Characteristics and breeding of 'Yeoreumhyang2ho': a new blackish-brown variety of Flammulina velutipes that is adaptable to high temperature (고온성 흑갈색 팽이버섯 신품종 '여름향2호'의 육성 및 특성)

  • A new blackish-brown variety of Flammulina velutipes, 'Yeoreumhyang2ho,' which was bred by crossing two monokaryons isolated from the 'Heukhyang' and 'Garlmoe' varieties, showed adaptability to high temperature. During bottle cultivation, the temperature was maintained at $16^{\circ}C$ and the variety displayed good productivity compared to the control ('Heukhyang'), without necessitating low temperature ($4^{\circ}C$) treatment. The period necessary for mycelial and fruit body growth, and primordia formation was 24 days, 8 days, and 8 days, respectively. The total cultivation period was 40 days, which was 3 days shorter than that of the control. Particularly, primordia formation took 2 days less than the control and showed initial uniformity. The yield of 'Yeoreumhyang2ho' was 154 g per 850 ml bottle, which was 8% higher than the control yield. The ${\beta}-glucan$ content was 67.9 g per 100 g dry weight, which is approximately 1.1-2.2 times the amount of ${\beta}-glucan$ in control, the golden and white mushrooms that are available in the market.

Comparison of Fruiting Body Characteristics Among Shiitake Varieties Developed by National Institute of Forest Science Using Sawdust Block Cultivation Method (사각블럭 배지에서 국립산림과학원 개발 표고 품종의 자실체 특성 비교)

  • Six strains among the shiitake varieties developed by the National Institute of Forest Science were used to select suitable varieties for sawdust block cultivation method. After 100 days cultivation (60 days in dark and 40 days in light), the weight loss rate of the sawdust block media was 14.4% (12.8% to 15.7%) in average. After 5 to 7 days of fruiting process, 2~36 primordia per medium were formed. The number of primordium formation was in the following order: Manchuhyang> Sansanhyang> Sanbaekhyang> Bambithyang> Sangjanghyang> Taehyanggo. The stipe length was 41~59 mm as followed by Taehyanggo (59 mm), Sanbackhyang (56 mm), Bambithyang (54 mm), Manchuhyang (53 mm), Sanjanghyang (45 mm) and Sansanhyang (41 mm). Among the strains, Sansanhyang had short and thick stipe. The average weight of fruiting bodies was 33~77 g as following order of Taehyanggo (77 g), Bambithyang (51 g), Sansanhyang (50 g), Manchuhyang (41 g), Sanjanghyang (34 g) and Sanbackhyang (33 g). Mushroom production per 2 kg medium was the highest in Sanbackhyang (473 g) followed by Bambithyang (287 g), Manchuhyang (175 g), Sansanhyang (109 g), Sanjanghyang (78 g) and Taehyanggo (60 g). The hardness of the cap was 1,218 to 1,839 g/10 mm. Taehyanggo was the hardest (1,839 g/10 mm) and Bambithyang was the softest (1,218 g/10 mm). In conclusion, in the cultivation condition of sawdust block medium, mushroom production was good in Sanbackhyang and Bambithyang. Sansanhyang showed good in terms of fruiting body shape.

Genetic Variation in Mutants Induced by Gamma Ray in Hypsizigus marmoreus (느티만가닥버섯에서 감마선에 의한 돌연변이체들의 유전적 변이)

  • This research was carried out to evaluate whether gamma ray is a useful tool for breeding new strains of mushrooms. For this research, 5 mutant groups, 20 strains of Hypsizigus marmoreus, 2 strains of Lyophyllum decastes, and 1 strain of Lyophyllum shimeji were used. Monokaryon spores from one variety of H. marmoreus were irradiated with 50~2,000 Gy of gamma ray. The propriety dose was 50~200 Gy for mutagenesis. Mutant monokaryon mycelia crossed each order to become dikaryon mycelia. The internal transcribed spacer (ITS) regions of rDNA were amplified using PCR, and the products were sequenced. The sequences of the ITS regions (16 partial rDNA, complete ITS1, 5.8 rDNA and partial rDNA) were analyzed by PCR, and strains of H. marmoreus, L. decastes, and L. shimeji were auto-sequenced. The lengths of the sequenced ITSs were 1,052~1,143 nucleotides. Genetic matrices were calculated using Nei-Li's genetic distance coefficient based on ITS sequence. The dissimilarities were 0~3.35% in strains of H. Hypsizigus. In addition, a phylogenetic tree was constructed based on ITS sequences using the neighbor-joining (NJ) method. The phylogenetic tree revealed that 23 strains and 5 mutant groups were divided into 12 clusters; the mutant groups fell into different clusters. These results show that mushroom spores were mutated effectively by gamma ray; therefore, gamma ray could be a useful tool for breeding new strains of mushrooms.

  • The characteristics of monosporous isolates of winter mushroom [Flammulina velutipes (Curt. ex Fr.)] were investigated to obtain useful breeding materials. Within monokaryons from cultivars which have a white colored fruitbody showed narrow genetic variation, while domestic strains which have a brown colored fruitbody showed wide variation. The mating type of the white strains was A1A2B1B2 genotype, but that of the domestic brown strains were A3A4B3B4. In intra-crossing, the genetic stability of dikaryons mated by monokaryons from white strain was less than that of parents. While in brown strain, dikaryons with high yield and color variation were obtained.

Identification of Mating Type Loci and Development of SCAR Marker Genetically Linked to the B3 Locus in Pleurotus eryngii

  • In order to estimate how diverse the mating types in Pleurotus eryngii from different regions are, pairings between monokaryons derived from inter- and intra-groups were done. Sixteen and 15 alleles were identified at loci A and B from the 12 strains. In the P. eryngii KNR2312, widely used for commercial production, four mating loci, A3, A4, B3, and B4, were determined. Those loci, except A3, were found in 4 strains out of 12 strains. To improve breeding efficiency, especially in mating type determination, RAPD and BSA were performed to screen for a mating type specific marker. The SCAR marker 13-$2_{2100}$ was developed based on the RAPD-derived sequence typing B3 locus. The sequence analysis of 13-$2_{2100}$ revealed that it contained a conserved domain, the STE3 super-family, and consensus sequences like the TATA box and GC box. It seems likely that the SCAR marker region is a part of the pheromone receptor gene.

Characterization of Newly Bred Cordyceps militaris Strains for Higher Production of Cordycepin through HPLC and URP-PCR Analysis

  • Cordyceps militaris, a member of Ascomycota, a mushroom referred to as caterpillar Dong-chung-ha-cho, is commercially valuable because of its high content of bioactive substances, including cordycepin, and its potential for artificial cultivation. Cordycepin (3'-deoxyadenosine) is highly associated with the pharmacological effects of C. militaris. C. militaris is heterothallic in that two mating-type loci, idiomorph MAT1-1 and MAT1-2, exist discretely in two different spores. In this study, nine C. militaris strains were mated with each other to prepare newly bred strains that produced a larger amount of cordycepin than the parent strains. Nine strains of C. militaris were identified by comparing the internal transcribed spacer sequence, and a total of 12 single spores were isolated from the nine strains of C. militaris. After the MAT idiomorph was confirmed by PCR, 36 mating combinations were performed with six single spores with MAT1-1 and the others with MAT1-2. Eight mating combinations were successfully mated, producing stroma with perithecia. Cordycepin content analysis of all strains by high-performance liquid chromatography revealed that the KASP4-bred strain produced the maximum cordycepin among all strains, regardless of the medium and stroma parts. Finally, universal rice primer-PCR was performed to demonstrate that the bred strains were genetically different from the parental strains and new C. militaris strains. These results may be related to the recombination of genes during mating. The newly produced strains can be used to meet the industrial demand for cordycepin. In addition, breeding through mating suggests the possibility of producing numerous cordycepin-producing C. militaris strains.

The Growth Characteristics of Pleurotus eryngii (큰느타리버섯의 자실체 생육특성)

  • In this study, we investigated the properties of incubation and growing of Pleurotus eryngii in addition to the mycological properties to use them as basic data for breeding. The speed of mycelial growth on the MCM was faster than on the PDA. The biomass in the PDB broth culture was higher than in the MCM and YMG broth culture. KNR2515 and KNR2516 required 19 days for growth of mycelia on commercial sawdust media. KNR2503 required 6.5 days and 15.3 days for pin-heading and harvesting, respectively. In morphological properties by the mushroom, the heights of KNR2312 and KNR2322 were 122.7 and 121.0 mm, respectively. The thickness of KNR2322 and KNR2513 were 39.8 mm and 31.3 mm, respectively. The weight of KNR2524's fruiting body was 36.3 g, which is good as wild strain. The quality of fruiting body of KNR2503 was 4.0 in comparison to the score 7 of commercially cultivated strains. KNR2512 had the darkest color of pileus with L value 43.6. The slow growing strains, KNR2511, KNR2513, and KNR2512 had the bright pileus with L value 80. In morphological characteristics, KNR2511, KNR2513, and KNR2515 had white lamellar and plane pileus. The three strains are supposed to be the same group and KNR2516 and KNR2518 appeared to be related to the group. The commercially cultivated strains had convex pileus, KNR2502, KNR2503, KNR2504, KNR2521, and KNR2525 had infundibuliform, and the other strains had plane pileus. Several strains were valuable for breeding, JNR2503 for growth rate, KNR2512 for pileus color, and KNR2312, KNR2322, KNR2503, and KNR2513 for the quality.

Characteristics of a new Hypsizigus marmoreus variety 'Yunseul' with little bitterness and high yield (쓴 맛이 적은 다수성 느티만가닥버섯 신품종 '윤슬')

  • This study was conducted to diversify the cultivation of mushroom items and develop a competitive variety of Hypsizigus marmoreus. We focused on developing Hypsizigus marmoreus with lower bitterness, likable shape, and high yield. We have collected and tested characteristics of genetic resources from domestic and abroad since 2019. Breeding (2019), characterization (2020), productivity test (2021), and farm demonstration test (2021) have been sequentially conducted. We bred the new variety 'Yunseul' with unique traits. The optimal temperature for mycelial and fruit body growth were 22-25℃ and 15-18℃, respectively. It was similar to the control variety (Mangadak-2Ho) in the pileus form (hemispherical shape) and the cultivation period. However, it was thinner and longer than the control variety with the pileus diameter, thickness, stipe diameter, and length being 19.9 mm, 7.0 mm, 9.4 mm, and 86.3 mm, respectively. The effective number of fruit bodies was 47.8 in bottle cultivation, which was more than that of the control variety. The yield was 197.4 g/bottle (1,100 cc), which was 30% higher than the yield of the control variety, 151.9 g/bottle (1,100 cc). The parent and control varieties were also incubated alongside the new variety. The somatic incompatibility line was distinct. The band pattern in the mycelial DNA PCR reaction was different from that of the parent and control varieties, confirming the hybrid species.

Development and Application of a Cleaved Amplified Polymorphic Sequence Marker for Discriminating A Mating Type Alleles of Lentinula edodes (표고 A 교배형 구분을 위한 CAPS 마커의 개발 및 적용)

  • Lentinula edodes is one of the most widely consumed edible mushrooms in Korea. Mating in L. edodes is regulated by a tetrapolar system, and two unlinked genetic loci, A and B, are known to be major determinants of the mating types, as reported in other heterothallic basidiomycetes. The A locus of L. edodes encodes a pair of homeodomain (HD) transcription factors. The highly variable N-termini of these HD transcription factors contribute to the diversity among the A mating types. In this study, we developed a cleaved amplified polymorphic sequence (CAPS) marker to discriminate 11 different A mating type alleles predominant among both cultivated and wild strains. Amplification of the variable region of the A locus followed by digestion with HaeIII and EcoRI restriction enzymes enabled successful discrimination among the 11 A mating type alleles. We also evaluated the applicability of this method in the identification of two A mating types of a dikaryotic strain.

Vertical Transmission of RNA Mycoviruses in Lentinula edodes (표고에서의 RNA 바이러스 수직감염)

  • Lentinula edodes is an important commercial mushroom and there have been several reports of viral infections in L. edodes. Two mycoviruses (LeV-HKB and LeNSRV1) were detected in Sanbaekhyang (NIFoS 2778) and Taehyanggo (NIFoS 4317), the sawdust-cultivated commercial strains. The vertical transmission rates of the viruses were investigated by detecting the viruses in 80 monokaryotic strains derived from basidiospores isolated from the fruiting bodies of each strain. Most of the monokaryotic strains were infected with the virus and the two viruses showed different levels of meiotic stability, with LeV-HKB showing higher meiotic stability than LeNSRV1. Therefore, it seems that the vertical transmission mechanism of mycoviruses is different depending on the virus species. We also examined the mycelial growth rate of the monokaryotic strains and compared the growth rate according to virus infection status. Although there was no statistically significant correlation between viral infection and mycelial growth rate, we found that the average growth rate was reduced by additional virus infection. We expect our data to contribute to a greater understanding of the mechanism of the vertical transmission of mycoviruses, and promoting breeding using virus-free monokaryotic strains.
