The effects of the composition of a mixture containing food waste compost (FWC), rice bran (RB), and oak sawdust (SD) on the antler-type fruiting body (FB) yield of Ganoderma lucidum were studied. Experiments were performed using 0 (control), 5, 10, 15, 20, 25, 30, 35, and 40% (w/w) FWC added to a basal growth medium consisting of 20% (w/w) RB and 80% (w/w) SD. The content of 15% FWC gave the highest FB yield ($27.0{\pm}1.3$ g/bottle), which was 44% higher than the yield ($18.6{\pm}2.8$ g/bottle) of the control treatment. However, FWC contents of 20~40% showed reduced yield (2.4~23.0 g/bottle), partly because FWC had a high Na concentration (0.6%). These results demonstrate the potential for use of FWC as a component of a growth medium for production of G. lucidum FBs.
느타리버섯의 생산단가를 절감할 목적으로 한의원 등에서 폐기처리 되고 있는 한방슬러지를 수거하여 느타리 버섯 배지의 개발을 행하였다. 한방재료의 일반성분을 검토한 결과 조회분 함량은 홍화대와 지리오갈피가 11.6%와 10.1%를 나타내어 대조구인 폐면의 9.6% 보다 높게 나타났다. 조지방 함량은 모든 시험구에서 대조구인 폐면보다 높게 나타났다. 조단백 함량은 폐면배지에서는 9.8%로 검출되었으나 야생 쑥, 지리오갈피, 한방슬러지 및 홍화대에서는 각각 14.9%, 13.9%, 13.4% 및 11.5%로 대조구보다 높게 나타났다. 한방슬러지, 야생쑥 및 지리오갈피의 pH는 5.27~5.72 범위로 폐면이 나타낸 pH 5.70과 거의 같은 수준이었다. 느타리버섯의 균사 성장에 미치는 각 한방재료의 첨가농도는 3% 및 6%의 첨가보다 9%의 첨가구에서 느타리버섯 균사의 성장이 양호하였다. 그러나 한방재료의 첨가는 $P.$$tolaassi$의 생육 및 $Trichoderma$$sp.$의 생육에는 크게 영향을 미치지 못하였다. 폐면배지에 9%의 한방슬러지를 첨가하여 실시한 현장 실험결과 버섯의 균사생장은 폐면배지에서 보다 약 2~3일 정도 빠르게 나타났고, 버섯의 수확량은 10~15% 정도 증수되었으며 또한 버섯의 대 및 갓 등이 균일하고 우수하였다.
A fungal strain, capable of solubilizing insoluble phosphate under diverse temperature, pH and salt conditions was isolated from Waste Mushroom bed of Agaricus bisporus in South Korea. Based on 18S rRNA analysis, the strain was identified as Aspergillus awamori bxq33110. The strain showed maximum phosphate solubilization in AYG medium (525 ${\mu}g\;mL^{-1}$) followed by NBRIP medium (515 ${\mu}g\;mL^{-1}$). The strain solubilized $Ca_3(PO_4)_2$ to a greater extent and rock phosphate and $FePO_4$ to a certain extent. However $AlPO_4$ solubilizing ability of the strain was found to be very low. Glucose at the rate of 2% ($561{\mu}g\;mL^{-1}$) was found be the best carbon source for Aspergillus awamori bxq33110 to solubilize maximum amount of phosphate. However, no significant difference ($P{\leq}0.05$) in phosphorus solubilization was found between 1% and 2% glucose concentrations. $(NH_4)_2SO_4$ was the best nitrogen source for Aspergillus awamori bxq33110 followed by $NH_4Cl$ and $NH_4NO_3$. At pH 7, temperature $30^{\circ}C$ and 5% salt concentration (674 ${\mu}g\;mL^{-1}$) were found to be the optimal conditions for insoluble phosphate solubilization. However, strain Aspergillus awamori bxq33110 was shown to have the ability to solublize phosphate under different stress conditions at $30-40^{\circ}C$ temperature, pH 7-10 and 0-10% salt concentrations indicating it's potential to be used as bio-inoculants in different environmental conditions.
Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.
This study was performed to study utilization of separated carbohydrates as well as separation, following analysis of the major components and separation of the carbohydrates in waste liquors of SP, KP, ASAM and AS. As for chemical characteristics of cultivated media waste, the total content of nitrogen, phosphoric acid, and potassium was 6.93%; organic matter content was 67.78%; pH and EC were 7.20 and 1.46 ds/m, respectively. Organic acids identified during stabilization included citric acid, oxalic acid, fumaric acid, and acetic acid, which was highest. During the stabilization of cultivated media waste, temperature decreased and pH increased on the 15th day, and the germination index was 88 after the period, indicating that at least 15 days of stabilization is needed when cultivated media waste is used for growth material of crop. As for germination characteristics, when the mixture rate of cultivated media waste was 20% and under, germination of cucumber and oriental melon was favorable without difference compared to the control plots. For lettuce and Chinese cabbage, no statistically significant difference was found when the rate was 10% and under compared to the control plots. As for seedling quality after 20 days of sowing, cucumber, oriental melon, and Chinese cabbage showed favorable growth only when the mixture rate of cultivated media waste was 10% and under; lettuce showed lower growth characteristics regardless of mixture rate when compared to the control plots. Cultivated media waste may be significantly useful for cucumber and oriental melon, but not for Chinese cabbage and lettuce.
본 연구는 느타리버섯 재배에 있어 필수공정인 살균공정을 생략하기 위하여 $Ca(OH)_2$를 처리한 배지(이하 무살균 배지라 칭 함)의 주원료와 두께를 달리하여 지리산 야생주 No.1의 종균을 접종한 후 균사의 활착, 잡균의 오염 수준, 균이 (원기)발생 소요일, 수확일을 조사한 결과와 수확된 느타리버섯의 일반성분, 무기물과 아미노산을 분석한 결과는 다음과 같다. 가열살균공정을 생략하기 위한 $Ca(OH)_2$[순도95%]의 처리수준은 건배지의 5%(w/w)이었고, 처리초기의 pH는 11이었으며 균일하게 30분간 혼화시키면 pH 9가 되었다. 무살균배지의 pH가 8과 7일 때 잡균이 각각 50%와 100% 발생되었다. 무살균 배지의 주원료가 폐면보다 옥수수 속대를 사용할 때 느타리버섯의 생산량이 $2,030\;ton\;ha^{-1}$까지 증대되었다. 무살균 배지에 균사를 배양시 균사체 배양종료일은 30일이었고, 그로부터 $2{\sim}3$일이 경과되면 균이가 발생되었다. 무살균 배지의 두께를 $70{\sim}80\;mm$로 치상하고, 온도 $22{\sim}26^{\circ}C$, 배지습도 65%, 배지의 pH $6.5{\sim}7.0$ 조건일 때 균이 발생이 가장 빨라 $22{\sim}28$일이 소요되었고, 배지두께가 두꺼울 수록균이 발생은 지연되나 느타리버섯 수량은 증가하였다. 무살균배지에서 수확한 버섯의 Ca 함량은 1차 수확 버섯이 2차 수확 버섯보다 $30.3\;mg\;kg^{-1}$이 더 많았다. 느타리버섯 자루의 굵기에 따른 아미노산 함량은 1차 수확 버섯의 갓과 자루에서 각각 $498.2{\sim}498.8\;mg\;kg^{-1}$과 $411.16{\sim}343.9\;mg\;kg^{-1}$이었고, 2차 수확 버섯의 갓과 자루에서 각각 $402.37{\sim}498.2\;mg\;kg^{-1}$과 $442.6{\sim}470.4\;mg\;kg^{-1}$ 범위이었다. 이상의 결과는 느타리버섯 재배를 할 때 $Ca(OH)_2$ 처리에 의한 무살균배지의 이용할 수 있음을 시사하고 있다.
화학적으로 안정한 다환 방향족 염료을 페수처리시설이나 천연적인 분해에 통해서 제거시키는 것이 비효율적인 것으로 알려져 있다. 다환 방향족 염료를 제거를 위하여 우리는 여러 버섯균류의 조사하였다. 한국의 야산에서 채집된 230종의 버섯으로부터 조직분리를 통하여 100개 버섯 균을 분리하였다. 염료(Bromophenol Blue, Congo Red, 혹은 Methylene Blue)를 함유한 배지에 분리된 버섯 균을 10%로 접종한 후 7일 동안 정치배양 혹은 진탕배양하여 염료제거능이 우수한 6 종의 버섯 균을 선발하였다. 이렇게 선발된 버섯 균을 대상으로 다핵방향족 염료를 제거하는 능력을 조사하였다. 모든 버섯 균이 진탕배양 시보다 정치배양 시 더 우수한 염료제거 능을 보였다. 염료에 따른 염료제거능은 대부분의 경우에서 Methylene Blue, Bromophenol Blue, Congo Red 순으로 증가하였다. 위 세 염료 중에서 Congo Red가 가장 많이 흡착되었다.
A total of 35 phosphate solubilizing bacterial strains were isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam and screened for the production of indole acetic acid (IAA). The best IAA producing strain was identified as Pantoea rodasii using 16S rRNA analysis. In addition to the IAA production, this strain could act as an efficient phosphate solubilizer (1100 ${\mu}g$$ml^{-1}$ after 5 days of incubation) also. The selected strain was cultured under different conditions in order to assess the optimum conditions for maximum IAA production. The nutrient broth (NB) medium was recorded as the best medium, where the maximum IAA production (229 ${\mu}g$$ml^{-1}$) was recorded at the start of stationary phase (12 hours after inoculation) of the bacteria growth. The performance of the strain was found to be maximum at the temperature of $30^{\circ}C$ followed by $25^{\circ}C$. IAA production was found to be increased with increasing tryptophan concentration (from 0.1 to 0.6%), however beyond this limit, a slight reduction in IAA production was observed. The strains' ability to produce IAA was further confirmed by extraction of crude IAA and subsequent TLC analysis. A specific spot from the extracted IAA preparation was found corresponding with the standard spot of IAA with same $R_f$ value. The results of HPLC analysis conducted in identifying and quantifying the IAA production more precisely, are in agreement with the results of the assessment done with colorimetric method. As revealed by the results of the pot experiment, the isolated strain could significantly enhance the growth (as measured by shoot and root growth) of mung bean plants compared to that of non-inoculated plants. Therefore it can be concluded that the present strain, Pantoea rodasii has great potential to be used as bio-inoculants.
This study was undertaken to examine the feasibility of black locust (Robinia pseudoacacia) as substrates for several edible mushrooms. For the cultivation of several edible and/or medicinal fungi on black locust, optimum bulk densities, synthetic or semisynthetic additives, natural additives and pretreatment methods were investigated. Fruit body yields of the fungi on various sawdust media composed of different wood species were also analyzed for testing the capability of black locust as a substrate for mushroom production. Mycelial growths decreased proportional when the bulk density increased. The most suitable carbon and nitrogen sources as additives to promote the mycelial growth were sucrose (2%, w/w) and ammonium phosphate (0.2%, w/w) respectively. When corn-powder and beer-waste as natural additives were added to sawdust of black locust showed the significant growth of mycelia. And the optimum mixing ratio was 10:2:1 (sawdust: corn-powder: beer-waste, w/w). Black locust after cold water treatment showed the outstanding mycelial growths. Any significant changes of pH, moisture content (%) and dry-weight losses (%) could not be found among culture substrates (sawdust of black locust, oak and poplar wood) examined before and after harvesting of fruit bodies. Yield of fruit bodies on black locust culture media were comparable with those culture media composed with oak and poplar wood. The present work indicated strongly the potentiality of black locust as raw materials for edible and medicinal mushrooms.
