Two series of 2-(5-nitro-2-furyl)- and 2-(1-methyl-5-nitro-1H-imidazol-2-yl)-5-propyl, allyl and propargyl)thio-1,3,4-thiadiazoles (6a-f) and 2-(5-nitro-2-furyl)- and 2-(1-methyl-5-nitro-1 H-imidazol-2-yl)-5-(nitrobenzyl)thio-1,3,4-thiadiazole derivatives (8a-f) have been synthesized and evaluated against Mycobacterium tuberculosis, as part of the TAACF TB screening program under direction of the US National Institute of Health, the NIAID division. Primary screening was conducted at a single concentration, 6.25 $\mu\textrm{g}$mL$^{-1}$ , against M. tuberculosis H$_{37}$ Rv in BACTEC 12B medium, using the Microplate Alamar Blue Assay (MABA). The minimum inhibitory concentration (MIC) was determined for the compounds that demonstrated $\geq$90% growth inhibition in the primary screening. A varying degree of antituberculosis activity (from 0-97% of growth inhibition) was observed with the alkylthio series (6a-f), and the nitroimidazole derivative with a propylthio group (6b) and the nitrofuran derivative with a propargylthio group (6e), were the most active compounds (MIC=3.13 and 1.56 /$\mu\textrm{g}$mL$^{-1}$ , respectively). Among the nitrobenzylthio derivatives (8a-f), all the ortho, meta and para nitrobenzyl isomers in the nitrofuran series exhibited good antituberculosis activity (MIC=3.13 $\mu\textrm{g}$mL$^{-1}$ ), while the corresponding nitroimidazole analogues were completely inactive (Inhibition=0%).

Objectives: Irrigants are imperative in endodontic therapy for the elimination of pathogens from the infected root canal. The present study compared the antimicrobial efficacy of octenidine dihydrochloride (OCT) with chlorhexidine (CHX) and sodium hypochlorite (NaOCl) against Staphylococcus epidermidis (S. epidermidis) for root canal disinfection. Materials and Methods: The minimum inhibitory concentration (MIC) was obtained using serial dilution method. The agar diffusion method was then used to determine the zones of inhibition for each irrigant. Lastly, forty 6-mm dentin blocks were prepared from human mandibular premolars and inoculated with S. epidermidis. Samples were randomly divided into 4 groups of 10 blocks and irrigated for 3 minutes with saline (control), 2% CHX, 3% NaOCl, or 0.1% OCT. Dentin samples were then collected immediately for microbial analysis, including an analysis of colony-forming units (CFUs). Results: The MICs of each tested irrigant were 0.05% for CHX, 0.25% for NaOCl, and 0.0125% for OCT. All tested irrigants showed concentration-dependent increase in zones of inhibition, and 3% NaOCl showed the largest zone of inhibition amongst all tested irrigants (p < 0.05). There were no significant differences among the CFU measurements of 2% CHX, 3% NaOCl, and 0.1% OCT showing complete elimination of S. epidermidis in all samples. Conclusions: This study showed that OCT was comparable to or even more effective than CHX and NaOCl, demonstrating antimicrobial activity at low concentrations against S. epidermidis.
Purpose : The study objective was to assess the antibacterial activity of essential oil of basil against S. mutans and P. gingivalis and to explore its potential to prevent dental caries and peridontal disease. Method : Essential oil of basil, extracted using steam distillation, was diluted with triple distilled water and Tween 20 to generate samples at various concentration, that is 30%, 50%, and 70% (v/v). Strains of S. mutans and P. gingivalis were incubated in the medium under anaerobic condition. Broth microdilution susceptibility testing and plate incubation diffusion were utilized to determine the minimum inhibitory concentration (MIC) and to measure antibacterial activity, respectively. Result : An upsurge in antibacterial activity was seen to correlate with and increase in the concentration used for both bacterial strains, but was more significant with S. mutans. A statistically significant growth inhibition effect and reduction in the number of colonies was also observed with both strains dependent on the concentration used following 24 hours of incubation. Conclusion : Thus, the current study finding was that essential oil of basil was effective against dental caries and periodontal disease and could be used in dentifrice to help prevent oral disease.
담배(Nicotiana tabacum cv. BY4)의 이배체식물과 담배의 약으로부터 유도된 반수체식물의 잎절편을 사용하여 2.4-D를 단독톄처리하여 캘러스를 유도하였다. 캘러스증식은 이배체에서는 2.0 mg/L 2.4-D와 1.0mg/L 카이네틴의 조합처리구에서, 반수체의 경우에 있어서는 2.0mg/L 2.4-D와 0.1mg/L BAP 조합처리구에서 실시하였으며, 현탁배양도 동일조건으로 실시하였다. 현탁배양 세포괴를 배양 15-20일째가 되면 0.8% low melting agarose와 혼합하여 카드a뮴이 0-2,000 $\mu$M의 농도로 처리된 선발배지에 평판배양하여 카드뮴저항성 colony(cell clump)를 조사관찰하였다. 그 결과 본 실험에서는 이배체는 세포의 성장에 있어서 최하저해농도(MIC : Minimum Inhibition Concentration)는 Cd$^{++}$ 300 $\mu$M이었으며 반수체는 Cd$^{++}$ 200 $\mu$M이었다. 이때 반수체 및 이배체의 MIC에서 선발된 세포주를 카드뮴 500, 1,000, 2,000 $\mu$M 농도로 옳겨 경화시킨 후 카드뮴저항성을 선발하였고 그 특성을 조사하였다. 그 결과 카드뮴 저항성을 띤 반수체 및 이배체유래 캘러스의 경우에는 각각 카드a뮴의 농도가 증가할수록 생중량은 감소하였으나, 건조중량은 오히려 증가하였다. 그리고 각각 선발된 세포주내의 카드뮴의 함량을 AAS를 사용하여 측정하였던 바, 카드뮴의 농도가 높을수록 캘러스에 축적된 카드뮴의 양이 높게 나타났다.
수종의 한약계를 70% 메탄올로 추출하여 식중독 세균인 Staphylococcus aureus에 대한 항균활성으로 생균수의 측정, 생육저해환의 측정, 최소저해농도와 최소사멸농도를 조사한 결과 이들 추출물의 Staphylococcus aureus에 대한 항균활성의 크기는 Rubus coreanus > Sanguisorba officinalis > Eriobotrya japonica > Prunus mume > Crataegus pinnatifida로 나타났다. 생육저해환은 복분자가 16.5 mm, 지유 14.3 mm, 오매 14.0 mm, 산사 12.7 mm로 측정되었다. MIC는 지유가 5mg / ml로 가장 낮았으며, MBC는 복분자가 10mg / ml로 나타났다. 실험에 사용된 한약재 추출물중 복분자, 비파엽, 산사, 오매, 지유는 Staphylococcus aureus에 대해서 강한 항균활성을 나타내었다. 따라서 이들 추출물을 이용한 천연보존료를 개발하여 식품에 적용한다면 그 기대효과가 상당히 클 것으로 생각된다.
In the study reported here, the antimicrobial and antioxidant activities of the ethanol leaf extract of Dendropanax morbiferus Lev. from Jeju Island was investigated. Of the 14 strains of 12 species of microorganisms tested, the extract exhibited antimicrobial activity observed against seven Gram-positive bacteria of four species, but not against six Gram-negative bacteria and a yeast strain. Using the disc diffusion method, the diameter of the inhibition zone increased with application of the extract with every strain and the highest growth inhibition was exhibited with Staphylococcus aureus KCTC 1916 at 5 mg/ml. The minimal inhibitory concentration of the extract (MIC) by turbidity was 2.5 mg/ml against Bacillus cereus KACC 12672 and 15 mg/ml when with Enterococcus faecalis KCTC 3206. The minimum bacterial concentration (MBC) values defined as being $${\geq_-}99.9%$$ reduction in viable cells against the tested strains was higher than the MIC values. Time killing curves using the optimum MIC were performed on seven strains incubated for 48 hr. The growth of B. cereus KACC 12672 was detected after 12 hr and no significant growth was found in the others strains after 48 hr (p<0.05). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity of the ethanol leaf extract was similar to that of butylated hydroxyanisole (BHA) at the same concentration. These results indicate that leaf extract of D. morbiferus Lev. can be utilized as a natural preservative and an antioxidant.
Objectives: Pleurotus eryngii is used both for edible and medicinal purposes, and has a physiological activity. The purpose of this study is to investigate the antibacterial effect of Pleurotus eryngii against six oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Methods: The antibacterial activities of various extracts of Pleurotus eryngii were examined by disc diffusion assay and minimum inhibitory concentration (MIC). The disc diffusion assay was performed by putting a paper disc soaked in extracts on plates inoculated bacterial cultures. The MIC of these extracts was determined by using a broth microdilution assay at a concentration ranging between 0.03 mg/ml to 15.00 mg/ml. The growth inhibition effect of extracts was measured at 600 nm for 24 hrs. Results: The antibacterial activity was confirmed against all six tested bacteria at Pleurotus eryngii ethyl acetate extract by the disc diffusion method. Acetone extract showed the antibacterial activity only against 4 strains containing Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, and Actinomyces viscosus. In ethanol extract, no activity was observed against other strains except Staphylococcus aureus. MIC values of ethyl acetate extract were the same, 7.50 mg/ml in all tested bacteria. Conclusions: Pleurotus eryngii exhibited the antibacterial activity against oral pathogens (Staphylococcus aureus, Streptococcus criceti, Streptococcus mutans, Streptococcus ratti, Streptococcus sobrinus, and Actinomyces viscosus). Thus, Pleurotus eryngii may be considered as a natural antibacterial agent for treatment of dental diseases.
단삼으로부터 항균성 물질을 추출하여 어류질병에 대한 천연 항균약제로서의 이용 가능성을 타진하기 위하여 어류질병세균에 대한 항균효과, 최소증식억제농도, 최소살균농도 및 어류사료에 적용 시험 등을 실시하였다. 단삼 추출물은 그람양성균에 대하여 강한 항균활성을 나타내었으나, 그람음성균에 대하여는 항균활성이 약하였다. 단삼 추출물의 그람양성균에 대한 최소증식억제농도는 13.4 ~40.3 $\mu\textrm{g}$/ml이었으며, 67.2~403.0 $\mu\textrm{g}$/ml의 농도에서는 살균작용을 보였다. 그러나 그람음성균인 Edwardsiella tarta에 대하여는 403.0 $\mu\textrm{g}$/ml를 첨가하여도 살균작용은 물론 균 증식을 억제하지도 못하였다. 사료를 단삼 추출액에 침지시 적정 침지시간은 10분이었고, 단삼 추출물에 침지시킨 사료는 사용된 모든 어류질병세균의 증식을 억제하여 10.5~14.5 mm의 저해환을 형성하였으며, 특히 Streptococcus 속 및 Vibrio anguillarum에 대하여 강한 항균활성을 나타내었다. 또한, 사료 중량에 따른 추출물 흡수량은 식 Y=2.4953X+3.3276 ($R^2$=0.9999)으로 아주 밀접한 상관관계를 나타내었으며, 단삼추출물에 침지시킨 사료의 항균활성은 저장온도 10~35$^{\circ}C$에서 28일까지 안정하였다.
Objective : Infectious diseases are a growing problem worldwide by Methicillin-resistant Staphylococcus aureus (MRSA). Daehwangmokdan-tang is one of the oriental medicine prescriptions contained in Principles and Practice of Eastern Medicine. This study investigated the antibacterial activity of EtOH 70% extracts of Daehwangmokdan-tang (DMT) which prescription is composed of oriental medicine against MRSA. Methods : The antimicrobial activity and active concentration of MRSA were verified by measuring the minimum inhibitory concentration (MIC) of DMT. In addition, the effects of the disease were checked by treating the existing antibiotics and large ethanol extract in parallel, and the extent of growth suppression was checked over time. In addition, cell membrane permeability experiment confirmed the effect of large DMT on the immunity mechanism of MRSA. Results : TThe minimum inhibitory concentration of DMT against MRSA is 500 ~ 2000 ㎍/㎖ by broth dilution method. In the checkerboard method, the combinations of DMT with antibiotics has partial synergistic effect or synergy effect and DMT markedly reduced the MICs of the antibiotics oxacillin (OX), gentamicin (GEN) against MRSA. In the inhibition of resistance mechanism of DMT against MRSA, the expression of resistance gene and protein about β-lactam antibiotic was reduced. Also, we observed the effect of DMT about cell membrane permeability against MRSA, and confirmed that DMT suppressed growth of strains by increasing cell membrane permeability and energy metabolism. Conclusion : Basis on the result, we speculate that DMT may be useful for the treatment of MRSA infections when used in combination with β-lactam antibiotic.
Objective : The purpose of this study is to investigate the antibacterial activity of Saussurea lappa for the applications of herb-based extracts to both cosmetic and medicinal industries. Methods : Antibacterial activity of ethanol extract of Saussutes lappa (ESL) was assessed using agar diffusion and broth dilution methods, and determined by whether clear zone was formed around paper disc and in terms of the size (mm) of clear zone, Results : ESL provided activity against Staphylococcus aureus KCTC 1927 in concentration of 1 mg/ml with an clear zone of 16 mm, and showed an activity against Staphylococcus epidermids KCTC 1917 in concentration of 1 mg/ml with an clear zone of 18mm. Moreover, the minimum inhibitory concentration (MIC) of ESL against S. sureus and S. epidermidls were 1 mg/ml for both bacterial species. However, ESL showed no growth inhibition against Pseudomonas aeruginosa KCTC 12513, Enterobactet aerogenes KCTC 2190, Escherichia coli KCTC 2571, Salmonella typhimurium KCTC 1925 and Propionibacterium acnes KCCM 41747. Conclusions : Antibacterial activity of ESL against Staphylococcus sureus and Staphylococcus epidermids causing eye and skin diseases was proved. The result suggest that ESL may be useful as a natural preservative on behalf of synthetic preservatives.
