Cosmeceutical Properties of Fructan (Levan) Produced by Zymomonas mobilis

  • Fructan, a polysaccharide existing in plants or produced by microorganisms, is a sugar polymer of fructose with $\beta$-2,6 linkages. In this study, we investigated some cosmeceutical properties of Fructan such as moisturizing effect, cell proliferation effect, anti-inflammation effect and cell cytotoxicity. Zymomonas mobilis, a microorganism producing Fructan, was cultured in a medium containing 10% sucrose and 2% yeast extract as main components for 24 hours at 37$^{\circ}C$ and pH 7. Fructan was obtained by precipitation from the cultured medium by adding alcohol (alcohol ratio of 1:3) after removing the enzyme by centrifuging. Fructan exhibited almost same moisturizing effect as hyaluronic acid and cell proliferation effect on human fibroblast and keratinocyte as well. Moreover, on cell proliferation test on bio-artificial skin constructed by 3-dimensional(3-D) culture after inducing primary skin inflammation with 0.5% sodium lauryl sulfate (SLS), the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan exhibited higher cell proliferation than the 3-D artificial skin treated with SLS only. On anti-inflammation test on 3-D artificial skin evaluated by measuring secreted quantity of interleukin-1$\alpha$ (IL-1$\alpha$) which is a pre-inflammatory mediator induced by SLS, the quantity of IL-1$\alpha$on the 3-D artificial skin treated with 0.01 mg/ml, 0.05mg/ml of Fructan was less than the one on the 3-D artificial skin treated with SLS only. As a result of these studies, Fructan has anti-inflammation effect against inflammatory reaction by a skin irritant as well as cell proliferation effect in bio-artificial skin. Fructan was also evaluated as a safe material without any toxicity in safety tests using fibroblasts and animals.

Enhancing the Effect of Aronia Extract on Hyaluronic Acid Synthesis through Liposome Formation

  • Background and objective: Aronia melanocarpa, called black chokeberry, is a natural product belonging to the family rosaceae, and is known to contain polyphenolic antioxidants including cyanidin-3-galactoside, cyanidin-3-arabinoside, cyanidin-3-xyloside, and cyanidin-3-glucoside Because of the abundance of anthocyanins, Aronia has been studied to be used in various industries. Methods: Aronia melanocarpa extract was treated 24 hours a day to RAW 264.7 cells with inflammations induced by LPS. After extracting total RNA, the amount of inflammatory cytokine expression was measured using RT-PCR. After processing the Aronia liposome using Aronia extract and the layer-by-layer electrostatic deposition method in keratinocyte cells at the same time, we checked the synthesis of Hyaluronic acid enhanced through the formation of Aronia liposome using ELISA. Results: The treatment of Aronia extract in inflammation-induced RAW 264.7 cells conducted to check the anti-inflammatory efficacy of Aronia extract inhibited inflammatory cytokines including TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased the mRNA expression of HAS2 genes related to moisturizing. Based on the anti-inflammatory and moisturizing effect of Aronia extract, the Aronia liposome technology was introduced to Aronia extract to produce Aronia liposome. Conclusion: The liposome formation of Aronia extract is expected to be used as a functional material in treating various inflammatory skin diseases by controlling the moisture content of the corneocytes by increasing the expression rate of genes associated with the synthesis of hyaluronic acid, while retaining the efficacy of its components.

Cosmetical Properties of Polysccharides from the Root Bark of Ulmus davidiana var. Japonica (유근피로부터 추출한 다당류의 화장료적 특성)

  • The fruit or root of Ulmus davidiana var. Japonica and its extract has traditionally been used as anti inflammatation medicines as well as a remedy of the digestive ulcer for a long time. Ulmus davidiana var. Japonica has also been used to cure palliative and diuresis, because it contains lot of mucus and tannin. In this study, we examined the moisturizing effect and anti-inflammation effect of polysaccharides extracted from Ulmus davidiana var. Japonica to investigated whether it can be used as a cosmetic ingredient or not. The polysaccharides were extracted from Ulmus davidiana var. Japonica. Then the hydrolyzed was obtained through the hydrolysis. It has been ascertained that the polysaccharides we mainly composed of rhamnose, galactose, and glucose through HPLC experiments, and it has 20,000 of the molecular weight and 89.3 dL/g of intrinsic viscosity In a moisturizing test of measuring water loss in a desiccator and moisture content by Comeometer CM820, Ulmus davidiana root extract showed a similar moisturizing effect as hyaluronic acid. And its anti-inflammation effect on carrageenin-induced edema evaluated by measuring the thickness of swelling over paw induced by carrageenin (25 uL of $2\%$ carrageenin saline injection), the $\%$ swelling over paw treated with Ulmus davidiana root extract was much less than that of paw treated with carrageenin only. The anti-inflammatory effect of Ulmus davidiana root extract was almost similar to that of ketoprofen. These results can say that Ulmus davidiana root extract can be effectively used as a cosmetic ingredient.

Effects of 1-tetradecanol and β-sitosterol Isolated from Dendropanax morbifera Lev. on Skin Whitening, Moisturizing and Preventing Hair Loss (미백, 보습 및 탈모방지에 대한 황칠나무(Dendropanax modifera Lev.)에서 분리한 1-tetradecanol, β-sitosterol의 효과)

  • Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea and has been used in folk medicine. However, the effects of Dendropanax morbifera Lev. on skin biology remain to be elucidated. In this study, we isolated 1-tetradecanol and ${\beta}$-sitosterol from the n-hexane fraction of Dendropanax mobifera Lev. and To investigate the whitening effect of the fraction, we tested the inhibition of tyrosinase activity of 1-tetradecanol. The results show that the inhibitory effect of the 1-tetradecanol was higher than water extract and n-hexane fraction. And 1-tetradecanol significantly reduced melanin contents of B16F10 cells compared to more than water extract and n-haxane fraction dose-dependantly without cell cytotoxicitiy (below $100{\mu}g/mL$). We also investigated the skin moisturizing effect using HR-1 hairless mice. The transepidermal water loss (TEWL) in the 1-tetradecanol treated group was significantly smaller than that in the other groups. To investigate the effect of the preventing hair loss by ${\beta}$-sitosterol, we observed HR-1 hairless mice through periodic growth feature. The results suggest that hair loss of mice by ${\beta}$-sitosterol was delayed and it's hair density showed the highest. These data provide evidence that Dendropanax morbifera Lev. may be a potent candidate for the improvement of both skin whitening, moisturizing and alopecia from the point of cosmetic industry view.

Antibaterial Activity and Skin Moisturizing Effect of Cedrela sinensis A. Juss Shoots Extracts (참죽나무 새순 추출물의 항균 활성과 피부 보습효과에 관한 연구)

  • In this study, we investigated the antibacterial activity and the skin moisturizing effect of cream contaning Cedrela sinensis A. Juss shoots extracts. MIC values of ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts on P. acnes and S. aureus were 0.13 %, 0.03% respectively and P. ovale and E. coli were 0.25 % equally. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes and S. aureus were higher than the methyl paraben. The cream containing the ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts was formulated and its skin moisturizing effect was evaluated in a clinical trial. The skin hydration and transepidermal water loss (TEWL) were investigated after topical application of the cream. The skin hydrating effect of the cream containing Cedrela sinensis A. Juss shoots extract was 10 ~ 15 % higher than the placebo cream, and transepidermal water loss was decreased to 6.7 g/$m^2h$ in the subjects applied with cream containing the ethyl acetate fraction of Cedrela sinensis A. Juss shoots extracts. These results indicate that ethyl acetate fraction from Cedrela sinensis A. Juss shoots extracts could be applicable to new functional antibacterial cosmetics.

Anti-wrinkle Effect of Pressure Sensitive Adhesive Hydrogel Patches Containing Ulmi Cortex Extract (유백피 추출물을 함유한 하이드로겔 패치의 주름 억제 효과)

  • The decreasing effect of wrinkle on the pressure sensitive adhesive hydrogel patches containing ulmi cortex extract and sorbitol as the drug for anti-wrinkle were investigated. In this study, hydrogels were prepared by the crosslinking reaction of acrylic polymers and aluminum ions produced by L(+)-tartaric acid hydrolysis of the dihydroxy aluminum aminoacetates. The inhibition concentration of ulmi cortex extract on the collagenase exhibited at 0.01%. Furthermore, the moisturizing effect of hydrogel patches formulated with sorbitol was higher than that without it. In vivo animal test in hairless mouse showed that the ulmi cortex-loaded hydrogel patches had about 31.2% of anti-wrinkle effect compared to blank (before attaching the patches). Human test showed that only 33% of subjects showed the decreasing of wrinkle during 8 weeks. In conclusion, the model pressure sensitive adhesive hydrogel patches in this study would be pharmaceutically applicable for the wrinkle treatment on the facial skin.

Moisturization and Transdermal Penetration Characteristics of PEGimpregnated Aloe vera Gel from DIS Processing (DIS에 의한 Polyethylene Glycol 함침 알로에 베라 겔의 보습 및 경피흡수 특성)

  • This study was carried out to investigate the in vitro and in vivo moisturizing properties and percutaneous absorption of PEG-impregnated Aloe vera gel. The PEG-i-Aloe gel was obtained from dewatering and impregnation by soaking (DIS) of Aloe vera leaf slice. The moisturizing property of the obtained sample was evaluated by moisture determination using gravimetric method in desiccator under different RH% and by water sorption-desorption test on human skin. The transdermal penetration characteristics of PEG-i-Aloe gel was investigated by Franz diffusion cell in vitro transdermal absorption method. PEG-i-Aloe gel had high moisture retention ability and could significantly lead the enhancing skin hydration status as well as reducing the skin water loss due to the film formation as a skin barrier. The skin penetration rate of PEGi- Aloe gel at steady state was 9.76 ${\mu}g/(h{\cdot}cm^2)$ and the quantity of the transdermal absorption was 144 ${\mu}g/cm^2$ in 9 hr. The penetration mechanism was well fitted with Higuchi model ($R^2$ = 0.974-0.994). The results show that PEG-i-Aloe gel has the significant moisturizing effect and strong penetration of the animal skin. It could be used as the moisturizing additive in cosmetic skin products.

In vitro anti-skin-aging effects of dried pomegranate concentrated powder

  • Purpose : In this study, we intended to observe the anti-wrinkle and moisturizing effects of dried pomegranate juice concentration powder (PCP) using in vitro test. Materials and methods : Antioxidant effects of PCP were determined by free radical scavenging capacity (DPPH assay) and the cytotoxicity of PCP was examined in human keratinocyte (HaCaT) and human primary dermal fibroblast-neonatal (HDF) cells. To investigate the moisturizing effect of PCP, hyaluronan synthesis was examined in HaCaT cells. Activity of procollagen production were assessed in HDF cells and elastase inhibition properties of PCP were evaluated in cell free condition, to determine their anti-wrinkle effects. Metalloproteinase 1 (MMP-1) activity was also assessed following UVB irradiation, in the current in vitro experiment. Results : No PCP treatment related significant cytotoxic effects were demonstrated against to the both HDF and HaCaT cells. PCP showed favorable free radical scavenging activities in dose-dependent manner. In PCP-treated HaCaT cells, hyaluronan synthesis was non-significantly but markedly increased, and pro-collagen productions were significantly increased in HDF cells, at all three different concentrations (0.25, 0.75 and 1 mg/ml), and elastase inhibitory activities were observed by PCP treatment. A significant decrease in UVB-induced MMP-1 activity was also observed in 1 mg/ml PCP-treated HDF cells as compared to those of UVB-exposed cells. Conclusions : Taken together, these results suggest that PCP has favorable antioxidant, anti-wrinkle and moisturizing effects without meaningful cytotoxicity on HDF and HaCaT cell lines.

  • This study described about a liquid crystalline technology that is used in cosmetics industry. Various intermediate phases may exist between solid and liquid. At high surfactant concentration, several liquid crystalline phases can be made to have formed. Although molecular arrangement with crystallization is not regular, it is known that more relative regular state is liquid crystalline or meso-phase than liquid phase. Usually, it described in detail about manufacturing method that explained about a kind of liquid crystalline technology in cosmetics, a new liquid crystalline technology, and makes liquid crystalline. Specialty, it introduced about kind of special an emulsifier to form liquid crystalline. There were hydrogenated lecithin, ceramide, dipalmitoylhydroxyproline, DEA-cetyl phosphate, Gemini-surfactant in representative raw material to form liquid crystalline. Liquid crystalline extent that used polarization microscope to observe formation, and appears best from 400times, 600times, 1,000times well appeared. Also, droplet particle size that liquid crystal is made best 1.0-10.0$\mu\textrm{m}$ be. General emulsion more than superior result that measures the skin moisturizing effect to take advantage of liquid crystalline technology of vitamin was seen. As presence at a clinical result, wave and general emulsion more than superior result (more than 20%) that measures skin moisturizing effect about liquid crystalline of vitamin B$\_$5/ were seen (ANOMA t-test, p<0.05)

Moisturizing Effect and Durability of Sun Protection Factor (UVA/B) Activity with Multiple Emulsion (W/O/W) System (멀티플 에멀전(W/O/W) 시스템을 이용한 자외선차단성능(UVA/B)의 내수성과 보습효과)

  • This study is to evaluate moisturizing effect and durability of UV A/B blocking activity with multiple (W/O/W) emulsion system. Most of the sun protective products come to be hot issue having both high SPF and long-lasting activity as using special products when is going out, mountain climbing and sports. Also, many consumers prefer the products which have the excellent waterproofing activity of sun care cosmetics as well as the non-sticky feeling that carried out the study of the sensorial science and texture preference. Therefore, development of the specific formulation using this multiple (W/O/W) emulsion technology, it has O/W type hydro skin feel having soft and moist texture when it is treated on the skin. Finally, this formulation is instantly changed to W/O type feel after adsorbed into the skin. The purpose of this study is to get high SPF lasting effect having high water resistance tactivity with high functional multiple (W/O/W) emulsion cream. We used major ingredients, UV-B absorbers were selected with ethylhexyl methoxycinnamate, isoamyl-p-methoxycinnamate, ethylhexylsalicylate, and octocrylene, UV-A absorbers were selected with butylmethoxydibenzoylmethane, bis-ethylhexyloxyphenol methoxy phenyltriazine. SPF effect of O/W type cream was 34.1. SPF effect of W/O/W type cream was 40.6 (increased about 19%). Water resistance effect after 4 hours, SPF effect of O/W type cream was 3.6 (quickly drop down). SPF effect of W/O/W type cream having 81.0 % waterproofing effect was 32.7 (decreased about SPF 7.9). Moisturizing effect of O/W cream at first was superior comparing multiple emulsion. But after 3 hours quickly was drop-down. Moisturizing effect of multiple emulsion was high comparing O/W type and other sun block creams after 4 hours was constantly maintaining water-content.
