Characteristics of The Wastewater Treatment Processes for The Removal of Dyes in Aqueous Solution(2) - Ozonation or ACF Adsorption Treatment of Reactive Dyes - (수용액 중의 염료 제거를 위한 폐수처리공정의 특성(2) - 반응성염료의 오존산화 및 섬유상활성탄 흡착 처리 -)

  • This study was carried out to treat the aqueous solutions containing reactive dyes(RB19, RR120 and RY179) by the Ozone demand flask method and adsorption process using activated carbon fiber(ACF) which are one of the main pollutants in dye wastewater. Ozone oxidation of three kinds of the reactive dyes was examined to investigate the reactivity of dyes with ozone, competition reaction and ozone utilization on various conditions for single- and multi-solute dye solution. Concentration of dyes was decreased continuously with increasing ozone dosage in the single-solute dye solutions. Competition quotient values were calculated to investigate the preferential oxidation of individual dyes in multi-solute dye solutions. Competition quotients(CQi) and values of the overall utilization efficiency, ${\eta}O_3$, were increased at 40mg/l of ozone dosage in multi-solute dye solutions. ACF(A-15) has much larger specific surface area$(1,584m^2/g-ACF)$ in comparison with granular activated carbon adsorbent (F400, $1,125m^2/g-GAC$), which is commonly used, and most of pores were found to be micropores with pore radius of 2nm and below. It was found that RB19 was most easily adsorbed among the dyes in this study. In the case of PCP (p-chlorophenol) and sucrose, which are single component adsorbate, adsorption capacities of ACF(A-15) were in good agreement with the batch adsorption measurement, and saturation time predicted of ACF columns for these components was also well agreed with practically measured time. But in the case of reactive dyes, which have relatively high molecular weight and aggregated with multi-components, adsorption capacities or saturation time predicted were not agreed with practically measured values.

Syntheses of Polysiloxane-Bridged Dinuclear Metallocenes and Their Catalytic Activities

  • The polysiloxane-bridged dinuclear metallocenes $[(SiMe_2O)_n-SiMe_2(C_5H_4)_2][(C_9H_7)ZrCl_2]_2$ (n=1 (7), 2 (8), 3 (9)) have been generated as a model complex for the immobilized metallocene at silica surface by treating the respective disodium salts of the ligands with 2 equivalents of $(C_9H_7)ZrCl_3$ in THF. All three complexes are characterized by $^1H$ NMR and measurement of metal content through ICP-MS. It turned out that the values of ${\Delta}{\delta}=[{\delta}_d-{\delta}_p]$, the chemical shift difference between the distal $({\delta}_d)$ and proximal $({\delta}_p)$ protons, for the produced dinuclear compounds (0.47 for 7, 0.49 for 8, and 0.5 for 9) were larger than the Δδ value of the known ansa-type complex holding the same ligand as a chelating one, that is just the opposite to the normal trend. In order to compare polymerization behavior of the dinuclear metallocene with the corresponding mononuclear metallocene, (Cp)$(C_9H_7)ZrCl_2$ was separately prepared. To investigate the catalytic properties of the dinuclear complexes and mononuclear metallocenes ethylene polymerization has been conducted in the presence of MMAO. The polymerization results display the typical activity dependence on polymerization temperature for all complexes. The most important feature is that the polymers from the dinuclear metallocenes represent enormously improved molecular weight compared with the polymer from the corresponding mononuclear metallocene. In addition, the influence of the nature of the bridging ligand upon the reactivities of the dinuclear metallocenes has also been observed.

Intramolecular Disulfide Bonds for Biogenesis of Calcium Homeostasis Modulator 1 Ion Channel Are Dispensable for Voltage-Dependent Activation

  • Calcium homeostasis modulator 1 (CALHM1) is a membrane protein with four transmembrane helices that form an octameric ion channel with voltage-dependent activation. There are four conserved cysteine (Cys) residues in the extracellular domain that form two intramolecular disulfide bonds. We investigated the roles of C42-C127 and C44-C161 in human CALHM1 channel biogenesis and the ionic current (ICALHM1). Replacing Cys with Ser or Ala abolished the membrane trafficking as well as ICALHM1. Immunoblotting analysis revealed dithiothreitol-sensitive multimeric CALHM1, which was markedly reduced in C44S and C161S, but preserved in C42S and C127S. The mixed expression of C42S and wild-type did not show a dominant-negative effect. While the heteromeric assembly of CALHM1 and CALHM3 formed active ion channels, the co-expression of C42S and CALHM3 did not produce functional channels. Despite the critical structural role of the extracellular cysteine residues, a treatment with the membrane-impermeable reducing agent tris(2-carboxyethyl) phosphine (TCEP, 2 mM) did not affect ICALHM1 for up to 30 min. Interestingly, incubation with TCEP (2 mM) for 2-6 h reduced both ICALHM1 and the surface expression of CALHM1 in a time-dependent manner. We propose that the intramolecular disulfide bonds are essential for folding, oligomerization, trafficking and maintenance of CALHM1 in the plasma membrane, but dispensable for the voltage-dependent activation once expressed on the plasma membrane.

Optimization, Purification, and Characterization of Haloalkaline Serine Protease from a Haloalkaliphilic Archaeon Natrialba hulunbeirensis Strain WNHS14

  • The present study addresses isolation, optimization, partial purification, and characterization of a haloalkaline serine protease from a newly isolated haloarchaeal strain isolated from Wadi El Natrun in Egypt. We expected that a two-step sequential statistical approach (one variable at a time, followed by response surface methodology) might maximize the production of the haloalkaline serine protease. The enzyme was partially purified using Hiprep 16/60 sephacryl S-100 HR gel filtration column. Molecular identification revealed the newly isolated haloarchaeon to be Natrialba hulunbeirensis strain WNHS14. Among several tested physicochemical determinants, casamino acids, KCl, and NaCl showed the most significant effects on enzyme production as determined from results of the One-Variable-At-A-time (OVAT) study. The BoxBehnken design localized the optimal levels of the three key determinants; casamino acids, KCl, and NaCl to be 0.5% (w/v), 0.02% (w/v), and 15% (w/v), respectively, obtaining 62.9 U/ml as the maximal amount of protease produced after treatment at 40℃, and pH 9 for 9 days with 6-fold enhancement in yield. The enzyme was partially purified after size exclusion chromatography with specific activity, purification fold, and yield of 1282.63 U/mg, 8.9, and 23%, respectively. The enzyme showed its maximal activity at pH, temperature, and NaCl concentration optima of 10, 75℃, and 2 M, respectively. Phenylmethylsulfonyl fluoride (PMSF, 5 mM) completely inhibited enzyme activity.

Improved Coating of PEDOT : PSS onto CVD Graphene by the Addition of PVA (PVA의 첨가에 의한 CVD 그래핀상 PEDOT : PSS의 코팅성 향상)

  • We successfully coated poly(3,4-ethylenedioxythiophene) : poly(styrene sulfonate) (PEDOT : PSS) on CVD graphene by adding poly(vinyl alcohol) (PVA) to PEDOT : PSS. Extensive studies on the wettability of coating solutions and electrical properties of formed films led us to conclude that PVA with 89% of the degree of saponification and the molecular weight of less than $100,000gmol^{-1}$ produced optimum results. Furthermore, the optimum content of PVA was found to be 5% of PEDOT : PSS by the solid weight. The film coated by PEDOT : PSS with PVA on CVD graphene displayed a conspicuous improvement in the surface roughness, adhesive property, bending durability and stability in resistance at $160^{\circ}C$, compared to those of using CVD graphene films.

Review on Graphene Oxide-based Nanofiltration Membrane (산화그래핀 기반 나노여과막의 최신 연구동향)

  • Various two-dimensional nano materials such as graphene, zeolite, and metal-organic framework have been utilized to develop an ultra-thin high-performance membrane for water purification, gas separation, and so on. Particularly, in the case of graphene oxide, synthesis methods and thin film coating techniques have been accumulated and established since early 2000s, therefore graphene oxide has been rapidly applied to membrane field. The multi-layered graphene oxide thin film can filter molecules separately by the molecular sieving of interlayer spacing between adjacent layers, and it is also possible to separate various materials depending on the surface functional groups or the degree of interaction to intercalated materials. This review mainly focuses on the nanofiltration application of graphene oxide. The major factors affecting the separation performance of graphene oxide membrane in solvent are summarized and other technical elements required for the commercialization of graphene oxide membranes will be discussed including stability issue and fabrication method.

Viator vitreocola gen. et sp. nov. (Stylonematophyceae), a new red alga on drift glass debris in Oregon and Washington, USA

  • A new encrusting red alga was found growing abundantly on glass debris items that drifted ashore along the coasts of Oregon and Washington. These included discarded fluorescent tubes, incandescent light bulbs, capped liquor bottles, and ball-shaped fishing-net floats. Field collections and unialgal cultures of the alga revealed that it consisted of two morphological phases: a young loosely aggregated turf and a mature consolidated mucilaginous crust. The turf phase consisted of a basal layer of globose cells that produced erect, rarely branched, uniseriate to multiseriate filaments up to $500{\mu}m$ long with closely spaced cells lacking pit-plugs. These filaments expanded in size from their bases to their tips and released single cells as spores. At maturity, a second phase of growth occurred that produced a consolidated crust, up to $370{\mu}m$ thick. It consisted of a basal layer of small, tightly appressed ellipsoidal-to-elongate cells that generated a mucilaginous perithallial matrix containing a second type of filament with irregularly spaced cells often undergoing binary division. At the matrix surface, the original filaments continued to grow and release spores but often also eroded. Individual cells, examined using confocal microscopy and SYBR Green staining, were found to contain a central nucleus, a single highly lobed peripheral chloroplast without a pyrenoid, and numerous chloroplast nucleoids. Morphological data from field and culture isolates and molecular data (rbcL, psbA, and SSU) show that this alga is a new genus and species which we name Viator vitreocola, "a traveller on glass."

Adsorption of Cesium and Strontium Ions in Aqueous Phase Using Porous Metal Organic Frameworks Connected with Functional Group (작용기 적용 다공성 금속 유기골격체를 이용한 수중 세슘 및 스트론튬 이온의 흡착 제거)

  • In the current study, MIL-101(Cr)-SO3H[HCl] as metal-organic frameworks (MOFs) was fabricated via a hydrothermal method. The physicochemical properties of the synthesized material were characterized using XRD, FT-IR, FE-SEM, TEM, and BET surface area analysis. The XRD diffraction pattern of the prepared MIL-101(Cr)-SO3H[HCl] was similar to previously reported patterns of MIL-101(Cr) type materials, indicating successful synthesis of MIL-101(Cr)-SO3H[HCl]. The FT-IR spectrum revealed the molecular structure and functional groups of the synthesized MIL-101(Cr)-SO3H[HCl]. FE-SEM and TEM images indicated the formation of rectangular parallelopiped structures in the prepared MIL-101(Cr)-SO3H[HCl]. Furthermore, the EDS spectrum showed that the synthesized material consisted of the elements of Cr, O, S, and C. The fabricated MIL-101(Cr)-SO3H[HCl] was then employed as an adsorbent for the removal of Sr2+ and Cs+ from aqueous solutions. The adsorption kinetics and adsorption isotherm models were studied in detail. The maximum adsorption capacities of MIL-101(Cr)-SO3H[HCl] for Sr2+ and Cs+ according to pH (3, 5.3~5.8, 10) were 35.05, 43.35, and 79.72 mg/g and 78.58, 74.58, and 169.74 mg/g, respectively. These results demonstrate the potential of the synthesized MOFs, which can be effectively applied as an adsorbent for the removal of Sr2+ and Cs+ ions from aqueous solutions and other diverse applications.

Effect of length of alkyl chain consisting of fluorine and carbon in self-assembled monolayers

  • We investigated the interfacial properties of fluorocarbon self-assembled monolayers (FC-SAMs) with different alkyl chain lengths. It was found that the substrate characteristics were changed rapidly with the fabrication time and temperature of the SAM. FC-3SAM, which has the shortest alkyl chain in this study, showed a contact angle of $54.1^{\circ}$ when it was fabricated in an electric oven at $60^{\circ}C$ for the first minute. The FC-3SAM showed a contact angle of up to $76.9^{\circ}$ when it was fabricated in an electric oven at the same temperature condition for 180 minutes. FC-10SAM, which has the longest alkyl chain in this study, showed a contact angle of $64.7^{\circ}$ when it was fabricated at a temperature condition of $60^{\circ}C$ for 1 minute, and a contact angle of $98.7^{\circ}C$ at a temperature condition of $60^{\circ}C$ for 180 minutes. It was found that the FC-10SAM shows an increased contact angle and hydrophobic properties due to a well-aligned molecular structure resulting from a strong van der Waals force. In contrast, the FC-3SAM shows a small contact angle due to the intermolecular disorder resulting from a weak van der Waals force. The average roughness of FC-SAMs was investigated using AFM. The surface roughness of FC-SAMs, which verifies the results of contact angle, was confirmed. At a fabrication time of 120 minutes, the FC-10SAM showed an improvement in average roughness by 62% compared to that of FC-3SAM due to its good alignment.

An Emergence of Equine-Like G3P[8] Rotaviruses Associated with Acute Gastroenteritis in Hospitalized Children in Thailand, 2016-2018

  • Rotavirus A (RVA) is recognized as a major etiology responsible for the development of acute gastroenteritis in children worldwide. The purpose of the present study was to perform the molecular characterization of RVA. A total of 323 stool specimens collected from hospitalized children with acute gastroenteritis in Chiang Rai, Thailand, in 2016-2018 were identified for G- and P-genotypes through RT-PCR analysis. RVA was more prevalent in 2017-2018 (37.8%) than in 2016-2017 (23.2%). The seasonal peak of RVA occurred from March to April. G3P[8] was predominant in 2016-2017 (90.6%) and 2017-2018 (58.6%). Other genotypes including G1P[8], G8P[8], G9P[8], and mixed infections were also identified. G3P[8] strains clustered together in the same lineage with other novel human equine-like G3P[8] strains previously identified in multiple countries and presented a genotype 2 constellation (G3-P[8]-I2-R2-C2-M2-A2-N2-T2-E2-H2). Several amino acid differences were observed in the antigenic epitopes of the VP7 and VP8* capsid proteins of the equine-like G3P[8] compared with those of the RVA vaccine strains. The homology modeling of the VP7 and VP8* capsid proteins of the equine-like G3P[8] strains evidently exhibited that these residue differences were present on the surface-exposed area of the capsid structure. The emergence of the equine-like G3P[8] strains in Thailand indicates the rapid spread of strains with human and animal gene segments. Continuous surveillance for RVA is essential to monitor genotypes and genetic diversity, which will provide useful information for selecting rotavirus strains to develop a safe and effective RVA vaccine that is efficacious against multiple genotypes and variants.
