In 2004, bacterial spot-causing xanthomonads (BSX) were reclassified into 4 species-Xanthomonas euvesicatoria, X. vesicatoria, X. perforans, and X. gardneri. Bacterial spot disease on pepper plant in Korea is known to be caused by both X. axonopodis pv. vesicatoria and X. vesicatoria. Here, we reidentified the pathogen causing bacterial spots on pepper plant based on the new classification. Accordingly, 72 pathogenic isolates were obtained from the lesions on pepper plants at 42 different locations. All isolates were negative for pectolytic activity. Five isolates were positive for amylolytic activity. All of the Korean pepper isolates had a 32 kDa-protein unique to X. euvesicatoria and had the same band pattern of the rpoB gene as that of X. euvesicatoria and X. perforans as indicated by PCR-restriction fragment length polymorphism analysis. A phylogenetic tree of 16S rDNA sequences showed that all of the Korean pepper plant isolates fit into the same group as did all the reference strains of X. euvesicatoria and X. perforans. A phylogenetic tree of the nucleotide sequences of 3 housekeeping genes-gapA, gyrB, and lepA showed that all of the Korean pepper plant isolates fit into the same group as did all of the references strains of X. euvesicatoria. Based on the phenotypic and genotypic characteristics, we identified the pathogen as X. euvesicatoria. Neither X. vesicatoria, the known pathogen of pepper bacterial spot, nor X. perforans, the known pathogen of tomato plant, was isolated. Thus, we suggest that the pathogen causing bacterial spot disease of pepper plants in Korea is X. euvesicatoria.
IPP isomerase (Iso) and Limonene synthase (Limo) are important enzymes in terpenoids biosynthesis pathway. The wild type and each metabolically engineered (Iso and Limo) transgenic spearmint (Mentha spicata Linne) plants were compared for their growth patterns and the contents of essential oil in in vitro culture media. The profile of terpenoid metabolites was obtained from the essential oil of the metabolically engineered transgenic spearmint, which was extracted using a modified SDE method, by GC-MS analysis. The growth of wild spearmint was more profuse in B5 culture medium than in other media. Significant differences in leaf and root growth patterns were observed between metabolically engineered transgenic and wild type spearmint plants. The leaves of the transgenic spearmint plants were slightly elongated but were dramatically narrower than those of wild type spearmints. The content of essential oil of transgenic spearmint was different slightly depending on the target terpenoid genes. The content of essential oils in Limo transgenic plants was higher than that of Iso, except for transgenic plant in B5 medium. The transgenic spearmint produced more terpenoids than the wild type. Iso spearmint extracts showed eleven terpenoids and a phenylpropane, while Limo spearmint extracts contained nine terpenoids. However, extracts from the wild type showed the presence of only four terpenoids.

Objectives : This study was conducted to investigate candidate materials as anti-inflammatory agent from extracts of Korean medicinal plants in Hwaak mountain. Ligustrum obtusifolium (LO) is a Korea medicinal plants that commonly used for robustness and hemostasis. It has been reported that LO has exhibited anti-ischemic, anti-oxidative, anti-hypolipidemic, anti-tumor and hypoglycemic effects. However, LO has not been previously reported to have an anti-inflammatory effect. Therefore, we have evaluated the anti-inflammatory effects of LO and its underlying molecular mechanisms in LPS-induced RAW 264.7 macrophages. Methods : Cell viability was determined by MTT assay in RAW 264.7 macrophages. Nitric Oxide (NO) was measured with Griess reagent and pro-inflammatory cytokines were detected by ELISA in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Protein expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and p65 subunit of nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) were determined by Western blot analysis. Results : Among 15 extracts of Korean medicinal plants tested, Ligustrum obtusifolium (LO) showed the inhibition of NO production without cytotoxicity. LO reduced the expression levels of iNOS and COX-2 proteins in LPS-simulated RAW 264.7 macrophages in dose-dependent manner. Consistent with these data, LO inhibited the productions of $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$ in LPS-simulated RAW 264.7 macrophages. Furthermore, LO attenuated the LPS-induced nuclear translocation of p65 $NF-{\kappa}B$ in RAW 264.7 macrophages involving suppression of $NF-{\kappa}B$ activation. Conclusions : Taken together, these results suggest that the anti-inflammatory effects of LO is associated with regulation of inflammatory mediators via inhibition of $NF-{\kappa}B$ activation in LPS-treated RAW 264.7 macrophages.

Human erythropoietin (EPO) is a leading product in the biopharmaceutical market, but functional EPO has only been produced in mammalian cells, which limits its application and drives up the production costs. Using plants to produce human proteins may be an alternative way to reduce the cost. However, a recent report demonstrated that overexpression of the human EPO gene (EPO) in tobacco or Arabidopsis rendered males sterile and retarded vegetative growth, which raises concern whether EPO might interfere with hormone levels in transgenic plants. In the present study, we demonstrated that overexpressing EPO with additional 5'-His tag and 3' ER-retention peptides in tobacco did not cause any developmental defect compared to GUS plants. With our method, all 20 transgenic plants grew on selective medium and, further confirmed by PCR, were fertile. Most of them grew similarly compared to GUS plants. Only one transgenic plant (EPO2) was shorter in plant height but had twice the life span compared to other transgenic plants. When 11 randomly selected EPO plants, along with the abnormal plant EPO2, were subjected to RT-PCR analysis, all of them had detectable EPO transcripts. However, their protein levels varied considerably; seven of them had detectable EPO proteins analyzed by western blot. Our results indicate that overexpressing human EPO protein in plants does not have detrimental effects on growth and development. Our transformation systems allow us to further explore the possibility of glycoengineering tobacco plants for producing functional EPO and its derivatives.

The HCV gene was expressed in potato plants under the control of the constitutive CaMV 355 promoter or tuber-specific patatin promoter. Solanum tuberosum plants carrying a plant expression vector harboring the encoding region of HCV gene were generated by Agrobacterium tumefaciens-mediated in vitro transformation methods. The presence of HCV gene in the plant genome was detected by PCR and DNA hybridization experiments. We obtained the 5 lines of transgenic potato with the pMBPHCV construct and 4 lines of transgenic potato with the pATHCV construct. The HCV transgenic stably integrated into the potato genome, as well as their transcription. HCV mRNA was identified in leaf and tuber tissues of transgenic plants by Northern blot analysis. The transgenic potato plants produced the expected transcript, and the corresponding HCV protein accumulated in individual transgenic plants.
Despite the remarkable advancements of the modern medicine, the traditional system of medicine (TSM) still serves as a potential primary health care modality in the in low- and middle-income countries. The recent reports suggest that there is a renewed interest has been observed towards TSM in the developed countries too, because of the adverse side-effect of modern medicines. Medicinal plants have been widely serving as a rich source of therapeutic agent. Ethiopia is one of the most reserves rich countries in the world. It is renowned for well-diversified and natural resources in terms of its unique flora and fauna. Ethiopian deep-rooted tradition and culture largely depends on the usage of plants for their religious ceremonies, impressive festivals, traditional medicinal uses and other basic necessities. The present scrutiny is an attempt to understand the omnipotent nature of an Ethiopian Ethnomedicinal plant called Tinjute [vernacular name (local native language, Amharic); Otostegia integrifolia]. There are several studies suggest that Tinjute can be used as a natural medicine or health-promoting agents for various disorders and ailments. Nevertheless, in Ethiopia, it is renowned as an insect repellent to drive-away insect vector of diseases, particularly mosquitoes in the early evening. However, there are many more issues and challenges which must be urgently addressed to scientifically formulate various potent, efficacious, safe and highly selective phytotherapeutic agents and insects' repellent from the Tinjute plant in the near future.

The effects of crude extracts from medicinal plants on biological activity were investigated. The crude ethanol extract of H. paucistipula inhibited the growth of the Gram positive bacterium Bacillus subtilis ATCC 19659, (2 mm inhibition zone at 150 ㎍/disc) and the dermatophyte Trichophyton mentagrophytes ATCC 28185, (7 mm inhibition zone at 150 ㎍/disc), and toxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, (IC/sub 50/ 2.48 ㎍/㎖ at 75 ㎍/disc). This crude ethanol extract of H. paucistipula is the strongest antimicrobial and cytotoxic activities against P388 murine leukaemia cells ATCC CCL 46 P388D1.

The purpose of this was investigate the possibility of medicinal plants application as an edible functional food resources. We arried out to develop a beverage by using Nelumbo nucifera and we examined the nutritional characterics. Also we examined the effects of drink on physiological function in aorta relation. Thus, the effect of developed beverage on phenylephrine induced contraction of isolated rat thoracic aorta. Contractile force was measured with force displacement thansducer under 1.5g loading tension. Brix, pH and titratable acidity of developed drink were 12.25%, 4.5 and 0.092%. The approximate nutritional composition of beverage was carbohydrate, 9.40%, crude protein, 0.30%, crude fat, 0.25% and ash, 0.15%. Also insoluble fiber and soluble fiber were 1.30%, 0.80%. Developed beverage contained Na(11.45 mg%), K(6.87 mg%), Ca(2.53 mg%). The contraction forces by injection of phenylephrine in isolated thoracic aorta were significantly law in each beverage treatment groups compared with control group. These results that developed drink with medicinal plants can be used as a functional material to decrease aorta contraction.

(-)-Yatein is a lignan of the dibenzyl-butyrolactone type. that has been isolated from some Cupressaceae plants. It was reported that (-)-yatein. isolated from plants. showed different activities from the synthetic yatein [3]. Hence. the enantiosetective determination of yatein from synthetic materials and natural products would be necessary. (omitted)

Anti-Pseudomonas aeruginosa activities for ethanol extract of 34 medicinal plants widely used in the folk medicine were evaluated to screening of anti-Pseudomonas aeruginosa agents. The ethanol extracts of Gardenia jasminoides, Arctium lappa, Citrus unshiu, and Phellodendron amurense showed antimicrobial activities against P. aeruginosa ATCC 27853. The ethanol extracts of Gardenia jasminoides among these medicinal plants showed significant antimicrobial activity against P. aeruginosa. Therefore, we expect that these medicinal plants will be useful for nature antimicrobial agent against P. aeruginosa in future.
