Immunostimulation Effects of Cell Wall Components Isolated from Lactobacillus plantarum

  • Immunostimulation effects of the cell wall components isolated from Lactobacillus plantarum were investigated by studying the macrophage s tumorcidal activity, splenocyte proliferation, anticomplementary activity and the inhibition of peritoneal tumor cell growth measured with ICR mice inoculated with sarcoma 180. The immunopotentiating cell wall components were a complex of peptidoglycan and exopolysaccharides. The tumorcidal activity of macrophage against Yacl and B16 tumor cells was enhanced when the cell wall components were added into the macrophage s culture medium. They also stimulated splenocytes to proliferate up to the same level as when the concanavalin A was added into the splenocyte's culture medium. The complementary activity was inhibited by 50% when the cell wall components were incubated with the sheep red blood cells treated with hemolysin and guinea pig complement. This result confirmed that the cell wall components had an antitumor effect, because the anticomplementary activity is usually accompanied by an antitumor activity at the same time. This fact was confirmed again by the inhibition of the growth of sarcoma 180 when the cell wall components were injected intraperitoneally into ICR mice inoculated with sarcoma 180. As a result, it is concluded that the cell wall components isolated from Lactobacillus plantarum had multifunctional immunostimulation effects in vitro and in vivo.

  • 최소 생산배지에 포함된 25mM 이상의 무기인(Inorganic phosphate)은 Cephalosporium acremonium의 생육은 증대시켰으나, cephalosporin C(CPC) 생산은 감소시켰다. Phosphate-starved 휴지세포계를 이용하여 균체내의 ATP, ADP와 AMP를 측정하였다. 그 결과로부터 무기인은 adenylated nucleotide의 농도의 비율로 표시되는 energy charge를 조절하여 CPC 생산성에 영향을 주는 것으로 밝혀졌다. 또한, 무기인에 의한 CPC 생합성은 repression 효과에 의한 것이 아니라 inhibition 효과에 의해 조절받는 것을 알 수 있었다.

Studies on the Constityents and Cultite of Kotean Basidiomycetes Anitumor Polysaccharides from the Cultured Mycelia of Some Basiodiomycetes

  • To develop potent anitumor polysac-charides from higher hungi of Korea the alcohol precipitated polysaccharides, which were obtained from hot water extracts of shake-cultured mycelia of eight species of basidiomycetes, were subjected to antitumor screening tests, using sarcoma 180 as a tumor cell. When administered i. p, at the dose level of 20mg/kg/day for ten days into the mice which were implanted with $1{times}10^{8}$ cells of sarcoma 180 twenty-four hour before the start of polysaccharide injection, the polysaccharides of Plutcus cervinus ISA-Pc-1004, Pleurotus pulmonarius, Hydnum repandum ISA-Hr-1006, and Laccaria laccata ISA-LI-1008 respectively showed inhibition ratios of 62.0%, 51.7%, 64.8%, and 57.8%. They were selected for further study, while those of Pancillus serotina ISA-Ps-1012, Laccaria amethystina ISA-La 1001, Pholiota adiposa ISA-Pa-1010, and Lepista spISA-Ls-1007 respectively showed inhibition ratios of -9.3%, 25.3%, 27.3% and 33.0%.

Comparison of Microbial Fungicides in Antagonistic Activities Related to the Biological Control of Phytophthora Blight in Chili Pepper Caused by Phytophthora capsici

  • Two similar microbial fungicides (termed as MA and MB) developed in a Korean biopesticide company were analyzed and compared each other in their biocontrol activities against the phytophthora blight of chili pepper caused by Phytophthora capsici. MA and MB contained the microbe Paenibacillus polymyxa and Bacillus subtilis, respectively, with concentrations over those posted on the microbial products. In comparison of the isolated microbes (termed as MAP from MA and MBB from MB) in the antagonistic activities against P. capsici was effective, prominently against zoospore germination, while MBB only significantly inhibited the mycelia growth of the pathogen. Some effectiveness of MAP and MBB was noted in the inhibition of zoosporangium formation and zoospore release from zoosporangia; however, no such large difference between MAP and MBB was noted. In a pot experiment, MA reduced the severity of the phytophthora blight more than MB, suggesting that the disease control efficacy would be more attributable to the inhibition of zoospore germination than mycelia growth of P. capsici. These results also suggest that the similar microbes MA and MB targeting different points in the life cycle of the pathogen differ in the disease control efficacies. Therefore, to develop microbial fungicides it is required to examine the targeting points in the pathogen's life cycle as well as the action mode of antagonistic microorganisms.

소금 첨가에 따른 도라지 발효 특성과 미생물 변화 및 항비만 효능 평가 (Effect of Platycodon grandiflorum Fermentation with Salt on Fermentation Characteristics, Microbial Change and Anti-obesity Activity)

  • Objectives: This study investigated the effect on microbial ecology, fermentation characteristics and anti-obesity of Platycodon grandiflorum (PG) fermentation with salt. Methods: PG was fermented for four weeks with 2.5% salt and the characteristics of fermented PG were performed by measuring pH, total sugar content, viable bacteria number and microbial profiling. Also, we measured total polyphenol, flavonoid and the percent of inhibition of lipase activity and lipid accumulation. Results: Salt added to PG for fermentation had an effect on pH, total sugar, total and the number of lactic acid bacteria. Total sugar and pH were reduced and number of total and lactic acid bacteria were increased after fermentation. The majority of bacteria for fermentation were Lactobacillus plantarum, Leuconostoc psedomesenteroides and Lactococcus lactis subspecies lactis regardless of salt addition. However, microbial compositions were altered by added salt and additional bacteria including Weissella koreensis, W. viridescens, Lactobacillus sakei and Lactobacillus cuvatus were found in fermented PG with salt. Total flavonoid was increased in fermented PG and lipid accumulation on HepG2 cells treated with fermented PG was reduced regardless of salt addition. Moreover, fermented PG without salt suppressed lipase activity. Conclusions: Addition of salt for PG fermentation had influence on fermentation characteristics including pH and sugar content as well as number of bacteria and microbial composition. In addition, fermented PG showed anti-obesity effect by increasing flavonoid content and inhibition of lipase activity and lipid accumulation.

  • The most extensively used synthetic antioxidants are butylated hydroxyanisole (BHA), butylated hydroxy toluene (BHT) and tert-butylhydroquinone (TBHQ). However BHT and BRA have been suspented of being responsible for liver damage andcar cinogenesis. Therefore, the importance of the search and exploitation of natural antioxidant, especially of plant origin, has greatly increased in recent years. Plant contain a wide variety of chemicals that have potent biological effects. As a result, there has been a growing interest in the use of herbs as a source of therapeutic drugs. The aim of this study was to assess the antioxidant and cosmeceutical of natural materials. The antioxidant and cosmeceutical activity of natural materials were investigated by hydroxyl radical scavenging, superoxide dismutase (SOD) -like, xanthine oxidase inhibition, tyrosinase inhibition, anti-microbial and astringent.

Inhibition of Homodimerization of Toll-like Receptor 4 by 6-Shogaol

  • Toll-like receptors (TLRs) play a critical role in sensing microbial components and inducing innate immune and inflammatory responses by recognizing invading microbial pathogens. Lipopolysaccharide-induced dimerization of TLR4 is required for the activation of downstream signaling pathways including nuclear factor-kappa B ($NF-{\kappa}B$). Therefore, TLR4 dimerization may be an early regulatory event in activating ligand-induced signaling pathways and induction of subsequent immune responses. Here, we report biochemical evidence that 6-shogaol, the most bioactive component of ginger, inhibits lipopolysaccharide-induced dimerization of TLR4 resulting in the inhibition of $NF-{\kappa}B$ activation and the expression of cyclooxygenase-2. Furthermore, we demonstrate that 6-shogaol can directly inhibit TLR-mediated signaling pathways at the receptor level. These results suggest that 6-shogaol can modulate TLR-mediated inflammatory responses, which may influence the risk of chronic inflammatory diseases.

삼림토양의 미생물군집과 아밀라아제 활성에 관한 연구 (Studies on the Microbial Population and the Amylase Activity of the Forest Soil)

  • Soil condition, total number of bacteria, soil amylase activity and microbial biomass $(CO_2-C)$ were measured at soil of different forest types. And the difference of the allelopathic effect was determined between fresh leaf extract of Quercus acutissima and Pinus rigida to the bacteria isolated from soil of different forest types. 1. Total number of bacteria in Carpinus laxiflora forest soil was 4~7 times larger than that in pinus desiflora forest soil. 2. Soil amylase activity was positively correlated with total number of soil bacteria and soil organic matter content. The amylase activity at F layer was 4~5 times larger than that at H layer, and that at H layer was 2~4 times larger than that at A layer. 3. Seasonal changes of microbial biomass showed a peak in summer, and vertical distribution of microbial biomass decreased with increasing soil depth. The microbial biomass in Pinus densiflora forest soil was larger than that in Quercus serrata forest soil. 4. Fresh leaf extract of Pinus rigida and Quercus acutissima showed an acceleration or inhibition effect on the growth of soil bacteria, and that of !. acutissima inhibited larger number of soil bacterial strains than that of P. rigida. 4.2% and 25% of soil bacterial strains isolated from soil of P. rigida and Q. acutissima forests were inhibited by fresh leaf extract of P. rigida and Q. acutissima, respectively.

Methionyl-tRNA Synthetase Regulates Lifespan in Drosophila

  • Methionyl-tRNA synthetase (MRS) is essential for translation. MRS mutants reduce global translation, which usually increases lifespan in various genetic models. However, we found that MRS inhibited Drosophila reduced lifespan despite of the reduced protein synthesis. Microarray analysis with MRS inhibited Drosophila revealed significant changes in inflammatory and immune response genes. Especially, the expression of anti-microbial peptides (AMPs) genes was reduced. When we measured the expression levels of AMP genes during aging, those were getting increased in the control flies but reduced in MRS inhibition flies age-dependently. Interestingly, in the germ-free condition, the maximum lifespan was increased in MRS inhibition flies compared with that of the conventional condition. These findings suggest that the lifespan of MRS inhibition flies is reduced due to the down-regulated AMPs expression in Drosophila.

Toxicity Evaluation of Complex Metal Mixtures Using Reduced Metal Concentrations: Application to Iron Oxidation by Acidithiobacillus ferrooxidans

  • In this study, we investigated the inhibition effects of single and mixed heavy metal ions ($Zn^{2+},\;Ni^{2+},\;Cu^{2+},\;and\;Cd^{2+}$) on iron oxidation by Acidithiobacillus ferrooxidans. Effects of metals on the iron oxidation activity of A. ferrooxidans are categorized into four types of patterns according to its oxidation behavior. The results indicated that the inhibition effects of the metals on the iron oxidation activity were noncompetitive inhibitions. We proposed a reduced inhibition model, along with the reduced inhibition constant ($\alpha_i$), which was derived from the inhibition constant ($K_I$) of individual metals and represented the tolerance of a given inhibitor relative to that of a reference inhibitor. This model was used to evaluate the toxicity effect (inhibition effect) of metals on the iron oxidation activity of A. ferrooxidans. The model revealed that the iron oxidation behavior of the metals, regardless of metal systems (single, binary, ternary, or quaternary), is closely matched to that of any reference inhibitor at the same reduced inhibition concentration, $[I]_{reduced}$, which defines the ratio of the inhibitor concentration to the reduced inhibition constant. The model demonstrated that single metal systems and mixed metal systems with the same reduced inhibitor concentrations have similar toxic effects on microbial activity.
