The Floristic Study of Mt. Bonghwa (Yanggu-gun, Gangwon-do) Area Adjacent to the Korean Demilitarized Zone (DMZ에 인접한 봉화산 일대의 관속식물 분포 연구)

  • Mt. Bonghwa reaches a height of 800 m (2,870 ft) and stands on the flank of Mt. Daeam-san, which has an elevation of 1,304 m (4,278 ft). It is important to survey the flora of Bonghwa-san in that it is a way to make out what kind of plant species exist in DMZ due to the geographical characteristics of Bonghwa-san adjacent to Korean DMZ. Therefore this study was conducted to elucidate the distribution of vascular plants and their value of Bonghwa-san. The vascular plants that were collected 10 times from May 2015 to April 2017 consisted of a total of 525 taxa; 93 families, 309genera, 458 species, 4 subspecies, 58 varieties and 5 forms. The observed rare plant species as designated by Korea Forest Service were 12 taxa including Echinosophora koreensis, Epimedium koreanum, Anemone koraiensis etc. Endemic species were 16 taxa containing Cirsium setidens, Heloniopsis koreana, Salvia chanryoenica etc. 31 taxa were recorded as naturalized plants and their naturalized rate and urban index were noted as 5.8% and 7.25%. Among the surveyed 525 taxa, edible, medicinal, pasturing, ornamental, timber, dye, fiber and industrial plants included 213 taxa, 172 taxa, 196 taxa, 50 taxa, 23 taxa, 10 taxa, 6 taxa and 3 taxa, respectively.

Comparative Study of Protocorm-like body and Multiple Shoots from Dendrobium Candidum on Biological Activities (철피석곡의 원괴체상구체 및 다신초 추출물의 생리활성에 관한 비교 연구)

  • Dendrobium candidum is one of the well-known orchid on traditional and rare medicinal herb in China. We artificially cultured protocorm-like body and multiple shoots using the bioreactor culture system from Dendrobium candidum and experimented an efficacy as a cosmetic ingredient for skin care. Several studies previously have reported anti-tumor, wound healing and immunological function of Dendrobium candidum but other efficacies were not well known. In the present study, we investigated the cosmetic efficacy to know applicable value of protocorm-like body and multiple shoots cultured from Dendrobium candidum as a cosmetic ingredient. The biological activities of extracts from protocorm-like body, multiple shoots, aerial part and underground part of Dendrobium candidum were investigated. Results were found that extracts of protocorm-like body are superior to other extracts (underground part, aerial part and multiple shoots extracts) on anti-oxidant effect. Also, protocorm-like body extract contained the phenolic and flavonoid compounds more than aerial part, underground part and multiple shoots extracts. In addition, we investigated skin whitening effect related to whitening of skin. In tyrosinase activity and melanin synthesis assay, multiple shoots extract is superior to other extracts (aerial part, underground part and protocorm-like body), on inhibitory effect of tyrosinase activity and melanin synthesis respectively. These results indicate that the protocorm-like body and multiple shoots extracts cultured from Dendrobium candidum presents a new possibility of being applicable to skin improvement as a cosmetic ingredient.

Physiological responses involved in reactive oxygen species (ROS) of rice plant under alone or multi artificial stress conditions

  • The Earth's climate is rapidly changing because of increasing carbon dioxide content in atmosphere so, climate prediction models anticipate that earth surface temperature will rise by 3 to $5^{\circ}C$ in next 50 to 100 years. Therefore, frequency of un-expected weather events such as drought, salinity, low or high temperature and flooding etc. will be increasing worldwide. Furthermore, increased atmosphere temperature can influence pests and pathogens spread as well. Therefore, to protect enormous grain loss from unexpected weather conditions, studies related with combine stress conditions like abiotic plus biotic stress condition are really required. Thus, our research focused on physiological responses under combined abiotic and biotic stress condition in rice plant. To induce uniform stress condition, we used NaCl (100 mM) and salicylic acid (0.5 and 1.0 mM SA) as each stress a stimulator. Each artificial abiotic and biotic stress inducer was applied to hydroponically grown rice seedlings alone or together for four day. The data were collected in a time-dependent manner [1, 2, 3 and 4 day(s) after treatment (DAT)] and were matched with our anticipation that shoot length and shoot fresh weight was decreased in solo and combined abiotic and biotic stress condition. The lipid peroxidation content was significantly increased ($1.5{\pm}0.2$ to $2.7{\pm}0.1mg$ mg of $MDA\;g^{-1}FW$) in the first two days in both stress exposed plants, and showed the opposite trend ($0.5{\pm}0.01$ to $0.1{\pm}0.001mg$ of $MDA\;g^{-1}FW$) in last two days under multi stress condition. Superoxide dismutase (SOD) activity did not showed difference in only biotic stress condition (alone 0.5 and 1.0 mM SA) as compared to control however, it was significantly increased in multi stress condition or solo abiotic stress condition whereas, catalase (CAT), and ascorbate peroxidase (APX) activities were significantly decreased in solo biotic and combined abiotic and biotic condition. In particular, both enzymes activities were more decreased in multi stress condition as compared to solo biotic stress condition. The results for relative mRNA expression level of CAT and APX enzymes were in agreement with results of spectrophotometric values. Correlation value between each stress condition and phenotypic data showed that biotic stress condition showed high correlation with activity of CAT and APX whilst, abiotic stress condition revealed significant correlation with SOD activity.

Fragrance, Chemical Composition and Toxicity of the Essential Oil in Erect Bur-marigold (Bidens tripartita L.) (가막사리 (Bidens tripartita L.) 정유의 향취, 화학성분 및 세포독성)

  • The essential oil was extracted by steam distillation from the aerial part of erect bur-marigold (Bidens tripartita L.), one of the noxious weed in paddy field. The composition of the essential oil was analyzed by gas chromatography-mass spectrometry. The fragrance of the essential oil was green, herbal, oily, spicy. There were 42 constituents in the essential oil:17 hydrocarbons, 6 alcohols, 6 acetates, 5 N-containing compounds, 3 ethers, 3 ketones, 1 lactone and 1 S-containing compound. Major constituents were ${\alpha}$-phellandrene (22.50%), ${\alpha}$-pinene (22.21%), 2,4-dimethyl (2,5-dimethylphenyl) methyl ester benzoic acid (15.11%), limonene (10.66%), ${\beta}$-pinene (35.43%), and ${\beta}$-cubebene (5.27%). The $IC_{50}$ value in MTT assay using HaCaT keratinocyte cell line was 0.018%. However, attachment of patch with 0.1% of the erect bur-marigold essential oil for 24 hr did not show any skin toxicity. Overall results of this study suggest that the essential oil of erect bur-marigold could be used as a source for the development of perfumery industrial products.

Chemical Compositions of Glycoprotein and Chondroitin Sulfates from Sea Cucumber(Stichopus japonicus) (해산 극피동물 중의 당단백질의 특성과 이용 I. 해삼 당단백질 및 황산콘드로이친의 화학조성과 특성)

  • To elucidate food value and medicinal effect of sea cucumbers, sugar composition of those gly-coprotein and chondroitin sulfate was studied. The contents of sulfate esters in sea cucumbers were 1.21%(blue), 0.90%(red) and 1.19%(black). Predominant carbohydrates were identified as fucose, glucose, D-mannuronic acid and N-acetylglucosamine, and those amount was more than 80% to total carbo-hydrate, while the minor sugar composition was ribose, mannose, galactose, N-acetylgalactosamine and D-glucuronic acid. Also, the major carbohydrate moiety of glycoproteins of sea cucumbers was revealed as fucose, mannose, N-acetylglucosamine, glucose and ribose, and those amount was more than 86% to total carbohydrate. Galactose, N-acetylgalactosamine, D-glucuronic acid and mannuronic acid were minor carbohydrate moiety. The contents of sulfate esters in glycoproteins were 0.96% for blue sea cucumber, 1.15% for red sea cucumber and 1.13% for black sea cucumber, while those in chondroitin sulfates were 3.52%(blue), 3.60%(red) and 3.72%(black). The carbohydrate moiety of chondroitin sulfate was identified as N-acetylgalactosamine (73~ 87%), fucose (7~15%) and D-glucuronic acid(5~12%). As the base on the IR spectrum of strong absorption appeared in 1240$cm^{-1}$ / for stretching vibrations in S=0 group and weak absorptions in 850$cm^{-1}$ / and 820$cm^{-1}$ /for stretching vibrations in C-0-S group, chondroitin sulfates had sulfate group which was bound to $C_4$in fucose.

Effects of Artemisia Capillaris Thunberg on Apoptosis in HeLa Cells (사철쑥의 HeLa 세포고사 효과)

  • This study analyzes the apoptosis of HeLa cells to see if we can use the Artemisia capillaris Thunberg for the prevention of chronic degenerative diseases. We used the HeLa cells to see what effects the A. capillaris Thunberg had on apoptosis of the cancer cells. We checked the cell activity, cell morphological change, DNA fragmentation, and DNA content after administering 0, 100, 500, 1000, and $2000{\mu}g/ml$ methanol, ethyl acetate, n-butanol extract of the A. capillaris Thunberg. As for the cell viability, the increase of concentration of methanol and ethyl acetate decreased the survival rate of the cell, but the phenomenon was much weakened in n-butanol extract and was not observed in aqueous extract. The higher the density of the methanol, ethyl acetate, n-butanol and aqueous extract was, the lower the survival rate of the HeLa cell was. These extracts obstructed the cell cohesion and caused the blebbing of he cell membrane and fragmentation of the nucleus, both of which are symptoms of apoptosis. Laddering-pattern DNA fragmentation was observed in the groups that were treated with the $1000{\mu}g/ml$ and $2000{\mu}g/ml$ of methanol extract. The DNA content of the cells apoptosis measured by fluorescent-activated cell sorter (FACS) increased as the density of the methanol, ethyl acetate and butanol extract increased. The result of the study shows that A. capillaris Thunberg fosters the apoptosis of HeLa cells, which suggests that the A. capillaris Thunberg has a great potential value as food additives, medicinal supplements for patients with chronic diseases, and preventive measures against cancer.

Current status and prospects of chrysanthemum genomics (국화 유전체 연구의 동향)

  • Chrysanthemum is one of the top floriculture species with ornamental and medicinal value. Although chrysanthemum breeding program has contributed to the development of various cultivars so far, it needs to be advanced from the traditional phenotype-based selection to marker-assisted selection (molecular breeding) as shown in major cereal and vegetable crops. Molecular breeding relies on trait-linked molecular markers identified from genetic, molecular, and genomic studies. However, these studies in chrysanthemum are significantly hampered by the reproductive, genetic, and genomic properties of chrysanthemum such as self-incompatibility, inbreeding depression, allohexaploid, heterozygosity, and gigantic genome size. Nevertheless, several genetic studies have constructed genetic linkage maps and identified molecular markers linked to important traits of flower, leaf, and plant architecture. With progress in sequencing technology, chrysanthemum transcriptome has been sequenced to construct reference gene set and identify genes responsible for developments or induced by biotic or abiotic stresses. Recently, a genome sequencing project has been launched on a diploid wild Chrysanthemum species. The massive sequencing information would serve as fundamental resources for molecular breeding of chrysanthemum. In this review, we summarized the current status of molecular genetics and genomics in chrysanthemum and briefly discussed future prospects.

The Antioxidative and Antimicrobial Activities of the three Species of Leeks(Allium tuberosun R.) Ethanol Extracts (품종별 부추 추출물의 항산화성 및 항균성)

  • The antioxidative and antimicrobial activities were carried on the leeks (Allium tuberosun R.) ethanol extracts in order to find out new food functional components. Three species of leeks used in this study were Chinese leek(long type, LL), general leek(medium type, LM), and medicinal leek (short type, LS). Total amounts of polyphenol compounds in LS was shown as the highest (436.60mg%) value. All of ethanol extracts of these leeks were shown to be had good electron donating ability(EDA) and nitrite scavenging activity. Specialty, the ethanol extract of LS(LSEx) had the highest EDA 30.47% and nitrite scavenging activity 77.24% and the lowest was LMEx. The antioxidative activities of these ethanol extracts on the corn germ oil were measured by peroxide values(POV) and conjugated diene values (CDV) storaging for 30 days at $60{\pm}2^{\circ}C$. The antioxidative activities of these extracts by POV and CDV were determined as following order as LSEx> LLEx) LMEx. The antioxidative activities of all extract were presented as high tendency by increasing adding amounts (0.02%>0.05%>0.1%). When the antioxldative activities were compared with BHT and ${\alpha}-tocopherol$, the degree of the antioxidative activities of these extracts were certified as lower than BHT and higher than ${\alpha}-tocopherol$. And also LSEx, LMEx, and LLEx had antimicrobial effects on the several micro organisms, especially the effect on the Pseudomonas aeruginasa was remarkable. While LMEx had shown inhibit effect on most of micro organism used in this study.

Effects on Growth and Medicinal Ingredients of Platycodon grandiflorum Radix by Organic and Chemical Fertilizers (유기질 및 화학비료 시용수준이 도라지의 생육 및 약용성분에 미치는 영향)

  • This study was done to elucidate effects on organic and chemical fertilizers for growth characteristics, saponin content and antioxidant activity in Platycodin grandiflorum Ridix root. Chemical and mixed organic fertilizers were treated at levels of 50%, 100% and 200% based on nitrogen 3 kg/10a before transplant of Platycodin grandiflorum Ridix. As a result, when 50% mixed organic and chemical fertilizer plots were treated to P. grandiflorum Ridix root, the root length showed the highest scores of 26.9 and 26.5 cm, respectively. Root width exhibited the highest scores of 25.6 mm and 25.2 mm in 50% mixed organic and chemical fertilizer plots, respectively. The number of fine-roots and fresh weight presented the highest values in 50% mixed organic fertilizer plot as 26.7 and 50.4 g, respectively. The content of platycodin D3 was ranged in 183.4~321.7 mg/100 g, which the highest value was observed in 100% mixed organic fertilizer plot. The content of saponin except for deapioplatycodin D presented higher amount in the mixed organic fertilizer plots. Treatment of 200% chemical fertilizer presented to be withered in transplant cultivation of 3 year-old roots.

A Segmentation Study of Temple Food for the Global Convergence - Focusing on Recognition and Preference - (글로벌 융복합을 위한 사찰음식 세분화 연구 - 인식과 선호도를 중심으로 -)

  • The purpose of this study is to investigate the recognition and preference for temple food between Korea and foreign national temple-stay participants. In order to achieve the research purpose, 220 research questionnaires were distributed to Korean and foreigners who have participated in a temple-stay. T-test and ANOVA analyses were performed for this study. The findings are summarized as follows. The highest recognition item for Korean temple food that the subjects perceived was 'I think Korean temple food is a medicinal food.'(4.31 points). In the value recognition score for Korean temple food, Asians(4.58 points) are relatively higher than Korean (4.23 points), North American (4.13 points) and European (3.94 points) participants. Participants in Asia appeared relatively higher than the others in the preference score on Korean temple food. The higher globalization strategy items of Korean temple food that the subjects perceived were 'Korean temple food needs a storytelling marketing'(3.94 points) and 'Korean temple food needs a modernization of cookware'(3.90 points).
