in this study, the cloned rfaE gene was added to the S. typhimurium rfaE mutant strain SL1102 (rfaE543), which makes heptose-deficient LPS and has a deep-rough phenotype. The complementation created a smooth phenotype in the SL1102 strain. The sensitivity of SL1102 to bacteriophages was also recovered to that of wild-type strain, indicating that LPS is used as the receptor for bacteriophage infection. The permeability barrier of SL1102 to hydrophobic antibiotics such as novobiocin and baicalin was restored to that of the wild-type, suggesting that antibiotic resistance of the wild-type strain is highly correlated with their LPS. Through an agar diffusion assay, the growth-inhibition activity of baicalin was fully observed in the mutant SL1102 strain. However, only a half of the inhibitory activity was detected in the rfaE complemented SL1102 strain. Furthermore, the LPS produced by the rfaE-complemented SL1102 strain was indistinguishable from LPS biosynthesis of smooth strains.

  • 노랑 느타리버섯의 유전 연구 및 다른 느타리종과의 융합을 위해서 노랑 느타리의 담자포자에 자외선을 조사하여 영양요구성 균주를 선발하였다. 노랑 느타리 포자에 UV를 조사한 결과 UV조사시간이 $40{\sim}70$초이고 생존율 $0.4{\sim}1.4%$ 일때 많은 mutants를 얻을 수 있었고, 선발된 14 auxotrophic mutants의 유전 marker는 amino acid-requiring strain 3균주, nucleic acid base-requiring strain 1 균주, vitamin-requiring strain 3 균주, amino acid/nucleic acid base-requiring strain 2 균주, amino acid/vitamin-requiring strain 5 균주로 되었다.

Rifampicin과 Ofloxacin에 내성인 Bifidobacterium bifidum 균주의 개발 (Development of Bifidobacterium bifidum Strains Resistant to Rifampicin and Ofloxacin)

  • Bifidobacterium bifidum, one strain of medical preparation being on the market for human intestinal disorders, was sensitive to rifampicin and fluoroquinolones. If this preparation is taken with rifampicin and fluoroquinolones, its therapeutic effect can't be expected. Serial passage of B. bifidum RFR61, which was obtained by MNNG mutation method, on agar with 2-fold minimal inhibitory concentration of ofloxacin produced B. bifidum OFR9 with minimal inhibitory contentrations of fluoroquinolones up to $4{\sim}256-fold$ higher than that for the original strain. B. bifidum OFR9 produced almost the same amount of organic acid as parental strain. This strain showed growth inhibitory activity against E. coli NM522, Shigella dysenteriae ATCC9752 and E. coli 078. No inactivations of rifampicin and ofloxacin by this resistant mutant strain were found.

영양요구성 대장균 변이주를 이용한 L-트립토판 생산 (Production of L-Tryptophan by Auxotrophs Derived from Analogue- resistant Mutants of Escherichia coli)

  • 트립토판 생산성을 향상시키기 위해 대장균 변이 주인 SB1007로부터 sulfanilamide 내성 균주를 유도하여 SA 3-39-16출 분리하고 이 균주로부터 tyrosine 영양요구주인 TY-90을 개발하였다. 시험관배양시 TY-90균주는 모균주보다 트립토판 생산량이 우수하였으나 발효조 배양시에는 모균주에 비해 낮은 양의 트립토판이 축적되었다. 발효조에서의 생산성 증대를 인해 또 다른 대장균 변이주인 SB2756으로부터 새로운 변이주의 개발을 모색하였다. 먼저 이중 영양요구성 균주인 SB2756의 revertant를 분리하여 그중 2,000mg/$\ell$ 농도의 $\beta$-thenylalanine에 내성을 갖는 TA-40-10을 분리하였고 이 균주로부터 phenylalanine 영향요구주인 TP-4를 개발하였다. 이 TP-4균주는 발효조에서 71시간 배양시 3.7g/$\ell$의 L-tryptophan을 생산하였다.

Deletion of cg1360 Affects ATP Synthase Function and Enhances Production of L-Valine in Corynebacterium glutamicum

  • Bacterial ATP synthases drive ATP synthesis by a rotary mechanism, and play a vital role in physiology and cell metabolism. Corynebacterium glutamicum is well known as an industrial workhorse for amino acid production, and its ATP synthase operon contains eight structural genes and two adjacent genes, cg1360 and cg1361. So far, the physiological functions of Cg1360 (GenBank CAF19908) and Cg1361 (GenBank CAF19909) remain unclear. Here, we showed that Cg1360 was a hydrophobic protein with four transmembrane helices (TMHs), while no TMH was found in Cg1361. Deletion of cg1360, but not cg1361, led to significantly reduced cell growth using glucose and acetic acid as carbon sources, reduced F1 portions in the membrane, reduced ATP-driven proton-pumping activity and ATPase activity, suggesting that Cg1360 plays an important role in ATP synthase function. The intracellular ATP concentration in the ${\Delta}cg1360$ mutant was decreased to 72% of the wild type, while the NADH and NADPH levels in the ${\Delta}cg1360$ mutant were increased by 29% and 26%, respectively. However, the ${\Delta}cg1361$ mutant exhibited comparable intracellular ATP, NADH and NADPH levels with the wild-type strain. Moreover, the effect of cg1360 deletion on L-valine production was examined in the L-valine-producing V-10 strain. The final production of L-valine in the $V-10-{\Delta}cg1360$ mutant reached $9.2{\pm}0.3g/l$ in shake flasks, which was 14% higher than that of the V-10 strain. Thus, Cg1360 can be used as an effective engineering target by altering energy metabolism for the enhancement of amino acid production in C. glutamicum.

Oleic Acid 내성균주로부터 Tylosin 생산 (Tylosin Production by Mutant Resistant to Oleic Acid)

  • Streptomyces fradiae로부터 Tylosin을 효율적으로 생산하기 위해 지방산 내성균주을 분리 했다. 여러 지방산중에서 oleic acid 1.6 g/l 이상이 첨가될 때 세포 성장이 완전히 저해 되었다. 그러나 oleic arid 1.2 g/l에서 얻어진 TM-224-1 균주는 최대 균체농도와 tylosin 생산이 얻어졌다. 또한 oleic acid 소비속도는 parent strain 비해 3.8배 증가했다. Oleic acid 내성균주, TM-224-1 균주와 parent strain을 이용해서 jar fermentor에서 균체농도, tylosin 생산, 그리고 rapeseed oil 소비를 표준조건하에 5일동안 비교하였다. TM-224-1 균주을 이용할 경우 균체 농도는 초기에 parent strain비해 증가했으나 배양중반부터는 감소하기 시작했다. rapeseed oil 소비의 경우는 거의 비슷했다. 그러나 Tylosin 생산 수율은 parent strain 비해 약 3.2배 증가했다.

Monascus purpureus로부터 다량의 색소생성 변이주의 분리 및 색소생성의 최적조건 (Isolation of Pigment Overproducing Mutant from Monascus purpureus and Optimization of Pigment Production)

  • Monascus purpureus KCCM 60016을 이용하여 다량의 색소생산 변이주를 개발하고 생산조건에 대하여 조사하였다. 자외선 조사로 M. purpureus KCCM 60016으로부터 돌연변이를 유도한 결과, 색소생성이 우수한 P-57 변이주를 분리하였다. 분리된 P-57 변이주는 고체 배양에서 현미를 기질로 했을 때 색소생성이 가장 우수한 것으로 나타났으며, 색소생성의 최적 배양조건은 배양온도 $30^{\circ}C,\;90\%$의 배양습도에서 30일간 배양이 가장 우수했다. 이상의 조건에서 배양한 적색색소, 오렌지색소와 황색색소의 양은 각각 160.0 unit, 193.6 unit와 141.6 units-나타났다.

  • The enzyme-producing potentials of industrially important strains of Aspergillus spp. were studied. Irradiation of three original isolates of Aspergillus oryzae to ultra-violet rays resulted in the production of mutants which differed from the parent riboflavin and vitamin $B_{12}$ in culture media. 1. Irradition three strains of Aspergillus oryzae to ultraviolet light produced mutants and two strains of them were selected for soy sauce brewing. 2. The two strains are the physiological mutants of Aspergillus oryzae. Both were found to have superior enzyme activity to their relatives. 3. Aspergillus oryzae UV-induced mutant 172-722 and 569-713 were more powerful than others in the production of riboflavin and vitamin $B_{12}$. The enzyme activity of these strain were high and decreased only slightly even in 20 percent solution of NaCl. 4. Aspergillus oryzae UV-induced mutant 172-722 had more powerful protease producibility in wheat bran media than in modified Czapek's solution. On the contrary, Aspergillus oryzae UV-induced mutant 569-713 had more powerful producibility of saccharogenic and dextrinogenic amylase in modified Czapek's solution than in mold bran. 5. Aspergillus oryzae UV-induced mutant 172-722 formed the spore rapidly and Aspergillus oryzae UV-induced mutant 569-713 did ordinarily. 6. It is found from the results that Aspergillus oryzae UV-induced mutant 172-722 is valuable material for the manufacture of soy sauce because of its high protease activity in 20 percent solution of NaCl. Aspergillus oryzae UV-induced mutant 569-713 is suitable for soy bean mash and for fermented red pepper sauce for its high saccharogenic and dextrinogenic amylase activity in 20 percent solution of sodium chloride.

Acinetobacter sp. B-W의 2, 3-dihydroxybenzoic acid 생산과 항생제 저항성에 미치는 플라스미드 제거 효과 (Effect of plasmid curing on the 2, 3-dihydroxybenzoic acid production and antibiotic resistance of Acinetobacter sp. B-W)

  • 시드로포어인 2, 3-dihydroxybenzoic acid (DHB)를 생산하는 Acinetobacter sp. B-W의 플라스미드를 분석한 결과, 20 kb 크기의 플라스미드를 함유하였다. 배양 온도 $43^{\circ}C$ 가 플라스미드가 제거된 돌연변이체 생산에 효과적이었다. 이 돌연변이체는 2,3-DHB 생산 능력을 소실하였으며, chrome azurol S (CAS) 아가 배지에서 시드로포어 생산이 검출되지 않았다. 포도당과 황산 망간을 함유한 배지에서 $28^{\circ}C$로 3일간 배양한 B-W 원 균주와 돌연변이체의 배양 상등액의 pH는 각각 4.5와 8.5로 나타났다. 돌연변이체에서는 ampicillin, actinomycin D, bacitracin, lincomycin과 vancomycin 같은 항생제에 대한 저항성이 사라졌으며, 이러한 항생제에 대한 최소 억제 농도(MIC)가 급격하게 감소하였다. B-W 균주에서 분리한 플라스미드로 대장균을 형질전환시킨 결과, 원 균주와 같은 크기의 플라스미드가 이 형질전환 대장균에서 발견되었다. 플라스미드가 제거된 돌연변이체에서는 플라스미드가 발견되지 않았다. 20 kb 크기의 플라스미드에 2,3-DHB 생산 유전자와 여러 항생제 저항성 유전자가 자리잡고 있는 것으로 추정된다.

Isolation and characterization of Vitreoscilla mutant defective in catalase-peroxidase hydroperoxidase I

  • Mutants of an obligate aerobic bacterium, Vitreoscilla, that have deficiency in heat-labile catalase-peroxidase hydroperoxidase I (HPI) were created by EMS treatment. The catalase-peroxidase HPI-deficient mutant showed substantially lower peroxidase activity in exponential and mid-stationary phase compared with the wild type strain. In late stationary phase, the mutant exhibited no peroxidase activity. Peroxidase deficiency in the mutant was revealed by polyacrylamide gels stained for peroxidase activity. Characteristically, catalase levels in the mutant increased about 14- and 8-fold during growth in the exponential and stationary phases, respectively, compared to those in the wild type, suggesting a compensatory effect for protection from $H_2O_2$ toxicity. The mutant showed differences in physiology from the wild type: retardation in growth rate and decrease in oxygen consumption. Both the wild type and the catalase-peroxidase HPI-deficient mutant of Vitreoscilla had lower growth rates in media containing increasing $H_2O_2$ concentrations. However, the mutant exhibited an additionally decreased growth rate after 6 to 8 h of growth compared to the wild type. The wild type was resistent up to 20 mM $H_2O_2$, whereas the mutant was very sensitive to high concentrations of exogenous $H_2O_2$. Although elevated catalase levels would provide protection of the bacteria from the deleterious effect of $H_2O_2$, it did not appear to be complete. Cell-free extracts of the mutant showed decreased NADH oxidation rates and higher accumulation of $H_2O_2$ during this oxidation. These results may account for the impaired growth and earlier onset of death phase by the catalase-peroxidase HPI-deficient mutant of Vitreoscilla.
