To investigate changes in protein and total lipid contents, seed storage protein pattern, and fatty acid composition of germination perilla(Perilla frutescens) seeds. Also, the corresponding value components in cotyledons, hypocotyles and roots were measured according to germination stage. The results were summarized as follows ; During germination, pertein and total lipid contents of Yepsilldalggae and Kwangyang cultivar were decreased continuously. In particular, protein contents rapidly decreased to the 3 days after germination(DAG), and then total lipid contents rapidly decreased between 3 DAG and 10 DAG. In changes of protein and total lipid contents of cotyledons, protein contents of Yeupsildalggae was increased during the germination, but Kwangyang cultivar was decreased during the same periods. The total lipids contents of Yeupsildalggae and Kwangyang cultivar were decreased during the germination. According to SDS-PAGE analysis, there was no detectible polypeptide bands on the gel before seed germination suggesting that this may be due to the rapid degradation of the storage proteins in the mature seed by hydrolyttic enzymes during the stage. During germinatation , the polypeptide band with 27$\sim$28KD of Yeupsildalggae and Kwangyang cultivar were accumulated gradually. In changes of fatty acid composition of total lipid of Yeupsildalggae and Kwangyang cultivar , saturated fatty acids such as palmitic acid and stearic acid increased during the germination. On the other hand, unsaturated fatty acid such as linoleic acid and linolenic acid decreased during the same periods. However, oleic acid increased to the 5 DAG, and then was repidly decreased.
Seed germination is a biological process that is affected by a variety of genetic and environmental factors. The cardinal temperature and thermal time are required for germination. The principal objective of this study was to identify and characterize variations in the base, optimum, and maximum germination temperatures of Haloxylon aphyllum L. from two seed sources, in order to establish models for use in predicting seeding dates. Mature H. aphyllum seeds were germinated at temperatures between 5 and $35^{\circ}C$. The germination behavior of H. aphyllum seeds to different temperature regimens in light was evaluated over a temperature range of $5-35^{\circ}C$ at intervals of $5^{\circ}C$. The rate of germination increased between base and optimum thermal conditions, and decreased between optimum and maximum thermal conditions; the germination rate varied in a linear fashion at both sub-optimal and supra-optimal temperatures. The linear regression fit the range of germination rates at $5^{\circ}C$ to $25^{\circ}C$ and $25^{\circ}C$ to $30^{\circ}C$, and thus the base temperature, optimum temperature, and maximum temperature for the germination of H. aphyllum were measured to be $0.6^{\circ}C$, $25.69^{\circ}C$, $37.90^{\circ}C$, and $1.76^{\circ}C$, $21.56^{\circ}C$, $37.90^{\circ}C$ for Qom and the Fars dune desert respectively.

Ethanol extract of 20 botanicals in 3 concentrations i.e., 2.5%, 5% and 10% were evaluated for their efficacy to improve reproductive performance of mulberry silkworm (Bombyx mod L.) through feeding botanical enriched mulberry leaves during $2^{nd}$ day of $4^{th}$ age as well as $1^{st}$ and $3^{rd}$ day of $5^{th}$ age larvae. The preliminary screening was done by ranking the botanicals based on influence in most important reproductive contributing parameters such as single cocoon weight, effective rate of rearing, pupation rate, fecundity and recovery of eggs/kg cocoon. Five botanicals viz., Asparagus recemosus, Achyranthes aspera, Tribulus terrestris, Withania somnifera, Parthenium histerophorus ranked first were further evaluated at 8% 5% and 3% concentrations to confirm their effectiveness and standardize effective concentration. Among the rearing parameters, mature larval weight, shell ratio and number of cocoons/kg were not significantly influenced by the treatments. However, reproductive parameters such as effective rate of rearing (wt), pupation rate (%), fecundity (No.) and egg recovery (g)/kg cocoon were significantly (P<0.05) influenced by the botanicals. Higher fecundity and egg recovery were obtained on treatment with Withania somnifera irrespective of concentrations. However at 5% concentration, pupation rate, fecundity as well as egg recovery were found significantly higher than that of other concentrations, confirming its effectiveness for improving reproductive efficiency.

The ecological background of a restoration project is complex and difficult to betermine without experimentation. A useful context for experiments is the well-studied process of natural succession, because the factors that drive or inhibit succession are also at work during reclamation (a form of primary succession) and restoration (which often resembles secondary succession). Using experimental studies on urban wasteland reclamation, we have tested for factors that stimulate or inhibit succession during early phases of woodland development in the Northeastern United states. The emphasis has been on mutualisms (seed dispersal, pollination, and mycorrhizae) and microsite limitations in the recruitment, growth, and reproduction of woody plants. Using plantings of seeds, seedlings, and clusters of reproductively mature plants on abandoned landfills, we have observed that (1) soil microsite deficiencies lead to very poor germination (<0.1$\%$) and seedling survival (<0.01$\%$) of most native species; (2) seed dispersal by birds is a significant and reliable source of woody plant recruitment; however (3) proximity effects are strong, with most (up to 95$\%$) of seed rain falling in the vicinity of planted clusters that are closest to putative seed sources; and (4) remnant natural woodlands are critical components of the recruitment process. To emphasize the last point, in one case, we found that the destruction of approximately 50$\%$ of nearby natural woodland vegetation led to a commensurate decline in seed rain. In another case, we found that the species richness of recruits was strictly limited by the species composition of nearby source plant communities, with no evidence of community enrichment by long distance dispersal over 5 years. We conclude from these results that the size and proximity of remnant natural populations are critical considerations when planning reclamation and restoration programs that rely on natural successional processes.
We had already reported the successful germination for green pods of purple lady's slipper orchid (Cypripedium macranthos Sw.). The green pod methods is to take immature seeds in green capsules, sterilize the capsule, and take out the sterile seeds. This method, however, needs very critical time of harvest. The critical time of seed harvest changes depending upon the species, condition of the specimen, and climatic influence, and the right time lies between 5 and 12 weeks after fertilization. In this study, the mature seeds were collected after 120-130 days with hand-polination of lady's slipper orchids. Mature seeds are usually dormant and it has to be overcome, either with hormone or storing the seeds near freezing for two or three months to break dormancy. The seeds were first surface sterilized with 70% ethanol and then transferred 1% NaOCl for 10-15 minutes, followed by rinses 3 times with sterilized distilled water. The cypripedium seeds consists of an embryo within a seed coat known as a testa. The testa is water repellent and the seed has a large air space between the embryo and testa so the seed tends to float on water. We had resolved the problems with vacuum pump to soak water into the testa before sterilization. The seeds were placed on liquid or agar solidified germination media. Cultures were incubated at $24{\pm}1^{\circ}C$ in dark. The seeds were germinated in 6-8 weeks in liquid suspension culture (germination percentage over 18%); however, the seeds on agar solidified media took more than 5 months to germinate and the germination percentage less than 5%. The most effective media for liquid culture was 1/4 strength Murashige and Skoog (MS) medium with 50 ml/l coconut water ($4brix^{\circ}$) at pH 5.8.
들잔디(Zoysia japonica Steud.)의 최적 조직 배양 조건을 확립하기 위하여 성숙종자로부터 최적 캘러스 유도 및 효율적인 식물체 재분화에 미치는 몇 가지 요인의 영향을 조사하였다. 성숙종자로부터 배발생 캘러스 유도율은 3mg/L 2,4-D가 첨가된 MS 배지에서 85%로 가장 높았으며 NAA 또는 IAA를 첨가한 처리구보다 높게 나타났다. 배발생 캘러스로부터 식물체 재분화는 0.1 mg/L 2,4-D와 5 mg/L BA가 첨가된 N6 배지에서 73.3%로 가장 높게 나타났다. 기본배지의 종류에 따른 배양효율의 차이는 배발생 캘러스 유도에는 MS 배지가, 식물체 재분화에는 N6배지가 가장 효과적이었다. 재분화 된 식물체는 1/2 MS rooting 배지에서 뿌리를 형성되어 pot에 이식하였을 때 정상적으로 분화되어 생장하였다. 본 연구를 통하여 확립된 단기간 고효율 재분화 시스템은 분자육종을 통한 신품종 들잔디의 개발에 유용하게 응용되어질 수 있을 것이다.
유자의 성숙종자를 MS 기본배지에 치상하여 종자의 내피에서 주심조직 유래의 희고 부서지기 쉬운 배발생캘러스가 1.2%의 빈도로 형성되었다. 이 캘러스는 1 mg/L 2,4-D를 첨가한 MS배지에서 증식되었다. 증식된 캘러스를 0.1 mg/L kinetin을 첨가한 MS 배지에 옮겼을 때 많은 수의 체세포배가 형성되었다. 배발생캘러스를 1 mg/L 2,4-D를 첨가한 액체 배지에 넣어 배발생 현탁배양계를 확립하였다. 배양된 현탁배양세포를 0.5 mg/L ABA를 첨가한 고체배지에 평판하였을 때 높은 빈도로 체세포배로 발달하였으며 MS 기본배지 혹은 1 mg/L kinetin 첨가배지에서 소식물체로 발달하였다. 소식물체는 성공적으로 토양으로 옮겨서 온실에서 육성되었다.
티모시의 4가지 품종을 이용하여 성숙종자로 부터 캘러스 유도 및 식물체 재분화 효율을 조사하였다. 4주 동안 배양한 종자로부터 캘러스 유도율은 Colt, Chair, Richmond, Hokuo 순으로 캘러스 유도율을 보였고, 식물체로의 재분화 또한 캘러스 유도율과 같은 Colt > Chair > Richmond > Hokuo 순으로 나타났다. 기본 배지의 종류에 따른 배양 효율의 차이는 품종별 차이는 있으나 MS 배지가, 식물체 재분화는 N6 배지와 MS 배지가 SH 배지에 비해 효과적이었다. 전체적으로는 캘러스 유도율이 높은 품종이 식물체 재분화율도 높은 경향을 나타내었다. 본 연구를 통하여 밝혀진 캘러스 유도와 식물체 재분화율이 우수한 Colt 품종은 유용유전자 도입을 통한 신품종 개발에 유용하게 이용될 수 있을 것이다.
유채에 있어서 유묘의 저온감응 처리가 하계고온포장에서의 좌지 방지에 미치는 효과를 조사하여 유채 세대단축 기술의 보완과 우량종자의 비격리 순수채종 방안을 검토코자 시험을 실시하였던 바 그 결과를 요약하면 다음과 같다. 1. 유채 유묘의 저온록체 춘화처리후 하계고온의 일반포장조건에서 재배했을 때 추태, 개화단축 효과는 조생, 중생종에서 추태까지 34일, 개화까지 52일 소요했으며 만생종은 추태까지 44일, 개화까지는 73일을 소요했다. 2. 성숙까지의 전생육기간에서는 조생, 중생종들이 85-88일이 필요했으며 만생종에서는 108일이 소요되어 표준재배 227-240일 보다는 67%-55% 단축할 수 있었다. 3. Winter형의 B.napus에서 5월 이후의 고온재배에서는 전부 좌지하던 것이 국내품종도 록체춘화저온처리에서는 모든 공시품종이 완전 추태ㆍ개화, 성숙할 수 있었다. 4. 저온록체 처리후 하계고온재배시의 유채 주요형질들은 초장, 수장에 있어서 보통기재배보다 50% 내외로 빈약하였으며 수량형질인 분지수와 1수협수에서는 특히 적어서 보통재배시의 1/10 정도로 분지수 2-3개, 협수 4-7협에 불과했다. 5. 그러나 결실립수, 결실비율, 1,000립중 등에서는 다소 표준추파재배보다 떨어지나 큰 차이는 없었다. 6. 저온처리 하계고온 재배시의 주당종실중과 10a당 수량성에서는 1주당 8.8g으로 표준재배의 34.5g의 26%밖에 되지 않으며 10a당 수량성은 평균 53kg로 표준추파재배의 230kg에 비해 23%에 불과하였다. 7. 수량성이 10a당 불과 53kg 내외이긴하나 이같은 채종량은 육묘이식으로 100㏊ 이상의 (1,000배 증식율) 배경면적에 보급 증식할 수 있을 뿐만 아니라 혼매원이 없는 하계재배로 강실, 격리재배를 하지 않고도 순정한 채종이 가능하게 되었다.
