Microcystis sp. is one of the most common harmful cyanobacteria that release toxic substances. Counting algal cells is often used for effective control of harmful algal blooms. However, Microcystis sp. is commonly observed as a colony, so counting individual cells is challenging, as it requires significant time and labor. It is urgent to develop an accurate, simple, and rapid method for counting algal cells for regulatory purposes, estimating the status of blooms, and practicing proper management of water resources. The flow cytometer and microscope (FlowCAM), which is a dynamic imaging particle analyzer, can provide a promising alternative for rapid and simple cell counting. However, there is no accurate method for counting individual cells within a Microcystis colony. Furthermore, cell counting based on two-dimensional images may yield inaccurate results and underestimate the number of algal cells in a colony. In this study, a three-dimensional cell counting approach using a novel model algorithm was developed for counting individual cells in a Microcystis colony using a FlowCAM. The developed model algorithm showed satisfactory performance for Microcystis sp. cell counting in water samples collected from two rivers, and can be used for algal management in fresh water systems.
Microcystis blooms threaten ecosystem function and cause substantial economic losses. Microorganismbased methods, mainly using cyanobactericidal bacteria, are considered one of the most ecologically sound methods to control Microcystis blooms. This study focused on gaining genomic insights into Paucibacter aquatile DH15 that exhibited excellent cyanobactericidal effects against Microcystis. Additionally, a pan-genome analysis of the genus Paucibacter was conducted to enhance our understanding of the ecophysiological significance of this genus. Based on phylogenomic analyses, strain DH15 was classified as a member of the species Paucibacter aquatile. The genome analysis supported that strain DH15 can effectively destroy Microcystis, possibly due to the specific genes involved in the flagellar synthesis, cell wall degradation, and the production of cyanobactericidal compounds. The pan-genome analysis revealed the diversity and adaptability of the genus Paucibacter, highlighting its potential to absorb external genetic elements. Paucibacter species were anticipated to play a vital role in the ecosystem by potentially providing essential nutrients, such as vitamins B7, B12, and heme, to auxotrophic microbial groups. Overall, our findings contribute to understanding the molecular mechanisms underlying the action of cyanobactericidal bacteria against Microcystis and shed light on the ecological significance of the genus Paucibacter.
Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of pine needle extract (PNE) to control nuisance algae by allelopathic inhibition will be environmentally friendly and promising. PNE removed successfully upto 98% of M. aeruginosa at the following optimal conditions: pH 7, $25^{\circ}C$ of temperature, 100 rpm of mixing rate, 5 min of mixing time. These results was indicated that the amount of 1 g/L PNE was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 0.5 g/L of initial concentration of PNE. Different substrate inhibition models were fitted to the kinetic data and found the Luong model was best. The model predicted kinetic parameters were in agreement with the experimental findings. The natural extract, PNE, can be a promising inhibition due to its high efficiency and low dose requirements.

Three out of fourteen Microcystis-dominant cyanobacterial blooms in Central India were found to be toxic to mice ($LD_{50}$ ranging from 35-450 mg bloom dry mass/kg body weight). The liver architecture of the treated mice showed characteristic symptoms of hepatotoxicity relative to the untreated controls, with increased enzyme activities of serum lactate dehydrogenase (LDH), serum glutamate oxaloacetate transaminase (SGOT), alkaline phosphatase (ALP), and serum glutamate pyruvate transaminase (SGPT). RP-HPLC revealed the presence of microcystin-LR, microcystin-RR, and desmethyl microcystin-RR in the given region to maximum amounts of 390, 1,030, and $860{\mu}g/g$ bloom dry weight, respectively, corresponding to a maximum of 2.8 mg/l microcystin-LR in the lake water. Further confirmation of the microcystin variants was conducted using a MALDI-TOF MS analysis.
We evaluated the field application feasibility that biologically derived substances (Naphthoquinone derivate: NQ 2-0) can be used for the eco-friendly mitigation of natural harmful cyanobacterial blooms in freshwater. We conducted a 30 ton scale mesocosm experiment to investigate the effects of NQ 2-0 on biotic and abiotic factors in water collected from Gi-heung reservoir. In the mesocosm experiments, the abundance of Microcystis sp. was continuously increased in the control. However, the Microcystis sp. cell density was sharply decreased on the $10^{th}$ day. In the treatment, NQ 2-0 showed the strong and selective algicidal activity toward the target cyanobacteria (Microcystis sp.). Accordingly, the algicidal activity of NQ 2-0 compound increased gradually until $10^{th}$, $15^{th}$ days and algal biomass was decreased to 99.4 and 100 %, respectively. NQ 2-0 compound was not only selective algicidal activity but also the growth of other phytoplankton and increased the Shannon-Wiener diversity index of phytoplankton. In the mesocosm experiments, the dynamics of biotic (bacteria, heterotrophic nanoflagellate, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. These results suggest that NQ 2-0 could be a selective and ecologically safe algicide to mitigate harmful cyanobacterial blooms. In addition, it is believed that NQ 2-0 will play a major role in forming a healthy aquatic ecosystem by facilitating habitat and food supply of aquatic organisms.
Microcystis aeruginosa (M. aeruginosa) is a cyanobacterium species that can form harmful algal blooms in freshwater bodies worldwide. The use of Artemisia asiatica extracts to control M. aeruginosa inhibition will be environmentally friendly and promising. Artemisia asiatica extracts removed successfully upto 88% of M. aeruginosa pH 8 at $25^{\circ}C$ of temperature. These results was indicated that the amount of 2.24 g/L Artemisia asiatica extracts was removed 1g dryweight/L of M. aeruginosa. The kinetic data showed substrate inhibition kinetics and maximum growth rate was obtained when the M. aeruginosa was grown in medium containing 2.5 g/L of initial concentration of Artemisia asiatica extracts. In the various growth control models, Luong model showed the highest correlation coefficient of 0.9916. Therefore, the Luong model was the most suitable control model for the growth control of M. aruginosa using Artemisia asiatica extracts. In conclusion, the growth control of M. aruginosa using Artemisia asiatica extracts can be applied in the field without controlling the temperature and pH of rivers and streams, and it is possible to control the growth of M. aruginosa efficiently in a short time. The natural extract, Artemisia asiatica extracts, can be a promising inhibition due to its high efficiency and low dose requirements.
In order to establish an axenic (bacteria-free) culture of Microcystis aeruginosa NIER 10039 isolated from a Korean reservoir, the culture was subjected to sequential treatment, including ultrasonication, washing, and addition of antibiotics. Three broad-spectrum antibiotics, namely, kanamycin, ampicillin, and imipenem, were applied separately in that order. Axenicity of the culture was confirmed by cultivation on bacterial media and observation under epifluorescence and scanning electron microscopes. We are the first to establish an axenic culture of a Microcystis strain isolated from Korean reservoirs and can be used in physiological and molecular studies to control toxic Microcystis blooms.
Stable isotope tracers were first applied to evaluate the Microcystis cell assimilation efficiency of bivalves, since the past identification method has been limited to tracking the changes of each chl-a, clearity, and nutrient. The filter-feeders (Sinanodonta woodiana and Unio douglasiae) were assessed under the condition of cyanobacteria (Microcystis aeruginosa) blooms through an in mesocosm experiment using $^{13}C$ and $^{15}N$ dual isotope tracers. chl-a concentration in the treatment mesocosm was dramatically decreased after the beginning of the second day, ranging from 116 to $66{\mu}g\;L^{-1}$. In addition, the incorporated $^{13}C$ and $^{15}N$ atom % in the S. woodiana bivalve showed higher values than U. douglasiae bivalves. The results demonstrate that U. douglasiae has less capacity to assimilate toxic cyanobacteria derived from diet. Our results therefore also indicate that S. woodiana can eliminate the toxin more rapidly than U. douglasiae, having a larger detoxification capacity.
Bloom-forming toxic cyanobacteria Microcystis spp. are common in the summer season in temperate freshwater ecosystems. Often, it leads to the degradation of water quality and affects the quality of drinking water. In a previous study, NQ (naphthoquinone) compounds were shown to be effective, selective, and ecologically safe algicides for Microcystis spp. blooms. To analyze the superiority of developed NQ derivatives, we conducted a microcosm experiment using clay, which is frequently used in South Korea. Similar to previous studies, the NQ 40 and NQ 2-0 compounds showed high algicidal activities of 99.9% and 99.6%, respectively, on Microcystis spp. at low concentrations (≥1 μM) and enhanced phytoplankton species diversity. However, when treated with clay, a temporary algicidal effect was seen at the beginning of the experiment that gradually increased at the end. In addition, treatment with the NQ compounds did not affect either the abiotic or biological factors, and similar trends were observed with the control. These results showed that the NQ 2-0 compound was more effective, with no ecosystem disturbance, and more economical than the currently used clay. These results suggest that NQ 2-0 compound could be a selective, economically and ecologically safe algicide to mitigate harmful cyanobacterial blooms in the field.
Microcystis aeruginosa, a freshwater cyanobacteria species known to be one of the most predominant species responsible for cyanobacterial harmful algal blooms (CyanoHABs). It has been frequently associated with the contamination of neurotoxins and peptide hepatotoxins, such as microcystin and lipopolysaccharides-LPSs. CyanoHABs control technologies so far put in place do not provide a fundamental solution and cause secondary pollution linked with the control measures. For this study, algicidal peptides, which have been reported to be non-toxic and to have antimicrobial properties, were employed for the development of novel eco-friendly control against CyanoHABs. The four peptides (CMA1, CMA2, HPA3P, and HPA3NT3) selected in this study showed significant algicidal effects against M. aeruginosa cells inducing cell aggregation and flotation. Moreover, the newly generated peptides (K160242-5) with certain modifications also displayed high algicidal activity. The algicidal activity of the peptides was found to depend on the concentrations and structures of each of amino acid. The results of this study suggested a novel possibility of CyanoHABs control using the non-toxic algicidal peptides.
