The use of microalgal biomass as feedstock for biofuels has been discussed for decades as it provides a sustainable approach to producing fuels for the future. Nonetheless, its feasibility has not been established yet and various aspects of biomass applications such as CO2 biofixation should also be explored. Therefore, in this study, the CO2 biofixation and lipid/carbohydrate production potential of Chlorella sp. ABC-001 were examined under various nitrogen concentrations. The highest biomass productivity and CO2 biofixation rate of 0.422 g/l/d and 0.683 g/l/d, respectively, were achieved under a nitrogen-rich condition (15 mM nitrate). Carbohydrate content was generally proportional to initial nitrate concentration and showed the highest value of 41.5% with 15 mM. However, lipid content showed an inverse relationship with nitrogen supplementation and showed the highest value of 47.4% with 2.5 mM. In consideration as feedstock for biofuels (bioethanol, biodiesel, and biogas), the sum of carbohydrate and lipid contents were examined and the highest value of 79.6% was achieved under low nitrogen condition (2.5 mM). For lipid-based biofuel production, low nitrogen supplementation should be pursued. However, considering the lower feasibility of biodiesel, pursuing CO2 biofixation and the production of carbohydrate-based fuels under nitrogen-rich condition might be more rational. Thus, nitrogen status as a cultivation strategy must be optimized according to the objective, and this was confirmed with the promising alga Chlorella sp. ABC-001.
In traditional mixotrophic cultures of microalgae, all the inorganic nutrients and organic carbon sources are supplied in the medium before inoculation. In this study, however, an alternative approach was adopted in Haematococcus pluvialis Flotow, a microalga capable of growing mixotrophically on sodium acetate (Na-Ac). First, the cells were grown under 75 ${\mu}Mol$ photons $m^{-2}s^{-1}$ phototrophically without Na-Ac until the stationary phase and then exposed to five different light regimes by the addition of Na-Ac (e.g., dark, 20, 40, 75, and 150 ${\mu}Mol$ photons $m^{-2}s^{-1}$). Dry weight (DW), pigments, and especially cell number in alternative mixotrophy (AM) were higher than traditional mixotrophy (TM). Cell number in AM almost doubled up from 21.7 to $42.9{\times}10^4$ cells/ml during 5-day exposure to Na-Ac, whereas the increase was only 1.2-fold in TM. Maximum cell density was reached in 75 ${\mu}Mol$ photons $m^{-2}s^{-1}$ among the light intensities tested. We propose that Na-Ac in TM of H. pluvialis can not be utilized as efficiently as in AM. With this respect, AM has several advantages against TM such as a much higher cell density in a batch culture period and minimized risk of contamination owing to the shorter exposure of cells to organic carbon sources. In consequence, this method may be used for other strains of the species, and even for the other microalgal species able to grow mixotrophically.
In the present study, the biomass productivity of two green microalgae (Chlorella sp. and Dunaliella salina DCCBC2) were assessed in a 12 L tubular photobioreactor under optimum culture conditions. In the batch culture optimization process, the Chlorella sp. biomass was obtained as 1.2 g/L under atmospheric air as a sole $CO_2$ source and other culture conditions as follows: light intensity, temperature, pH, $NH_4Cl$ and $K_2HPO_4$ were 100 ${\mu}E/m^2/s$, $27^{\circ}C$, 7.0, 20.0 mM and 2.0 mM, respectively. On the other hand, 2.9 g/L of D. salina DCCBC2 biomass production was observed under the following conditions: light intensity, temperature, pH, $KNO_3$ and $K_2HPO_4$were 80 ${\mu}E/m^2/s$, $27^{\circ}C$, 8.0, 3.0 mM and 0.025 mM, respectively. At 1% $CO_2$ supply to the reactor, the Chlorella sp. production was reached 1.53 g/L with 25% increment under the same operating conditions. In addition, the maximum D. salina DCCBC2 biomass was observed as 3.40 g/L at 3% $CO_2$ concentration. Based on the aforementioned optimized conditions, the dilution rate and maximal biomass productivity of Chlorella sp. and D. salina DCCBC2 in the continuous cultivation were 0.4/d and 0.6 g/L/d and 0.6/d and 1.5 g/L/d, respectively.
Fucoxanthin has been reported as bioactive compounds exhibiting strong antioxidant, anticancer and anti-inflammatory activities. Owing to its a wide range of applications and potentials, commercial production of fucoxanthin from algae has been attracted many attentions. Although, most of seaweeds and diatoms contain fucoxanthin as major carotenoid contents, low productivity of fucoxanthin still hinder the industrial application. Here, we newly isolated and identified indigenous marine diatom Odontella sp. BS-003 as a resource of fucoxanthin production. The characterization, optimization and production of the fucoxanthin, along with other bioactive compound omega-3 fatty acid from odontella sp. BS-003 were analyzed in this study, and the results represented optimal culture condition (two-fold silicate containing F/2 medium) significantly enhanced the algal biomass productivity. The maximum biomass (1.83 g/L), fucoxanthin (3.88 mg/g), along with omega-3 fatty acid (10 %, w/w) were obtained from the 10 L of photobioreactor. Based on the results, it is speculated that the microalga Odontella sp. BS-003 can be a promising natural resource for the production of bioactive compounds.
Nitrogen is one of the most critical nutrients affecting cell growth and biochemical composition of microalgae, ultimately determining the lipid or carbohydrate productivity for biofuels. In order to investigate the effect of nitrogen sources on the cell growth and biochemical composition of the marine microalga Tetraselmis sp., nine different N sources, including NaNO3, KNO3, NH4NO3, NH4HCO3, NH4Cl, CH3COONH4, urea, glycine, and yeast extract were compared at the given concentration of 8.82 mM. Higher biomass concentration was achieved under organic nitrogen sources, such as yeast extract (2.23 g L−1) and glycine (1.62 g L−1), compared to nitrate- (1.45 g L−1) or ammonium-N (0.98 g L−1). All ammonium sources showed an inhibition of cell growth, but accumulated higher lipids, showing a maximum content of 28.3% in ammonium bicarbonate. When Tetraselmis sp. was cultivated using yeast extract, the highest lipid productivity of 36.0 mg L−1 d−1 was achieved, followed by glycine 21.5 mg L−1 d−1 and nitrate 19.9 mg L−1 d−1. Ammonium bicarbonate resulted in the lowest lipid productivity of 14.4 mg L−1 d−1. The major fatty acids in Tetraselmis sp. were palmitic, oleic, linoleic and linolenic acids, regardless of the nutritional compositions, indicating the suitability of this species for biodiesel production.
The diatom Melosira nummuloides is a microalga that is widely distributed in freshwater and seawater is used is used in the production of silicon and fucoxanthin. The objective of this experimental study was to determine the effects of diatom powder on the physiology of olive flounder Paralichthys olivaceus. In four feeding groups consuming 0%, 1%, 2% and 3% diatom powder. After 8 weeks of feeding, we investigated P. olivaceus growth rate, feed efficiency rate, survival rate, anti-oxidant enzyme rate, non-specific immune activity and immune gene expression. The rates of growth rate, feed efficiency rate and survival were significantly higher for olive flounder in all diatom groups than in the control. The results for anti-oxidant enzyme, superoxide dismutase and catalase showed no significance, but glutathione was significant, depending on the concentration of diatom addition. The galectin and lysozymes of immune genes were increased in the control group. Galectin and lysozymes were thought to have increased due to infections by from pathogens during the experiment period. These results suggest that the addition of diatoms to olive flounder diets is effective in enhancing growth rate and innate immunity.
Purpose: Poly-N-acetyl glucosamine(PGlcNAc) nanofiber-based materials, produced by a marine microalga, have been characterized as effective hemostatic and angiogenic agents. The similarity between PGlcNAc patch and the natural extracellular matrix allows it to support new healthy tissue growth in an injured area and to encourage fluid absorption. In this study, we hypothesized that a poly-N-acetyl glucosamine fiber patch(PGlcNAc patch) may enhance wound healing in the db/db mouse. Methods: PGlcNAc patches were applied on one square centimeter, full-thickness, skin wounds in the db/db mouse model. Wounds(n=15 per group) were dressed with a PGlcNAc nanofiber patch for 1 hour(1 h), 24 hours(24 h) or left untreated(NT). After the application time, patches were removed and wounds were allowed to heal spontaneously. The rate of wound closure was evaluated by digital analysis of unclosed wound area in course of time. At day 10, wounds(n=7 per group) were harvested and quantified with immunohistochemical markers of proliferation(Ki-67) and vascularization (platelet endothelial cell adhesion molecule, PECAM-1). Results: Wounds dressed with PGlcNAc patches for 1 hour closed faster than control wounds, reaching 90% closure in 16.6 days, nine days faster than untreated wounds. Granulation tissue showed higher levels of proliferation and vascularization following 1 h treatment than the 24 h and NT groups. In addition to its hemostatic properties, the PGlcNAc material also appears to accelerate wound closure in healing-impaired genetically diabetic mice. Conclusion: This material, with its combination of hemostatic and wound healing properties, has the potential to be effective agent for the treatment of complicated wounds.
The kinetics of growing microalga, Spirulina platensis was investigated to treat swine wastes with optimum growth conditions. Temperature was varied from 15 to 40$\circ$C at three different light intensities, 6 W/m$^{2}$, 12 W/m$^{2}$ and 24 W/m$^{2}$. The specific growth rate was increased as temperature increased up to 30$\circ$C. The activation energy was estimated as 13.5 kcal/mol by an Arrhenius relationship. 0.24 (1/day) of specific growth rate was obtained from batch cultivation with 30% swine wastes, compared to 0.31 (1/day) from clean culture. It was found that Spirulina platensis was able to reduce 70-93% of PO$_{4}$$^{3-}$ -P, 67-93% of inorganic nitrogen, 80-90% of COD and 37-56% organic nitrogen by adding various concentrations of swine wastes for 12 days of batch cultivation. Rate constants for removing nitrates and phosphates in treating swine wastes were estimated as 0.17 (1/day) and 0.14 (1/day) in the first order reaction, respectively. 1.52 (g/L) of maximum cell density was maintained at 0.20 (1/day) of dilution rate in continuous culture, adding 20% swine wastes for 30 days. The chemical composition of the biomass obtained from the process showed 58.7% of protein, 11.0% of lipid and 15.6% of ash.

To increase the availability of microalgae as producers of valuable compounds, it is necessary to develop novel systems for gene expression regulation. Among the diverse expression systems available in microalgae, none are designed to induce expression by low temperature. In this study, we explored a cold-inducible system using the antifreeze protein (AFP) promoter from a polar diatom, Chaetoceros neogracile. A vector containing the CnAFP promoter (pCnAFP) was generated to regulate nuclear gene expression, and reporter genes (Gaussia luciferase (GLuc) and mVenus fluorescent protein (mVenus)) were successfully expressed in the model microalga, Chlamydomonas reinhardtii. In particular, under the control of pCnAFP, the expression of these genes was increased at low temperature, unlike pAR1, a promoter that is widely used for gene expression in C. reinhardtii. Promoter truncation assays showed that cold inducibility was still present even when pCnAFP was shortened to 600 bp, indicating the presence of a low-temperature response element between -600 and -477 bp. Our results show the availability of new heterologous gene expression systems with cold-inducible promoters and the possibility to find novel low-temperature response factors in microalgae. Through further improvement, this cold-inducible promoter could be used to develop more efficient expression tools.

The three speices of miroalgae (Chlorella vulgaris, Dunaliella salina and Porphyridium purpureum) were immobilized in Ca-alginate capsules as a basic study for development of economic cultivation process, and then were cultivated in an air-bubble column bioreactor. Under the batch culture of aerobic conditions, the thickness of the capsule membrane and $CO_2$ supply did not affect the growth of the immobilized microalga, Chlorella vulgaris. Cell concentration of immobilized microalgae in the capsule was higher than those of imobilized microalgae in beads and free cells. The cell concentration of microencapsulated Dunaliella salina was greater about 5 times than that of free cells. Based on these results, it is concluded that the application of microencapsulation technology to the culture of microalgae was an effective method for high-density cultivation.
