Background: Based on outstanding progresses in animal experiments, vaccines for some human tumors have been developed. However, clinical effects of these vaccines have been far below than expected. This discrepancy might come from differences between animal models and human patients with respect to immunocompetency. The immune status of mice after tumor inoculation has not been well studied, which make us cautious in interpreting and applying the results from mice to human. We evaluated cell-mediated immune responses in mice after tumor cell inoculation. Methods: Mice were inoculated with TA3Ha, CT26, or 4T1. Delayed-type hypersensitivity (DTH) responses were induced 2-4 weeks after inoculation using 2,4-dinitro-1-fluorobenzene as an antigen. The relationships between the severity of DTH responses and the duration of tumor inoculation or the size of tumor mass were analyzed. Results: In T A3Ha groups, DTH response was elevated 2 weeks after inoculation, but depressed after 4 weeks, compared to the control group. When analyzed based on the sizes of tumor masses elicited, DTH responses were inversely related to the mass size, especially in those greater than 10 mm in diameter. In CT26 groups, while the duration after inoculation did not affect the severity of DTH responses, those with large mass showed depressed responses regardless the duration of inoculation. 4T1 cells grew so slowly that the size of tumor mass was small even 4 weeks after inoculation, and this group showed much higher DTH responses compared to that of tumor-free group. Conclusion: At least in an experimental setting where tumor model was induced by inoculating tumor cell lines into immunologically competent mice, the host immune response was elevated in early stage, and then depressed in late stage when the mass grew over a critical size.
모발은 피부를 둘러싸고 있는 피부 부속기관이며, 외부로부터 들어오는 물리적, 화학적 자극에 대하여 보호하고, 미용 효과 등의 기능을 가지고 있다. 모발은 피부의 표피와 진피의 사이에서 발생하며, 모발성장주기를 가지고 있다. 모발성장주기는 모발이 성장하는 시기(anagen)와 성장을 끝내는 시기(catagen), 성장을 멈추고 휴식하는 시기(telogen)로 구분할 수 있다. 모발성장기 중, 성장기의 모낭 부위에는 많은 성장인자들이 분포하고 있다는 것이 이미 보고된 바 있다. 뇌하수체에서 분비되는 인간 성장 호르몬은 신진대사를 수행하는 신호를 전달하며, 기관의 성장을 촉진하는 역할을 수행한다. 성장호르몬은 혈류를 따라 온몸으로 이동되며, 여러 가지의 성장인자들을 활성하거나 억제하는 조절 작용을 한다는 연구결과가 보고된 바 있다. 본 연구에서는 인간성장호르몬을 유전자재조합기술을 이용하여 관찰이 용이하도록 6개의 히스티딘 아미노산을 첨가하였으며, 나노크기의 리포좀으로 캡슐화한 LhGH를 (주)리제론으로 부터 받아 사용하여 마우스(C57BL6/CrN)에서 LhGH에 의한 모발의 성장 관계를 알아보았다. 8주령의 마우스 3마리씩 증류수, Liposome, Minoxidil, His hGH, LhGH를 실험의 목적에 따라 20일, 6주 동안 도포하였고, 피부 속의 침투 경로는 형광면역반응법을 이용하여 형광현미경으로 관찰하였다. 또한, 광학현미경과 주사전자현미경을 이용하여 모발 수의 변화를 비교하여 보았다. 마우스 피부에 도포한 LhGH는 피부표피의 모공과 모발을 따라 피부 안으로 이동되는 것이 광학현미경으로 관찰되었다. 또한 LhGH는 성별에 관계없이 모발 수를 증가시켰으며, 피부 밖으로 도출된 모발 수를 증가시킴을 확인하였다. 자연적으로 탈모된 마우스에서도 발모를 증진시켰으며, 발모 후 3주 동안 새로운 탈모현상을 억제하였다. 이러한 결과로 LhGH는 마우스(C57BL/6CrN)에서 모발의 수를 증가시키며, 탈모된 피부에서 모발의 재성장을 촉진시키는 효과가 있다고 생각한다.
Purpose: Pseudomonas aeruginosa is an etiologic agent in serious wound infection. Pseudomonas aeruginosa infection is problematic because this organism is resistant to many antimicrobial drugs. The purpose of this study was to compare the bactericidal effect of commonly used topical agents and their effect on wound healing. Methods: Pseudomonas aeruginosa-infected full-thickness skin defect was developed on the mouse to compare 3 commonly used topical agents-Betadine, 2% Gentamicin solution and 0.3% Acetic acid with the control group. Wound size change, bacterial colony counts and histologic findings of each groups were analyzed. Results: The wound size decreased in all treated groups as compared with the control group. However, there was no statistical difference. Gentamicin solution group was showed the lowest bacterial colony count and statistically significant difference compared with the control group(p=0.032). Other treated groups were also effective against Pseudomonas aeruginosa, but not different statistically. Histologic findings revealed that epithelialization, granulation tissue formation and microvessel proliferation were increased and necrosis and inflammation were decreased in all treated groups compared to the control group, but not different statistically. Betadine group significantly increased granulation tissue formation compared to the control group (p= 0.041). Conclusion: There is no universal topical agent that enhances most aspects of wound healing while simultaneously decreasing the bacterial concentration. However, Gentamicin solution may be an optimal topical agent for Pseudomonas aeruginosa infected wound. Further study should experiment on human with Gentamicin solution to confirm a effect on Pseudomonas aeruginosa infected wound for clinical applications.
Objectives: This study investigated the effects of water extract of Coix lacrymajobi var. mayuen on obesity and its associated factors in high fat diet (HFD) induced obese mice. Methods: Male C5BL/6 mice were randomly assigned to 4 groups, normal group (chow diet), a HFD, HFD with water extract of Coix lacrymajobi var. mayuen 100 mg/kg (W-Coix), and HFD with phentermine 5 mg/kg (Phen) as positive control. Body weight, food intake, fasting blood glucose were checked every week and then organs, blood serums were collected after 16 weeks of treatment. Results: Body weight and adipocyte size were significantly lesser in W-Coix than in HFD group; however energy efficiency in W-Coix were not different with HFD. The oral glucose tolerance test, serum glucose and insulin were significantly decreased in W-Coix, also lipid accumulations in liver tissue and lipid levels (total cholesterol, high density lipoprotein-cholesterol) were improved with W-Coix treatment. However skeletal muscle loss with HFD was not changed in W-Coix compared with HFD group. Conclusions: The W-Coix treatment decreased body weight, adipocyte size and it is associated with improved glucose and lipid metabolism. Further studies are needed to know the mechanism of antiobesity in W-Coix.
Background: Nanoparticles (NPs) prepared from biodegradable polymers, such as poly (D,L-lactic acid-co-glycolic acid) (PLGA), have been studied as vehicles for the delivery of antigens to phagocytes. This paper describes the preparation of antigen-loaded PLGA-NPs for efficient cross-priming. Methods: NPs containing a similar amount of ovalbumin (OVA) but different sizes were produced using a micromixer-based W/O/W solvent evaporation procedure, and the efficiency of the NPs to induce the cross-presentation of OVA peptides were examined in dendritic cells (DCs). Cellular uptake and biodistribution studies were performed using fluorescein isothiocyanate (FITC)-loaded NPs in mice. Results: The NPs in the range of $1.1{\sim}1.4{\mu}m$ in size were the most and almost equally efficient in inducing the cross-presentation of OVA peptides via $H-2K^b$ molecules. Cellular uptake and biodistribution studies showed that opsonization of the NPs with mouse IgG greatly increased the percentage of FITC-positive cells in the spleen and lymph nodes. The major cell type of FITC-positive cells in the spleen was macrophages, whereas that of lymph nodes was DCs. Conclusion: These results show that IgG-opsonized PLGA-NPs with a mean size of $1.1{\mu}m$ would be the choice of biodegradable carriers for the targeted-delivery of protein antigens for cross-priming in vivo.
터치 인터페이스 입력방식에서 메뉴를 마우스를 이용하여 입력하는 것 보다 터치입력 방식이 의도하지 않는 터치와 입력의 어려움이 증가하고 있다. 본 연구에서는 버튼메뉴의 최소 형상버튼을 기준으로 형태와 크기변화에 따라서 사용성 평가를 분석하고자 한다. 적용방법으로는 터치 입력방식의 인터페이스 디자인에서 활용 할 수 있도록 회귀 모형식과 인터페이스 인간 수행 평가방법의 대표적인 모형인 피츠의 법칙(Fitts' Law)의 비교분석을 통해서 더욱 효과적인 정량적 수행예측 평가 모형식 적용을 통해서 가능성을 확인하고자 했다. 따라서 터치 환경에서 적용할 수 있는 버튼의 형상과 크기변화에 따른 사용자의 사용시간을 고려한 수행예측시간을 디자인에 반영할 수 있다.
With the development and widespread application of online shopping, the number of online consumers has increased. With one click of a mouse, people can buy anything they want without going out and have it sent right to the doors. As consumers benefit from online shopping, people are becoming more concerned about protecting their privacy. In the group buying scenario described in our paper, online shopping was regarded as intra-group communication. To protect the sensitive information of consumers, the polynomial-based encryption key sharing method (Piao et al., 2013; Piao and Kim, 2018) can be applied to online shopping communication. In this paper, we analyze security problems by using a polynomial-based scheme in the following ways : First, in Kamal's attack, they said it does not provide perfect forward and backward secrecy when the members leave or join the group because the secret key can be broken in polynomial time. Second, for simultaneous equations, the leaving node will compute the new secret key if it can be confirmed that the updated new polynomial is recomputed. Third, using Newton's method, attackers can successively find better approximations to the roots of a function. Fourth, the Berlekamp Algorithm can factor polynomials over finite fields and solve the root of the polynomial. Fifth, for a brute-force attack, if the key size is small, brute force can be used to find the root of the polynomial, we need to make a key with appropriately large size to prevent brute force attacks. According to these analyses, we finally recommend the use of a relatively reasonable hash-based mechanism that solves all of the possible security problems and is the most suitable mechanism for our application. The study of adequate and suitable protective methods of consumer security will have academic significance and provide the practical implications.
Ursolic acid (UA), a bioactive triterpene acid, has been known to increase collagen content in human skin in addition to other actions such as anti-inflammatory, skin-tumor prevention and anti-invasion. However, it is poorly soluble in water. Therefore, we firstly prepared microemulsion system with benzyl alcohol, ethanol and Cremophor EL, RH 40 and Brij 35 as surfactant in order to increase solubility of UA and then prepared microemulsion was dispersed in o/w cream base for the topical delivery of UA in an effort to improve anti-wrinkle effect. The pseudo-ternary phase diagrams were developed and various microemulsion formulations were prepared using benzyl alcohol as an oil, Cremophor EL, RH 40 and Brij 35 as a surfactant. The droplet size of microemulsions was characterized by dynamic light scattering. The accumulation of VA in the skin from topical cream was evaluated in vitro using hairless mouse skins. The mean droplet size was $26.8{\pm}6.6$ nm for microemulsions II with Cremophor EL. All UA creams showed pseudoplastic flow and hysterisis loop in their rheogram, depending on the type of materials added in topical creams. The in vitro accumulation data demonstrated the UA topical cream prepared with the combination of Poloxamer 407 and Xanthan gum as a copolymer showed higher accumulation percentage than those prepared with either Poloxamer 407 or Xanthan gum. These results suggest that UA topical cream using microemulsion systems may be promising for the topical delivery of UA.

Development of a biomarker for predicting tumor-treatment efficacy is a matter of great concern, to reduce time, medical expense, and effort in oncology therapy. In a preclinical study, we hypothesized that the blood-flow parameter based on laser speckle flowmetry (LSF) could be a potential indicator to estimate the efficacy of breast-cancer treatment. To verify this hypothesis, a 13762-MAT-B-III rat breast tumor was grown in a dorsal skinfold window chamber applied to a nude mouse, and the change in blood flow rate (BFR) - or the speckle flow index (SFI) is used together as the same meaning in this manuscript - was longitudinally monitored during tumor growth and metronomic cyclophosphamide treatment. Based on the daily LSF angiogram, several BFR parameters (baseline SFI, normalized SFI, and △rBFR) were compared to tumor size in the normal, treated, and untreated tumor groups. Despite the incomplete tumor treatment, we found that the daily changes in all BFR parameters tended to have partially positive correlation with tumor size. Moreover, we observed that the changes in baseline SFI and normalized SFI responded one day earlier than the tumor shrinkage during chemotherapy. However, daily variations in the hypercapnia-induced △rBFR lagged tumor shrinkage by one day. This study would contribute not only to evaluating tumor vascular response to treatment, but also to monitoring blood-flow-mediated diseases (in brain, skin, and retina) by using LSF in preclinical settings.
Purpose: In order to determine the amount of wound healing, objective sequential assessments of changes in wound size and depth are essential. Although a variety of measurements for wound healing have been proposed, a gold standard for quantifying day-to-day changes in healing has not been established. We present here a simple and non-invasive wound measurement method that quantitatively and accurately documents changes of the size of a raw surface and the volume of a soft tissue defect using a stereoimage optical topometer(SOT) system. Methods: Using a 5mm diameter biopsy punch, four circular wounds were created on abdominal area of a diabetic mouse. Photographs were taken using SOT system at baseline, 5th day and 10th postoperative day. The wound margin was traced on a digitalized photo and evaluated the area and the volume of the wound by SOT system. Results: The SOT system calculated a mean wound surface of $15.93{\pm}0.29mm^2$ and volume of $827.50{\pm}88.86$ intensity/pixel${\times}$area(I/PA) immediately after wounding. On the 5th day after the operation wound surface declined by $10.73mm^2$ and on the 10th day declined by $5.95mm^2$. The wound volume also declined from 827.50 I/PA to 161.75 I/PA and 30.50 I/PA on 0, 5th and 10th day, respectively. Conclusion: The SOT system described in this study represents a reliable, simple, practical, and non-invasive technique to accurately monitor and evaluate wound healing.
