Applications of Transposon-Based Gene Delivery System in Bacteria

  • Mobile genetic segments, or transposons, are also referred to as jumping genes as they can shift from one position in the genome to another, thus inducing a chromosomal mutation. According to the target site-specificity of the transposon during a transposition event, the result is either the insertion of a gene of interest at a specific chromosomal site, or the creation of knockout mutants. The former situation includes the integration of conjugative transposons via site-specific recombination, several transposons preferring a target site of a conserved AT-rich sequence, and Tn7 being site-specifically inserted at attTn7, the downstream of the essential glmS gene. The latter situation is exploited for random mutagenesis in many prokaryotes, including IS (insertion sequence) elements, mariner, Mu, Tn3 derivatives (Tn4430 and Tn917), Tn5, modified Tn7, Tn10, Tn552, and Ty1, enabling a variety of genetic manipulations. Randomly inserted transposons have been previously employed for a variety of applications such as genetic footprinting, gene transcriptional and translational fusion, signature-tagged mutagenesis (STM), DNA or cDNA sequencing, transposon site hybridization (TraSH), and scanning linker mutagenesis (SLM). Therefore, transposon-mediated genetic engineering is a valuable discipline for the study of bacterial physiology and pathogenesis in living hosts.

  • This paper examines critical successful elements related to the management of new product development process. For this, we have selected a VK-X100, slim bar type cellular phone, produced by VK mobile co., which was thought as a successful product development case and then analysed the case from a concept driven strategic product development's point of view. We have performed in-depth interviews with project managers, engineers and designers to get information on the product development process and analysed huge amount of data obtained from web-sites, global market indicators, and related firms' materials to find a changing trend of market conditions. The findings suggest that the successful new Product development and management is shaped by a number of different factors, such as the business environments, firm's technological level, firm's deep market insight capabilities, nature of the product development activities, and so on. Finally, we suggested the limitation of our research and further research subjects that would be required to support our findings.

A Broadband Microstrip Array Antenna for 3G Smart Antenna System Testbed

  • A compact and broadband $4{\times}1$ array antenna was developed for 3G smart antenna system testbed. The $4{\times}1$ uniform linear array antenna was designed to operate at 1.885 to 2.2GHz with a total bandwidth of 315MHz. The array elements were based on the novel broadband L-probe fed inverted hybrid E-H (LIEH) shaped microstrip patch, which offers 22% size reduction to the conventional rectangular microstrip patch antenna. For steering the antenna beam, a commercial variable attenuator (KAT1D04SA002), a variable phase shifter (KPH350SC00) with four units each, and the corporate 4-ways Wilkinson power divider which was fabricated in-house were integrated to form the beamforming feed network. The developed antenna has an impedance bandwidth of 17.32% ($VSWR{\leq}1.5$), 21.78% ($VSWR{\leq}2$) with respect to center frequency 2.02GHz and with an achievable gain of 11.9dBi. The design antenna offer a broadband, compact and mobile solution for a 3G smart antenna testbed to fully characterized the IMT-2000 radio specifications and system performances.

  • This paper suggests a PDA-based scientific traffic accident investigation system in order to improve the reliability and accuracy of a traffic accident investigation procedure. In order to fulfill this objective, a standard accident sheet with 32 elements to be used in the PDA system was developed through a reference review and an expert survey. Using this information, the required functionalities and realization procedure of the PDA system for traffic accident investigation were presented including mobile network environment and GPS/GIS design. A prototype of PDA system is presented and more improved and efficientmanagement is highly expected by applying the system developed in this paper.

  • In this paper, we implement and evaluate the performance of a vector-based rasterization algorithm for 3D graphics by using a SIMD (single instruction multiple data) many-core processor architecture. In addition, we evaluate the impact of a data-per-processing elements (DPE) ratio that is defined as the amount of data directly mapped to each processing element (PE) within many-core in terms of performance, energy efficiency, and area efficiency. For the experiment, we utilize seven different PE configurations by varying the DPE ratio (or the number PEs), which are implemented in the same 130 nm CMOS technology with a 500 MHz clock frequency. Experimental results indicate that the optimal PE configuration is achieved as the DPE ratio is in the range from 16,384 to 256 (or the number of PEs is in the range from 16 and 1,024), which meets the requirements of mobile devices in terms of the optimal performance and efficiency.

  • Paved road dust emissions were investigated 14 times on 12 main roads in Seo-Cho Gu, Seoul, Korea by vehicle-based mobile sampling system(Road Dust Monitoring System) during September to December 2011. Also, fourteen heavy metals present in the dust samples were analyzed by ICP. ICP analysis showed that one of major source of the road dust would be urban construction. A large amount of silt was found, which might be originated mainly from building construction and open beds of trees. Trace element and pollution indices of heavy metals(Cd, Cu, Ni, Pb, Zn) on the roads adjacent to the commercial area had higher concentrations than those on the roads adjacent to the construction and residential areas because of traffic density and heavy traffic.

Trimethoprim Resistance by Class I Integron in Vibrio parahaemolyticus from a Fish Farm (어류 양식장에서 분리한 Vibrio parahaemolyticus의 Class I Integron에 의한 Trimethoprim 내성)

  • A trimethoprim resistant Vibrio parahaemolyticus, which cause acute gastroenteritis in humans, was isolated from farmed fish and seawater. The resistance profiles of isolated V. parahaemolyticus and their correlation with mobile elements were investigated. All of the V. parahaemolyticus were resistance to both rifampin and trimethoprim. The presence of class I integron was confirmed by PCR. PCR-amplified inserted gene cassettes contained aminoglycoside aac6-II, rifampin arr-3 and trimethoprim dfrA27 resistance genes. This study indicated that class I integron mainly contributed to the circulation of trimethoprim resistance determinants in V. parahaemolyticus.

  • One of the trends on information storage device is focused on the development of micro-optical devices with OFH (Optical Flying Head). Many different types of sliders for OFH and optical component systems have been introduced in the literature. However, the important issue on the mechanical system, which consists of suspension, swing arm and VCM (Voice Coil Motor) part has not been discussed up to date. In this research, we analyses the suspension, which is used for the small form factor MO drive. The analysis is in process two ways. One is probabilistic analysis, another is dynamic shock analysis. Probabilistic analysis is a technique you can use to assess the effect of uncertain input parameters and assumptions on your analysis model. Using a probabilistic analysis you can and out how much the results of a finite elements analysis are affected by uncertainties in the model. Dynamic shock analysis is used for the mobile applications. The mechanical robustness of the suspension simulating the shock responses of a disk-suspension is proposed in this paper.

  • The recent issue of optical pickup actuators is to apply optical storage devices to mobile devices such as a cellular phone and PDA. It requires actuators to become smaller than conventional types. As the size becomes smaller, the magnetic force is reduced and the assembly of optical pickup actuators becomes more difficult. In addition, its dynamic characteristics are changed. In this paper, methods to improve magnetic forces and dynamic characteristics are suggested and the optimal result of the plate spring design is obtained. A diamond shape magnet and the fine pattern coil (FPC) are used to improve magnetic forces and damping elements are attached to decrease the peak magnitude of the mode instead of using structural damping, mostly for the purpose of improving the accuracy of the finite element simulation. To get more stable dynamic characteristics than conventional ones, a plate spring is applied to the optical pickup actuator and it is optimized with topology and parameter optimization to obtain the concept and the detail design, respectively.

  • This paper aims at analyzing the traffic of CDMA base station. RTD(Round Trip Delay) method, used for the study, is one of the developed tool for calculating the call setup time between the BTS(Base Station TransceiverSubsystem) and mobile station.We compare the calculated call setup time in air with the field experiments.And we suggest the RTD method for dividing the traffic of the connected repeater from that of the BTS, andwe can testify it by the experiment which analyze the difference of the received time between the base stationand the repeater including the forced delay elements.
