The Experimental Study on Anti-bacterial Potency of Jinpi-san on Staphylococcus aureus & Staphylococcus epidermidis. (진피산(秦皮散)이 Staphylococcus aureus와 Staphylococcus epidermidis에 미치는 항균효과에 대한 실험적 연구)

  • Objective : This experimental study was performed to investigate the effect of Jinpi-san on Staphylococcus aureus(S. aureus) and Staphylococcus epidermidis(S. epidermidis) that induce keratitis. Methods : Minimal inhibitory concentration(MIC) was measured by dropping to $50{\mu}l$ according to density Jinpi-san(100%, 10%, 1%, 0.1 %). Anti-bacterial potency was measured by the size of inhibition zone with change of volume. Results : 1. MIC on S. aureus in Jinpi-san was $40{\mu}l$ undiluted solution. 2. MIC on S. epidermidis in Jinpi-san was $20{\mu}l$ undiluted solution. Conclusions : These results indicate that Jinpi-san can be used to cure S. aureus, S. epidermidis that induce eye disease(keratitis). If further study is performed, the use of eye drops will be valuable and beneficial in the clinical medicines.

Effect of Silver Ion Solution on the Inhibition of Microcystis Growth (은이온 수용액의 Microcystis 생장 억제 효과)

  • The effect of silver ion solution on the growth of Microcystis aeruginosa UTEX 2388 (cyanobacterium) and Chlorella sp. KCTC AG20136 (green alga) was investigated using separated and mixed culture in filtered natural water and BG11 medium. In separated culture, M. aeruginosa UTEX 2388 and Chlorella sp. KCTC AG20136 were found to be sensitive to 0.01 and 0.1 mg L$^{-1}$ of silver ion, respectively. Also, the silver ion concentrations for the growth inhibition of M. aeruginosa UTEX 2388 and Chlorella sp. KCTC AG20136 in the mixed culture were same in separated culture. Cyanobacteria were more sensitive to the silver ion solution than green algae. In bloom sample, the minimal inhibition concentration of silver ion solution for the low Chl-${\alpha}$ sample (110$\sim$190 ${\mu}g$ L$^{-1}$) and high Chl-${\alpha}$ sample (1,500$\sim$1,900 ${\mu}g$ L$^{-1}$) was about 0.1 and 3.0 mg L$^{-1}$, respectively. The silver ion concentration for the inhibition of algal bloom sample was affected by the algal biomass. In order to use silver ion solution for the control of algal bloom, the silver ion concentration must be determined in consideration of a minimal effect on the environment.

Cytotoxicity and Antimicrobial Effects of Extracts from Salvia miltiorrhiza (단삼 추출물의 세포독성과 항균효과)

  • This study was carried out to evaluate cytotoxic effects of Salvia miltiorrhiza extracts on NIH 3T3 fibroblasts and KB cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The comparison of $IC_{50}$ of values of Salvia miltiorrhiza extracts in KB cell lines showed that their susceptibility to these extracts decreased in the following order: hexane extract > chloroform extract > methanol extract> dichloromethane extract > ethyl acetate extract>ethanol extract by the MTT method. The dried roots of Salvia miltiorrhiza was extracted several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentrations (MIC's) of the extract against microorganisms were also examined. Amtimicrobial activity of ketoconazol as reference was compared to those of extracts of hexane, chloroform, dichloromethane, ethyl acetate, ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi. These results suggest that the hexane and chloroform soluble extracts of Salvia miltiorrhiza may be a valuable choice for the studies on the tumor cell lines and growth inhibition activity.

Inhibition of Growth and Collagenase Activity of the Extract from Salvia miltiorrhiza against Microorganisms Causing Periodontal Diseases (단삼(Salvia miltiorrhiza) 추출물의 치주질환유발 세균의 생육억제 및 Collagenase 저해 활성)

  • This study was carried out to evaluate the inhibition of growth and collagenase activity of Salvia miltiorrhiza Bunge against microorganisms causing periodontal diseases. The ethanol extract of Salvia miltiorrhiza showed sig-nificant growth inhibition against microorganisms causing periodontal diseases. Ethanol extract was further fractionated with organic solvents in the order of hexane, chloroform and ethyl acetate. Among the fractions tested, the hexane fraction showed the highest cell growth inhibition. The minimal inhibitory concentration (MIC) of the extract against C. curvus, C. rectus, E. corrodens, F nucleatum, P. gingivalis, P. intermedia and W. succinogenes were 200, 50, 50, 250, 150, 250 and 200 ${\mu}g$/ml, respectively. The inhibition of collagenase activity by organic solvent fractions were higher than that of minocycline, and the inhibition ratio of collagenase activity was $88.2{\pm}2.1$ % in the chloroform fraction.

Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora (작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.

Effects of Solvents and Extracting Condition on the Antimicrobial Activity of Salviae miltiorrhizae Radix (Saliva miltiorrhiza ) Extract (용매와 추출조건에 따른 단삼 (Salvia mitiorrhiza) 추출물의 항균력)

  • In order to develop a natural food preservative, dried salviae miltiorrhizae radix (Salvia miltiorrhiza) was extracted with several solvents, and then antimicrobial activity was investigated. The optimum extracting condition for the antimicrobial sustance from the sample, minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial ethanol extract from the sample was the strongest compared to those of other solvent extracts such as n-hexane, acetone, butanol, methanol and water. the optimum extractingcondition for antimicrobial substance from the sample was shaking extraction for 2 hours at room temperature incase that 10 volumes of absolute ethanol was added to crushed Saliva Miltiorrhiza. The ethanol extract had strong growth inhibition activity against Gram-positive Bacteria (MIC, 3.13-50$\mu\textrm{g}$/ml) such as B. cereus, B, subtilis, L. minocytogenes, S. aureus, Sc. Mutans. Among Grampositive bacteria tested, Bacillus species was the most susceptibile to the extracted substance. The antimicrobial activity of the ethanol extract from the sample was weak to Gram -negative bacteria yeasts, for example MIC for Gram-negative bacteria and yeasts was 0.8mg/ml and 0.4-0.8mg/ml , respectively.

  • Objectives : This study was designed to identify the effects of Salvia Miltiorrhiza or antibacterial activity against Streptococcus pneumoniae ATCC 33400. Methods : The ethanol- and water-extracts of more than 80 oriental herbal medicines were investigated by the Kirby-Bauer method to determine their inhibitory effect on growth of S. pneumoniae ATCC 33400 in vitro. Of thorn, Salvia Miltiorrhiza was selected. Results : The ethanol-soluble extract of Salvia Miltiorrhiza showed relatively high antibacterial activity against S. pneumoniae ATCC 33400. However, the water-soluble extract of Salvia Miltionrhiza showed no antibacterial activity. The ethanolic extract was forker fractionated with organic solvents such as hexane, chloroform, and ethyl acetate in that order. Among the fraction tested, the chloroform fraction showed the highest antibacterial activity, when the ethanol-soluble extract of Salvia Miltiorrhiza minimal inhibitory concentration (MIC) was 25${\mu}g/ml$. Conclusions : Further study should be carried out to identity of the cell growth inhibition effects of S. pneumoniae ATCC 33400.

The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial methanol extract from the sample was the strongest compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The methanol extract had strong growth inhibition activity against gram-postive bacteria (MIC, $25~50{\;}{\mu\textrm{g}}/ml$) such as B. subtilis. Among gram-positive bacteria tested, B. subtilis was the most susceptible to the extracted substance. The antimicrobial activity of the methanol extract from the sample had strong growth inhibition activity against gram-negative bacteria such as P. aeruginosa (MIC, $25{\;}{\mu\textrm{g}}$/ml).

  • One subfraction from the hexane fraction of Acri graminei Rhizoma, the E4 fraction which is mainly consisted of acorenone, showed a potential inhibitory activity against chloramphenicol acetyltransferase (CAT) of S. aureus SA2 that is a multidrug-resistant strain to 10 usual antibiotics. The combination therapy of this fraction with chloramphenicol resulted in reduction of the minimal inhibitory concentration from 128 $\mu\textrm{g}$/ml to 8 $\mu\textrm{g}$/ml. The E4 fraction also revealed to prevent the induction of CAT from this strain.

The Effect of Anti-microbial and the Inhibitory Effect of Biofilm Formation and Inflammatory Factors Production of Perillae semen Supercritical Fluid Extracts (초임계 자소자추출물의 항균효과와 바이오필름, 염증매개인자 생성 억제 효능)

  • In this study, we assessed anti-oxidant activity, anti-microbial, inhibition of biofilm formation and inflammatory factors(nitric oxide, interleukin-6, interleukin-8) inhibitory effect of Perillae semen hydrothermal extract(PSW) and three kinds of Perillae semen supercritical fluid extract(PSSs) extracted by controlling temperature with no variation of pressure. Compared with PSW, PSSs had significantly lower minimal inhibitory concentrations(MICs) against Staphylococcus aureus(S. aureus) and the ability of PSSs to inhibit formation of biofilm was also superior. PSSs reduce the production of inflammatory mediator and inflammatory cytokines significantly compared to PSW. We suggest, therefore, Perillae semen supercritical fluid 45℃ extract which showed the best anti-microbial, inhibition of biofilm formation, and inhibition of inflammatory factors production among the supercritical fluid extracts could be used for protecting patients with atopic dermatitis from pruritus and transepidermal water loss as a functional ingredient from nature.
