Effects of Age and Gender on the Viability and Stem Cell Markers, mRNA, and Protein Expression of Bone Marrow-Derived Stem Cells Cultured in Growth Media

  • Purpose: Bone marrow has long been a source of primary cells. This study was performed to evaluate the effects of age and sex on the cellular viability and expression of stem cell markers of mRNA and on the protein expression of bone marrow stem cells (BMSCs) derived from healthy donors. Materials and Methods: Stem cells were isolated from human bone marrow and plated in culture plates. The shape of the BMSCs was observed under inverted microscope. Quantitative cellular viability was evaluated using a Cell-Counting Kit-8 assay. The expression of stem cell surface markers was tested and a series of quantitative real-time polymerase chain reaction (qRT-PCR) and Western blot was performed to evaluate the expression in each group. Result: The shapes of the cells at 20s, 30s, and 50s were similar to each other. No significant changes in cellular viability were noted among different age groups or sex groups. The BMSCs expressed CD44, CD73, and CD90 surface markers but did not express CD14 and CD34. There were no noticeable differences in CD surface markers among the different age groups. The expressions of CD surface markers were similar between men and women. No significant differences in the secretion of vascular endothelial growth factors (VEGFs) were noted at Day 3 between different age groups. qRT-PCR regarding the expression showed differences between the age groups. However, Western blot analysis showed a decrease in expression but did not reach statistical significance (P>0.05). Conclusion: This study clearly showed no significant differences in shape, cell viability, expression of stem cell surface markers, or secretion of human VEGF among different age groups. However, western blot analysis showed a tendency of age-related decrease which did not reach statistical significance. Collectively, autologous or allogeneic BMSCs should be meticulously applied to obtain optimal results regarding age and sex.

  • 인터넷의 폭발적인 성장에 힘입어 '정보사회', '가상공간'이라는 용어가 더 더욱 혼란스럽게, 그렇지만 당연시되어 사용되고 있다. 현대세계의 핵심적 특성으로서 '정보'와 '미디어'에 일부 사회이론가와 대부분의 일반인은 매료되어 있으며, '지식이 가치의 원천이고 세계를 움직이는 것은 정보이다.' 라는 식의 검증되지 않은 주장을 쉽게 하고 있다. 이에 보드리야르는 현대문화를 기호(signs)의 문화로 정의하고, 우리는 기호와 의미화 양식으로 둘러싸여 있으며, 모더니티의 주장과 같이 기호의 배후에 어떤 실체가 있는 것이 아니라 단지 기호만이 존재한다고 주장한다. 따라서 우리는 비진정성(inauthenticity)을 벗어날 수 없으며 그렇게 할 수 있다고 주장하는 것은 전혀 타당하지 못하다는 것이다. 사람들이 기호가 시뮬라시옹에 불과하다는 것을 닫고 있다면 무엇이든 괜찮게 된다 - 그리고 아무것도 괜찮지 않게 된다. 그리하여 보드리야르의 결론, 즉 "우리는 아무 것도 볼 것이 없는 이미지를 대량으로 만들어낸다. 오늘날 대부분의 이미지는... 아무 것도 볼 것이 없는 문자 그대로 이미지이다." 라는 것에 도달하게 된다. 따라서, 이러한 세계는 의미 없이 보여지고 경험되기 위한 그리고 즐겨지기 위한 기호를 가지게 되는 포스트모던한 세계이며, 이러한 예들은 인터넷등 미디어에서 폭발적으로 보여지고 있다. 즉, 관객들은 아무 것도 보거나 듣지 않으며 단지 현대를 특징 짓는 볼거리들을 경험할 뿐이라고 결론 내릴 수 있다. 본 연구는 정보와 미디어의 새로운 디자인 패러다임에 정보의 긍정적 사회 현상뿐 아니라, 비판적 사상을 검토해 봄으로써 미디어를 바라보는 디자인 철학과 방향에 도움이 되고자 하는데 그 목적이 있다.

독일에서의 시청점유율제한을 통한 다양성 보호 연구 (Protection of Diversity through the Control of the Delimitation of the Audience Share in German Television)

  • 독일의 방송국가협약은 독일기본법에서 보호하고 이는 '긍정적 질서'의 보호를 위해 방송자본 집중을 규제한다. 이러한 규제는 다양한 의견의 형성을 보호하고, 국가와 사회이익집단으로부터 독립적인 보편적 방송서비스를 가능하게 하기 위해서이다. 그러나 경제적 경쟁은 방송분야에서의 자본집중을 초래할 위험이 있다. 특히 경제적 경쟁은 다양한 보도적 경쟁을 저해한다. 특히 보도의 다양성은 서로 다른 여론을 적절하게 반영해 낼 수 있는 제도적 장치가 마련되어야만 가능하다. 독일의 방송국가협약 제26조는 방송사업자의 시청점유율 상한선을 제한함으로써, 방송시장에서 내용의 다양성과 소유의 다양성을 보장하고, 결과적으로 의견의 다양성을 보호한다. 구체적으로 시청점유율 30%를 초과했거나 시청점유율이 25%에 불과하지만 인접 미디어시장에서 시장지배적 사업자인 경우에는 의견의 다양성을 침해한다고 본다. 그럼에도 불구하고 시청점유율 제한은 미디어 사이의 합산 영향력 산출방식의 모호함과 시사정보 프로그램과 교양오락프로그램에서의 시청점유율을 동일한 무게로 평가하는 등의 한계가 있다.

메탄올 작업장 노출 평가에의 EU REACH 프로토콜 적용: Targeted Risk Assessment (The Application of an EU REACH Protocol to the Occupational Exposure Assessment of Methanol: Targeted Risk Assessment)

  • Background: The European Centre for Ecotoxicology and Toxicology of Chemicals' Targeted Risk Assessment (ECETOC TRA) tool has been recognized by EU REACH as a preferred approach for calculating worker health risks from chemicals. Objectives: The applicability of the ECETOC TRA to occupational exposure estimation from industrial uses of methanol was studied by inputting surveyed and varied parameters for TRA estimation as well as through comparison with measured data. Methods: Information on uses of methanol was collected from seven working environment monitoring reports along with the measured exposure data. Input parameters for TRA estimation such as operating conditions (OCs), risk management measures (RMMs) and process categories (PROCs) were surveyed. To compare with measured exposures, parameters from the surveyed conditions of ventilation but no use of respiratory protection were applied. Results: PROCs 4, 5, 8a, 10, and 15 were assigned to ten uses of methanol. The uses include as a solvent for manufacturing sun cream, surfactants, dyestuffs, films and adhesives. Methanol was also used as a component in a release agent, hardening media and mold wash for cast products as well as a component of hard-coating solution and a viscosity-controlling agent for manufacturing glass lenses. PROC 8a and PROC 10 of a cast product manufacturer without LEV (local exhaust ventilation) and general ventilation as well as no respiratory protection resulted in the highest exposure to methanol. Assuming the identical worst OCs and RMMs for all uses, exposures from PROC 5, 8a, and 10 were the same and the highest followed by PROC 4 and 15. The estimation resulted in higher exposures in nine uses except one use where measured exposure approximated exposures without RMMs. Conclusions: The role of ECETOC TRA as a conservative exposure assessment tool was confirmed by comparison with measured data. Moreover, it can guide which RMMs should be applied for the safe use of methanol.

Effect of Garlic Oil on Fatty Acid Accumulation and Glycerol-3-Phosphate Dehydrogenase Activity in Differentiating Adipocytes

  • Garlic oil (GAR, Allium sativum L.) has been studied as a feed additive to improve animal production performance and decrease methane emission in ruminants. The present study was designed to determine the possible effect of GAR on fatty acid composition and accumulation in animal fat tissue using a cell model. 3T3-L1 preadipocytes at $2{\times}10^{4}\;mL^{-1}$ were seeded to 24-well plates and allowed to proliferate to reach confluence. The cells were then treated with media containing 0, 2.5, 5, 10, 20 and 40 $\mu{g}$ $mL^{-1}$ of GAR during the differentiation period for 8 days. Media containing dexamethasone, methyl-isobutylxanthine and insulin was applied during the first 2 days of the early differentiation period. On day 8 sub-sets of the wells were stained with oil red-O and the remaining cells were harvested for determination of glycerol-3-phosphate dehydrogenase [EC 1.1.1.8] (GPDH) activity (n = 6) and cellular fatty acid concentration (n = 6). It was found that supplementation of GAR increased (p<0.05) the ratio of monounsaturated fatty acids/saturated fatty acids in the adipocytes and showed inhibitory effect (p<0.05) on the post-confluent proliferation. With relative low dosage, GAR (5-20 $\mu{g}$ $mL^{-1}$) increased (p<0.05) the GPDH activity without affecting the cellular fatty acid concentration, while a high dosage (40 $\mu{g}$ $mL^{-1}$) inhibited (p<0.05) fatty acid accumulation and decreased GPDH activity. Supplementation of GAR had an effect on cell post-confluent proliferation, differentiation and fatty acid accumulation. However, the effect may be diverse and depends on the dose applied.

  • Developments and changes in digital environment based on computer technologies and internet networks enabled us interactive communication for planning and execution through the freedom of 'connection', 'sharing', and 'conversion'. The subject of this study, interactive commercial, might be the core of success based on creation which stems from communication design, revolution in design and technology, and conversion. Communication in modern economy is not optional, but essential. Furthermore, like evolution process up to Augmented Reality from developments of mobile communication technology, or concept in paradigm change, digital interactive commercial means the core of success, not a part of it. Therefore, in order to achieve success in the global competition structure, mobile communication design, revolution, and creation which produce added value shall be working as a base. In order to reach effective communication process performance of interactive commercial media, this study would like to propose building a media commercial system which is realized by human senses. Using this, standardized methods for interactive commercial which can be used together with augmented reality based on digital technology, and for using digital robot contents are available, along with methodology system of their use. In addition, the ultimate goal of this study aims to discover new factors through reorganization of precedent analysis and adaptation based on academic achievement and practical application. And thus, it wants to contribute to practical use of interactive commercial out of augmented reality-based communication in which technology, marketing, and design are integrated to be used in real sense.

3차원 공간 상의 신체 부위 드래깅을 통한 캐릭터 애니메이션 제어 (Dragging Body Parts in 3D Space to Direct Animated Characters)

  • 본 논문은 캐릭터의 특정 신체 부위를 3차원 가상 환경 안의 원하는 위치로 드래깅 함으로써 미래의 동작 시퀀스를 지시하는 새로운 대화형 기법을 제시한다. 캐릭터의 동작 시퀀스는 미리 촬영된 동작 데이터의 세부 구간들을 모션 그래프 구조에 따라 시간적으로 재배열 함으로써 합성한다. 손 동작 추적 장치에 의하여 사용자의 3차원 위치 입력이 주어지면, 모션 그래프를 공간 상에 펼침으로써 캐릭터의 현재 상태로부터 도달 가능한 잠재적 미래 상태들을 샘플링 하고, 선택된 신체 부위와 입력 위치 간의 거리가감소하는 미래 상태를 검출하여 해당 자세를 사용자에게 제시한다. 연속적으로 검출된 자세 간의 변화를 최소화 함으로써 사용자가 입력 위치를 변화 시킴에 따라 어떤 상태가 검출될지 예상하기 용이하도록 하고, 결과적으로 원하는 미래 상태에 신속하게 도달하도록 한다. 제안된 방법의유용성은 브레이크 댄스, 권투, 농구 등의 동작 데이터를 이용한 실험을 통하여 확인할 수 있었다.

Zika Virus on YouTube: An Analysis of English-language Video Content by Source

  • Objectives: The purpose of this study was to describe the source, length, number of views, and content of the most widely viewed Zika virus (ZIKV)-related YouTube videos. We hypothesized that ZIKV-related videos uploaded by different sources contained different content. Methods: The 100 most viewed English ZIKV-related videos were manually coded and analyzed statistically. Results: Among the 100 videos, there were 43 consumer-generated videos, 38 Internet-based news videos, 15 TV-based news videos, and 4 professional videos. Internet news sources captured over two-thirds of the total of 8 894 505 views. Compared with consumer-generated videos, Internet-based news videos were more likely to mention the impact of ZIKV on babies (odds ratio [OR], 6.25; 95% confidence interval [CI], 1.64 to 23.76), the number of cases in Latin America (OR, 5.63; 95% CI, 1.47 to 21.52); and ZIKV in Africa (OR, 2.56; 95% CI, 1.04 to 6.31). Compared with consumer-generated videos, TV-based news videos were more likely to express anxiety or fear of catching ZIKV (OR, 6.67; 95% CI, 1.36 to 32.70); to highlight fear of ZIKV among members of the public (OR, 7.45; 95% CI, 1.20 to 46.16); and to discuss avoiding pregnancy (OR, 3.88; 95% CI, 1.13 to 13.25). Conclusions: Public health agencies should establish a larger presence on YouTube to reach more people with evidence-based information about ZIKV.

이토토요의 도서관 공간에 나타나는 창조적 인터페이스요소에 관한 연구 - 타마미술대학신도서관을 사례로 - (A Study on the Creative Interface Factors of Library Space on Ito Toyo - Focus on the Tama Art University New Libraries -)

  • The importance of communication which is oriented to interaction, fusion and connection is more increased in the post-information society. The information environment is already realized as interface to reach immediately and freely to the databased information and knowledge. The contemporary libraries execute very important roles as an intellectual infra of modern times to deliver the great human achievement and create new culture. In the mean time, the rapid information-oriented society and the development of digital technology bring out big change in the traditional dispatch and receipt method of information. For this reason, libraries as the community space of visitors integrates physical space - electronic space and reconstructs new temporary culture and value to build the new interface that connects human to human, human to space and human to information. On the premise of these awareness, this study analysis the creative interface factor of Tama Art University New Libraries in Ito Toyo in order to research the function and the application method of modern libraries. And the results are follows. 1)We can make sure the significance of communication and the integration consciousness between digital technology and analogue sensitivity that is the characteristic of the post-information society. 2)We can classify for the creative interface factors by the character as space factor(Arcade, Gallery, Lounge, AV Booth, Cafe), facility/equipment factor(Laboratory, Media, Bar, Info Shelf, Media Seat) and furniture factor(Carrel, Desk, Mag Table, XI Shelf). 3)We can see that these interface factors change the stream of library users and that the library is constructed for library users to create and discovery something easily from books and modern technologies. And we can realize also that these factors are creative installation to lead out naturally the latent liberty sense of users.

점토광물(粘土鑛物)에 의한 Nitrofen의 흡착(吸着) 및 활성(活性) 변화(變化) (Adsorption and Activity Changes of Nitrofen by Clay Minerals)

  • 점토광물(粘土鑛物)의 종류(種類)에 따른 Nitrofen의 흡착(吸着)과 6종(種)의 점토광물(粘土鑛物)과 소맥(小麥)을 사용(使用)하여 점토(粘土)현탁액 중(中)에서 Nitrofen의 활성변화(活性變化)를 조사(調査)하였다. Nitrofen은 점토광물(粘土鑛物)과의 현탁액 중(中)에서 4시간(時間)의 진탕으로 거의 평형농도(平衡濃度)에 이르렀다. 시료량(試料量)이 많을수록 Nitrofen의 흡착량(吸着量)이 증가(增加)되었으나, 시료(試料) : 용액(溶液)의 비율(比率)이 작은 편이 단위중량당(單位重量當) 흡착량(吸着量)이 많았다. 점토광물별(粘土鑛物別)로 볼 때 Nitrofen의 흡착량(吸着量)에는 큰 차이(差異)가 없었으나 Na-montmorillonite가 다소 많이 흡착(吸着)하였으며 Nitrofen의 흡착(吸着)에 pH의 영향(影響)은 거의 없었다. Growth media에 점토광물(粘土鑛物)을 첨가(添加)한 결과(結果), montmorillonite첨가(添加)가 소맥(小麥)의 저해율(沮害率)을 감소(減少)시키는데 가장 효과적(效果的)이었다.
