Ethacrynic Acid and Citral Suppressed the All Trans Retinoid-Induced Monocyte Chemoattractant Protein-1 Production in Human Dermal Fibroblasts

  • Skin irritation caused by retinol and retinoic acid results in mild erythema called as retinoid dermatitis. To develop compounds modulating the retinoid dermatitis, we tried to establish the screening method for retinoid dermatitis. At first we examined the inflammatory cytokine profile in neonatal human dermal fibroblasts which are known to be one of main site of retinoid action. As a result, interleukin-8 (IL-8) and monocytes chemoattractant protein-1 (MCP-1) were significantly produced by all trans retinoic acid (ATRA) and all trans retinol (ATROL) in dermal fibroblasts. Especially the production of MCP-1 was more than that of IL-8. The production of MCP-1 by retinoid was dose-dependently increased, continuing up to 24 hrs. After then using ethacrynic acid (ECA) known to reduce mouse ear edema induced by ATRA, we checked whether ECA suppressed the production of MCP-1. As a result, ECA effectively suppressed the production of MCP-1 in the ATRA- or ATROL-treated-fibroblasts. These results suggested that screening method effectively reflects the in vivo anti-inflammatory activity of ECA. It was reported that citral inhibited the enzyme involved in the conversion of ATROL to ATRA. We showed that citral suppressed the production of MCP-1 in ATROL-treated fibroblasts. We expect these finding might be helpful to find useful compounds modulating the side effects of retinoid or retinoid dermatitis.

흰쥐의 피부화상 후 저강도 레이저 조사가 표피성장인자의 발현에 미치는 영향 (The Effects of Low Power Laser for the Expression of Epithelial Growth Factor in the Burned Skin of the Rats)

  • This study was performed to investigate the effect of low power laser irradiation on epidermal growth factor(EGF) expression in the burned skin of the rats. Burns of about 3cm in diameter were created with 75'c water on the back of the rats, and the lesion of experimental group were irradiated on days 1, 2, and 3 postwounding. Control tensions were not irradiated. After burns, low power laser irradiation was applied by using 1000Hz, 830nm GaAlAs (Gallium-aluminum- arsenide) semiconductor diode laser. The expression of epidermal growth factor evaluated immunohistochemistry on mouse anti-EGF. The results of this study were as follows 1. In expression of EGF, the lesion of experimental group made EGF to more induce significantly than control tensions. 2. EGF immunoreactivity in burned skin were increased markedly 3 days after burns, and increased gradually from 1 day to 2 days in burns which is laser irradiation These data suggest that low power laser have wound healing effect in the burned skin of the rats.

발효홍삼 추출물의 광노화 피부 보호 효과 (Protective Effect of Fermented Red Ginseng Extracts on Photoaging Skin of Induced by UVB in Hairless Mice)

  • This study is designed to investigate the protective effects of fermented Red Ginseng (FRG) against photoaging in vitro and in vivo. UVB was irradiated to the human keratinocyte HaCaT cell and dorsal skin of SKH-1 mice for the induction of photoaging. After treatment of non-fermented red ginseng (NRG), fermented red ginseng (FRG), and fortified fermented red ginseng (FFRG) to the UVB irradiated HaCaT cell, ROS production and activity of MMP-9 were examined by DCFC-DA assay and gelatin zymographic assay respectively. UVB irradiated SKH-1 mice were treated with NRG, FRG, and FFRG via oral(300 mg/Kg B.W./day) and topical($100{\mu}{\ell}/mouse/day$) route.All of NRG, FRG, and FFRG had significantly reduced the intracellular ROS production elicited by UVB, among them FRG slightly more reduced the ROS production than NRG and FFRG. FFRG had slightly more reduced the MMP-9 activity in UVB irradiated HaCaT cells than NRG and FFRG in high dose. Oral and topical treatment of NRG, FRG, and FFRG had decreased the expression of matrix metalloproteinase-2, -3, and -9 in dorsal skin of UVB irradiated mice. Among them, inhibitory effect of FRG on the expression of MMP-2 was apparent. We speculate that FRG has therapeutic potentials on the UVB irradiated photoaging.

Comparison of irradiated and non-irradiated acellular dermal matrices in breast reconstruction under radiotherapy

  • Background Acellular dermal matrices (ADMs) have become an essential material for implant-based breast reconstruction. No previous studies have evaluated the effects of sterility of ADM under conditions of radiation. This study compared sterile (irradiated) and aseptic (non-irradiated) ADMs to determine which would better endure radiotherapy. Methods Eighteen male Balb/C mice were assigned to the control group with no irradiation (group 1) or one of two other groups with a radiation intensity of 10 Gy (group 2) or 20 Gy (group 3). Both sterile and aseptic ADMs were inserted into the back of each mouse. The residual volume of the ADM (measured using three-dimensional photography), cell incorporation, α-smooth muscle actin expression, and connective tissue growth factor expression were evaluated. The thickness and CD3 expression of the skin were measured 4 and 8 weeks after radiation. Results In groups 2 and 3, irradiated ADMs had a significantly larger residual volume than the non-irradiated ADMs after 8 weeks (P<0.05). No significant differences were found in cell incorporation and the amount of fibrosis between irradiated and non-irradiated ADMs. The skin was significantly thicker in the non-irradiated ADMs than in the irradiated ADMs in group 3 (P<0.05). CD3 staining showed significantly fewer inflammatory cells in the skin of irradiated ADMs than in non-irradiated ADMs in all three groups after 4 and 8 weeks (P<0.05). Conclusions Under radiation exposure, irradiated ADMs were more durable, with less volume decrease and less deposition of collagen fibers and inflammatory reactions in the skin than in non-irradiated ADMs.

p-coumaric acid, an active ingredient of Panax ginseng, ameliolates atopic dermatitis-like skin lesions through inhibition of thymic stromal lymphopoietin in mice

  • Background: Atopic dermatitis (AD) is associated with chronic skin inflammatory reactions. p-coumaric acid (pCA) is an active ingredient of Panax ginseng Meyer (Araliaceae). Methods: Here, we estimated an anti-AD effect of pCA on activated mast cells, activated splenocytes, and a mouse model of AD. Cytokines levels were measured by ELISA and protein activation was analyzed by Western blotting. 2,4-dinitrofluorobenzene (DNFB) was used to induce AD-like skin lesions. Results: The treatment with pCA suppressed the productions and mRNA expressions of thymic stromal lymphopoietin (TSLP), TNF-α, IL-6, and IL-1β in HMC-1 cells. pCA downregulated the expressions of RIP2 and caspase-1, phosphorylated-(p)p38/pJNK/pERK, and pIKKβ/pIkBα/NF-κB in HMC-1 cells. pCA also decreased the productions of TSLP, TNF-α, IL-6, IL-4, and IFN-γ in the supernatant of stimulated splenic cells. Comparing to DNFB-sensitized control group, pCA-treated group alleviated pathological changes of AD-like lesions. pCA decreased the proteins and mRNA expressions levels of TSLP, IL-6, and IL-4 in the skin lesions. Caspase-1 activation was also downregulated by pCA treatment in the AD-like lesions. The serum levels of histamine, IgE, TSLP, TNF-α, IL-6, and IL-4 were suppressed following treatment with pCA. Conclusion: This study suggests that pCA has the potential to improve AD by suppressing TSLP as well as inflammatory cytokines via blocking of caspase-1/NF-κB signal cascade.

Comparison of the presentation of atopic dermatitis induced by trinitrochlorobenzene and house dust mite in NC/Nga mice

  • Background: Atopic dermatitis (AD) is a common chronic inflammatory skin disease. To understand AD, there have been many trials establishing AD animal models. Although various trials to establish AD animal models have been existed, even the mechanisms of AD in animal models are not enough clarified. Objectives: This study assessed AD characteristics induced in Nishiki-nezumi Cinnamon/Nagoya (Nc/Nga) mice following trinitrochlorobenzene (TNCB) treatment for different periods and house dust mite (HDM) treatment to compare each model's immunological patterns, especially with cytokine antibody array tool. Methods: In this study, we exposed Nc/Nga mice to TNCB or HDM extract to induce AD. Nc/Nga mice were divided into 4 groups: control, TNCB 2 weeks-treated, TNCB 8 weeks-treated, and HDM-treated groups. After AD induction, all mice were evaluated by serum immunoglobulin E (IgE) concentration and serum cytokine antibody assays, scoring of skin lesions, scoring of scratching frequency, and histological analysis. Results: The results showed significant differences between groups in serum IgE concentration, skin lesion scores, and scratching frequency. The analysis results for serum cytokine antibody arrays showed that in the TNCB 8 weeks- and HDM-treated groups, but not in the TNCB 2 weeks-treated group, expressions of genes related to the immune response were enriched. Among the histological results, the skin lesions in the HDM-treated group were most similar to those of AD. Conclusions: We confirmed that immunological pattern of AD mice was markedly different between HDM and TNCB treated groups. In addition, the immunological pattern was quietly different dependent on TNCB treated duration.

강활과 벌사상자 추출물의 피부 미백효과 (Skin Whitening Effects of Angelica koreana and Cnidium monnieri Extracts)

  • 본 연구의 목적은 강활과 벌사상자 추출물에 의한 피부 멜라닌 형성 저해와 안정성과 자극성 시험을 통하여 기능성 화장품의 사용 물질 가능성을 평가하고자 하였다. 본 실험에 사용된 추출물은 White-$AK^{TM}$이고 INCI 명칭은 Osthole이다. White-$AK^{TM}$의 주성분은 쿠마린이고 EC50 값은 마우스 멜라노마 B16 세포 시험에서 2.7ppm이였다. White-$AK^{TM}$는 알부틴보다 100배 낮은 농도에서 저해 효과를 보였다. 미백 효과의 주 기작은 티로시나제-관련 유전자 발현을 저해하는 것으로 생각되었다. White-$AK^{TM}$ 5% 용액을 함유한 기본적인 에센스 처방을 피부에 적용 시 피부자극을 경감시켰다. White-$AK^{TM}$은 불투명한 용기에서 4주간 자외선 하에서 100%의 회수율을 보였고 $50^{\circ}C$에서 4주간 97%의 회수율을 보였다. 따라서 피부 미백효과와 자극성 경감효과가 있고 자외선 조건하에서 안정한 White-$AK^{TM}$은 화장품 처방에서 기능성 원료로 사용될 수 있을 것으로 사료된다.

라만 분광 피부 측정기를 이용한 기능성 화장품 성분의 in vivo 피부 투과 측정 및 in vitro 비교 평가 연구 (The Study on the Skin Penetration of Cosmetic Ingredient with in vivo Raman Spectroscopy and in vitro Franz Cell)

  • 현재, 기능성 화장품 성분의 피부흡수에 대한 연구 보고가 부족하며, 인체에서의 in vivo 피부 흡수는 거의 보고된 바 없다. 본 연구에서는 in vivo 라만 분광 피부 측정기를 이용하여 대표적 기능성 화장품 8종 성분의 인체 표피 투과 데이터를 수집 및 분석하고, 이를 in vitro 결과와 비교하여 주요 성분의 피부 흡수 자료를 확보하였다. 그리고 본 연구에 사용한 성분의 피부 투과도 측정한 결과를 보면, in vitro 평가에서 ascorbyl-2-glucoside, retinol, retinyl palmitate, 그리고 kojic acid가 우수한 피부 투과율을 보였으며, in vivo 평가에서는 retinol, vitamin C, 그리고 arbutin 이 우수한 것으로 나타났다. 반면, In vitro 평가와 in vivo 평가에서 가장 우수한 투과율을 보인 성분은 각각 ascorbyl-2-glucoside와 retinol이었다. 이러한 차이점은 in vitro 평가의 경우 무모생쥐 모델의 피부를 모두 관통하는 물질을 평가하였고, 라만 분광 피부 측정기의 경우는 표피의 각질층을 평가하였으므로 물질의 특성에 따라서 표피 및 진피층의 영향에 민감할 수 있기 때문인 것으로 판단된다. 그리고 대부분 물질의 투과 장벽은 각질층이므로 각질층에서 물질 이동을 살펴보는 것이 의미 있다고 생각된다. 결론적으로, 본 연구에서는 라만 분광 피부 측정기를 이용하여 대표적 기능성 화장품 성분의 피부 흡수에 대한 기초적인 자료를 확보하였으며, 따라서 이들 자료는 추후 많은 연구에 활용될 수 있을 것으로 생각된다.

사인추출물의 인슐린 유사 성장인자-3의 합성과 피부 노화 개선에 대한 효과 (Effects of Amomi, Semen Extract on Synthesis of Insulin-like Growth Factor-1 and Anti-wrinkle in Skin)

  • 본 연구에서 프로모터-리포터 분석방법을 통해 인슐린 유사 성장인자-1의 프로모터를 자극하는 천연물을 선별한 결과 사인 추출물이 가장 좋은 프로모터 자극효과를 나타냈으며, 무모생쥐에서 패쇄첩포 후 RT-PCR로 실험한 결과 IGF-1 mRNA를 $35\%$ 증가시키는 것으로 나타났다. 사인 추출물의 피부 주름개선 효과를 알아보기 위하여 인체 섬유아세포에서는 type-I collagen과 MMP-1 합성 변화를 관찰하였으며, 무모생쥐에서는 콜라젠의 증가와 진피 두께를 관찰하였다. 그 결과, 동위원소를 이용한 콜라젠 증가실험에서 type-I collagen은 $38\%$ 증가하였으며 무모생쥐에서 실시한 RT-PCR 결과에서는 mRNA가 $21\%$ 증가하는 것으로 관찰되었다. MMP-1 효소발현의 경우 ELISA 분석을 통해서 $63\%$의 높은 발현저해능을 확인하였고 Western blot에서도 발현이 저해되는 것을 확인하였다 추출물을 무모생쥐에 패쇄첩포 하였을 경우 대조군에 비해 진피 두께가 두꺼워지고 콜라젠 양도 증가되는 것으로 조직염색 관찰을 통해 확인하였다. 이상의 결과를 통해 사인추출물은 피부 주름개선에 좋은 효과를 나타내는 것으로 보이며, 여기에는 인슐린 유사성장 인자-I의 발현증가와 관련된 기전이 관여하는 것으로 판단된다.

Activation of melanogenesis by non-thermal atmospheric pressure plasma

  • Several reports have demonstrated the wide range of nonthermal plasma applications in biomedical field including cancers, diabetics, wound healing and cosmetics. Recently, it has been shown that plasma is able to modulate the p38 MAPK and JUN level in cells which has a crucial role in melanin synthesis and skin pigmentation. Therefore we investigated the effect of plasma on melanogenesis in-vitro using melanoma (B16F10) cells and in-vivo using mouse and zebra fish. To investigate the mechanism of plasma action, plasma device characteristics were measured, reactive species inside and outside the cells were detected, and western blot was performed to find the signaling pathway involved in melanin activation in-vitro and in-vivo. This is the first report presenting the role of nonthermal plasma for melanogenesis which provides a new perspective of plasma in the field of dermatology.
