Modulation of Pro-inflammatory and Anti-inflammatory Cytokines in the Fat by an Aloe Gel-based Formula, QDMC, Is Correlated with Altered Gut Microbiota

  • Abnormal inflammatory responses are closely associated with intestinal microbial dysbiosis. Oral administration of Qmatrix-diabetes-mellitus complex (QDMC), an Aloe gel-based formula, has been reported to improve inflammation in type 2 diabetic mice; however, the role of the gut microbiota in ameliorating efficacy of QDMC remains unclear. We investigated the effect of QDMC on the gut microbiota in a type 2 diabetic aged mouse model that was administered a high-fat diet. Proinflammatory (TNF-α and IL-6) and anti-inflammatory (IL-4 and IL-10) cytokine levels in the fat were normalized via oral administration of QDMC, and relative abundances of Bacteroides, Butyricimonas, Ruminococcus, and Mucispirillum were simultaneously significantly increased. The abundance of these bacteria was correlated to the expression levels of cytokines. Our findings suggest that the immunomodulatory activity of QDMC is partly mediated by the altered gut microbiota composition.

산사의 장내 미생물 조절을 통한 항비만 효과 (Anti-obesity Effect of Crataegus pinnatifida through Gut Microbiota Modulation in High-fat-diet Induced Obese Mice)

  • Objectives This study was performed to evaluate anti-obesity effects of Crataegus pinnatifida (CP) on high-fat-diet induced obese mice. Methods The experimental animals were divided into four groups: normal diet (NOR) group, high fat diet (HFD) group, HFD+Xenical (XEN) group, and HFD+CP (CP) group. NOR group was fed a normal diet and the other three groups were fed high fat diet during the experiment. After the first two weeks of diet, XEN group and CP group were administered with XEN or CP for seven weeks, respectively. After that, we measured body weight, liver weight, fat weight, food intake, and serum concentrations of lipids and liver enzymes. Also the liver, intestine, fat tissue was removed to estimate the obesity-related mRNA expressions and the stool sample was collected to analyze the gut microbiota. Results We found that body weight, fat weight, and triglyceride level were decreased significantly in CP group compared to HFD group. Also CP significantly suppressed gene expressions associated with lipogenesis and inflammation, and increased gene expressions of browning of white adipose tissue and mitochondrial biogenesis. Moreover, it shifted the microbial diversity closer to that of NOR group and increased Firmicutes/Bacteriodetes ratio. Conclusions These results suggest that CP decrease body weight, fat weight and serum triglyceride. Also it inhibit inflammation and adipogenesis, altering gut microbial diversity and abundance. In conclusion, CP could be used as a therapeutic drug for obesity via gut microbiota modulation.

  • 비만은 에너지 섭취와 소비의 불균형으로 인해 유발되는 비정상적인 지방 축적으로, 근래에 다양한 만성질환을 초래하는 주요 국제 보건 문제로 부상하였다. 이러한 이유로, 비만 문제에 대한 여러 해결책들이 제시되고 있다. 에너지를 저장하며 내분비 작용을 하는 백색 지방과 달리 열을 생성하여 에너지를 발산하는 두 종류의 지방조직인 갈색 지방과 베이지색 지방이 성인에 존재하며 외부 자극에 의해 유도될 수 있다는 것이 밝혀진 이래로, 이들은 비만 치료의 유망한 표적으로서 각광받고 있다. 이러한 외부 자극 중, 인간 장관계에서 인간과 공존하는 장내 미생물총은 다양한 대사 작용에 참여하며, 이를 조절하는 것이 여러 대사 질환의 치료에 유력한 작용을 할 것으로 보인다. 따라서, 다양한 연구에서 항비만 치료가 장내 미생물 환경 전환이나 갈색 지방 조직 활성화에 미치는 영향에 초점을 맞추고 있다. 본 총설에서는 비만과 체중 증가, 대사 질환을 해소하는 것으로 알려진 자극과, 장내 미생물총의 변화나 갈색지방의 활성화를 야기하는 자극과, 이 자극들과 장내 미생물총의 조작, 지방조직의 갈색화 사이에서 알려져 있거나 있을 수 있는 상관관계를 중점적으로 다루고자 한다.

발효 김치가 흰쥐의 장내 미생물 형성에 미치는 영향 (Modulation of Intestinal Microbiota by Supplementation of Fermented Kimchi in Rats)

  • 장내 미생물은 숙주의 건강을 유지하는 데 중요한 역할을 하며, 식단에 의하여직접적으로 영향을 받아 조절된다. 김치는 식이 섬유와 젖산균(LAB)이 풍부한 발효 식품이다. 발효 김치가 장내 미생물의 구성에 미치는 영향을 조사하기 위하여 6주령의 수컷 Sprague-Dawley 흰쥐 45마리를 대상으로 기본 사료(CON), 발효 김치(FK)와 키토산 첨가 발효 김치(CFK)를 각각 4주간 급여 하였다. 체중과 사료 섭취량을 매주 측정하였으며, 미생물 분석은 장내용물 수집 후 pyrosequencing을 통하여 16S rRNA 유전자 분석으로 확인 하였다. FK 및 CFK군은 대조군에 비해 체중, 사료 효율 및 혈중 triglyceride 농도가 감소한 것으로 나타났다. 장내 미생물의 다양성은 대조군에 비해 FK와 CFK군 모두에서 증가하였다. 비만과 관련된 Firmicutes 미생물이 감소한 반면, 체중 감소와 관련된 Bacteroidetes 미생물이 증가하였다. 젖산균과 체중 감소 관련 박테리아 및 butyrate 생산 박테리아는 대조군에 비해 FK 및 CFK군에서 증가하였다. 발효 김치는 비만을 억제하고 장내 유익한 미생물의 성장을 촉진하였다.

The Ingestion of Dietary Prebiotic Alternatives during Lactation Promotes Intestinal Health by Modulation of Gut Microbiota

  • Palm kernel expeller (PKE), a by-product of palm oil extraction, contains higher amounts of fiber than corn and soybean meal, but offers low energy density, protein value, and amino acid (AA) composition, limiting its use for swine. Recently however, it was reported that dietary fiber has a positive effect on the gut microbiota of the host, and therefore it is necessary to study the effect of PKE feeding on the intestinal microbiota of swine. In this study, we investigated the effects of supplementation with PKE in lactation diets on the gut microbiota composition of lactating sows and their litters. A total of 12 sows were randomly assigned to two dietary treatment groups in a completely randomized design. The treatments were a diet based on corn-soybean meal (CON) and CON supplemented with 20% of PKE. Sow and piglet fecal samples were collected before farrowing, on days 7 and 28 (weaning) after farrowing, and on days 7 and 28 (weaning) after farrowing, respectively, to verify gut microbiota composition by pyrosequencing analysis. The beta-diversity result showed a significant difference only in weaning-stage piglets, but dietary PKE altered the gut microbiota in sows by increasing the abundance of Lactobacillus compared with CON. In piglets, dietary PKE decreased the abundance of opportunistic pathogen Proteus and increased the abundance of potentially beneficial bacteria, such as Prevotellaceae and Prevotella. Our results can be helpful in developing feeding strategies and support the beneficial effects of dietary PKE to improve the gut health of animals.

발효커피가 사람장내미생물에 미치는 영향 (Effects of fermented coffee on human gut microbiota)

  • 발효식품은 장 건강을 포함하여 건강상의 이로운점을 제공하는 건강기능식품으로 인식되고 있다. 따라서 본 연구는 Lactobacillus plantarum과 Bacillus amyloliquefaciens로 발효 된 커피원두가 건강한 사람의 장내미생물 생태에 미치는 영향을 조사하였다. 커피원두를 발효하여 플라보노이드와 폴리페놀과 같은 이로운 물질이 증가하였다. 또한 발효커피의 섭취로 인해 유의한 장내 미생물생태 및 물질대사 변화가 관찰되지 않았지만, 섬유소 분해 및 단쇄지방산을 생성하는 유익한 미생물이 증가하였다. 본 연구 결과는 발효커피 섭취로인해 장내미생물생태 및 물질대사를 유지하면서 유익한 미생물이 증가하였음을 확인하였다.

Multispecies probiotics alter fecal short-chain fatty acids and lactate levels in weaned pigs by modulating gut microbiota

  • Short-chain fatty acids (SCFAs) are metabolic products produced during the microbial fermentation of non-digestible fibers and play an important role in metabolic homeostasis and overall gut health. In this study, we investigated the effects of supplementation with multispecies probiotics (MSPs) containing Bacillus amyloliquefaciens, Limosilactobacillus reuteri, and Levilactobacillus brevis on the gut microbiota, and fecal SCFAs and lactate levels of weaned pigs. A total of 38 pigs weaned at 4 weeks of age were fed either a basal diet or a diet supplemented with MSPs for 6 weeks. MSP administration significantly increased the fecal concentrations of lactate (2.3-fold; p < 0.01), acetate (1.8-fold; p < 0.05), and formate (1.4-fold; p < 0.05). Moreover, MSP supplementation altered the gut microbiota of the pigs by significantly increasing the population of potentially beneficial bacteria such as Olsenella, Catonella, Catenibacterium, Acidaminococcus, and Ruminococcaceae. MSP supplementation also decreased the abundance of pathogenic bacteria such as Escherichia and Chlamydia. The modulation of the gut microbiota was observed to be strongly correlated with the changes in fecal SCFAs and lactate levels. Furthermore, we found changes in the functional pathways present within the gut, which supports our findings that MSP modulates the gut microbiota and SCFAs levels in pigs. The results support the potential use of MSPs to improve the gut health of animals by modulating SCFAs production.

  • Objective: In this study, we investigated the effects of Rubus coreanus-derived lactic acid bacteria (LAB) fermented feed (RC-LAB fermented feed) and three types of LAB (Lactobacillus plantarum, Lactobacillus acidophilus, Bifidobacterium animalis) on the expression of transcription factors and cytokines in Th1, Th2, Th17, and Treg cells in the intestinal lymph nodes and spleens of rats. In addition, the effect on intestinal microbiota composition and body weight was investigated. Methods: Five-week-old male rats were assigned to five treatments and eight replicates. The expression of transcription factors and cytokines of Th1, Th2, Th17, and Treg cells in the intestinal lymph nodes and spleens was analyzed using real-time reverse transcriptase polymerase chain reaction assays. Intestinal tract microbiota compositions were analyzed by next-generation sequencing and quantitative polymerase chain reaction assays. Results: RC-LAB fermented feed and three types of LAB increased the expression of transcription factors and cytokines in Th1, Treg cells and Galectin-9, but decreased in Th2 and Th17 cells. In addition, the intestinal microbiota composition changed, the body weight and Firmicutes to Bacteroidetes (F/B) ratio decreased, and the relative abundance of LAB increased. Conclusion: LAB fermented feed and three types of LAB showed an immune modulation effect by inducing T cell polarization and increased LAB in the intestinal microbiota.

Value of spray-dried plasma as a supplement to swine diets

  • One of the most powerful health management practices is the use of antibiotics, but their use is being restricted because of health safety issues. The swine industry has been looking for various alternatives to antibiotics and increasingly considers the use of dietary factors like feed ingredients, feed additives, feed formulation practices, or feeding methods, instead of using antibiotics to improve pig health and performance. Among other alternatives to antibiotics, spray-dried plasma may be a candidate. Spray-dried plasma is a blood product that provides bioavailable nutrients and physiologically active components such as immunoglobulins, glycoproteins, growth factors, peptides, etc. It is an excellent protein source with balanced and highly digestible amino acids. Several beneficial physiological activities depend on components of spray-dried plasma, such as immune competence (antibacterial activity), modulation of microbiota and/or immune system, integrity of intestinal barrier function, etc. These beneficial effects can contribute to improvement of pig performance and health by modulation of microbiota in the digestive tract and/or immune system. Therefore, it is suggested that spray-dried plasma has great potential as an antibiotics alternative.
